Effect of Pimpinella Brachycarpa Ethanol Extract on Chronically Ethanol-Induced Liver Damage in Rats (참나물이 만성적으로 알코올 유도된 흰쥐의 간손상에 미치는 영향)

  • This study was performed to investigate the effect of ethanol extract of Pimpinella brachycarpa(PBE) on chronically ethanol-induced hepatotoxicity in rat liver. Sprague-Dawley rats weighing 90-130 g were divided into 5 groups; normal group(NOR), ethanol(35%, 10 ml/kg) treated group(CON), PBE 200 mg/kg treated group(P1), PBE 200 mg/kg and ethanol treated group(P2), and PBE 400 mg/kg and ethanol treated group(P3). PBE was also fractionated by the following solvent: n-hexane, chloroform, ethylacetate and n-butanol. The antioxidative capacity of the n-hexane fraction was the highest among fractions and was similar to that of butylated hydroxytoluene(BHT). The body weight gain and feed intake of the rats were decreased by ethanol administration, but were gradually increased to the similar levels of the NOR group by administering PBE. The AST activity in serum elevated by ethanol was significantly decreased by administering the high dosage of PBE, but exerted no significant change on serum ALT activity. It was also observed that the hepatic activities of xanthine oxide(XO), catalase and glutathione peroxidase(GSH-Px) increased by ethanol were markedly decreased in the combined ethanol and PBE administered groups(P2 and P3), but not in the activity of superoxide dismutase(SOD) as compared with the CON group. The glutathione(GSH) contents were decreased by ethanol adminstration, however, increased after administering PBE. These results suggest that ethanol extract of Pimpinella brachycarpa has a possible positive effect on the liver function in hepatotoxicity-induced rats by ethanol administration.

Effects of Polygoni multiflori radix Liquors on the Memory Impairment of Rat (하수오 침출술이 흰쥐의 기억력 손상에 미치는 영향)

  • This study was carried out to investigate the effect of memory impairment in rats fed alcohol liquid diets. The rats were randomly divided into 4 groups. While the control(C) group received a diet containing 12% of the daily calories with isocarbohydrate, the other groups received a diet containing 12% of the daily total calories with 36% Soju (A), 25% Cynanchi wilfordii radix liquors (WA), and 25% Polygoni multiflori radix liquors (RA) for 6 weeks. After 6 weeks, ADH activity in the brain tissue of the Group A was found to be significantly lower than Group C but significantly higher than both Groups WA and RA(p<0.05). ADLH activity was revealed to be the highest in the Group WA (p<0.05). The concentration of acetaldehyde, a highly toxic metabolite, was the highest in the Group A, but its concentration decreased significantly if fed a liquid diet containing WA. The concentration of acetylcholine, which has a high correlation with memory impairment, was significantly lower in Group A, although Group RA group was the highest compared to the other groups (p<0.05). AChE activity of the Group A was higher than Group C but lower in Groups WA and RA (p<0.05).

고혈압환자 운동처방

  • 고혈압은 증상이 없이 나타나며, 심장질환이나 뇌질환과 같이 인체에 치명적인 손상을 입히기 때문에 '조용한 살인자(silent killer)'라고 불린다. 고혈압을 예방하기 위해서는 체중감소, 알코올 섭취의 감소, 규칙적인 운동, 염분 섭취의 감소가 중요하다. 그리고 혈압을 강하시키기 위해서는 약물치료보다는 행동수정요법과 운동요법이 선행되어야 한다고 하였다. 고혈압을 치료하기 위해서 속보나 조깅, 자전거, 수영과 같은 산소운동이 가장 좋다고 보고되고 있다. 그러나 화자들은 저마다의 생리적 특성이 다르다. 관절이 좋지 않은 사람, 허리에 통증이 있는 사람, 운동 중 혈압의 반응이 비정상적인 사람 등 다양하다. 본고에서는 고혈압 환자에게 권장되는 운동종목을 제시하고, 운동종목별 특성과 주의사항을 제시하고자 한다.

  • 본 논문의 목적은 음주섭취로 인한 혈중 알코올 농도에 따른 뇌의 활동도변화를 측정, 분석하는데 있다. 1차원 시계열데이터인 EEG신호는 생체 비선형 동역학 시스템으로부터 발생하는 Deterministic Nonlinear Chaos신호로써 무작위적인 신호와는 구분되어질 수 있다. EEG시계열데이터를 위상공간에 적절한 어트랙터로 재구성하여 상관차원 최대발산지수 등의 카오스 지수들을 추출하여보면 EEG시계열데이터가 무작위적인 계에서 발생하는 랜덤한 신호가 아닌 카오스계에서 기인함을 알 수 있고, 인간의 정신상태에 따른 뇌의 활동도를 정성적, 정량적으로 판별해 볼 수 있다. 이러한 카오스 분석방법을 토대로 음주전의 뇌의 활동도와 음주후 혈중알코올 농도에 따른 뇌의 활동도변화를 EEG의 카오스 지수들의 변화를 통해 분석해 보았다.

Effect of Monascus Pigment Extract on the Alcohol Metabolism in Rats (흰쥐에 있어서 홍국 색소 추출물이 알코올대사에 미치는 영향)

  • To investigate the alcohol metabolizing system in liver of rats drunken 10% ethanol with Monascus pigment extract (MPE), Sprague-Dawley male rats weighing about 250 g have been drunken 10% ethanol containing 1, 2.5 and 5% Monascus pigment extract for a month. Three groups of rats drunken 10% ethanol with MPE gained somewhat less body weight than normal group, but the changes of body weight was not significantly different among the former groups. All groups drunken MPE supplemented alcohol had no remarkable changes in liver function on the basis of liver weight/body weight, the serum levels of alanine aminotransferase and xanthine oxidase activity. 10% alcohol drunken animals (control group) showed significantly increased activity of hepatic alcohol dehydrogenase (ADH) by 87% compared with normal group and the animals drunken 1%, 2.5% and 5% MPE showed respectively 34%, 29% and 21% increased activity of hepatic ADH, whileas Km value of ADH in 1, 2.5 and 5% MPE group decreased by 40%, 30% and 19% respectively compared with the control, but Vmax showed no significant changes among MPE groups. In case of aldehyde dehydrogenase (ALDH), 1% MPE group showed significantly increased activity by 32% and 2.5% or 5% MPE group showed increasing tendency compared with control, and Km value in three experimental groups declined by 27% and no particular changes were found among those. Furthermore, Vmax value in 1, 2.5 and 5% MEP group increased by 88,56 and 22% respectively with the control. In the aspect of the area under the curve of a ethanol concentration versus time (AUC) profile obtained after administration of 10% alcohol with 1 or 5% MPE, the decreasing rate of AUC to the control was 18% in 1% MPE treated rats whereas 10% in 5% MPE group.

Effects of Fermented Milk Intake on Hepatic Antioxidative Systems in Alcohol treated Rats (알코올 투여한 쥐에서 발효유의 섭취가 간조직 내 항산화 체계에 미치는 영향)

  • Effects of fermented milk, $Kupffer's^{\circledR}$, intake on hepatic antioxidative systems were investigated in rats fed ethanol (3 g/kg B.W.) for 2 weeks. Serum AST and ALT were $88.7{\pm}6.5\;and\;41.2{\pm}4.1IU/L$ in control group, $114.6{\pm}7.1\;and\;64.7{\pm}3.8IU/L$ in alcohol group, and $94.0{\pm}5.5\;and\;44.7{\pm}5.3IU/L$ in fermented milk (FM) group, respectively. Fermented milk intake decreased hepatic glutathione peroxidase and superoxide dismutase activities of FM group to level of control group (p<0.05). Glutathione S-transferase activity of fermented milk group increased by 122% compared to control group. These results suggest antioxidative activities of lactic acid bacteria and ingredients in $Kupffer's^{\circledR}$ improve antioxidative system in alcohol-treated rats.

Modulation of Ethanol-Induced P450 Enzyme Activities and Antioxidants in Mice by Hordeum vulgare Extract (만성 알코올 급여 흰쥐에서 보리 추출물 섭취가 Cytochrome P450 효소 조절 및 항산화계에 미치는 영향)

  • The effects of methanol extract of barely (Hordeum vulgare) on alcohol-induced damages of liver were investigated in male ICR mice. Mice were divided into three groups, control, ethanol, and ethanol plus 0.15% of barley extract. After four weeks of ethanol feeding, ethanol group significantly increased the P450 content, CYP2E1 and CYP1A2 enzyme activities, whereas ethanol plus barely group markedly decreased to levels similar to control group. Catalase activity in ethanol group was significantly lower than that in control group; however, ethanol plus barely group stimulated catalase activity as well as SOD activity significantly. These results indicated that barely extract modulated P450 enzymes for ethanol-induced liver damage and might be useful in developing functional food for alcoholic liver damage.

Comparison of Attitudinal Beliefs regarding Smoking and Antioxidant Vitamins Status in the College Male Smokers and Non-smokers (일부 남자 대학생 흡연자와 비흡연자의 흡연에 대한 인식 및 항산화 비타민 수준 비교)

  • The purpose of this study was to investigate of attitudinal beliefs regarding smoking, and antioxidant vitamins status between smokers and non-smokers in college male students. Attitudinal beliefs regarding smoking and antioxidant vitamins intake were investigated by a questionnaire. The result obtained were as follows: Anthropometric measurements were no significant differences between smokers and non-smokers. The attitudinal beliefs regarding smoking of smokers were different from those of non-smokers(p<0.001). Compared with non-smokers, smokers responded more positively on advantage of smoking, while they felt less negatively on disadvantage of smoking. Systolic blood pressure(SBP) and diatolic blood pressure(DBP) of smokers were significantly higher than those of non-smokers(p<0.05). Although antioxidant vitamins intake was not significantly different between smokers and non-smokers, plasma level of vitamin C of smokers was significantly lower than that of non-smokers(p<0.05). However, plasma level of vitamin E was not significantly different between smokers and non-smokers.

Protective Effect of Citrus unshiu Peel Extract on Ethanol-Induced Fatty Liver in Rats (흰쥐에서 감귤과피 추출물의 알코올성 지방간 개선 작용)

  • This study investigated whether or not Citrus unshiu peel extract (CPE) affects fat accumulation in livers of rats fed an ethanol-containing liquid diet. Initially, male Sprague-Dawley rats were housed individually in stainless steel, wire-bottomed cages with free access to a Lieber-Decarli control liquid diet. Rats were divided by body weight into three groups of eight each: one group of rats was fed the Lieber-Decarli control liquid diet devoid of ethanol (control), another was fed the Lieber-Decarli ethanol diet (ethanol), and third was fed the same ethanol diet except containing CPE. All three groups were fed their respective diets for 6 weeks. Serum and liver lipids were analyzed and liver histology performed. Body weight did not differ among the groups over the 6-wk duration. Histology images showed that CPE administration significantly improved fat accumulation in livers, which was induced by ethanol diet. Serum levels of transaminases and lipids also were reduced by CPE consumption. Taken together, the results indicate that CPE may protect ethanol-induced fatty liver by lowering fat accumulation in both the liver and blood. The protective effects of CPE appear to be due to its phenolic contents.

Effects of Scopoletin Supplementation on Insulin Resistance and Antioxidant Defense System in Chronic Alcohol-Fed Rats (Scopoletin 보충이 만성 알코올을 급여한 흰쥐의 인슐린저항성 및 항산화방어계에 미치는 영향)

  • This study investigated the effects of scopoletin (6-methoxy-7-hydroxycoumarin) supplementation on insulin resistance and the antioxidant defense system in chronic alcohol-fed rats. Rats were fed a Lieber-Decarli liquid diet containing 5% ethanol with or without two doses of scopoletin (0.01 and 0.05 g/L) for 8 weeks. Pair-fed rats received an isocaloric carbohydrate liquid diet. Chronic alcohol did not affect fasting serum glucose levels, although it induced glucose intolerance and hyperinsulinemia compared with the pair-fed group and led to insulin resistance. Both doses of scopoletin similarly improved glucose intolerance, serum insulin level, and insulin resistance. Scopoletin supplementation significantly activated phosphatidyl inositol 3-kinase, which was inhibited by chronic alcohol. Two doses of scopoletin up-regulated hepatic mRNA expression and activity of glucokinase as well as down-regulated mRNA expression and activity of glucose-6-phosphatase compared with the alcohol control group. Both doses of scopoletin significantly reduced cytochrome P450 2E1 activity and elevated aldehyde dehydrogenase 2 activity, resulting in a lower serum acetaldehyde level compared with the alcohol control group. Chronic alcohol suppressed hepatic mRNA expression and activities of antioxidant enzymes such as superoxide dismutase, catalase, and glutathione peroxidase; however, they were reversed by scopoletin supplementation, which reduced hydrogen peroxide and lipid peroxide levels in the liver. These results indicate that dietary scopoletin attenuated chronic alcohol-induced insulin resistance and activated the antioxidant defense system through regulation of hepatic gene expression in glucose and antioxidant metabolism.
