Synthesis and Use of a Ligand for the Extraction of Uranium (I) (우라늄 추출을 위한 리간드의 합성 및 응용 (제 1 보))

  • The ligand, 2,10-dibenzyl-4,6,8-trioxo-3,9-diaza undecane dioic acid(DTDA) for the extraction of uranium was synthesized under dry nitrogen from phenylalanine and 3-oxoglutaric acid. Extraction was performed by stirring a solution of DTDA in dichloromethane for 1 hour with an aqueous solution of $UO_2(ClO_4)_2{\cdot}6H_2O$ at various pH values and at different $DTDA/UO_2{^{2+}}$ molar ratios. Extraction efficiency reaches a maximum when the pH of the aqueous phase was ca 8.0. The extraction percentage was affected by concentration of DTDA and increases with the $DTDA/UO_2{^{2+}}$ molar ratio to complete extraction with a 4 fold excess of DTDA. The high selectivity of the DTDA for uranium was ascertained by competition experiments with other cations. The bound uranyl ion was quantitatively liberated within few minutes from the organic phase by treatment with an aqueous 1M HCI solution and DTDA was recovered very satisfactorily from the organic phase. The values of the over-all formation constants of the complex between uranyl ion and DTDA were determined to be the following : ${\beta}_1=1.20{\times}10^5\;,\;{\beta}_2=1.01{\times}10^8$.

  • The chemical attractants for sea eels, Astroconger myriaster, were identified by using fish traps in their natural habitat around namhae island, southern part of Korea. The ethyl alcohol extracts of anchovy, Engraulis japonica, which was a very good baits for sea eels showed higher attraction activities than residuals of it. Because sea eels showed high attraction activities to the free amino acid or nucleotides of synthetic anchovy extract, the succeeding experiment was conducted to identify the major chemical attractants among 9 free amino acid and 2 nucleotides by omission test. L-Glutamic acid, L-glycine, taurine, L-methionine or IMP showed highest attraction activities among 11 chemical compounds. But sea eels showed significantly weaker attraction activities to the synthethic extracts and single component than to the natural extract.

  • Antheraea pernyi silkworm is one of typical wild silkworms, which spins a tawny color cocoon. The cocoon has been used as a resource for textile material due to strong chemical stability and good mechanical properties. In this study, to increase the solubility efficiency of A. pernyi silk fibroin, the composition of dissolution solvent were examined. Calcium chloride tertiary system, normally used for dissolution of Bombyx mori silk fibroin, does not act on A. pernyi silk fibroin. Calcium nitrate system dissolves A. pernyi silk fibroin, and calcium nitrate ethanol system do more easily than calcium nitrate system. Amino acid composition of A. pernyi silk fibroin obtained after dissolution is mainly composed of alanine, glycine, and serine. A. pernyi silk fibroin would be used for non-textile applications near future.

Aroma Compounds Produced by the Yeast Hansenula saturnus var. saturnus Isolated from Soil (토양에서 분리한 Hansenula saturnus var. saturnus에 의한 휘발성 방향성분의 생성)

  • A yeast strain producing fruity-floral aroma was isolated from soil and identified as Hansenula saturnus var. saturnus. Glucose was found to be the best carbon source and sodium nitrate or phenylalanine as nitrogen source in terms of the nature and the intensity of the aroma produced by the isolated yeast. Seventeen compounds, mainly esters and alcohols, were identified in the ether-pentane extract of the culture broth by gas chromatography and/or coupled gas chromatography-mass spectrometry. Ethyl alcohol, isobutyl alcohol, isoamyl alcohol, phenethyl alcohol and their acetate esters together with ethyl caprylate were the major compounds in the aroma concentrate. Three unusual compounds, dibutyl disulfide, 3-methyl pentanoic acid and methyl pentanoate were also tentatively identified in the culture broth of the isolated yeast.

Effects of Low Temperature on the IAA Level in Etiolated Pea(Pisum sativum L. var. Sparkle) Seedlings (백색 완두유묘 중의 IAA 수준에 미치는 저온의 영향)

  • Little is known about the regulation of IAA level in plants under low temperature. On exposing 6-day-old etiolated pea(Pisum sativum L. var. Sparkle) seedlings grown at $25^{\circ}$ to $5^{\circ}$ for 3 days, free IAA level decreased from 17.6 to 4.9 ng/g fresh weight(that is, from 68 to 10 pmol/shoot) and total IAA level from 113.2 to 60.8 ng/g FW. The level of free tryptophan, precusor of IAA, also decreased from 583 to 414 nmol/g FW and that of free phenylalanine and tyrosine, products of shikimic acid pathway along with tryptophan, decreased from 3,168 to 2,327 nmol/g FW. Thus low temperature was found to down-regulate the level of endogenous IAA, in part, by reducing tryptophan biosynthesis in etiolated pea shoots.

  • In this study, a new quantitative analytical method has been developed for the rapid determination of acylcarnitines and amino acids in human blood using electrospray ionization / tandem mass spectrometry (ESI-MS/MS). Acylarmitines and amino acids were analyzed by tandem mass spectrometry after conversion to their butylesters through treatment with 3N butanolic hydrogen chloride. Acylcrnitines were analyzed using precursor 85 ion scan and alanine, valine, leucine/isoleucine, methionine, phenylalanine, tyrosine, aspartic acid and glutamic acid were analyzed using neutral loss 102 scan, ornitine and citrulline were analyzed neutral loss 119 scan, glycine was analyzed using neutral loss 56 scan, arginine was analyzed using neutral loss 161 scan and argininosuccinic acid was analyzed product ion 459 scan. This method reduced sample preparation time compared to that with conventional amino acid analyzer and liquid chromatography, with high sensitivity and good reproducibility.

Physicochemical Composition of Trichosanthes kirilowii Maximowicz (하늘타리의 이화학적 조성)

  • Although each part of Trichosanthes kirilowii is frequently used as medicinal herbs, study on the chemical composition is not sufficient. It was found that sarcocarp consists of 70% of carbohydrate, 13% of crude protein, 5% of crude fat, 6% of crude fiber and 6% of crude ash; seed consists of 62.59% of carbohydrate, 12.75% of crude protein, 14.80% of crude fat, 6.50% of crude fiber and 3.36% of crude ash; and root consists of 89.40% of carbohydrate, 4.10% of crude protein, 0.50% of crude fat, 3.50% of crude fiber and 2.50% of crude ash. Sarcocarp and seed contain fifteen kinds of amino acids such as Asp, Ser, Glu, Gly, His, Arg, Thr, Ala, Pro, Try, Cys, Met, Val, Leu, and Phe. Sarcocarp contain large quantities of phenylalanine and glycine and seed does not contain leucine but lysine. The mineral contents in Trichoxanthes kirilowii are 0.55% Ca, 0.91% Mg, 10.29% Na, and 0.17% K.

Expression Pattern of Acetyl Xylan Esterase of Streptomyces coelicolor A3(2) in Escherichia coli (Escherichia coli에서의 Streptomyces coelicolor A3(2)의 acetyl xylan esterase 발현 양상)

  • We cloned a gene encoding acetyl xylan esterase(axeA) of Streptomyces coelicolor A3(2) and studied its expression pattern in Escherichia coli. The full sequence of axeA was amplified by PCR. Sequence analysis of the PCR product revealed an open reading frame of 1,008 nucleotides encoding a protein consisted of 335 amino acid residues, with a calculated molecular mass of about 38 kDa. The base sequence showed 98% homology to the same gene of Streptomyces lividans. Two different kinds of acetyl xylan esterases were produced in Escherichia coli(pLacI) by IPTG induction; their molecular weights were 38 kDa and 34 kDa, respectively. Of these, 38 kDa protein seemed to be a total protein holding N-terminal signal peptide region, whereas 34 kDa protein seemed to be a matured protein without signal peptide which was produced by peptide bond cleavage between two amino acid residues of alanine 41 and alanine 42.

Transport of Some Solutes in Blood Plasma Through Poly(2-Hydroxyethyl Methacrylate) Hydrogel Membrane (혈장내 염의 Poly(2-Hydroxyethyl Methacrylate) 격막 투과현상)

  • The relative permeabilities, distribution coefficients and diffusion coefficient of some salts which are important components in blood plasma through a poly(HEMA) membrane were measured. The crosslinker which was used for preparing the membrane was tetraethylene glycol dimethacrylate(TEGDMA), the weight percentage of the latter was about 2.8. We found that the diffusion coefficients ($D_m$) of the solutes decrease exponentially with increasing molecular weight, and also that $D_m$'s decrease linearly (except urea) with cylindrical radius (a). These facts were explained by a sieve pore flow model. The relative permeability and diffusion coefficient of urea at various temperature were larger than those of other solutes such as glycine, ${\beta}$-alanine, D-glucose, saccharose and maleic acid. The result indicates that the poly(HEMA) membrane might be suitable for hemodialysis application.

  • The two dissociation constants of three amino acid, glycine, alanine, and tryptophan were measured by the conductometric method in the temperature from 15 to 35$^{\circ}$C and pressure up to 2,500bar in aqueous solution. The both dissociation constants were increased as the temperature increased but the pressure effect were not same as the temperature effect. The first constants were increased as the pressure increased but second constants were decreased except tryptophan. The characteristic properties of these amino acids were discussed from the thermodynamic properties of the dissociation reaction. The substituted effects of the reaction were deduced from Hammett reaction and substituted constants which were calculated from the dissociation constants.
