Analysis of Androgen Receptor Gene by Capillary Gel Eelectrophoresis (모세관 젤 전기영동을 이용한 안드로젠 수용체 유전자 분석)

  • gel electrophoresis (CGE) with poly(ethylene oxide) has been applied to the measurement of CAG repeat number in Androgen receptor (AR) gene related to male infertility. Non-linear regression analysis was performed using the standard X174 RF/Hae III, 100bp step ladder DNA in order to investigate the exact number of CAG repeat. For 79 Korean infertile males and 89 controls, CAG repeats at exon 1 in AR gene was compared and analyzed by CGE. It turned out that CAG repeat numbers were 24.972.6 range, 17-29) for the infertile male, and 23.992.4 range, 18-29) for the control, respectively. P value (0.018) was less then 0.05, meaning that the result was statistically meaningful.

Estrogen, Androgen, and Retinoic Acid Hormone Activity of Ginseng Total Saponin (인삼 총 사포닌의 에스트로젠, 안드로젠, 레티노익산 호르몬 수용체에 대한 활성)

  • Alternative or complementary medicine plays an important role in health care system. Ginseng, being one of the most popular oriental herbs, is believed to contain various steroid hormone activity. Ginseng has been demonstrated pharmacological effect in the cardiovascular, endocrine, central nervous, and immune system. Our objective was to study that total saponin might mediate some of their actions by binding to the steroid hormone receptor, as they share many of the actions of steroid hormone in various physiological system. Using total saponin from Panax Ginseng, we have studied the possibility of total saponin being a potential estrogen receptor, androgen receptor, and retinoic acid receptor ligand. Total saponin activated the transcription of both the estrogen and androgen responsive luciferase reporter plasmids at a concentration of 100$\mu\textrm{g}$/ml in COS cells transiently transfected with the corresponding receptor and hormone responsive receptor plasmids. And total saponin caused a concentration-dependent stimulation of estrogen receptor. Total saponin increased the expression of estrogen responsive c-fos proto-oncogene at the protein level in MCF7 cells at 24 h treatment as examined by Western analysis. The c-fos induction was used as a specific marker of estrogen responsiveness. This activation was inhibited by the specific estrogen receptor antagonist, ICI 182,780. However, total saponin failed to activate the retinoic acid receptor in COS cells transiently transfected with the corresponding receptor and retinoic acid responsive reporter plasmids. These results show that total saponin is capable of activating estrogen and androgen receptors.

  • The inhibitory effect of histone acetyltransferase from methanol extract of Terminalia chebula Retz. fruit (TCME) was investigated in prostate cancer cell. TCME significantly inhibited histone acetyltransferase (HAT) activity by over 50% at 100 ${\mu}g/mL$ concentration. TCME treatment repressed androgen receptor (AR) mediated transcription, mRNA level of AR target genes, prostate specific antigen (PSA) and NKX-3.1, as well as AR acetylation. Finally, the prostate cancer cell viability was dramatically reduced by TCME treatment at 0~100 ${\mu}g/mL$ concentration. These results indicated that TCME, as a potent HAT inhibitor, could suppress prostate cancer cell growth by AR mediated transcription regulation.

Clonality Assay of Dysplastic Epithelial Lesions of the Stomach (위 이형성 상피 병변의 클론성에 대한 분자병리학적 연구)

  • Purpose: Dysplasia or flat adenoma of the stomach is regarded as a precancerous lesion. However, the frequency and the evolutionary process of malignant transformation of gastric dysplasia are still debated. In order to see whether the lesion was a monoclonal or a polyclonal proliferation, clonality was assayed by X-linked HUMARA polymorphism. Materials and Methods: DNA was extracted from the paraffin-embedded tissue of 16 consecutive cases of endoscopic biopsy, eight of which supplied both dysplastic and nondysplastic tissue for comparison. HUMARA was amplified by PCR with or without pretreatment with methylationsensitive restriction enzyme, HpaII. The amplification products were electrophoresed on polyacrylamide gel and silver-stained. Results: Among the 16 cases, 13 cases were informative and 3 cases noninformative. Of the 13 cases, one case showed skewed lyonization, rendering 12 cases to be analyzed further. A monoclonal band pattern was noted in 2 cases, and a polyclonal band pattern in 10 cases. A review of the histopathologies of the monoclonal and the polyclonal cases did not reveal features discriminating the two groups. Conclusion: These results suggest that gastric dysplasia is a disease entity heterogeneous in the genetic level, and many cases may be non-neoplastic.

Hair Revitalization Effects of Streptococcus sp. Strain Culture Medium Filtrate (HAIRCLETM) (Streptococcus sp. 배양 여과물(HAIRCLETM)의 모발 개선 효과)

  • Streptococcus sp. is generally known as a strain that produces hyaluronic acid. In this study, we focused on ingredients other than hyaluronic acid among several metabolites produced by Streptococcus sp. during cultivation. The hair experimental sample (HAIRCLETM) was prepared with Streptococcus sp. culture filtrate under the condition that hyaluronic acid was not produced and the effect on the hair was identified. Tensile strength of hair was tested with a universal testing machine (UTM). Surface properties of a hair were examined with an atomic force microscope (AFM). Our results suggest that HAIRCLETM could prevent damage to a hair such as destroyed-cuticle, increased-faction force, and broken-hair. Furthermore, hair-related experiments confirmed that HAIRCLETM could promote the expression of hair growth factors VEGF, HGF, and Wnt10b and mitigate male hair loss by inhibiting androgen receptors and TGF-β2 expression. In addition, it was confirmed that the expression of barrier-related factors (INV, CLDN-1) was increased for oxidative stress, and the COX-2 expression, an inflammatory factor, was inhibited, thereby improving damaged scalp barriers and alleviating induced inflammation.
