Design of Solid Lipid Nanoparticles to Improve Penetration of Niacinamide through Artificial Skin Tissue (나이아신아마이드의 인공피부조직 투과 개선을 위한 고형지질나노입자의 설계 연구)

  • In this study, Niacinamide (NI) was loaded into solid lipid nanoparticles (SLNs) and skin permeability was evaluated to improve skin permeability of NI, which was a skin whitening substance. NI was able to effectively load within SLN with a double-melting emulsification method, producing stable particles with average particle sizes of 263.30 to 436.93 nm and a zeta potential of -34.77 to -57.60 mV. Artificial skin tissue (SkinEthicTM RHE) derived from skin keratinocytes derived from human epidermal tissue was used for the skin permeation study of NI. Skin transmittance and deposition experiments of NI confirmed that all SLN formulations improved skin transmittance and deposition rates of NI, approximately 5.4 ~ 7.6 and 9.5 ~ 20.8 improvement over SLN applications. Therefore, SLN manufactured in this study have shown sufficient results to improve the skin permeability of the functional whitening substance, NI.

Effects of Supplementing Whole Oilseeds to Alcohol-Fermented Feedstuff Based on Rye Hay on in vitro Rumen Fermentation Characteristics (호밀 건초 급여하의 By-pass 전지종실을 첨가한 알코올 발효사료의 in vitro 발효특성)

  • This study was conducted to determine the effects of supplementing whole oilseeds to the alcohol fermented feedstuff based on rye hay on in vitro rumen fermentation of Hanwoo steers. Experiments were arranged with four treatment groups, T1 (without any oilseeds), T2 (supplemented with cottonseed), T3 (supplemented with linseed) and T4 (supplemented with soybean). The pH decreased at 6 and 12h in vitro incubation in T2 and at 3 and 12h in vitro incubation in T4 (p<0.05). However, no decrease of the pH was observed at any in vitro incubation in T3 (p>0.05). Ammonia concentration increased at 6h in vitro incubation in T2 and at 6 and 9 h in vitro incubation in T4 (p<0.05). Concentrations of acetate, butyrate and propionate increased at 3, 9 and 12h in vitro incubation in T4 and at 3h in vitro incubation in T3 (p<0.05). This study indicates that the supplementation of whole oilseeds to alcohol fermented feedstuff based on rye kay can improve the in vitro rumen fermentation.

Comparison of Reduced Acrylamide Formation in Chips Fermented with Different Cultivar Potatoes by Bacillus subtilis (Bacillus subtilis를 이용한 발효 감자 칩의 감자 품종에 따른 아크릴아마이드 저감화 비교)

  • Acrylamide is a probable human carcinogen and can be formed during frying of starchy foods. The objective of this study was to investigate the effects of acrylamide reduction in storage potato chips by fermentation of Bacillus subtilis and compare its usefulness with four different cultivars of storage potatoes (Goeun, Atlantic, Saebong, and Jinsun). Potato slices were fermented by B. subtilis at $37^{\circ}C$ for 0, 2, and 4 h and fried at $180^{\circ}C$ for 115 s. The total sugar contents of the fermented potato slices did not change compared to the control. However, reducing sugar contents increased in the fermentation solution containing potato slices. Asparagine contents of Saebong and Jinsun potato slices decreased with fermentation time. Color values of the fermented potato chips were improved compared to those of non-fermented potato chips. The highest $L^*$ value was found in Saebong (57.4), followed by Goeun (56.7), Jinsun (52.5), and Atlantic (48.8) after 4 h of fermentation. Potatoes stored for 240 days generated considerable amounts of acrylamide, ranging from 4.99 to 10.38 ppm, after frying. Four hours of fermentation reduced acrylamide formation in all potato chips. The lowest acrylamide content was found in Saebong (0.77 ppm), followed by Jinsun (1.21 ppm), Goeun (1.76 ppm), and Atlantic (4.09 ppm). In conclusion, fermentation of storage potatoes by B. subtilis can effectively lower acrylamide formation during frying of potato chips.

Preparation and Characteristics of Liquid Silicone Rubber Using Polyorganosiloxane Modified with Dimethylacrylamide (디메틸아크릴아마이드 변성 폴리오가노실록산을 이용한 액상 실리콘 고무의 제조와 그 특성)

  • ${\alpha}$,$\omega$-Hydrogen polyorganosiloxane(HPMDMS) prepolymer was prepared from equilibrium polymerization ofoctamethylcyclotetrasiloxane, 1,3,5-trimethylcyclotrisiloxane, 1,3,5,7-tetravinyl-1,3,5,7-tetramethyl-cyclotetrasiloxane, and 1,1,3,3-tetramethyl disiloxane as an end-blocker in the presence of tetramethylammonium siloxanolate as a catalyst. Polyorganosiloxane modified with dimethylacrylamide(APMDMS) was prepared by hydrosilylation of HPMDMS with dimethylacrylamide in the presence of Pt catalyst, and followed by coordination of metal oxide (APMDMS-MO), such as NiO and FeO, to the amide moieties of the resulting polymer. The chemical structures of HPMDMS and APMDMS were confirmed by FT-IR and $^1$H-NMR analysis. Liquid silicone rubber containing metal oxide composite (LSRMO) was prepared by compounding APMDMS-MO, ${\alpha}$,$\omega$-vinylpolydimethylsiloxane, and a catalyst in a high speed dissolver. The thermal conductivity of LSRMO composite was determined to be 0.29 W/mK, and the volume resistivity exhibited a lower value than that of LSR composite. The mechanical and thermal properties of LSRMO and LSR composite were measured by UTM and TGA.

Analysis and Decrease of Cyanogenic Glucosides in Flaxseed (아마인에 함유된 시안배당체의 분석과 저감화)

  • Linustatin and neolinustatin in flaxseed are called cyanogenic glucosides which produce acetone, hydrogen cyanide (HCN) and sugar when hydrolyzed. The HCN is the chemical whose toxin is being issued in food safety. Linustatin and neolinustatin were found in flaxseed by 206.5 and 174.2 mg/100 g, respectively using a RI-HPLC and LC/MSn analysis of $80\%$ methanol extraction. Also, HCN (137.38 mg/kg) was found in flaxseed distillate by autohydrolysis and IC analysis. More than 85$\%$ of linustatin, neolinustatin and HCN were removed when flaxseed was heated for more than 2 hours in $200^{\circ}C$. Especially, HCN was decreased to $98\%$ by heating only 30 min in $200^{\circ}C$.

Quality characteristics, acrylamide content, and antioxidant activities of Nurungji prepared using different grains (곡물 종류를 달리하여 제조한 누룽지의 품질 특성, 아크릴아마이드 함량 및 항산화 활성)

  • We determined the quality characteristics, acrylamide concentration, and antioxidant activity of Nurungji prepared using white rice, brown rice, gamma-aminobutyric acid (GABA) brown rice, oats, and barley. The moisture content of Nurungji prepared using white rice was the lowest (1.48%) and the highest (6.53%) was obtained in barley Nurungji. The brightness (L*) of white rice Nurungji was the highest, whereas that of GABA brown rice Nurungji was the lowest. The acrylamide concentration was the lowest (37.24 ㎍/g) in white rice Nurungji and the highest (255.50 ㎍/g) in oats Nurungji. The levels of total polyphenol and total flavonoid were the lowest in white rice Nurungji and high in oats and GABA brown rice Nurungji. The antioxidant activity was higher in Nurungji prepared using oats, GABA brown rice, brown rice, and barley than that using white rice. It is necessary to select an appropriate grain when preparing Nurungji by considering the amount of acrylamide produced.

Effect of amaranth seed extracts on glycemic control in HepG2 cells (HepG2 세포에서 아마란스 종자 에탄올 추출물이 포도당 흡수 조절에 미치는 효과)

  • Purpose: This study was carried out to investigate the effect of amaranth seed extracts on glycemic regulation in HepG2 cells. The 80% ethanol extracts of amaranth seeds were used to evaluate α-amylase and α-glucosidase activities, cell viability, glucose uptake and messenger RNA (mRNA) expression levels of acetyl-CoA carboxylase (ACC), glucose transporter (GLUT)-2, GLUT-4, insulin receptor substrate (IRS)-1 and IRS-2. Methods: The samples were prepared and divided into 4 groups, including germinated black amaranth (GBA), black amaranth (BA), germinated yellow amaranth (GYA) and yellow amaranth (YA). Glucose hydrolytic enzyme, α-amylase and α-glucosidase activities were examined using a proper protocol. In addition, cell viability was measured by MTT assay. Glucose uptake in cells was measured using an assay kit. The mRNA expression levels of ACC, GLUT-2, GLUT-4, IRS-1 and IRS-2 were measured by reverse transcription polymerase chain reaction. Results: The inhibitory activities of α-amylase and α-glucosidase were highly observed in GBA, followed by BA, GYA and YA. Similar results were observed for glucose. The GBA effect was similar compared to the positive control group. The mRNA expression levels of ACC, GLUT-2, GLUT-4, IRS-1, and IRS-2 were significantly increased. The potential hypoglycemic effects of amaranth seed extracts were observed due to the increase in glucose metabolic enzyme activity, and glucose uptake was mediated through the upregulation of ACC, GLUT-2, GLUT-4, IRS-1, and IRS-2 expression levels. Conclusion: Our findings suggest that the amaranth seed is a potential candidate to prevent a diabetes. The present study demonstrated the possibility of using amaranth seeds, especially GBA and BA for glycemic control.

  • 역삼투막에 이용되고 있는 고분자물질은 셀루로오스 아세테이트(CA)를 비롯하여 방향족 폴리아마이드가 주로 이용되고 있으며, 최근에는 폴리설펀막의 지지층 위에 계면중합법을 응용한 복합막이 주류를 이루고 있다. 여기서는 가장 많이 이용되고 있는 셀루로오스아세테이트를 이용한 역삼투 투과 특성에 관하여 연구하였다.
