Studies on the Effects of Copper on the Lactate Dehydrogenase and Esterase Isozymes in Various Tissues of Carassius carassius (붕어(Carassius carassius)의 조직내 젖산수소이탈효소와 에스테라아제 아이소자임에 미치는 동의 영향에 관한 연구)

  • In order to elucidate the effects of copper on Corassius carassius, the following were studied: 1) lactate dehydrogenase isozyme patterns by cellulose acetate electrophoresis, 2) LDH activity and copper effect on LDH enzyme system y spectrophotometry, 3) esterase isozyme patterns by agar thin layer electrophoresis, 4) hemoglobin patterns by starch gel electrophoresis, and 5) histological study. 1. There were two bands of LDH isozymes (LDH-3 and LDH-5) in the gill, three bands (LDH-2, LDH-4, and LDH-5) in the liver, and two bands (LDH-3 and LDH-4) in the muscle of the normal fish. The LDH-1 bond was not found in the above three tissues. When the fish were exposed to copper, LDH-3 appeared in the liver, LDH-5 in the muscle, but no new LDH band appeared in the gill. 2. The sepcific activities of the LDH were lowest in the gill and highest in the muscle of the normal fish, and they were gradually decreassed in the gill and highest in the muscle of the normal fish, and they were gradually decreased in the liver and mucle except in the gill from 1-day to 10-day exposure to copper. It indicates that LDH activities in the liver and muscle of the fish were inhibited by copper. 3. Through in vitro experiment, it is clear that the decrease of the LDH activities of the liver and muscle of the fish exposed to copper is mainly caused by the inhibition on the M-LDH in the fish. 4. The numbers of the esterase isozyme bands of the gill, liver, muscle, blood, brain, and kidney of the normal fish were 3, 6, 2, 2, 2, and 2 respectively, and these numbers were the same as those exposed to copper. The relative mobilities of the esterase bands in the gill, liver, blood, and kidney of the exposed group were different from those of the control. 5. There was one hemoglobin band on the anode in the normal fish. It seems that the nobility of hemoglobin band of the fish exposed to copper was slightly faster than that of the normal fish. 6. The normal gill lamellae of the fish consisted of centrally located pillar cells and a number of mucus cells. When the fish were exposed to copper, the epithelial layer was divorced first, disintegrated, and then destroyed completely. 7. The liver of the normal fish had prominent central veins, cords of hepatic cells, and sinusoids. When the fish were exposed to copper, numerous droplets of fat appeared in the cells around the central vein of the liver. It is assumed that the fatty droplets were accumulated by the lesion due to fatty metamorphosis of the liver caused by copper. 8. There was no histological difference between the muscle of the normal fish and that of the fish exposed to copper. 9. In the normal fish, the tubules of the kidney were surrounded by hemopoetic tissues. However, the kidney tissue of the fish exposed to copper received some damage on the proximal tubules. Since the tubule cells were reduced in height, the lumens of the tubules were enlarged. Consequently many proximal tubules exhibited some pink-stained granular casts and various stages of degeneration.

  • The present study was conducted to research some Actinosporeans species in Oligochaetes (Branchiura sowerbyi). From April to May, 2004, Oligochaetes were sampled from the Daewi Reservoir located in Kunsan, Jeonbuk and a drainageway of the fish farm located in Wanju, Jeonbuk. Tow species of Actinosporeans not reported yet in Korea, Raabeia sp. and Triactinomyxon sp., were detected from Oligochaetes. Actinosporeans detected are recorded to Raabeia sp. E and Triactinomyxon sp. F. A total of 200 Oligochaetes were investigated from two sites, and 28 individuals (14%) were found infected: 27 individuals (13.5%) with Raabeia sp. E and one individual (0.5%) with Triactinomyxon sp. F.

Infestation of Colpoda-like ciliate in the yellowtail Seriola quinqueradiata (양식산 방어에 기생한 Colpoda-like ciliate)

  • In a survey of net-caged yellowtail Seriola quinqueradiata in the southern parts of Korean Peninsula, we found infestations of ciliate in the gills of the moribund fish during the abnormally elevated temperature. The ciliate was very similar to genus Colpoda Muller, 1773 on morphological and reproductive characteristics. Histopathological changes revealed that the ciliate invaded gills only, causing extensive destruction of the gill filaments including lamella without any clinical signs found in the other organs and tissues. This is the first record of the parasitic ciliate in cultured yellowtail.

  • The suitable tag was investigated based on the cumulative mortality, blood chemistry and histopathology of olive flounder, Paralichthys olivaceus. The dissection of opercle, 50% dissection of pelvic fin and 100% dissection of pelvic fin was used in the experiment. Cumulative mortality of dissection of opercle group was high. The dissection of opercle and 100% pelvic fin dissection groups was showing histological changes after 13days of tagging: purulent inflammation in the liver, fibrous inflammation in the body kidney, hyperplastic ellipsoide capillary in the spleen. As the results, 50% pelvic fin cutting group is the most effective out of them.

  • White spot disease by Cryptocaryon irritans broke out among cultured river puffer (Takifugu obscurus) cultured in sea water in Oct. 2001 and May 2004. Many white spots were easily observed on the gills with the naked eyes. The parasites attached on the gills at the base of the gill lamellae. The trophonts penetrated the interlamellar epithelium. Epithelial cell proliferation in interlamellar spaces occurred. PAS- positive mucous cells were prominent in the proliferated epithelium.

  • Acanthochondria spirigera is redescribed based on the specimens recovered from the Lophiomus setigerus in the southern coast of Korea. This parasitic copepoda is distinguished from congeners by the possession of spirally twisted egg-strings. Acanthochondria spirigera(Chondracanthidae, Poecilostomatoida) is the first recorded species from Korea. This parasitic copepod was not found on the fish in May. Relative density and prevalence also decreased from January to May. A. spirigera live symbiotically within the pharyngeal cavity of the L. setigerus, and eat oesophageal mucous string of their hosts. The male of the parasitic copepoda is dwarf, and found on the ventral surface of genital segment of the female.

  • The ultrastructure of the gills of Oplegnathus fasciatus was examined by means of light and transmission electron microscopes. The gills have primary and secondary filaments (lamellae). The following cells are identified and described : epithelial cell, pillar cell, chloride cell and mucose cell. The simple epithelial layer consists of squamous epithelium containing a large nucleus and the surface is covered with some of microridges. The lamella pillar structures are characterized by axial microtubules and lateral membrane interdigitations. Chloride cells contain a lot of mitochondria and specifically developed tubular systems. The rough endoplasimic reticulum and golgi complex, and some of mucous granules were observed in immature mucous cells. The mature mucous cells were AB-PAS positive, globular in shape, and had mucous granules of similar size with various electron densities.

  • Changes of gill tissue and body composition of grey mullet (Mugil cephalus) and Nile tilapia (Oreochromis niloticus) by the manipulation of salinity were observed in a recirculating rearing system. Salinity was increased from $0\%_{\circ}$ to $33\%_{\circ}$for 1 day and remained far 15days, thereafter salinity was decreased from $33\%_{\circ}$ to $0\%_{\circ{$ for 1 day and remained for other 15 days. Any morphological differences of gill lamella in grey mullet were not observed in seawater and freshwater. However, on day 2 in exposure to seawater, Nile tilapia showed the edema and bloodclot in gill lamella. In the case of grey mullet, mitochondria and tubular system of chloride cell were more densely packed according to the salinity increase. The whole body of grey mullet showed no significant differences in moisture content during experimental period. However, moisture content in whole body of the Nile tilapia was signincantly decreased when exposed to seawater. Protein content in whole body of grey mullet showed no significant difference between beginning and day 15 of the experiment.

Fine Structure and Histopathological Changes Exposed to Acute High Salinity of the Gill of Japanese Clam, Corbicula japonica (일본재첩, Corbicula japonica 아가미의 정상구조와 고염분 급성노출에 의한 조직병리학적 변화)

  • This study was performed to observe ultrastructure of the gill and to ascertain the effect of salinity on histopathological and ultrastructural changes in the gill of the Japanese clam, Corbicula japonica. Experimental period was 7 days. Experimental groups consisted of control, 5, 10, 20 psu. $LC_{50}$ (96 h.) by the probit was 19.55 psu. Mortality was significantly different from the control (p < 0.05). Inner demibranch of the gill of C. japonica was wider 1.37 times than outer demibranch (p < 0.001). The filament zone on the plica can be distinguished by the six epithelial celll cell; frontal ciliated epithelium ($7{\mu}m$), latero-frontal ciliated epithelium ($5{\mu}m$), postlatero-frontal epithelim ($3{\times}8{\mu}m$), and lateral ciliated epithelium ($5{\mu}m$) in the frontal zone, endothelial cellin the intermediate zone, and abfrontal cell in the abfrontal zone. It had one type of secretory cell that was filled with fibrous substances of low electron density. The gill of C. japonica exposed to 5 psu for 7 days was observed partially disappearance of the cilia, and glycogen granule in the filament. In the 10 psu, gill appeared partially modification of epithelial cell and destruction of the glycocalyx. Gill exposed to 20 psu was extended nuclus of the ciliated epithelial cell, destruction of the organelles, and observed glycogen granules infiltration and numerous vacuoles. Moreover, more than 50% filaments were observed that come out chitinous rod from disappearance of epithelial cell in the filament. Therefore, the destruction of the cilia and epithelial cell induce physiological activity and it may be leading directly to death.

Survival and Histological Changes in Gill of the Rockfish, Sebastes schlegeli Following Exposure to Naphthalene (나프탈렌에 노출된 조피볼락의 생존 및 아가미의 조직학적 변화)

  • Rockfish, Sebastes schlegeli (total length; $10.36{\pm}0.49cm$, total weight; $16.28{\pm}1.86g$, N; 290) were exposed to various concentrations of naphthalene for 28 days. Exposure concentrations of naphthalene established control, ethanol (solvent) control, 0.5, 1.0, 1.5 and 2.0 mg Nap $L^{-1}$. After exposure, We observed survival rate, and degree of tissue change (DTC) in gill under optical microscopy. Survival rate of the rockfish was more than 90% in control, ethanol control, 0.5 and 1.0 mg Nap $L^{-1}$, whereas it decreased in 1.5 and 2.0 mg Nap $L^{-1}$ (respectively 80%, 62.2%). In histological observation of gill, hyperplasia of epithelial cells observed in all exposure groups. But no showed increase of DTC which was related to concentration. Whereas, DTC at fusion of gill lamellar, lamellar telangiectasia, stasis, aneurysm and necrosis showed dose dependent increase. Especially, fusion of gill lamellar, lamellar telangiectasia and stasis observed at more 1.0 mg Nap $L^{-1}$, and aneurysm and necrosis at more 1.5 mg Nap $L^{-1}$. These results showed naphthalene caused survival and severe change to the gill of the rockfish which was related to exposure concentration.
