Recently, phytochemicals in whole grains have received increasing attention because the increased consumption of whole grains and whole grain products has been associated with a reduced risk of chronic diseases. Among the phytochemicals in whole wheat, arabinoxylan influences various physiological activities and can aid the treatment of numerous health conditions, either directly or indirectly. In this study, 614 wheat germplasms, collected from the world, were cultivated in 2018 and harvested in 2019 in South Korea. The qualities of these whole grains, including seed protein content, were evaluated using the NIR spectrophotometric method, and arabinoxylan content was determined using enzymatic methods. The ash content was 0.36 to 2.80% and protein content was 7.66 to 20. The SDS-sedimentation for gluten complex ability ranged from 26.85 to 80.63 mL, and protein content showed a high correlation with SDS-sedimentation in the germplasms. Verify the result of the enzymatic method of arabinoxylan, the reliability of the experimental method was determined through repeatability, reproducibility, and recovery. The average value of the 614 resources was 51.64 mg/g of whole grain, and when classified according to the country of origin, the South Korean origin resources tended to have a higher content compared to the genetic resources of other countries. These results are expected to be used as basic data for setting standards for whole grain quality in wheat breeding systems.
Relatively high indoor CO2 concentration (>1,000 ppm) has a negative impact on human health. In this work, indoor CO2 adsorbent was developed by impregnating KOH or K2CO3 on commercial activated carbon, named as KOH/AC and K2CO3/AC. Commercial activated carbon (AC) showed relatively high BET surface area (929 m2/g) whereas KOH/AC and K2CO3/AC presented lower BET surface area of 13.6 m2/g and 289 m2/g. Two experimental methods of TGA (2,000 ppmCO2, weight basis) and chamber test (initial concentration: 2,000 ppmCO2, CO2 IR analyzer) were used to investigate the adsorption capacity. KOH/AC and K2CO3/AC exhibited similar adsorption capacities (145~150 mgCO2/g), higher than K2CO3/Al+Si supports adsorbent (84.1 mgCO2/gsample). Similarly, chamber test also showed similar trend. Both KOH/AC and K2CO3/AC represented higher adsorption capacities (KOH/AC: 93.5 mgCO2/g K2CO3/AC: 94.5 mgCO2/gsample) K2CO3/Al+Si supports. This is due to the KOH or K2CO3 impregnation increased alkaline active sites (chemical adsorption), which is beneficial for CO2 adsorption. In addition, the regeneration test results showed both K-based adsorbents pose a good regeneration and reusability. Finally, the current study suggested that both KOH/AC and K2CO3/AC have a great potential to be used as CO2 adsorbent for indoor CO2 adsorption.

The present study was designed to observe the thermal dynamics of core and skin temperatures during sub-maximal treadmill exercise; to investigate the effect of regional body cooling during short rest after the treadmill exercise on the thermal dynamics. Three conditions (No cooling, Head/Hand cooling, Leg cooling) were simulated in a climatic chamber at 24±1℃ and 50±5%RH. Subjects performed two bouts of treadmill exercise at a rate of 80%HRmax followed by rest. Body cooling with a hood, long gloves, and a blanket that circulated water set at 15℃ was assigned during two bouts of rest. The results showed that (1) rectal temperature (Tre) did not show significant difference between three conditions; (2) Skin temperatures had specific features, depending on body regions. In particular, the initial fall phenomena of skin temperatures at the onset of exercise were noteworthy in the chest, thigh, calf, and finger tip. Of these, the most significant initial fall was found in finger temperature (Tfing). (3) During the period of the initial fall in skin temperatures, Tre gradually increased. (4) The magnitude of the fall of Tfing at the onset of 2nd running was on average 4.8, 5.1 and 3.4℃ for Control, HH cooling, and Leg cooling, respectively (p<0.05). The initial drop of Tfing at the onset of running was maintained for an average of 8.1, 7.9 and 6.3 minutes for Control, HH cooling, and Leg cooling, with no significant differences. In conclusion, the initial fall phenomena at the onset of treadmill exercise reflected non-thermal factors, as opposed to internal thermal status. The magnitude of the initial fall in Tfing was affected by legs cooling. Therefore, the initial fall phenomenon should be considered when interpreting the thermal status of the shell during heavy works/exercises that assigned with intermittent regional body cooling.
This study was carried out to evaluate the potential for preselection of transgenic embryos prior to transfer into recipient animals. In these experiments, I used a 3.2 kb transgene which contained the neomycin resistance gene (neo) and lac Z gene driven by the $\beta$ actin promoter (iacZ Ineo). Oocytes were aspirated from abattoir ovaries, matured in TCM-199 supplemented with 10% fetal bovine serum (FBS), 5 ${\mu}\textrm{g}$/ml LH, 0.5 ${\mu}\textrm{g}$/ml FSH, 100 unit/ml penicillin, and 100 ${\mu}\textrm{g}$/ml streptomycin for 22 to 24 hrs then inseminated with a sperm suspension of 1 X 10$^6$ sperm/ml containing 5 ${\mu}\textrm{g}$/ml of heparin. At 18-20 hrs after insemination, cumulus cells were removed by vortexing and pronuclei of centrifuged zygotes microinjected with the lacZ/neo construct (3 ng/$\mu$l). All cultures were carried out in CR1aa with transfected BRL monolayers containing 3 mg/ml BSA, 20 $\mu$/ml NEM amino acids and 40 $\mu$I/ml BME amino acids. To identify the appropriate concentration of G418 for selection, non-microinjected zygotes were cultured in the presence of 0, 50, 100 and 200 $\mu$g/ml of G418. After 8 days of culture in these treatments, 44/145 (30.3%), 13/150 (8. 7%), 1/151 (0.7%) and 0/134 (0.0%) devel-oped to the blastocyst stage in 0, 50, 100 and 200 $\mu$g/ml of G418, respectively. A total of 1,127 zygotes were microinjected and placed into culture (without G418) and subsequently 710 (63.0%) cleaved. At 48 hrs post-injection, embryos ($\geq$2-cell) were randomly assigned to control (medium alone) or G418 (100 ${\mu}\textrm{g}$/ml) treatments. A control culture of 740 non-microinjected embryos from the same replicates of embryos were also placed into control medium. After 8 days in culture, 54/343 (15.7%) and 22/367 (6.0 %) of the microinjected embryos developed to the blastocyst stage in control and G418 media, respectively. A total of 151/740 (27.2%) of the non-microinjected embryos placed in the control medium developed to the blastocyst stage. The blastocysts in the control treatment had a mean of 70.7 ${\pm}$ 4.7 cells of which 23.1 ${\pm}$ 2.6 (32.7%) stained for $\beta$-Gal activity. B1astocysts in the G418 treatment had a mean of 48.8${\pm}$7.5 cells of which 40.3 ${\pm}$ 4.1 (82.6%) stained for $\beta$-Gal ac tivity (P<0.05). In the control treatment 26 of 30 (87.0%) blastocysts had some cells with $\beta$-Gal activity while all of the blastocysts in the G418 treatment had some cell with $\beta$-Gal ac tivity (P<0.05). However, ICM colonies in either control or G418 treatments were not expressed any epiblast cell except of trophetoderm celIs. The $\beta$-actin promoter/lacZ gene may not be e expression or silence expression in epiblast cells These results clearly show an enrichment of blastocysts expressing the transgene in the majority of their cells after culture in the presence of G418. The exogeneous gene was not express a and silence in ICM colonies especiallly epiblast cells except of trophectederm cells. Even though the higher rate cell number expressed of exogeneous gene in the G418 treatments, a total cell number was decrease significantly (P<0.05) of which might be also drop of the establishment of ES like-cell colonies and production of transgenic animals. However, futher studies need to determine the viability of these selected embryos and the avability of production of transgenic offspring.

The sea urchin has been used as sea food in many countries. This species has also been an important organism of embryological studies for more than a century. In recent years, sea urchin embryos are being used as testing materials for toxicity of pollutants and toxins. Usefulness of sea urchin embryos as experimental models comes from the easiness in obtaining sea urchin samples and a lot of gametes, in rearing embryos in the laboratory, in observing the cellular movement and organ formation during the embryogenesis and in manipulating blastomeres and genetic maferials. The sea urchin in itself is a key organism for the understanding of deuterostome evolution from the protostomes and of indirect development of marine invertebrates which undergo the planktotrophic larval stage. A fertilized sea urchin egg goes through rapid cleavage and becomes a 60 cell embryo 7hr after fertilization. It then develops into a morula, a blastula, a gastrula and finally a pluteus larva approximately 70 hr after fertilization. At the 60 cell stage, the embryo comprises of five territories that express territory-speciflc genes and later form different organs. Micromeres at the vegetal pole ingress into the blastoceol and become the primary mesenchyme cells(PMCs). PMCs express genes involved in skeletogenesis such as SM30, SM37, SM50, PM27, msp130. Among the genes, SM37 and SM50 are considered to be members of a gene family which is characterized by early blastula expression, Glycine-Proline-Glutamine rich repeat structures and spicule matrix forming basic proteins. Genetic studies on the sea urchin embryos help understand the molecular basis of indirect development of marine invertebrates and also of the biomineralization common to the animal kingdom.

In animal studies, Magnesium (Mg) - incorporated oxidized implants showed significant enhancement of the bone response. This prospective clinical trial was performed to investigate the success rate, implant stability and marginal bone loss of Mg oxidized clinical implant. The experimental protocol was approved by Institutional Review Board of the Gangneung-Wonju National University Dental Hospital. Fifty healthy patients had partial edentulism were included in this study. Mg oxidized clinical implants (Implant M, Shinhung, Korea) were installed and restored with conventional protocol. The patients were recalled at 1, 3, 6 months after functional loading. Implant stability quotient (ISQ) was measured and periapical radiographic images were obtained. Amount of marginal bone loss was calculated with calibrated images from periapical radiographs. Repeated measured analysis of variance and post hoc Tukey test were used to compare the mean ISQ and bone level. A total of 101 implants were analyzed. The mean ISQ values increased continuously with time lapse from 68.4 at fixture installation to 71.5 at 6 months after loading. Implant stability was correlated with gender, fixture diameter, bone quality and implant sites. The mean marginal bone loss during 6 months after loading was 0.26 mm. There was no failed implant and six-month success rate was 100%. Within the limitations of this study, the six-month success rate of Mg oxidized implant was satisfactory. The implant stability and marginal bone level were excellent. However, further longer clinical studies will be needed to confirm the success of Mg oxidized clinical implant.

AOAC Mouse Bioassay Analysis (MBA) has been the gold standard for the analysis of paralytic shellfish poisoning toxin (PSP toxin) for more than 50 years. However, this method has inaccurate limit of quantification and cannot be used to determine toxic profiles. An HPLC method (PCOX) was optimized for Korean shellfish to establish an alternative or supplementary method for PSP analysis and was intended to be used for the official monitoring and regulation of food. The recovery rate of the PCOX method was 83.5-112.1% and the limit of quantification for total toxin was about $8.6{\mu}g$/100 g. A long-term comparison study showed a good correlation of the PCOX results with the AOAC MBA results: the correlation factors were 0.9534 and 0.9109 for oyster and mussel matrices, respectively. The PCOX method may be used as an alternative or supplementary method for AOAC MBA to monitor the occurrence of PSP and to analyze PSP toxin profile in oysters and mussels.
The purpose of this study was to evaluate the filtering-feeding effect of a freshwater mussel (Corbicula leana) on the phytoplankton communities in two lakes with different trophic conditions between June and September, 2000. Manipulation experiments were conducted with two treatments (the control and mussel addition), and each established in duplicate 10 l chambers. Both ambient nutrient (TN, TP) and chlorophyll-a concentrations were significantly (p<0.01) higher in Lake Ilgam than Lake Soyang. Cyanophytes (Microcystis, Oscillatoria, Lyngbya and Dactylococcopis) consistently dominated algal community in Lake llgam, while flagellated algae (Dinobryon divergence, Mallomonas, Rhodomonas) and cyanophytes (Microcystis)dominated in Lake Soyang. The net exponential death rate ($R\;=\;day^{-1}$) of total phytoplankton in the mussel treatment ranged $1.70{\sim}7.39$ and $0.38{\sim}1.64$ in Lakes Soyang and Ilgam, respectively. Mean filtering rate standardized by mussel AFDW ($ml\;mgAFDW^{1}\;h^{-1}$) was much higher in Lake Soyang ($1.70{\sim}3.06$) than in Lake Ilgam($0.24{\sim}0.88$0.24~o.88). Estimating FR per mussel, 1 mussel filtered $1.6{\sim}7.8\;l$ per day and $1.7{\sim}3.0\;l$ per day in Lakes Soyang and Ilgam, respectively. Based on tile C-flux tobiomass ratio, Corbicula leana consumed $0.8{\sim}4.4$ fold of phytoplankton standing stock in Lake Soyang, and $0.4{\sim}1.6$ fold in Lake Ilgam per day. Mussel feeding resulted in increase of SRP concentration by $30{\sim}50%$, compared with the control. The results of this study suggest that filter-feeding activity of Corbicula leana varies depending on the phytoplankton density and community composition. The high seston consumption rate of Corsicuja Jeaua even in a eutrophic lake suggests that biomanipulation approach using filter-feeding mussels can be used far wate rquality management in small eutrophic reservoirs.
The purposes of this study were to develop Hazard Analysis Critical Control Point plan applicable to cook/chilled Pork Bulkogi (broiled sliced pork with sauces) in school foodservice operations and to establish reasonable shelf-life limits by assessing food quality during chilled storage period of 5 days. During the product flow, time-temperature profile was recorded and microbiological analyses including mesophilic and psychrotrophic total plate counts, coliform, and fecal coliform and qualitative analyses of Salmonella and Listeria monocytogenes were done. Chemical analyses (pH, acid value, total volatile basic nitrogen), sensory evaluation, and quantitative analysis of thiamin were conducted for 5 days of chilled storage. The number of mesophiles in raw pork ($4.26{\pm}0.11\;Log\;CFU/g$), seasoning mixture ($5.97{\pm}O.04\;Log\;CFU/g$) and marinated pork ($5.56{\pm}0.21\;Log\;CFU/g$) were below the microbial standards for "requires further cooking" food items. Listeria monocytogenes was detected in seasoning mixture. After heating, the number of mesophiles ($5.17{\pm}0.04\;Log\;CFU/g$) were slightly reduced but it did not meet the microbial guidelines of $5\;Log\;CFU/g$ for "ready-to-eat" foods. No other microbes including pathogens were detected. By reheating the menu item after chilled storage, the number of mesophiles were reduced in every phase of 1st day ($4.62{\pm}0.22\;Log\;CFU/g$), 3rd day ($4.55{\pm}0.20\;Log\;CFU/g$) and 5th day ($4.25{\pm}0.16\;Log\;CFU/g$) of chilled storage, and the number of microbes was below the standard limits for "ready-to-eat" foods. At the fifth day of chilled storage, pH (p<0.05), acid value (p<0.01) and TVBN (p<0.05) showed significant increases. Sensory evaluation results did not show any significant change for 5 days of chilled storage. Thiamin content showed a decrease for 5 days of chilled storage. Consequently, the ideal shelflife recommended for Pork Bulkogi was within 3 days of chilled storage. CCPs for Pork Bulkogi were purchasing and receiving of raw meat and some seasoning ingredients, heating, chilling, chilled storage, reheating, and distribution.

This study was conducted to test a hypothesis that sensitivity of trees to ozone exposure was related to their growth rates. Two cultivars of Populus alba ${\times}$ P. glandulosa with different genetic growth potential were used for the comparison. Two clones(72-30, 72-16) of cultivar No. 4 with fast growing potential and three clones(71-28, 72-27, 72-19) of cultivar No. 2 with slow growing potential were propagated in early spring by cutting in $2-{\ell}$ plastic pots. They were grown outdoor for 5 months and exposed in late August for 30 days to 70 and 130ppb ozone in a open-top chambers(2.5m in diameter and 2m in height). Ozone concentration in a control chamber was maintained below 30ppb by filtering with activated charcoal. Each treatment was replicated twenty times. In a control chamber, cultivar No. 4 grew 73%, 64%, and 38% faster than cultivar No. 2 in leaf weight, root weight, and total dry weight, respectively. Visible injury was observed only in cultivar No. 4 in 130ppb treatment. Ozone treatment at both 70 and 130ppb decreased height growth, dry weight of leaf, root, and entire plants in all five clones. Particularly root growth was reduced by 39.7% and 13.8% in cultivar No. 4 and No. 2, respectively, in 70ppb treatment. Consequently, shoot/root ratio of cultivar No.4 was increased by 63.4%, while that of cultivar No.2 was increased by 22.1%. Stomatal conductance decreased more in cultivar No.4 than in cultivar No.2. Net photosynthesis of cultivar No.4 at 130ppb ozone decreased by 69.5%, while that of cultivar No.2 decreased by 31.5%. Above mentioned physiological responses of two cultivars to ozone strongly suggested that fast growing cultivar No.4 was more sensitive to ozone than slow growing cultivar No.2. It was concluded that sensitivity of trees to ozone exposure was closely related to their growth rates.
