Physiological Response of Rice to Metolachlor Herbicide (제초제(除草劑) Metolachlor에 대한 수도(水稻)의 생리적(生理的) 반응(反應))

  • The effect of metolachlor [2-chloro-N-(2-ethyl 1-6-methylphenyl)-N-(2-methoxy-l-methylethyl)-acetamide] on starch, sugar and protein content in relation with ${\alpha}$-amylase activity, and its interaction with GA in the germinating stage of rice were determined. The distinctive phytotoxic symptom was the failure of the primary leaf of rice to break the coleoptile. An inhibitory effect of metolachlor was nullified by the external application of GA $10^{-3}$M. A significantly slower starch degradation accompanying the lower content of sugars was observed in the gain of the metolachlor treated seedlings. Further, the total ${\alpha}$-amylase activity was significantly lower in the grain of rice seedlings treated with metolachlor than that of the untreated one, and the lower total ${\alpha}$-amylase activity could be due to an inhibition of ${\alpha}$-amylase formation.

Production of Virus-Free Stocks from Citrus Plant by the Shoot-Tip Grafting and Heat Treatment (열처리와 Shoot-Tip Grafting에 의한 감귤 바이러스 무독묘 생산)

  • Virus-free stocks was produced by the combination of the heat treatment of virus infected plant and shoot-tip grafting (STS). To produce virus-free stocks, the plants infected with citrus viruses were used for virus-free stock production using the modified method of STG in thermotherapy at $40^{\circ}C$ for 16 hours in the light, and at $30^{\circ}C$ for 8 hours of darkness for 4 weeks. Trifoliate orange (P. trifoliata) were used as rootstock seedling for STG. Percentages of virus-free stocks against citrus tristeza virus (CTV), satsuma dwarf virus (SDV) and citrus tatter leaf virus (CTLV) were $75.7\%,\;100.0,\%\;82.6\%$ respectively. Shoot tip size for successful STG were as small as possible. Less than $0.3\;\cal{mm}$ of shoot tips gave the hight efficiency of virus free plants but survival rates were low. And, survival rate after shoot-tip culture was analyzed and the rates were dependant on the cultivars; Yuzu cultivar showed the hight survival rate ($74.6\%$) and early satsuma mandarin (Iwasagi) was $13.3\%$ as the lowest cultivar. But citrus trees were not succeed to grown, turned brown, and died.

Characteristics of Cold Hardiness and Growth of Grapevines Grown under Rain Shelter Type Cultivation System in the Vineyard (간이비가림시설에서의 포도 내한성 발현 및 생육 특성)

  • There are variations among grapevine genotypes in the levels of tolerance to cold, and cold-hardiness of grapevine has been affected by the change of short-term environment during over-wintering. In this study, the responses of vines to cold in open field and rain-shelter system were investigated to obtain useful information in increasing the tolerance to cold in grape cultivation. Total carbohydrate content of bearing mother branches was higher in the rain-shelter system than in the open field, and lower in the branches of 'Muscat Bailey A' than in 'Campbell Early' and 'Kyoho'. Bud-burst and shoot growth were better in the rain-shelter system than in open field, whereas there is no significant difference among the treatments of net beside vines. There was also low incidence of gray mold in rain shelter system. Stilbene compounds such as t-piceid, resveratrol, piceatannol, c-piceid were accumulated in the cold-treated shoot from vine cuts harvested in rain shelter system. Genes of chalcone isomerase, manganese superoxide dismutase, proline rich protein 2, and temperature induced lipocalin were highly expressed in the cold-treated shoot from vine cuts harvested in rain shelter system. While there was not change of air temperature, but high reduction of wind speed in the rain shelter system compared to open field, and increase in the reduction of wind speed by net treatment. The damage of grapevines by cold in the extreme low temperature could be reduced by keeping them in the rain shelter system with net during winter season.

Comparative Analysis to Damage Reduction of Host Plant by Applying a Mating Disruptor of the Oriental Fruit Moth, Grapholita molesta in Two Different Cultivation Environments of Apple Orchard (재배환경이 상이한 사과원에서 복숭아순나방(Grapholita molesta) 교미교란제 처리에 의한 기주 피해 억제 효과 비교)

  • This study demonstrates a variability in efficacy of mating disruption against the oriental fruit moth, Grapholita molesta, populations infesting apples cultivated in differential environmental conditions. Throughout the growing seasons, trap catches of G. molesta male moths and damaged leaf and fruit were examined to evaluate the efficacy of a commercialized mating disruptor. Treatment of the mating disruptor significantly reduced the trap catches of male moth in treated plot, compared to those of the untreated plot. This reduced trap catches were significantly correlated with leaf and fruit damage. Moreover, the highest host damage occurred in June just after the highest overwintering adult peaks in both plots. The treatment of mating disruptors in Chungsong effectively disrupted the overwintering population in April and May, resulting in no noticeable host damage during the following growing seasons. However, there was a marked difference in host damage between two plots, especially in late seasons. Variation in the efficacy of mating disruption technology in terms of host damage appeared to be related with nearby pheromone-untreated orchards, which may result in the immigration of gravid females.

  • 본 연구는 유기종자 안전 생산기술 개발을 위한 효과적인 병해충 제어를 위해 대표적인 찰옥수수 품종인 미백2호와 흑미찰을 대상으로 병?해충 발생양상을 조사하였다. 2008년 유기농 찰옥수수 종자 생산지의 우점 비산포자의 종류 중Alternaria sp., Cladosporium sp., 및 Helminthosoprium sp.,은전 생육기간에 관찰되었다. 또한종자로부터 신초 출현 전후로 종자가 발아하지 않고 부패하거나 유묘의 시들음 증상, 심하면 고사하는 증상과 거세미나방에 의해 지재부가 절단된 전형적인 피해를 관찰할 수 있었고 발병주율은 14~6% 미만이었다. 특히, 포장내 조명나방 발생밀도는 2008년 트랩당 3.5~0.5 마리로 높게 나타났으며, 2009년 트랩당 0.5~6마리로 낮게 나타났다. 또한 피해주율은 2008년은 7.5(미백2호)~21(미흑찰)%였으며, 2009년은 1~46%로 매년 높은 피해주율을 보였으며 수확기까지 지속적으로 증가하였다. 옥수수잎마름병 (Leaf blight)의 발생 양상은 2008년도에 7%(미흑찰)~34%(미백2호)로 높게 나타났으며, 2009년은 6월18일 이후 초발생하여 발병주율이 8월 21일 (59%, 미흑찰)이후 급격히 증가하여 9월 25일이는 미흑찰 (69%)의 발병주율이 미백2호 (56%)보다 높게 나타났다. 옥수수깜부기병(Common Smut)의 발생 양상은 2008년도에 20주 중 2%~15%의 발병주율을 보였으며, 2009년은 초발생 시기가 7월17일로 2008년에 비해 17일 정도 늦추어졌으며 발병주율도 8% 미만 (9월 25일)으로 2008년에 비해 아주 낮게 나타났다. 옥수수깨씨무늬병 (Southern Leaf Spot)의 발생 양상은 2008년 20주 중 초기에 평균 11%(미흑찰)에서 증가하여 9월 19일 조사시에는 두 품종 모두 62%로 높게 나타났다. 2009년은 6월8일 이후 초발생하였으며 9월 25일까지 지속적 발병하여 약 86%의 발병 주율을 보였고 2008년에 비해 24% 이상 발병주율이 높았다.

Introduction of VP6 Gene into Potato Plant by Agrobacterium-mediated Transformation and Analysis of VP6 Expression in Transgenic Potatoes (Rotavirus VP6 유전자의 감자식물체내로의 도입과 형질전환체의 발현분석)

  • A VP6 fragments was subcloned with BamHI in the binary pMBP-1 vector under Califlower Mosaic Virus (CaMV) 355 promoter and neomycin phosphotransferase II (npt II) gene. The recombinant binary vector was mobilized into Agrobacterium-tumefaciens LBA4404 by the freeze-thaw method and potato (Solanum tubensum L. cv Desiree) was transformed by modified leaf-disc cocultivation. Shoots were induced on MS medium with 0.01 mg/L NAA, 0.1 mg/L GA$_3$, 2.0 mg/L Zeatin, 100.0 mg/L kanamycin, 500.0 mg/L carbenicillin. In order to identify the copy number of VP6 into potato plant, total genomic DNA was isolated from transgenic potato and analysed by Southern blotting. Genomic DNA and total mRNA analysis demonstrated the incorporation of the foreign gene into the potato genome, as well as their transcription.

Effect of Shade Net on Reduction of Freezing Damage at a Tea Garden (차밭에 설치된 차광망의 동해경감 효과)

  • In the result of effect on freezing damage reduction by the shade net colors and the shade rate to tea trees during wintering period, the high shade ratio decreased tea trees growth and increased freezing damage and 55% of shade ratio based on non treatment developed new leafs and green leaf productivity. By the shade net colors, colorless shade net (55% of shade type) treatment and green shade net treatment increased green leaf productivity and decreased damaged area compare to non treatment shade net and black shade net treatment. Colorless net shade treatment reduced over 50% of freezing damage and increased (10a) 68 kg for green leaf productivity compare to non treatment shade net. The colors of shade net treatments for reduction freezing damaged in order of Black < Green < Colorless but reduction of freezing damage was so high. In the relative microclimates, treatment shade nets were $0.7^{\circ}C$ lower, average relatively humidity 14.9% higher, soil temperature $0.6^{\circ}C$ lower and soil moisture 4.6% higher than non treatment shade net. And treatment shade nets decreased average wind speed 0.7 m/s and it showed us treatment net shades effected to excessive evapotranspiration and soil dry by wind and considered one of good solution for freezing damages.

Effect of Bedsoil on Cutting Propagation of Old Growth and Protected Tree of Salix chaenomeloides Kimura (노거수 및 보호수 왕버들 삽목증식에 미치는 상토의 영향)

  • Old growth and protected-trees are worth history and culturally in Korea. This research carried out to investigate habitat characterization and the vegetative propagation methods of Salix chaenomeloides(100-600 years old trees) in Gyeongsangnamdo Province. It has been preserved in good and grow naturally in area surrounding village(4 trees), riverside(1 tree) and field(1 tree). Cutting from old growth and protected trees was conducted using 10 kinds of bedsoils. It was survived above 93% in all bedsoils. Growth of cuttings is a distinct difference according to bedsoils and tree ages. Growth of cutting was high on CBS, followed mixed bedsoils(VPMP) including vermiculite, peatmoss and perlite. Root collar diameter was difference depend on tree ages and bedsoils. All trees are produced 1 to 4 branch. Root growth of cutting was 20 cm on CBS and VPMP, which is the best shoot growing on cutting. Bedsoil with physico-chemical characteristics of high moisturizing ability, high air permeability, high EC value and slightly acid may be suitable for efficiently cutting propagation of old growth and protected S. chaenomeloides. CBS and VPMP bedsoil are favorable for propagating this species by cuttings.

Propagation of Acorus gramineus from Seeds and In vitro Culture (종자 및 기내배양을 이용한 석창포 증식)

  • Rhizomes of Acorus gramineus Soland have been used as sedatives, analgesics, and stomachics in Korean medicine. Even though A. gramineus produced in Korea is known as having better efficacy of a remedy than that in China, its cultivation area has not increased because of the lack of seedlings. To solve this problem, seed propagation method was tested. Seeds were harvested and sowed three times in June, 2005. The best results showed at greenish yellow seed stage harvested in mid June, and its germination ratio was 90.7%. Seeds were well stored at $5^{\circ}C$ for 30 days. As for in vitro culture, multiple shoots were induced first in MS basal medium supplemented with $2.0mg{\cdot}L^{-1}$ NAA plus $0.1mg{\cdot}L^{-1}$ BA, and then roots were induced in MS basal medium containing $0.1mg{\cdot}L^{-1}$ BA or $0.5mg{\cdot}L^{-1}$ NAA plus $1.0mg{\cdot}L^{-1}$ BA. Growth of A. gramineus seedlings in Wonyesangto, potting material sold in markets for horticultural plants, was superior to vermiculite.

Proper Tree Vigor and Crop Load in High Density Planting System for 'Fuji'/M.9 Apple Trees (사과 '후지'/M.9 밀식 사과원의 성과기 적정 결실 및 수세 기준)

  • Proper tree vigor and crop load were determined for 'Fuji'/M.9 apple trees in high density planting system from 2001 to 2003. Leaf/fruit ratio was highly correlated to mean fruit weight (y=1.715x+205.02, $R^2=0.66^{**}$) and yield (y=-35.l56x+5963.7, $R^2=0.44^{**}$). In addition, there was a significant correlation between the number of leaves per tree and mean fruit weight. However, crop load did not affect tree growth, soluble solids content of fruit, and Hunter a value. To harvest the fruits heavier than 300 g without biennial bearing, it was appropriate to crop 55 to 64 fruits in a tree with 55 leaves per fruit of adult tree. The good indices for proper tree vigor could be 20 to 25 cm of mean shoot length and above 95% of shoot termination rate. Moreover, no secondary growth and 20 to 30% of spur formation could be the indices for highly productive tree vigor.
