Characteristics of Seed Germination and Potted Seedlings Growth of Endemic Species, Sambucus sieboldiana var. pendula and Sambucus sieboldiana for. xanthocarpa (특산식물 말오줌나무와 노랑말오줌나무의 종자발아 및 분화묘 재배특성)

  • This study was carried out to determine the effects of seed storage methods and pre-treatments on seed germination, soil types and shading conditions on potted seedlings growth of S. sieboldiana var. pendula and S. sieboldiana for. xanthocarpa, endemic species in Korea. The seed germination rate of S. sieboldiana var. pendula was the highest at 48.6% when seeds were treated with stratification for 60 days and then soaked in 100 ppm $GA_3$. And seed germination rate of S. sieboldiana for. xanthocarpa was the highest at 51.4% when seeds were pre-chilled for 30 days and then soaked in 100 ppm $GA_3$. And potted seedlings of S. sieboldiana var. pendula showed the best quality under 50% shading in bedsoil with the growth characteristics of plant height (41 cm), number of leaves(8), leaf width (16 cm), leaf length (18 cm), root length (22 cm), fresh weight(aerial part/root part; 20/6.9 g) and dry weight(aerial part/root part; 4.5/2.0 g). And potted seedlings of S. sieboldiana for. xanthocarpa showed the best quality under non-shading in bedsoil with the growth characteristics of plant height (39 cm), number of leaves(10), leaf width (12 cm), leaf length (15 cm), root length (22 cm), fresh weight (aerial part/root part; 21/13 g) and dry weight (aerial part/root part; 4.2/2.2 g). Therefore, seeds treated with stratification or prechilling and then soaked in $GA_3$ were effective in germination of S. sieboldiana var. pendula and S. sieboldiana for. xanthocarpa, and potted seedlings should be cultivated in bedsoil under non or 50% shading condition.

  • This study was conducted to develop a simple, rapid, and reliable method for in vitro propagation of disease-free and true-to-type clones from sucker explants of thornless blackberry (Rubus fruticosus L. ${\times}$ R. parvifolius L.). To induce multiple shoots, the sucker explants were sterilized in 1% NaOCl solution, and then were aseptically cultured on the full and 1/2 MS solid medium supplemented with BAP (0.1, 0.5, 1.0, 2.0 mg/L). After six weeks of culture, the highest frequency (85.4%) of shoot formation from sucker explants was obtained on the full-strength MS medium with 1.0 mg/L BAP. Node explants obtained from multiple shoots were cultured on the various media of full- or half-strength of AD, B5, MS, SH, QL, WPM media, respectively. After 30 days of culture, plant growth was good on the half-AD, half-QL medium. After 90 days of culture, plant growth was good on the full MS and full SH medium. The survival rate of the plantlets after transfer to plastic pots containing soil mixture (sand: soil: vermiculite was 1:1:1, vol.) in the greenhouse was 98%. The results indicate that a multiple-shoot procedure can be applied for an efficient mass propagation of Rubus fruticosus L. ${\times}$ R. parvifolius L.

Increment of Germanium Contents in Angelica keiskei Koidz. and Panax ginseng G.A. Meyer by In Vitro Propagation (명일엽(明日葉)(신선초(神仙草)) 및 인삼(人蔘)의 기내배양(器內培養)을 통한 Germanium 함량(含量) 증대(增大))

  • This study was carried out to find optimum concentration of germanium compounds and pH of medium on the induction and growth of callus from A. keiskei and P. ginseng and to intend to increase Ge. absorption by calli while those calli were subculturing on MS medium. Callus from a. keiskei was rarely induced under light condition. Under dark condition, callus in­duction from A. keiskei was good up to 5ppm, retarded at 50ppm of $GeO_2$, or C. E. Ge. O., and rarely done at 100 ppm of $GeO_2$ but was somewhat well at 100 ppm of C. E. Ge. O. The induction and growth of callus was good in order of pH 5. 7 > pH 5. 4 > pH 6. 0 Under light condition, the growth of callus induced from P. ginseng was poor at $1{\sim}10\;ppm$ of $GeO_2$, or C. E. Ge. O., but shooting from callus occurred frequently. Under dark condtion, the growth of callus from A. keiskei was good up to 5 ppm of $GeO_2$, or C. E. Ge. O. and was rarely done at 50 ppm of $GeO_2$, but was somewhat well even at 100 ppm of C. E. Ge. O. Shooting from callus occurred frequently in a. keiskei, especially at pH 5.7. The growth of callus from P. ginseng was poor at 10 ppm of $GeO_2$, or 50 ppm of C. E. Ge. O. Under dark condition, the amount of Ge absorption by callus induced from A. keiskei was much high­er than that from P. ginseng. The amount of Ge. absorption by callus treated with $GeO_2$, was higher than that treated with C. E. Ge. O.

A Study on the Plant Planning in Landscape Space Considering the Characteristics of the Gender Determination of Pine Tree (소나무 성 결정 요인의 특성을 고려한 조경공간 식재계획)

  • This study analyzed the components contained in the pine needles of first and second-year-olds to analyze the factors that the in vivo content of inorganic elements affects the sex determination of pine trees. In response, the plan for pine tree plant and maintenance was intended to be presented in consideration of the reproductive environment and physiological characteristics. The results are as follows. First, last year, when there were many encyclopedias, the analyzed N(%) content was found to be high. The nitrogen content of the previous year's soil was found to affect the production of the spheres the following year. This is believed to be possible to reduce the rate of Pine pollen produced in the new plant in the following year through a dispute over quality consumption in the spring of the previous year. Second, the weapons elements involved in the Pine cones and the generation of the Pine pollen in the new plant appeared to be P(%), K(%), Ca(%), and Fe(%). However, the nutrients from the previous year's leaves of Ca(%) and Fe(%) were found to have a low influence on the sex determination of first-year pine trees. Because Ca(%) and Fe(%) are not able to move nutrients accumulated in aging organs due to the nature of the components, feeding nutrients in the fall when the growth of the previous year's branches is reduced is expected to affect oral generation. Third, pine trees are extremely positive and Pine pollen is related to the time of the northeast wind. Therefore, it is believed that it would be good to be located in the northern direction, where the sunlight is good, in an outdoor space. In addition, it is important to use vaginal consumer products in spring and carry out a quarrel involving Mg and Fe during fall to reduce the effect of the Pine pollen, which is an outdoor plant. This is an important part of maintaining and managing pine trees in outdoor spaces as well as the sex determination of pine trees. This study suggested that plant planning, which derives the correlation between pine inorganic element content on sexual determination and takes into account the physiological characteristics of pine trees, is an important issue in the creation of outdoor space. Follow-up research on other factors affecting pine tree sex determination is expected.

Effect of Mixed Application of Cinosulfuron with Dymron on Rice (Cinosulfuron과 Dymron의 혼합처리(混合處理)가 수도(水稻)(Oryza sativa L.)의 초기생육(初期生育)에 미치는 영향(影響))

  • To investigate the safening effect of dymron against cinosulfuron, the responses of 104 rice cultivars and lines to single and mixture treatments of cinosulfuron [3-(4.6-dimethoxy-1,3,5-triazin-2-yl)-1-[2-(2-methoxyethoxy)-phenylsulfonyl]-urea] and dymron [1-(${\alpha}$, ${\alpha}$-dimethylbenzyl)-3-p-tolyl urea] were evaluated. The responses of 104 rice cultivars and lines to cinosulfuron were differed. Among the rice cultivars, growth of Calrose, Dasukei, Hwajinhyeo, and Milyang 105 was significantly inhibited by cinosulfuron but there was no inhibition in Daeseongbyeo, Iri 371, Jangseongbyeo, Taebaegbyeo and IR 74. The japonica types were generally inhibited more than the indica and indica${\times}$japonica types. When Kwangmyungbyeo was treated with the mixtures of cinosulfuron 48g and 96g ai/ha with dymron 450g ai/ha, the inhibition of plant height was significantly reduced regardless of dymron concentrations. With increase in cinosulfuron concentration, the safening effect of dymron against cinosulfuron was more evident in plant height than fresh weight. On the other hand, dymron did not have any safening effects of cinosulfuron on Echinochloa crus-galli at any concentrations. The mixtures of dymron and cinosulfuron produced antagonistic effects on the plant height regardless of rice cultivars and treatment concentrations. At cinosulfuron 24g ai/ha, the effects of the mixtures of cinosulfuron and brassinolide on the plant height of Kwangmyungbyeo and IR 74 were similar irrespective of brassinolide concentrations but the inhibition of Kwangmyungbyeo was greater than that of IR 74 as the concentrations of cinosulfuron increased. The effects of the mixtures of cinosulfuron at concentration of 96g ai/ha with various ABA levels on the plant height of both Kwangmyungbyeo and IR 74 showed synergistic effect regardless of ABA concentration. However, the response of fresh weight of both cultivars treated with cinosulfuron at 96g ai/ha with ABA synergisism was observed in Kwangyungbyeo while IR 74 showed antagonism.

Photomixotrophic Growth of Solanum tuberosum L. in vitro with Addition and Omission of Organic Materials at Thee Initial Sucrose Levels in the Medium (세 수준의 자당이 첨가된 배지에서 유기물의 첨가 유무에 따른 Solanum tuberosum L.의 기내 광혼합영양생장)

  • The most commonly used inorganic nutrient compositions such as Murashige & Skoog medium have been optimized for heterotrophic growth. Therefore, they may not be optimal for photomixotrophic and photoautotrophic growth of plantlets. In photomixotrophic micropropagation, emdium sugar level is often lowered, while light and $CO_2$ levels in vessel are raised, and chlorophyllous explants are used to facilitate photosynthetic carbon acquisition. In a factorial experiment effect of addition (+) and omission(_) of organic materials (OM, 0.5 g ${\cdot}$ $m^{-3}$ each of thiamine, nicotinic acid and pyridoxine and 100 ${\cdot}$ $m^{-3}$ myo-inositiol) combined with three sucrose levels (0, 15, and 30 kg ${\cdot}$ $m^{-3}$) was tested on the growth of potato plantlets. Each of nodal cuttings with a leaf was cultured on 0.1${\times}$$10^{-4}m^{-3}$) MS agar ( 8 kg ${\cdot}$ $m^{-3}$) medium (pH 5.80 before autoclave) in glass test tubes (100 mm${\times}$25mm) capped with a sheet of transparent film with a 6 mm diameter gas permeable filter (5.1 air exchanges ${\cdot}$$h^{-1}$). Cultures were maintained in a room for 27 days at $23^{\circ}C$, 50% RH, 350-450${\mu}mol\;{\codt}\;mol^{-1}CO_2$, 16 h ${\cdot}$ $d^{-1}$ photoperiod at 13${\mu}mol\;{\codt}\;m\;{\codt}\;s^{-1}$ PPFD provided by white cool fluorescent lamps. Growth of potato plantlet in the +OM and -OM treatments were similar, while medium pH was 0.2 scale lower in the latter. Dry weight, % dry matter, and stem diameter enhanced, while shoot to root dry weight ratio, leaf area, chlorophyll concentration per gram dry weight, and medium pH decreased with increasing initial sucrose level. Interaction between OM and sucrose levels was observed in shoot length and medium pH. Results indicate that OM can be omitted from the medium without detrimental effect while addition of sucrose was beneficial for the photomixotrophic growth of potato plantlets under raised light and $CO_2$.

Effect of IAA and Zeatin Riboside on Plantlet Induction from Leaf Disks of Solanum tuberosum L. and Variation of Regenerated Plants (IAA와 Zeatin Riboside가 감자의 엽절편체로부터의 식물체 유기 및 재분화개체의 변이에 미치는 영향)

  • Leaf disks from cultivar 'Kennebec' and one selection line (ND 860-2) were cultured on Murashige-Skoog medium with various combinations of indole acetic acid (IAA) and zeatin riboside. Shoots, roots and callus were induced at various combinations of plant growth regulator levels. The medium containing $3.5mg{\cdot}L^{-1}$ IAA and $4.0mg{\cdot}L^{-1}$ zeatin riboside produced the most plantlets. Rooted regenerants were grown in the greenhouse. The growth of regenerated plants obtained from the MS medium supplemented with $7.0mg{\cdot}L^{-1}$ IAA and $3.0mg{\cdot}L^{-1}$ zeatin riboside was significantly greater than those grown from nodal expalnts. In ND 860-2, a leaf chimera with chlorophyll deficient (light yellow) sectors was found in plants regenerated fiom leaf disks (grown on MS medium supplemented with $3.5mg{\cdot}L^{-1}$ IAA and $3.0mg{\cdot}L^{-1}$ zeatin riboside) but not in plants grown from nodal explants. The phenotypic variability was also observed in the tuber number, size and weight.

Sensitivity to Sterol Biosynthesis Inhibitors of Colletotrichum gloeosporioides Isolated from Persimmon in 2013 in Sangju, Gyeongsangbukdo (2013년 경북 상주지역 감나무로부터 분리한 Colletotrichum gloeosporioides 탄저병균의 스테롤 생합성 저해 살균제에 대한 감수성)

  • Colletotrichum gloeosporioides is one of the most serious pathogens of persimmon in Korea. In 2013, 67 isolates of C. gloeosporioides were isolated from infected fruits, leaf and twigs of persimmon (Diosprosi kaki) at Sangju, Gyeongsangbukdo and fungal responses against five fungicides (prochloraz manganese complex, tebuconazole, mancozeb+myclobutanil, fluquinconazole+prochloraz, and tebuconazole+tolyfluanid) were evaluated by their growth on the fungicide-medium. All isolates were inhibited mycelium growth on the medium with each recommended application concentration of flied. $EC_{50}$ (${\mu}g/ml$) of tebuconazole was from 0.02 to 1.04 and average was 0.31. $EC_{50}$ (${\mu}g/ml$) of prochloraz manganese complex was 0.02~0.23 average was 0.078. Average $EC_{50}$ values (${\mu}g/ml$) of Fluquinconazole+Prochloraz (FP) was 0.029. On the basis $EC_{50}$ (${\mu}g/ml$), the correlation coefficient (r) between tebuconazole and prochloraz manganese complex, prochloraz manganese complex and FP, tebuconazole and FP were 0.42, 0.44 and 0.27, respectively.

Effects of Light-emitting Diodes on In Vitro Growth of Virus-free Sweet Potato Plantlets (LED가 고구마 바이러스 무병묘의 기내 생장에 미치는 영향)

  • The in vitro growth of virus-free sweet potato [Ipomoea batatas (L.) Lam.] plantlets was investigated under different light sources: fluorescent lamp (control); red (660 nm), blue (460 nm), white light-emitting diodes (LED), and two mixtures of blue and red LED (R:B = 8:2, and 7:3). Single node explants (10 mm) of three cultivars ('Matnami', 'Shincheonmi', and 'Yeonhwangmi') were cultured on Murashige and Skoog medium supplemented with $0.2mg{\cdot}L^{-1}$ 6-benzyladenine for 4 weeks. Explants were exposed to $150{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ photosynthetic photon flux at a distance of 20 cm, constant temperature of $25^{\circ}C$, and under 16/8-h (day/night) photoperiod. Using the same method, the in vitro growth of 10 cultivars under red LED was also compared. After 3 weeks, vine length was highest in plantlets cultured under red LED, and lowest in plantlets cultured under blue LED. Fresh and dry weights were also greatest in plantlets cultured under red LED. Compared to the control, vine thickness was significantly higher in plantlets grown under white LED and the 7:3 R:B LED mixture. Significant differences were observed among the 10 cultivars grown under red LED. 'Matnami', 'Shincheonmi', and 'Shinhwangmi' all had excellent vine lengths, and fresh and dry weights. Compared to the control, vine elongation of sweet potato plantlets was most effective under red LED, and culture duration was about 1 week shorter.

Changes of Reserve Substances in the Bark of Stem and Root Mulberry (Morus alba L.) Graftages after Planting (뽕나무 묘목 식재후 지조 및 뿌리피부의 저장물질의 변화)

  • To make clear the changes of the amount of reserve substances in the bark of stem and root of mulberry graftages after planting, studies were carried with mulberry graftages classified into three groups of 7.0-7.9mm, 9.0-9.9mm and 11.0-11.9mm in diameter of stems at 3cm above their base. The results obtained were as follow : 1. The dry weight of stem and root bark gradually decreased with growth of new shoots till about five weeks after planting, and from then it increased. 2. The duration of new shoots growth depending on the reserve substance was limited for 6-7 weeks after planting. In no fertilized group, the length of new shoots increased up to 6-7 weeks, form then decrease. 3. Dry leaf yield three weeks after planting was found to be no difference in the same diameter of saplings between with and without fertilization. 4. The content of chlorophyll in the leaves was higher in thicker group and in fertilized group than thinner and no fertilized one, respectively. 5. There was no difference in the amount of carbohydrates of the bark of stem and root among the stem diameters. There was a transient increase in the amount of total sugar in the bark of stem and root in the first three weeks after planting, followed by gradual decline up to five weeks. The amount of reducing sugar in the bark of stem and root increased both in fertilized and in no fertilized group up to three weeks after planting, and then it decreased. There was a gradual decline in the content of starch in the bark of stem and root both in fertilized and in no fertilized group up to five weeks after planting, followed by gradual increase.
