Production of In-Vivo Embryos by Superovulation and Result of Transfer with Fresh or Frozen Embryos for Hanwoo and Holstein Cattle (한우 및 젖소에서 과배란 처리를 이용한 체내수정란 생산과 신선 및 동결 수정란 이식 결과)

  • To investigate the feasibility of embryo transfer technology to promote productivity of cattle, 36 cows(18 Holstein, 18 Hanwoo) were superovulated. Fresh embryos were transferred to 25 recipients(14 Holstein, 11 Hanwoo), whereas frozen embryos were transferred to 17 recipients(10 Holstein, 7 Hanwoo). Two embryos were transferred at a time to 13 recipients(9 Holstein, 4 Hanwoo) to produce twin calves. 1. 75.0% of donor cattle were reacted to hormonal treatment far superovulation. 2. The rate of embryo recovery by non-surgical method for Holstein and Hanwoo was 90.4 and 95.8% in comparison with numbers of corpus luteum. 3. Of all the ova collected non-surgically, the rate of viable blastocyst was 66.4% and the rate of transferrable blastocysts was 48.6%. 4. The rate of embryo collection by one-catheter method was 75.0%. 5. The rate of pregnancy/delivery following embryo transfer with fresh embryos was 60.0%. 6. The rate of pregnancy/delivery following embryo transfer with frozen embryos was 35.3%. 7. In embryo transfer to produce twin calves, the rate of pregnancy/delivery was 28.6% with fresh embryos and 16.7% with frozen embryos.

Development of Predictive Models of Listeria monocytogenes in Fresh-Cut Fruits and Vegetables (신선편의 냉장·냉동 과채류에서 Listeria monocytogenes의 예측모델 개발)

  • Processing fresh produce into fresh-cut products increases the risk of bacterial growth and contamination by breaking the exterior barrier of produce. Our objective in this study was to develop predictive models of Listeria monocytogenes in the fresh-cut salad, fresh-cut pineapple, and frozen mango. Predictive growth and survival models were developed to predict the change of L. monocytogenes populations in the fresh-cut salad (4, 10, 12, 13, 17, 25, and 36℃), fresh-cut pineapple (4, 10, 17, 25, 30, and 36℃), and frozen mango (-2, -10 and -18℃) as a function of temperature. The growth of L. monocytogenes in fresh-cut salad and pineapple was observed at above 13℃ and 10℃, respectively. The growth of L. monocytogenes in pineapple was faster than in salad. The delta value of L. monocytogenes in frozen mango increased as the storage temperature decreased. The results indicate that L. monocytogenes behave differently according to the physicochemical properties of fresh-cut fruits and vegetables. Since L. monocytogenes grow and survive well in refrigerated and frozen conditions, management programs and preventive controls for the processing of fresh-cut produce should be effectively implemented to enhance the safety of fresh-cut fruits and vegetables at retail markets.

Volatile Flavor Compounds of Korean Shiitake Mushroom(Lentinus edodes) (한국산 표고버섯의 휘발성 향기성분)

  • The volatile flavor compounds of fresh and dried shiitake mushrooms(Lentinus edodes) were extracted by simultaneous steam distillation-extraction apparatus, and analyzed by combined GC and GC-MS, and effects of pH on the formation of volatile compounds in fresh shiitake mushroom were investigated. Of the 29 compounds identified from fresh shiitake mushroom, the main volatile compound was 1-octen-3-ol comprising about 74.7% of the total volatiles and that in dried shiitake mushroom was 1, 2, 4-trithiolane comprising about 66.3%. With the exception of above two compounds, 3-octanone, 1-octen-3-one, 3-octanol, cis-2-octenal, n-octanol and cis-2-octenol as $C_8$ compounds were identified. Carbon disulfide, dimethyl disulfide, dimethyl trisulfide, 1-(methyl thio)-dimethyl disulfide, 1, 2, 4, 5-tetrathiane as sulfurous compounds were also identified. The formation of $C_8$ compounds in fresh shiitake mushroom during immersion was dominant in the range of pH 6.0 to 7.0, while the formation of sulfurous compounds in the range of pH 8.0 to 9.0.

Safety Effects against Nitrite and Nitrosamine as well as Anti-mutagenic Potentials of Kale and Angelica keiskei Vegetable Juices (케일과 신선초 채소즙의 안전성 및 항돌연변이 효과)

  • Vegetables contain high levels of nitrate, which can be converted to nitrite for reaction with secondary amines to form nitrosamines. In this study, we evaluated safety effects against nitrite and nitrosamine as well as anti-mutagenic activities of vegetable juices. To do this, the contents of nitrate, nitrite, and nitrosamines were determined in vegetable juices. The safety effects against nitrite and nitrosamine formation were also investigated under simulated human gastric conditions. The contents of nitrate and nitrite in common and organic kale and Angelica keiskei juices were 931~2,052 mg/kg and 13~82 mg/kg, respectively. However, seven kinds of nitrosamines were not detected in the vegetables juices. The nitrate content decreased when vegetable juices were digested under simulated human gastric conditions. Nitrosamine (N-nitrosodimethylamine) formation under simulated human gastric conditions was inhibited by addition of vegetable juices. In addition, vegetable juices, especially organically cultivated juices, showed anti-mutagenic effects in a Salmonella assay system. These results suggest that organically cultivated vegetable juices are a promising health-promoting source.

  • Onion has been reported to contain various organosulfur compounds which have antibiotic and anticarcinogenic properties and flavonoid like quercetin which is a valuable natural source of antioxidants. Carbohydrates in onion constitute about 80% of dry matter, and the major non-structural carbohydrate of onion bulb is fructo-oligosaccharides, well known as fructan, followed by glucose, fructose, and sucrose. The sugar concentration is associated with dormancy and storage life of onion, occurring as decrease in glucose, fructose and fructan, particularly towards the end of storage. Forced air pre-drying for 15-20 days at room temperature is an essential procedure to reduce freezing injury and sprouting, then onion bulbs can be stored at $0^{\circ}C$ for 6 months to control sprouting and decay. Bacterial soft rot caused by $Erwinia$ and $Pseudomonas$ is the main postharvest disease when the bulbs are infected with the bacteria and stored at room temperature. Browning in sliced onion is due to oxidation of phenolic compounds by polyphenol oxidase and it can be inhibited by citric acid treatment, packing with nitrogen gas, and polyethylene film.

A Mid-late Maturing, Lodging-Tolerant and Waxi rice Variety "Baegseolchal" (벼 중만생 내병 다수성 신품종 "백설찰")

  • "Baegseolchal" is a new Japonica type waxi rice variety developed and registered by the rice breeding team of Department of Rice and Winter Cereal Crop, NICS, RDA in 2007. This variety was derived from a across between Iksan435 which is highly tolerant to lodging and high yield potential, and Iksan425 which is resistance to multiple diseases. This variety has about 120 days of growth duration from transplanting to harvesting in middle, Honam and Youngnam plain of Korea. It is about 80 cm in culm length and tolerant to lodging. It is moderately resistant to blast, and resistance to bacterial blight pathogen races from $K_1$ to $K_3$, but susceptible to other major diseases and insect pests. "Baegseolchal" is midium-short grain and the milled rice exhibits high whiteness. The milled rice yield was about 5.37 MT/ha in local adaptability test for three years. "Baegseolchal" would be adaptable to plain areas south of the Chungnam Province of Korea.

Understanding the Effects of Different Edible Coating Materials on the Storability of 'Bing' Sweet Cherries

  • 'Bing' sweet cherries (P. avium L.) were coated with four different coating materials at 1% concentration ($Semperfresh^{(R)}$, calcium caseinate, chitosan, or TIC $Pretested^{(R)}$ colloid 911)and stored at $2.0^{\circ}C$ and 88% RH up to 35 days. The influence of different coating materials on the storability of fresh cherries was investigated. $Semperfresh^{(R)}$ coatings significantly improved overall quality of fresh cherries by decreasing weight loss and improving color stability, and chitosan-based coatings were effective in controlling mold incidence. However, colloid 911 and calcium caseinate coatings did not show significant benefit in preventing quality deterioration of fresh cherry during storage, probably due to their hydrophilic nature leads to exacerbated weight loss and shriveling with the possible interactions between coating materials and cherries epidermal layers.

Effect of Polypropylene Film Package and Storage Temperature on the Shelf-life Extension of Spinach (Spinacia oleracea L.) (시금치 포장 및 보관온도가 품질보존에 미치는 영향)

  • To extend the shelf-life of spinach after harvest, we investigated the effect of various packaging methods and storage temperature. In case of polypropylene film package, there was no weight loss, but in non-package, remarkably weight loss occurred as storage period extended and storage temperature risen. Content of vitamin C was decreased rapidly at early stage of storage, and decrease of vitamin C in low temperature storage was lower than that of room temperature storage, but its large difference according to packaging method was not observed. In hunter's value on spinach, b value in room temperature storage was higher than that of low temperature storage. In terms of freshness, shelf-life of spinach by low temperature storage $(1{\sim}3^{\circ}C)$ after PP film packaging lasted 34 days, and that by room temperature storage $(10{\sim}15^{\circ}C)$ after PP film packaging lasted 8 days, but that by room temperature storage after non-packaging lasted 3 days.

  • This study was performed to evaluate the effects of combined treatment of clove bud essential oil (CBEO) and mild heat (MH) on inactivation of Escherichia coli O157:H7 inoculated onto red oak leaves. Combined treatment of 0.2% CBEO with MH at $50^{\circ}C$ exhibited the highest inhibitory effect against E. coli O157:H7 among treatments, resulting in 1.45 log reduction compared with water washing treatment. In addition, inhibitory effect of the combined treatment was maintained during storage of red oak leaves at $4^{\circ}C$ for 9 days, showing 1.67~2.25 log reductions compared with non-treated samples. Thus, these results indicate that combined treatment with CBEO/MH can be used to ensure the microbiological safety of fresh leaf vegetables such as red oak leaves during storage.

Microbe and Quality Changes of Ready-to-Eat Lettuce during Storage at Different Temperatures (신선편이 양상추의 온도별 저장 중 미생물과 품질변화)

  • Microbe and quality changes of vacuum-packaged ready-to-eat lettuce were analyzed. While the vacuumpackaged lettuce after chlorine sanitizer were stored at $5^{\circ}C$, $15^{\circ}C$, and $25^{\circ}C$ for 7 days, viable numbers of total aerobic bacteria (TAB), coliform, E. coli, food-borne pathogens and lactic acids bacteria (LAB) were counted with gas production and sensory evaluation. Before the storage, only TAB of 2 log CFU/g and coliform of 1 log CFU/g were detected and LAB was not detected. TAB, coliform and LAB increased by 1 log CFU/g at $5^{\circ}C$ for 7 days without any detection of the pathogens. Sensory evaluations for off-flavour and crispness dropped to half the best value at 5 day storage. TAB and coliform increased by 3 log CFU/g and 2 log CFU/g, respectively, but LAB grew very actively by 4 log CFU/g under anaerobic environment and only B. cereus were detected after enrichment of the lettuce at $15^{\circ}C$ for 3 days. The evaluations for off-flavour and crispness were half the best value for 3 days. However, TAB and coliform increased by 3 log CFU/g, 1 log CFU/g, and 4 log CFU/g, respectively only at 1 day storage under $25^{\circ}C$. Also B. cereus were detected after enrichment and the sensory evaluation were half the best value within 1 day storage. Therefore, preservation at the lowest temperature possible is required for growth inhibition of the bacteria contaminated in the lettuce. Interestingly, LAB among them grew most actively under the anaerobic condition and the adulteration of lettuce might be closely related with the growth of LAB.
