Change of Peroxiredoxin-5 Expression by Curcumin Treatment in Cerebral Ischemia (허혈성 대뇌손상시 curcumin 투여에 의한 peroxiredoxin-5 발현의 변화)

  • Curcumin plays a protective role in brain injury through its anti-oxidant and anti-inflammatory activities. Moreover, peroxiredoxin-5 exerts a protective effect against oxidative stress. The aim of this study was to investigate whether curcumin modulated the peroxiredoxin-5 expression in focal cerebral ischemic animal model. Middle cerebral artery occlusion(MCAO) was performed to induce cerebral ischemic injury in rats. Adult male rats were injected intraperitoneally with vehicle or curcumin(50mg/kg B.W.) 1 h after MCAO and cerebral cortex tissues were collected 24 h after MCAO. Photographs of hematoxylin and eosin staining showed that MCAO induced necrotic changes with scalloped shrunken form and apoptotic changes with nuclear chromatin condensations. However, curcumin treatment attenuated MCAO-induced histopathological changes. Moreover, this study clearly showed that peroxiredoxin-5 expression was decreased in MCAO operated animal with vehicle using a proteomics approach. However, this decrease in peroxiredoxin-5 expression was attenuated by curcumin treatment. Reverse-transcription PCR and Western blot analyses confirmed that curcumin treatment alleviated the MCAO injury-induced decrease in peroxiredoxin-5 expression(p<0.05). These results demonstrated that curcumin regulates peroxiredoxin-5 expression in MCAO animal model. In conclusion, our findings suggest that curcumin exerts a neuroprotective effect in cerebral ischemia by attenuating the MCAO-induced decrease in peroxiredoxin-5 expression.

Neuroprotective effect by Juglandis semen-herbal acupuncture against $H_2O_2-induced$ apoptosis in human neuroblastoma, SH-SY5Y cells (과산화수소로 유발된 사람 신경아세포종의 아폽토시스에서 호도 약침액의 신경보호효과)

  • 목 적 : 과산화수소는 산화적 스트레스를 통해 아폽토시스를 유도하는 것으로 알려져 있다. 본 논문에서는 과산화수소로 유발된 신경아세포종 아폽토시스 과정에서 호도약침액의 효과를 관찰하였다. 방 법 : 과산화수소로 인한 신경아세포종의 아폽토시스에서 호도약침액의 효과를 알아보기 위해 배양 중인 신경아세포종에 과산화수소를 처리하고, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MIT)분석법, 4,6-diamidino-2-phenylindole (DAPI) 염색법, reverse transcription-polymerase chain reaction (RT-PCR), western blotting의 방법으로 확인하였다. 결 과 : 과산화수소로 인한 신경아세포종의 아폽토시스에서 호도약침액을 처리한 결과, 약침액을 처리한 세포의 생존이 약 30% 정도 증가하고, 핵 응축과 단편화를 막아주며, CASP3와 BAX단백질의 발현이 감소되었다. 결 론 : 이러한 결과로 호도약침액이 과산화수소로 인한 신경아세포종의 아폽토시스과정에서 보호효과를 나타내는 것으로 사료된다.

Neuroprotective and Anti-inflammatory Effects of Phenolic Compounds in Panax ginseng C.A. Meyer (인삼에 함유된 페놀성 선분의 신경세포보호 및 항염증 효과)

  • The six phenolic-compound (ascorbic acid, maltol, esculetin,p-coumaric acid, cinnamic acid, and quercetin) contents of Panax ginseng c.A. Meyer were determined in this study. The results showed that the ascorbic acid, cinnamic acid, and esculetin contents of Panax ginseng C.A. Meyer are higher than those of the other ingredients. Among these compounds, ascorbic acid and cinnamic acid significantly inhibited LPS-induced nitric oxide production in the RAW 264.7 cells. Cinnamic acid also effectively inhibited the oxidative damages in the human neuroblastoma SH-SY5Y cells. Although this study examined the neuroprotective and anti-inflammatory activities using only one kind of cells, its results suggest that cinnarnic acid potently contributes to the neuroprotective and anti-inflammatory properties of Panax ginseng C.A. Meyer.

Preventive mechanisms of Artemisia capillaries extract on ethanol-induced apoptosis in neuroblastoma cell line (균진약침액(菌蔯藥鍼液)이 에탄올로 유발된 신경아세포의 아폽토시스에 대한 보호 효과의 기전)

  • 목적 : 인진(茵蔯)이 SK-N-MC 신경아세포주에서 에탄올에 의해 유발된 아폽토시스에 대한 보호작용의 기전을 연구하였다. 방법 : SK-N-MC cell line에서의 세포 보호 기전을 알아보기 위하여 reverse transcription polymerase chain reaction (RT-PCR) 기법을 이용하여 bcl-2, bax 및 caspase-3의 변화를 관찰하였다. 결과 : RT-PCR을 이용하여 분석한 결과 SK-N-MC neuroblastoma에서 에탄올 처치는 bax, bcl-2 및 caspase-3 mRNA의 발현을 증가시켰다. 인진액의 전처치후 에탄올 처치한 신경 아세포에서는 에탄올에 의해서 증가된 bax와 caspase-3 mRNA 발현이 억제되었으나, bcl-2의 발현에는 유의한 증가를 나타내지 않았다. 결론 : 이상의 결과를 통하여 에탄올에 의해서 유발된 신경아세포의 아폽토시스에서 인진이 세포보호 효과가 있음이 확인되었고 그 기전은 bax와 caspase-3의 억제에 기인할 가능성을 시사한다고 할 수 있다.

Protective Effect against Neuronal Cell and Inhibitory Activity against Bacteria of Mulberry Fruit Extracts (오디 추출물의 신경세포 보호활성 및 항균활성)

  • As functional evaluation of mulberry fruits extracts, the protective effect on cerebral cell and antibacterial activities were carried. $1\%$ HCl-MeOH extract showed $37\%$ cytoprotective effect on hydrogen peroxide, also C3G identified mulberry fruits and cyanidin showed $52\%,\;76\%$, respectively, protective effects on oxygen-glucose deprivation (OGD). In the antibacterial activity of mulberry fruit extracts, MeOB-Cheongil extract showed the highest inhibitory activity. Salmonella typhimurium was shown inhibitory rate more than $70\%$ in all treatment groups. Also Klebsiella pneumoniae was shown inhibitory activity in all treatment groups.

Neuroprotective and Anti-inflammatory Effects of Bee Venom Acupuncture on MPTP-induced Mouse (MPTP 유발 파킨슨병 동물 모델에 대한 봉독약침의 신경보호 효과 및 항염증 효과)

  • 목적 : 이 연구는 MPTP 유발 파킨슨병 동물 모델에서 봉독약침의 신경보호 효과 및 항염증 효과를 확인하기 위해 시행되었다. 방법 : C57BL/6 mice에 신경독소인 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)를 하루에 2시간 간격으로 MPTP-HCl(20mg/kg per dose)을 4번 복강 내 주입하여 중뇌 흑질의 도파민 신경세포를 파괴한 파킨슨병 동물 모델을 유발하였다. 실험군은 MPTP군, MPTP 현종 BVA군, MPTP 곡지 BVA군, MPTP 신수 BVA군의 4군으로 하였다. 마지막 MPTP 투여 2시간 후에 1차로 봉독약침을 시술하고, 그 후 48시간 간격으로 총 5차 연속 시술하였다. 봉독약침액의 농도는 0.2mg/Kg으로 하였고, 경혈은 양측 현종($GB_{39}$), 곡지($LI_{11}$), 신수($BL_{23}$)를 사용했고, 주입량은 각 경혈당 양측으로 각 $20{\mu\ell}$씩 주입하였다. 항염증작용을 알아보기 위해 TH, MAC-1, iNOS HSP70을, 세포사멸에 대한 신경세포의 보호효과를 알아보기 위해 caspase-3을 면역조직화학법을 사용하여 실시하였다. 결과 : 실험 결과 MPTP 유발 파킨슨병 동물 모델에서 현종 곡지 신수혈에 대한 봉독약침은 TH-Immunoreactivity neuron의 감소와 microglial activation을 억제하였다. 봉독약침군 모두 효과를 보였으나 그 중 현종과 신수혈에서 특히 억제작용이 컸다. MAC-1에서는 현종혈이 억제작용이 컸다. HSP70-IR neuron은 곡지에서 유의한 억제작용을 보였으나, iNOS neuron은 모든 군에서 유의한 차이를 보이지 않았다. 또한 세포사멸억제여부 실험에서 봉독약침은 모두 억제작용을 보였으나 특히 곡지자침군에서 caspase-3 발현을 유의하게 억제하였다. 결론 : 이러한 결과는 봉독약침이 MPTP 투여로 인한 중뇌 흑질의 염증에 의한 도파민 신경세포 손상을, 염증을 억제함으로써 항염 효과를 나타냄을 알 수 있으며, 신경세포를 보호하는 활성이 있음을 보여줌과 동시에 세포사멸을 억제하는 활성이 있다고 사료된다.

Free radical scavenging activity and protective effect of three glycyrrhiza varieties against hydrogen peroxide-induced oxidative stress in C6 glial cells (종류별 감초의 라디칼 소거능 및 H2O2에 의한 C6 glial 세포의 산화적 스트레스 개선 효과)

  • Oxidative stress is common cause of neurodegenerative diseases. The purpose of this study is to investigate the in vitro free radical scavenging activity and protective effect of three Glycyrrhiza species including Glycyrrhiza uralensis, G. glabra, and a new variety of Glycyrrihza (Shinwongam, SW) against hydrogen peroxide-induced oxidative stress in C6 glial cells. In vitro assays, radical scavenging activities of G. uralensis, G. glabra, and SW against 2,2-diphenyl-1-picrylhydrazyl, ·OH, and O2- increased as concentration-dependent manner. In addition, the SW was found to contain the highest polyphenol and flavonoid contents. The treatment of H2O2 to C6 glial cell induced oxidative stress, whereas G. uralensis, G. glabra, and SW significantly increased the cell viability as dose-dependent manner. In particular, SW exerted stronger protective effect on H2O2-induced cytotoxicity, than G. uralensis and G. glabra. Furthermore, reactive oxygen species (ROS) formation was significantly elevated by H2O2 in C6 glial cells. However, treatments of G. uralensis, G. glabra, and SW decreased ROS formation. In addition, SW decreased pro-inflammatory related protein expression levels such as inducible nitric oxide synthase and cyclooxygenase-2, compared to H2O2-treated control group. These results indicated that G. uralensis and G. glavra, especially SW, may be useful for preventing from oxidative stress-induced neuronal damage by regulating inflammatory reaction.

Anti-oxidative Activity and the Protective Effect of Donkey's Bone and Skin Extracts on SK-N-SH Cells (당나귀 사골과 껍질의 항산화기능 및 SK-N-SH세포 보호효과)

  • The aims of this study were to determine antioxidation effect and neuroblastoma cell protection effect of donkey's bone and skin extracts (DBSE). DBSE was extracted by a pressure-cooker for 48 h and lyophilized. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was significantly increased with increased doses of DBSE and 40 mg/ml of DBSE showed 95.43% of the DPPH scavenging effect, which was equivalent to 1 mg/ml of vitamin C. The 2,2'-azino-bis (3-ethylbenzothiazoline-6- sulphonic acid) (ABTS) radical scavenging activity was also increased in a dose-dependent manner, and 20 mg/ml of DBSE showed 88.73% of the ABTS scavenging effect. The oxygen radical absorbance capacity (${\mu}M$ Trolox equivalent) of DBSE was significantly increased at a concentration of 10 mg/ml, which showed $132.53{\mu}M$ TE. The viability of oxidatively stressed brain cells induced by $500{\mu}M\;H_2O_2$ was protected by DBSE at concentrations greater than $50{\mu}M$. Cell viability after DBSE treatment at 50 and $100{\mu}g/ml$ was 53.78 and $54.34{\mu}M$ TE, respectively. There was no significant difference between both doses; however, 200 and $500{\mu}g/ml$ of DBSE showed 59.74 and 66.08% of cell viability, respectively indicating that DBSE protected SK-N-SH from oxidation stress. These results suggest that DBSE may have potential to be used as natural antioxidants in food industry, while in vivo evidence is necessary to support DBSE's in vitro-based antioxidative efficiency.

Antioxidant and Neuronal Cell Protective Effects of an Extract of Houttuynia cordata Thunb (a Culinary Herb) (어성초 추출물의 항산화 및 신경세포 보호효과)

  • The in vitro antioxidant activities and neuronal cell protective effects of 60% (w/v) methanolic extract from Houttuynia cordata were investigated. The contents of total phenolics and quercitrin in the extract were 17.71 mg/g and 75.80 ${\mu}g$/g, respectively. DPPH and ABTS radical-scavenging activities were 87.79% and 99.27%, respectively, when the extract was tested at 5 mg/ml. The FRAP (ferric reducing/antioxidant power) assay showed a dose-dependent increse in activity. In a cell viability assay using MTT, the extract protected against $H_2O_2$-induced neurotoxicity. Lactate dehydrogenase (LDH) leakage was also inhibited by the extract, as was lipid peroxidation as shown using the mouse brain homogenate test. These data indicate that a 60% (w/v) methanolic extract of Houttuynia cordata has in vitro antioxidant activities, and ingestion there of may reduce the risk of developing neurodegenerative disorders.

Neuroprotective effect of Deodeok (Codonopsis lanceolata) bud extracts in H2O2-stimulated SH-SY5Y cells (더덕순 에탄올 추출물의 신경세포 보호 효과)

  • Purpose: Deodeok (Codonopsis lanceolata) is generally used in conventional medicines and is considered to have remedial properties to cure several diseases. However, application of the C. lanceolata bud as a novel food ingredient has not been fully explored. Hydrogen peroxide (H2O2) is associated with the production of oxidative damage that results in mutagenesis, carcinogenesis, and cell death. This study examines the neuroprotective effect of C. lanceolate bud extracts (CLBE) on H2O2-stimulated apoptosis in SH-SY5Y cells. Methods: C. lanceolata bud of length 10 to 15 cm was collected and extracted using 70% ethanol. Cytotoxicity was evaluated by the EZ-cytox reagent, measurement of lactic dehydrogenase (LDH) release and reactive oxygen species (ROS). The morphological changes of the nuclei were determined using the Hoechst 33258 dye. Enzyme activities were analyzed using the caspase activity assay kit. Related protein expressions were quantified by the Western blot immunoassay in H2O2-stimulated SH-SY5Y cells. Results: Cell viability, LDH release and ROS generation, demonstrated neuroprotective effects of CLBE in H2O2-stimulated SH-SY5Y cells. The occurrence of apoptosis in H2O2-stimulated cells was confirmed by caspase activity, which was increased in H2O2-stimulated SH-SY5Y cells compared to the unexposed group. Pretreatment of CLBE was observed to inhibit the H2O2-stimulated apoptosis. In addition, exposure to CLBE resulted in increased expression of the Bcl-2 (B cell lymphoma 2) protein and decreased expression of the Bax (Bcl2 associated X) protein. Conclusion: This study shows that exposure to CLBE alleviates the H2O2-stimulated neuronal damage in SH-SY5Y cells. Our results indicate the potential application of CLBE in neurodegenerative disease therapy or prevention.
