Anti-obesity Effect of Chopped Sauce with Medicinal Plant Extracts (약용작물 추출물을 첨가한 다진양념의 비만 억제 효과)

  • 본 연구는 항비만 기능성을 가지면서 맛과 향이 증진된 분말형 다진양념(다데기)을 개발하기 위해 첨가할 수 있는 약용작물들의 선정 및 그 기능성을 평가하기 위해 실시되었다. 오미자, 진피, 감초, 녹차엽 등을 첨가군으로 선정하여 각각의 시료들을 8시간, $80^{\circ}C$ 증류수에서 추출 후 감압농축하여 추출물을 제조한 뒤 마우스 유래 adipocyte (3T3-L1)에 처리하여 세포내 지방 축적 억제 정도를 실험하였다. 시료 처리 배양 5일 후 오미자, 진피, 감초, 녹차엽 추출물을 처리한 모든 군에서 대조군과 비교시 20% 이상 세포내 중성지방 농도가 감소하였으며 특히 오미자 군에서는 30% 이상 감소를 보였다. C57BL/6 생쥐를 이용하여 고지방식이로 비만을 유도하면서 분말형 다진양념, 오미자, 진피, 감초, 녹차엽 추출물을 경구투여한 뒤 체중증가를 조사한 결과 8주 후 고지방식이와 다데기만을 경구투여한 군은 고지방식이군과 비교하여 체중감소경향만을 보였으나 분말형 다진양념과 추출물을 함께 경구투여한군에서는 유의적으로 체중이 감소하는 결과를 보였다. 이들 추출물을 혼합하여 제조한 분말형 다진양념은 관능검사에서도 고춧가루 특유의 풋내를 감소시켜 향미가 증진되는 것으로 평가되어 오미자, 진피, 감초, 녹차엽을 첨가한 다양한 항비만 기능성 양념 개발이 기대된다.

  • This study was orchestrated with the purpose of uncovering new nutraceutical resources possessing biological activities in the plant kingdom. To fulfill our objective, we analyzed several plant extracts and selected five species possessing powerful anti-oxidative activity. The anti-oxidative effect of these five plants, Apios fortunei Maxim., Colubrina arborescens Sarg., Croton caudatus Geiseler, Osmanthus matsumuranus Hayata and Schima noronhae Reinw. ethanol extracts were then evaluated by using in vitro assay, cell model system, and Western blot analysis of target proteins. As the results, all of them possessed the potent scavenging activity against 1,1-diphenyl-2-picryl hydrazyl (DPPH), similar with that of ascorbic acid, used as a common positive control. Moreover, they strongly inhibited hydrogen peroxide ($H_2O_2$)-induced reactive oxygen species (ROS), in a dose-dependent manner, in RAW 264.7 murine macrophage cells. Furthermore, they induced the protein expression of an anti-oxidative enzyme, heme oxygenase 1 (HO-1), and its upstream transcription factor, nuclear factor-E2-related factor 2 (Nrf2). Taken together, these results indicate that these five plants possess potent anti-oxidative activity and thus appear to be useful sources as potential anti-oxidant agents. Therefore, they might be utilized as promising materials in the field of nutraceuticals.

Biological activities of Brassica rapa (Turnip) callus extracts by plant cell culture technology (식물세포배양기술을 이용한 순무 캘러스 추출물의 생리활성)

  • The purpose of this study was to investigate biological activities of Brassica rapa (Turnip) plant callus extracts of Ganghwa-gun of Incheon city using water, ultrasonic wave and ethanol extractions to develop functional materials. DPPH radical scavenging activities of the callus extracts were increased in a concentration-dependent manner, as compared with control. The astringent effects of the ethanol extracts were higher, as compared to water and ultrasonic extracts. In the collagen synthesis assay, the ethanol extract showed significant anti-wrinkle effects of 59% and 78% at a concentration of 5 ppm and 10 ppm, respectively. These results suggested that water, ultrasonic wave and ethanol extracts of turnip plant calluses are natural antioxidant sources. Especially, the ethanol extract can be regarded as a functional, natural cosmetic material with astringent and anti-wrinkle effects.

Screening of Antiviral Activity from Natural Plants against Feline Calicivirus (Feline calicivirus에서 항바이러스 활성을 가지는 천연식물자원 탐색)

  • In an effort to discover an antiviral substance against noroviruse (NV), which causes gastroenteritis illness world-wide, several plants including spices and herbs were evaluated for their antiviral activities against feline calicivirus (FCV) as a surrogate for NV. Among them, methanolic extract of green tea (Camellia sinensis L.) exhibited significant antiviral activity against FCV. After treatment with green tea extract (3.13 mg/ml) for 1 hr, FCV was completely inactivated. The antiviral activity of green tea extract against FCV was also determined to be dose and time- dependent. The results obtained in this study suggested that green tea will be effective in the prevention of food-borne diseases caused by NV.

Effects of Hot Water Soluble Extract from Green Tea on the Lipid Metabolism and Antioxidant Effect in Rats fed Animal or Vegetable Protein and a Hypercholesterol Diet (녹차 열수추출물이 동$\cdot$식물성 단백질과 고콜레스테롤을 급여한 흰쥐의 지질대사와 항산화효과에 미치는 영향)

  • The aim of this study is to investigate the effects of hot water soluble extract from green tea on the components of serum and the liver and the antioxidative effects in accordance with the different protein types. For this purpose, four experimental groups were set up. As for the protein source casein, isolated soy protein was supplemented to the rats, together with hot water soluble extract from green tea. Four experimental groups kept eight Sprague-Dawley rats respectively. The CP group was supplemented with casein only, the CG group was supplemented with casein and hot water soluble extract from green tea, the ISP group was supplemented with isolated soy protein only, and the ISG group was supplemented with the isolated soy protein with hot water soluble extract from green tea. After 4 weeks of feeding with experimental diet, the levels of serum and liver lipid and antioxidant enzyme activity and TBARS in the liver were measured. The results are; 1. Weight gain and FER were higher in the casein group than in the isolated soy protein group in general. In the casein group, the weight gain and FER were reduced significantly when hot water soluble extract from green tea was supplemented (P < 0.05). There was no significant difference in feed intake. 2. In general, total cholesterol and LDL-cholesterol in the serum were higher in the casein group than in the isolated soy protein group, however just the concentration of LDL-cholesterol in the casein group was significantly lower when the hot water soluble extract from green tea was supplemented (P < 0.05). 3. Triglycerides in the liver were higher in the casein group than in the isolated soy protein group general, however when hot water soluble extract from green teas was supplemented only in the isolated soy protein group, the content of triglyceride in liver was significantly lower (P < 0.05). 4. There was no significant difference in antioxidant enzyme activity in the liver in all the groups, however the content of TBARS was low only in the casein group when hot water soluble extract from green tea was supplemented (P < 0.05).

  • 나노섬유를 제조하는 방법 중에는 상분리 현상을 이용한 방법, 자가 조립성을 이용한 방법, 템플레이트를 이용한 방법, 전기방사법이 있으며 특히 전기방사법은 연속적으로 균일한 나노섬유를 제조할 수 있다. 또한 전기방사법은 장비가 간단하며 고분자 blend ratio와 무기재료의 함량에 따라 뛰어난 특성을 나타내는 나노복합섬유를 만들 수 있다. 최근 식물에서 추출한 단백질을 전기방사법을 이용하여 나노입자 및 나노섬유를 제조하고 이를 의료 분야 등에 적용하기 위한 연구가 활발히 진행되고 있으며 이런 식물성 단백질은 동물성 단백질에 비하여 인체 적용이 용이하고 매장량이 풍부한 장점이 있다. 본 연구에서는 전기방사법을 이용하여 옥수수에서 추출한 단백질인 zein의 나노입자 및 나노섬유를 제조하였다. 또한 천연 추출물이 혼입된 복합 나노입자 및 나노섬유를 제조하여 zein이 가진 고유 특성 이외에 천연 추출물의 특성을 추가로 부여해서 더욱 발전된 나노입자 및 나노섬유를 제조하였다. 고분자 농도, 전압, 방사거리 등 다양한 공정변수를 조절하여 최적의 조건을 확립하였으며 제조된 나노입자 및 나노섬유는 field-emission type scanning electron microscope (FE-SEM), transmission electron microscopy (TEM), ultraviolet-visible spectroscopy (UV/vis), fourier transform infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC)를 이용하여 특성분석을 실시하였다.

Antimicrobial Activity of Some Plants Containing Allelochemicals (Allelochemicals 함유 식물의 항균 효과)

  • Korean native plants and naturalized plants were analyzed for allelochemicals, and their antimicrobial effects were studied. The difference in soluble solid contents between Korean native plants and naturalized species was not significant, and the Korean native plant, Solanum nigrum showed the highest soluble solid content of 90 mg/ml. The ethanol extract of the Korean native plant, Solanum nigrum showed antifungal activity to Aspergillus phoenicis KCTC 1228, with a clear zone of 18 mm, and spore formation was not observed from the treatment. The naturalized plants Ambrosia artemisiifolia var. elatior and Erigeron canadensis showed inhibition of spore formation and the clear zones were at 24 mm and 22 mm, respectively. The clear zones of Aspergillus phoenicis KCTC 1228 treated with ethanol extrats of Phytolacca americana and Rudbeckia bicolor were 22 mm and 19 mm, respectively, and spore formation was observed from the treatment. The Korean native plant, Solanum nigrum and naturalized plants, Phytolacca americana and Ambrosia artemisiifolia var. elatior showed antimicrobial activity against Bacillus sphiaericus 2362, and Bacillus sphiaericus 2297, Bacillus thuringiensis var. subtilis and Baicillus thuringiensis var. cereus. The antimicrobial activity of Ambrosia artemisiifolia var. elatior showed the largest clear zone of 32 mm against Bacillus thuringiensis var. subtilis. In general, the more soluble the solid contents of the extracts, the greater were the antifungal and antimicrobial activities. The phenolic compounds from the Korean native plant, Solanum nigrum and the naturalized species, Phytolacca americana and Ambrosia artemisiifolia var. elatior were analyzed by high performance liquid chromatography. Three phenolic compounds including hydroquinone were identified in Solanum nigrum. In contrast, five and seven phenolic compounds were identified in Phytolacca americana and Ambrosia artemisiifolia var. elatior, respectively. The antifungal activity against Aspergillus phoenicis KCTC 1228 was found to be due to the coumaric and benzoic compounds.

Bioactivities of Korean Ginkgo (Ginkgo biloba L.) Extract and Its Potential as a Natural Pesticide (은행나무 추출물의 생물활성 및 천연물농약으로 이용 가능성)

  • Bioactivities of Korean ginkgo (Ginkgo biloba L.) extract were investigated against several fungi, general bacteria and insect pests. Crude methanolic extracts of different parts of Korean ginkgo showed different bioactivities depending on the target organisms. The methanolic extract showed in vitro antimicrobial activity at dose of 200 ug per paper disc. The extract of ginkgo stalk was some higher than seed coat and root. The extract also showed a remarkable in vivo antifungal activity against green mold (Trichoderma harzianum) on compost surface of spawn bags and in vivo insecticidal activity to Nilaparvata lugens, Plutella xylostella and Tetranychus urticae. This study suggests that Korean ginkgo extracts have a potential as a natural pesticide.

Antimicrobial Effect of Phytochemicals to Listeria monocytogenes Isolated from Slaughterhouses

  • The objective of this study was to investigate the inhibitory effect of natural antimicrobials on Listeria monocytogenes isolated from Korean slaughterhouses. A mixture of 15 strains of L. monocytogenes at low (3 Log CFU/mL) or high (7 Log CFU/mL) concentration was exposed to various extracts (grapefruit seed extract, citrus fruit extract, ginger extract, pear extract, Japanese apricot concentrate, balloon flower extract, jujube extract, and omija extract) at $0.001-4.0{\mu}g/mL$. Ginger extract, pear extract, Japanese apricot concentrate, balloon flower extract, jujube extract, or omija extract showed no antimicrobial effects on high-concentration of L. monocytogenes (7 Log CFU/mL). However, grapefruit seed extract and citrus fruit extract showed antibacterial effects against L. monocytogenes at 3 and 7 Log CFU/mL with MBCs of 0.001 and $0.002{\mu}g/mL$, respectively. These results indicate that grapefruit seed extract and citrus fruits extract can be used to control L. monocytogenes as natural antimicrobials.
