스코폴라민으로 유도한 기억 손상 모델에서 용안육(龍眼肉)의 보호 효과 연구 (Memory Enhancing Effect of Longanae Arillus against Scopolamine-induced Amnesia in C57BL/6 Mice)

  • In this study, we have verified the memory and cognitive enhancing effect of Longanae Arillus, the fruit of Euphoria longana Lamarck, which has been used as a tonic and for the treatment of amnesia, insomnia, and palpitations in oriental medicine. To investigate the effect of Longanae Arillus water extract(LAE) on the memory and cognitive dysfunction, scopolamine (1 mg/kg, i.p.) was injected in C57BL/6 mice and several behavior tests including Y-maze, Morris water-maze, passive avoidance and fear conditioning tests were conducted. Administration of LAE (100 or 200 mg/kg/day, p.o.) effectively improved scopolamine-induced memory impairment and dysfunction. To further determine the possible molecule mechanism of LAE, we have examined the activity and/or mRNA expression of diverse proteins involved in the acetylcholine metabolism. LAE particularly increased the amount of acetylcholine in the cortex which was mediated by suppression of acetylcholine esterase (AchE) activity. In addition, LAE elevated the mRNA expression of muscarinic acetylcholine receptors (mAchRs) without affecting the mRNA levels of choline acetyltransferase (ChAT) and acetylcholine esterase (AchE). In another experiment, LAE effectively inhibited mRNA expression of pro-inflammatory cytokines such as tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-$1{\beta}$ (IL1-${\beta}$), which seemed to be mediated by inhibition of upstream transcription factor NF-${\kappa}B$ and extracellular-regulated kinase 1/2 (ERK1/2). These results demonstrate that Longanae Arillus can increase acetylcholine amount the cortex via regulation of AchE activity as well as mAchRs expression and decrease pro-inflammatory responses via inhibition of NF-${\kappa}B$ signaling pathway, thereby having therapeutic potential to improve memory and cognitive deficit in amnesia.

  • Cognitive impairment is considered to be key research topics in the field of neurodegenerative diseases and in understanding of learning and memory. In the present study, we investigated neuroprotective effects of Schisandra chinensis (SC) and Ribes fasciculatum (RF) extracts in hydrogen peroxide-induced neuronal cell death in vitro and scopolamine-induced cognitive impairment in Sprague Dawley® (SD) rat in vivo. Apoptotic cell death in neuroblastic PC12 cell line was induced by hydrogen peroxide for 1 hour at 100 μM. However, mixture of SC and RF treatment prevented peroxide induced PC12 cell death with no neurotoxic effects. For in vivo experiment, the effect of SC and RF extracts on scopolamine-induced cognitive impairment in SD rat was evaluated by spontaneous alternation behavior in Y-Maze test. After 30 min scopolamine injection, the scopolamine-induced rats presented significantly decreased % spontaneous alteration and acetylcholine level, compared to non-induced group. However, treatment of SC+RF extracts rescued the reduced % spontaneous alteration with acetylcholine concentration from hippocampus in scopolamine-induced rats. These results suggested that mixture of SC and RF extract may be a potential natural therapeutic agent for the prevention of cognitive impairment.

  • Objectives : In the present study, we assessed the effects of water extract of Ulmus davidiana(UED) on the learning and memory impairments induced by scopolamine in mice through its favorable acetylcholinesterase (AChE) activity and antioxidant effect. Methods : The memory and cognitive enhancing effect of the UDE was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. In addition, to examine the mechanism of UDE using acetylcholinesterase (AChE) and antioxidant activity. Results : The water extract of UDE (100, and 200 mg/kg) significantly reversed the scopolamine-induced cognitive impairments in the passive avoidance test (P < 0.05). Moreover, UDE (100, and 200 mg/kg) also improved escape latencies in training trials and increased swimming times and distances within the target zone of the Morris water maze (P < 0.05). On the Y-maze test, UDE (100, and 200 mg/kg) also significantly reversed scopolamine-induced cognitive impairments in mice (P < 0.05). In an in vitro study, UDE was found to inhibit acetylcholinesterase, changes in neurotrophic factor (CREB), and antioxidant activity in a dose-dependent manner. Conclusions : The water extract of UDE dramatically possesses the anti-amnestic and cognitive-enhancing activities related to the memory processes, and these activities were parallel to treatment duration and dependent on the learning models. These results suggest that the administration of UDE enhances learning and memory, and that this effect is partially mediated by ERK-CREB-BDNF signaling and the survival of immature neurons.

스코폴라민으로 유도된 기억력 손상에 대한 복신의 보호 효과 및 작용기전 연구 (Effect of Poria Cocos on the Scopolamine-induced Memory Impairment and Its Underlying Molecular Mechanism)

  • This study was performed to investigate the memory enhancing effect of Poria cocos Wolf (Hoelen cum radix) against scopolamine-induced amnesia in Sprague-Dawley (SD) rats. To induce amnesia, scopolamine (0.75 mg/kg) was intraperitonically injected into SD rats 30 min before starting behavior tests. We have conducted Morris water-maze and Y-maze tests to monitor learning and memory functions. Poria cocos effectively reversed scopolamine-induced memory impairment in SD rats which was represented by an improvement of mean escape latency in water-maze test and spontaneous alterations in Y-maze test. To elucidate possible molecule mechanism, we have measured mRNA as well as protein expression of acetylcholine esterase (AchE), choline acetyltransferase (ChAT), muscarinic acetylcholine receptor (mAchR), and brain-derived neurotrophic factor (BDNF) using RT-PCR and Western blot analysis, respectively. Poria cocos increased mRNA levels of ChAT and mAchR in rat hippocampus compared with those in the scopolamine-injected amnesic group. In addition, protein expression of ChAT and BDNF was also elevated by Poria cocos intake. Furthermore, as an upstrem regulator, the activation of cAMP response element-binding protein (CREB) was assessed by immunohistochemistry. In this immunohistochemical analysis, the phosphorylation of CREB (p-CREB) was reduced by scopolamine injection, which was restored back to control levels by administration of Poria cocos. These results suggest that Poria cocos may improve memory and cognitive deficit in amnesia and have therapeutic potentials through up-regulation of ChAT, mAchR, and BDNF, which seemed to be mediated by activation of CREB.

  • Perilla frutescens (P. frutescens) is an important herb used for many purposes such as medicinal, aromatic, and functional food in Asian countries and has beneficial effects such as antioxidant activity, anti-inflammation activity, anti-depression activity, and anxiolytic activity. However, there have been no studies on the protective effect of P. frutescens extract (PFE) on amnesia in vivo. The present study aimed to investigate whether PFE protects memory deficit using a scopolamine-induced mice model and elucidate the underlying mechanisms involved. The protective effect of PFE against scopolamine-induced memory deficits was investigated using Y-maze, passive avoidance, and Morris water maze tests. Furthermore, the potential mechanisms of PFE in improving memory capabilities related to the cholinergic system and antioxidant activity were examined. PFE significantly increased spontaneous alternation in the Y-maze test, step-through latency in the passive avoidance test, and swimming time in the target quadrant in the probe test when compared to the scopolamine-treated group. Likewise, PFE significantly decreased escapes latency in the Morris water maze test. PFE could not regulate cholinergic function in acetylcholine level and acetylcholine esterase activity. However, PFE increased DPPH radical scavenging activity dose-dependently and total polyphenol content was 127.7±1.2 ㎍ GAE/mg. The results showed that the PFE could be a preventive and/or therapeutic candidate for memory and cognitive dysfunction in Alzheimer's disease.

Scopolamine으로 인한 건망증 모델에서 sucralose의 기억력 및 인지기능 완화 효과 (Effects of sucralose on memory and cognitive function relief in a scopolamine-induced amnesia model)

  • Sucralose는 식품 분야에서 sucrose 대체물로 사용되며 세계적으로 승인된 무열량 고강도 인공 감미료이다. 하지만 감미료가 뇌에 미치는 영향에 대한 조사는 미비하여 본 연구에서 scopolamine으로 유발된 기억력 손상 동물모델에서 sucralose 단기 섭취가 인지 및 기억 보호 효과를 가지는지 확인하였다. Sucralose 2, 5, 10 mg/kg를 경구투여하였으며 30분 후 대조군, 약물군에 scopolamine (1mg/kg) 복강투여를 진행하였고 정상군에는 식염수로 복강투여 후 행동실험을 수행하였다. 결과적으로 Y-maze, passive avoidance, Morris water maze에서 대조군과 비교해 10% 이상의 인지기능이 회복되는 결과를 도출하였다. 또한, 전염증성 사이토카인을 측정한 결과 sucralose가 IL-6와 TNF-α를 30% 이상 억제하는 것을 보였으며 세포 내 신호 메커니즘으로 ERK-CREB의 발현량을 관찰한 결과 농도 의존적으로 증가하였다. 따라서 sucralose는 건망증 환자의 예방과 치료를 위한 기능성 식품과 관련이 있다고 시사된다.

${\alpha}_4{\beta}_2$ 니코틴성 아세틸콜린 수용체 영상 방사성리간드 $2-[^{18}F]fluoro-A85380$의 합성 및 평가 (Synthesis and Evaluation of $2-[^{18}F]Fluoro-A85380$, a Radioligand for ${\alpha}_4{\beta}_2$ Nicotinic Acetylcholine Receptor Imaging)

  • 목적: nAChRs의 ${\alpha}_4{\beta}_2$ 아형에 높은 친화력과 선택성을 갖는 리간드인 $2-[^{18}F]fluoro-A85380([^{18}F]1)$의 효율적인 합성과정을 연구하여 방사화학적 수율, 비방사능, 합성시간, 마우스에서 생물학적 평가의 측면에서 문헌에 보고된 결과들과 비교 평가하였다. 대상 밀 방법: 전구물질의 I을 $^{18}F$으로 치환한 다음 중간물질을 분리하지 않고 trifluoroacetic acid를 가하여 보호기를 제거하였다. 반응혼합액을 HPLC로 정제하여 얻은 방사성리간드($[^{18}F]1$)를 마우스의 꼬리 정맥에 주사한 후에 정해진 시간별로 6개의 뇌조직 (소뇌, 해마, 선조체, 피질, 시상, 상구)을 분리하여 무게를 측정하고 감마계수기를 이용하여 방사능을 측정하였다. 또한 스코폴라민, 메카밀아민, 사이티신, (-)니코틴, 비방사성 표준물질(1)을 마우스에 피하주사로 투여하고 방사성리간드 주사 후 90분에 6개의 뇌조직을 분리하여 무게 및 방사능을 측정하여 %ID/g으로 나타내었다. 이 결과를 대조군과 비교하였다. 결과: $^{18}F$ 표지 후 중간물질을 분리하지 않고 한 번의 HPLC 정제를 통하여 $[^{18}F]1$을 15-20%의 방사화학적 수율로 합성하였으며 비방사능은 $38-55GBq/{\mu}mol$이었다. 이 방사성리간드의 마우스 뇌에서의 분포결과, 주사 후 30분에 nAChRs가 풍부하게 분포되어 있는 시상과 상구에서 가장 많은 섭취를 보였고 이 수용체가 부족한 소뇌에는 거의 섭취되지 않았다. 여러 리간드를 사용하여 $[^{18}F]1$ 섭취의 억제효과를 본 실험에서는 nAChRs 작용제인 사이티신 및 (-)니코틴의 전처리가 시상 및 상구에서 $[^{18}F]1$의 섭취를 70-90% 감소시켰으며, 소뇌에서는 억제효과가 관측되지 않았다. 결론: 이 연구에서 사용된 방법으로 얻어진 $[^{18}F]1$의 방사화학적 수율, 비방사능, 뇌에서의 분포 그리고 여러 리간드에 의한 억제효과는 문헌에 보고된 결과와 유사하였다. 따라서 중간물질을 분리하지 않고 한번의 HPLC를 사용하는 방법이 $[^{18}F]1$의 상용생산에 유용하게 사용될 수 있을 것으로 기대한다.
