Optimization of industrial medium for the production of erythritol by candida magnoliae

  • Experiments were carried out to investigate the selection of industrial medium and optimization of fermentation process for the production of erythritol by Candida magnoliae SR101. In the batch fermentation, light steep water(LSW) was the best nitrogen source for the industrial production of erythritol. For the optimization of culture condition, the batch culture was performed. When the concentration of LSW was 65 mL/L in the defined medium containing 250 g/L of glucose, 44% of erythritol yield with 110 g/L of erythritol concentration and 0.66 g/L-hr of productivity, respectively were obtained. Two-stage fed-batch culture was performed to improve the volumetric productivity of erythritol. High density cell culture in the growth stage was performed by batch type with 100 g/L glucose and 500 mL/L LSW concentration, respectively. The cell yield was 0.72 g-cell/g-glucose. Productivity of erythritol was increased and concentration of organic acids such as gluconic acid and acetic acid were decreased when initial pH of 6.5 controlled by 28% ammonia water For increasing yield of erythritol, glucose concentration in the production stage was tested. 37% of total erythritol yield with 186 g/L of erythritol concentration and 1.66 g/L-hr of erythritol productivity were obtained when 820 g of glucose powder was directly added for making up 450 g/L of glucose at production stage.

Study on iodine Labelling (II) Efficient of Labelling Rose Bengal, Hippuran, and Human Serum Albumin in Small Scale

  • For efficient micro scale syntheses of Rose $Bengal-^{131}I$, $Hippuran-^{131}I$, and $H.S.A.-^{131}I$, the dependence of labelling yields on pH, on salt contents, and on the volume of buffer solution in the reaction mixtures as well as the reaction apparatus were studied. pH of 5.6 was optimum for preparation of both Rose $Bengal-_{131}I$ and Hippuran $-^{13}I$ but pH of 8.5 was optimum for preparation of $H.S.A.-^{131}I$. Salt in the reaction mixtures hindered drastically the formation of $Hippuran-^{131}I$ but it slightly increased the labelling yield of H.S.A.. The compactly closed reaction vessels were effective for preparations of both Rose $Bengal-^{131}I$and $Hippuran-^{131}I$ in small volume. Thereupon, the labelling procedures were modified to bring about higher labelling yields and better reproducibilities. By these newly established procedures, the labelling yields of Rose $Bengal-^{131}I$ and $Hippuran-^{131}I$ could be increased even with the home-produced sodium $iodide-^{131}I$ solution containing reducing agent.

Optimization of Production Yield for Neohesperidin by Response Surface Methodology (반응표면 분석법을 이용한 neohesperidin 생산 수율의 최적화)

  • Neohesperidin is a natural new nutrition sweetener, widely existing in plants of dry citrus peel, which can be derived from extraction. Since the sweetness is 1,300-1,500 times greater than that of sugar, neohesperidin are widely used in fruit juices, wines, beverages, bakeries and pharmaceutical formulations, and are particularly suitable for consumption by diabetic patients. However, the yield of extraction from citrus peel waste is very low. In this study optimal yield conditions were determinedusing response surface methodology (RSM) in order to increase the neohesperidin extraction yield. The critical factors for maximum extraction yield were selected extraction pressure ($x_1$), extraction time ($x_2$), and concentration of ethanol ($x_3$). As a result, the extraction yield was improved when the extracting pressure increased. The extraction yield also increased in a time-dependent manner. When adding ethanol as an assistance solvent to the supercritical carbon dioxide, extraction yield was increased as more ethanol concentration was added. Finally, the extraction yield of neohesperidin was improved to about 162.22% compared to ethanol extraction as a conventional method.

Fuzzy Logic Based Buffer Management Algorithm to Improve Performance of Internet Traffic over ATM Networks (ATM 네트워크에서 인터넷 트래픽 성능 향상을 위한 퍼지기반 버퍼 관리 알고리즘)

  • To support Internet traffic efficiently over ATM networks, Guaranteed Frame Rate(GFR) has been proposed in the ATM Forum. GFR provides minimum rate guarantees to VCs and allows any excess bandwidth in the network to be shared among the contending VCs in a fair manner. In this paper, we proposed a new fuzzy logic based buffer management algorithm that provides MCR guarantee and fair sharing to GFR VCs. A key feature of proposed algorithm is its ability to accept or drop a new incoming packet dynamically based on buffer condition and load ratio of VCs. This is achieved by using fuzzy logic controller for the production of a drop factor. Simulation results show that proposed scheme significantly improves fairness and TCP throughput compared with previous schemes.

Milling Condition and Harvesting Time for Improving Milling Recovery of Head Foxtail Millet Grain (완전미도정수율 향상을 위한 조(Setaria italica Beauvois) 도정방법 및 수확시기)

  • This study was conducted to establish proper milling condition and harvesting time for improving the milling recovery of head foxtail millet grain. Brown foxtail millet recovery and the percentage of head foxtail millet recovery were significantly different according to the number of hulling and milling time. Also, the interaction of these factors had influence on brown foxtail millet recovery and the percentage of head foxtail millet recovery. To improve the percentage of head foxtail millet recovery, it was effective to hull grain twice and mill for 1.30 minute. 1000-grain weight of brown foxtail millet, brown foxtail millet recovery, hardness of brown foxtail millet, milling recovery, and the percentage of milling recovery of head foxtail millet grain were significantly different according to harvesting day after heading. For improving the ratio of ripened grain and the percentage of recovery of head foxtail millet, it is recommended to harvest 45 days (accumulated temperature : $1,148^{\circ}C$) after heading (DAH) in early maturing Hwangkeumjo, 50 DAH (accumulated temperature : $1,150^{\circ}C$) in mid-late maturing Samdamae and Kyeongkwan1, and 55 DAH (accumulated temperature : $1,168^{\circ}C$) in late maturing Samdachal.

Effects of Additives on Yield of Coal Liquefaction (석탄액화시 첨가제에 의한 수율 향상 효과)

  • The effects of some additives (black liquor, NaOH, water and wood) on the conversion of coal and product were investigated in the lab-scale, high pressure reacting system around 375$^{\circ}C$. The addition of black liquor enhances the coal conversion yield about 38.6%, which is mainly due to NaOH in black liquor. Also, sulfur of the black liquor in coal liquefaction process evolved hydrogen sulfide, which causes the odor problem. Addition of water in coal liquefaction increased CO$_2$content in the gas phase, and low boiling range components in liquid products. Coprocessing of wood and coal at 400$^{\circ}C$ increased yield of liquid product about 8%, but higher temperature above 400$^{\circ}C$ reduced liquid product due to increase of gas products.

Development of Combined Method for Extraction of Sea Tangle (다시마의 효과적 추출을 위한 종합적인 추출방법의 개발)

  • Development for extraction method of sea tangle was investigated with addition of sucrose, NaCl, EDTA-2Na, SHMP, and hydrolysis with commercial polysaccharide hydrolyzing enzymes. The total solids and protein yields were significantly increased by enzymatic hydrolysis or addition of NaCl and EDTA-2Na during boiling. The extract prepared by enzymatic hydrolysis followed by 2 hrs of boiling in 1.5% NaCl solution was significantly improved in solids and protein yield from 24.1% and 13.9% of 2 hrs of water boiling to 44.6% and 32.2%, respectively. The combined method increased the amino-nitrogen content but reduced the turbidity and viscosity. Extracts with a high intensity of total and seaweed-like taste as revealed by sensory evalution were obtained by combined method.

  • An understanding of potential lumber cutting yields may lead to increased utilization of the lower grades of lumber in wood industry. Computer simulations of rough-mill operations require a lumber database as input to give reasonable estimates of such yields. The lumber database must contain detailed information regarding the location and type of defects, and then all manufacturing sequences can be tested with the same raw material. However, no suitable lumber database with mapped defects exists for Korean wood industry. In this study lumber databases of Pinus densiflora S. et Z and Quercus acutissima which are the main Korean domestic species were developed to prepare for coming era of "utilization of domestic species" in the near future. These databases were put into lumber cut-up simulation model(gang-rip-first simulator) to investigate the part yields. Gang-rip first simulation showed average part yields of 44.75% and 63.10% for Quercus acutissima and Pinus densiflora lumber database developed, respectively. In most cases process set-up of fixed blade best feed showed the best part yields and the level of acceptable defects could not make significant differences in part yields.

Effect of Pectinase Treatment on Extraction Yield of the Juice of Fragaria ananassa Duch. and the Quality Characteristics of Strawberry Wine during Ethanolic Fermentation (딸기 과즙의 수율과 딸기 발효주의 품질에 대한 pectinase 처리의 영향)

  • To develop a low-ethanol strawberry wine, the use of pectinase to improve the extraction yield of strawberry juice was investigated, and changes in physicochemical characteristics during ethanolic fermentation were assessed. The juice yield from strawberry fruit increased by 18.9% after Viscozyme L treatment (1,000 ppm, 30 min), compared with a control group, a greater increase than seen with other pectinases (17.5-18.7%). No significant quality differences were observed between control juice and juice prepared with enzyme treatment, indicating that neither physicochemical characteristics nor ethanol content during fermentation were affected by pectinase treatment. The major pigments of strawberry juice were cyanidin-3-glucoside and pelargonidin-3-glucoside, both of which are anthocyanins. The pigment level after enzyme treatment was slightly lower than that of the control group, at all fermentation times. We consider that the economics of strawberry wine manufacture may be increased by use of pectinase because juice level was increased, but no change in ethanol content or physicochemical characteristics was apparent.

  • Crease cut pearled baller was studied in relation to product and quality, and processing electricity requirement with various pearling yields. 1. Increasing the pearling rate resulted in increasing broken grain of cut-Polished barley. 2. The range of length/width ratio in cut-polished baller were from 2.09 to 2.13 and those of pearled barley were 1.36, respectively. 3. Cut-polished barley required more electricity than pearled barley at the same polishing rate. As pearling rate increased at first polishing before cutting, total electricity requirement of cut-polished barley reduced significantly.
