Automatic Bee-Counting System with Dual Infrared Sensor based on ICT (ICT 기반 이중 적외선 센서를 이용한 꿀벌 출입 자동 모니터링 시스템)

  • Honey bees are a vital part of the food chain as the most important pollinators for a broad palette of crops and wild plants. The climate change and colony collapse disorder (CCD) phenomenon make it challenging to develop ICT solutions to predict changes in beehive and alert about potential threats. In this paper, we report the test results of the bee-counting system which stands out against the previous analogues due to its comprehensive components including an improved dual infrared sensor to detect honey bees entering and leaving the hive, environmental sensors that measure ambient and interior, a wireless network with the bluetooth low energy (BLE) to transmit the sensing data in real time to the gateway, and a cloud which accumulate and analyze data. To assess the system accuracy, 3 persons manually counted the outgoing and incoming honey bees using the video record of 360-minute length. The difference between automatic and manual measurements for outgoing and incoming scores were 3.98% and 4.43% respectively. These differences are relatively lower than previous analogues, which inspires a vision that the tested system is a good candidate to use in precise apicultural industry, scientific research and education.

Preliminary Environmental Impact Assessments on Fish Compositions and the Ecological Health of Jeokbyeok River on the Road Construction of Muju-Geumsan Region (무주-금산간 도로건설에 따른 적벽강의 어류 종 조성 분석 및 생태건강도 사전환경성평가)

  • The objectives of the study were to evaluate fish compositions, endangered species, community structure, physical habitat, and general water quality for a preliminary environmental impact assessment of Jeokbyeok River on the road construction between two regions. Total number of species and total number of individuals, based on CPUE, were 23 and 1186, respectively. The endangered species (I, II) as the legal protection species were Pseudopungtungia nigra (79 samples) Gobiobotia brevibarba) (5) Gobiobotia macrocephala (2), indicating a requiring of endangered species conservation. In the meantime, exotic species and ecological disturbing species such as Micropterus salmoides and Lepomis macrochirus, were not present, indicating a well conserved area. According to fish community analysis, values of species diversity index were high (range: 0.788 - 1.030), and the dominance index were low (range: 0.097 - 0.183), indicating that the fish community in this area was maintained well without high dominacne by specific species. Also, fish analysis on tolerance guilds and trophic guilds showed that the proportions of sensitive species were largely exceeded the proportions of the tolerant species, while the proportions of insectivore species were largely exceeded the proportions of the omnivore species. This outcome suggests that the ecosystem was well maintained in terms of tolerance and trophic compositions (food chain). Ecological health, based on the multi-metric fish model of Fish Assessment Integrity (FAI), reflected those fish conditions. In other words, values of FAI model averaged 82.4, which means a "good condition" in the criteria of ecological health by the Minstry of Environment, Korea. In addition, general water quality and physical habitat analyses showed that the system was in good condition. Under these conditions, if the road constructions between the two regions happen in the future, inorganic suspended solids may increase in the waterbody, and this may result in indirect or direct influences on the physical habitats and food chain as well as fish compositions, so the ecological protections and prevention strategy from the soil erosion are required in the system.

Top-down Fish Biomanipulation Experiments on Algal Removal Effects (조류제거 효과에 대한 Top-down 어류 조작실험)

  • This study was to test algal removal efficiency by top-down fish biomanipulation experiments in the laboratory during Agust${\sim}$September 2000. We selected eight candidate fishes for the biomanipulation. We set up the experiments of eight fish-treatment tanks (3${\sim}$6 fishes) with initial chlorophyll-${\alpha}$ concentrations (CHL-${\alpha}$) of $100{\sim}120{\mu}g\;L^{-1}$ and one control tank including no fish with the same initial CHL-${\alpha}$. All tanks were maintained water quality of dissolved oxygen $(5.3{\sim}8.2mg\;L^{-1})$ and pH $(7.4{\sim}8.1)$ in the tests. During the biomanipulation, DO and pH in the treatments were lower than those of the control, while conductivity increased gradually in the treatments. Biomanipulation experiments showed that CHL-${\alpha}$ increased 13% and 0% (mean values of 8 fishes) in the controls and treatments, respectively. These results indicate that algal growth was maintained in the control and fish treatments, but the rate of CHL-${\alpha}$ in the treatments was lower than that of the control. The removal rates of bluegreens algae decreased 32% in the control, and 20% in treatments (mean values of 8 fishes) respectively, In other words, bluegreen algae showed greater growth rate in the fish treatments than the control and this was due to higher nutrients supplied from fish excretions. Overall, simple fish biomanipulation on algal control was not effective at all in these laboratory tests.

Approximation of Multiple Trait Effective Daughter Contribution by Dairy Proven Bulls for MACE (젖소 국제유전능력 평가를 위한 종모우별 다형질 Effective Daughter Contribution 추정)

  • This study was conducted to investigate the basic concept of multiple trait effective daughter contribution (MTEDC) for dairy cattle sires and calculate effective daughter contribution (EDC) by applying a five lactation multiple trait model using milk yield test records of daughters for the Multiple-trait Across Country Evaluation (MACE). Milk yield data and pedigree information of 301,551 cows that were the progeny of 2,046 Korean and imported dairy bulls were collected from the National Agricultural Cooperative Federation and used in this study. For MTEDC approximation, the reliability of the breeding value was separated based on parents average, own yield deviation and mate adjusted progeny contribution. EDC was then calculated by lactation using these reliabilities. The average number of recorded daughters per sire by lactations were 140.57, 94.24, 55.14, 29.20 and 14.06 from the first to fifth lactation, respectively. However, the average EDC per sire by lactation using the five lactation multiple trait model was 113.49, 89.28, 73.56, 54.02 and 35.08 from the first to fifth lactation, respectively, while the decrease of EDC in late lactations was comparably lower than the average number of recorded daughters per sire. These findings indicate that the availability of daughters without late lactation records is increased by genetic correlation using the multiple trait model. Owing to the relatedness between the EDC and reliability of the estimated breeding value for sire, understanding the MTEDC algorithm and continuous monitoring of EDC is required for correct MACE application of the five lactation multiple trait model.

Effects of pH of Washing Solution, Washing Frequency and Individual Bull on Proton Concentration in the Sperm Washed Solution and Sperm Acrosome Reaction (精子 洗滌液의 pH와 洗滌頻度 및 수소個體가 精子洗滌液內 水素이온濃度와 소 精子의 尖帽反應에 미치는 影響)

  • This study was carried out to investigate the effects of pH of sperm-washing solution, sperm-washing frequency and individual bull on concentration of hydrogen ion in sperm-washed solution and sperm acrosome reaction. The results obtained were as follow. 1. When bovine sperm was washed at 4 times with SHP solution and incubated, the difference of light absorbance between sperm-washing and sperm-washed solution(${\Delta}$Ao-${\Delta}$At) was higher in 2nd sperm-washed solution than that in the other washed solutions. 2. When sperm was thrice washed with SHP solutions of pH 5.99, 6.38, 6.78, 7.10, 7.40, 7.69, 8.15, 8.45, and 8.83, ${\Delta}$Ao-${\Delta}$At was significantly increased at pH7.69 to 8.83, and ${\Delta}$Ao-${\Delta}$At in 1st sperm washed solution was significantly higher than that in 2nd and 3rd sperm washed solution. 3. When sperm of Holstein, KNC and Hereford was thrice washed with SHP solutions of pH 5.99, 6.38, 6.78, 7.10, 7.40, 7.69, 8.15, 8.45, and 8.83, Holstein showed higher ${\Delta}$Ao-${\Delta}$At of sperm Washed solution than KNC and Hereford, and ${\Delta}$Ao-${\Delta}$At in sperm washed solution was significantly increased at 7.69 for Holstein and at 8.15 for KNC and Hereford, respectively. 4. When sperm was thrice with SHP solution of pH 6.8, 7.1 and 7.4, and then incubated in mTALP of pH 7.4 for 15 minutes, in 1st and 2nd sperm washing ${\Delta}$Ao-${\Delta}$At of sperm washed solution was significantly higher at pH 7.1 and 7.4 than at pH 6.8, and the sperm acrosome reaction of pH 6.8, 7.1 and 7.4 was 49.1, 68.8 and 72.9%, respectively. The sperm acrosome reaction of pH 7.1 and 7.4 was higher than that of pH 6.8.

Studies on Virus Diseases of Orchids in Korea (우리나라 란 바이러스에 관한 연구)

  • Orchids have been propagated vegetatively for a long time without adequate control measures against virus diseases in Korea. As a result, it is presumed that most of the orchid varieties in Korea may have been degenerated. Nevertheless there has been little work on the virus diseases of orchids in Korea. Therefore studies were initiated to isolate an4 characterize the orchid viruses occurring in Korea. The results obtained are summerized as follows. 1. Symptoms of virus diseases on orchid varieties can be grouped 1) mosaic, 2) necrotic streak with mosaic, 3) ring necrosis, 4) chlorotkc ring and 5) necrotic spot. 2. A total of 102 orchid plants representing 4 genera were investigated on the occurrence of Cymbidium mosaic virus and tobacco mosaic virus by serological agar-gel double diffusion test. The test revealed that approximately $45\%$ of the orchids were infected with Cymbidium mosaic virus. None of the plants were found to be infected with tobacco mosaic virus. 3. Local lesions appeared on the inoculated leaves of Chenopodium amaranticolor Cassia occidentalis and Datura stramonium 7-12 days after mechanical inoculation with Cymbidium mosaic virus. 4. Physical properties of the Cymbidium mosaic virus determined by inoculation on Chenopodium amaranticolor were as follows: Thermal inactivation Point; $75-80^{\circ}C$, dilution end Point; $10^{-5}-10^{-6}\%$ aging in vitro; 8 days. 5. Three different buffers at pH 7.0 and pH 9.0 were compared for the efficiency of agar-gel double diffusion test with Cymbidium mosaic virus. Phosphate, imidazol and tris buffer at pH 7.0 gave equally satisfactory results. 6. Electron microscopic examination of the Cymbidium mosaic virus revealed rod shaped particles measuring 460-580mu.

Development of a Field Oxygenation Device and Its Practice in the Oxygen Depleted Water Mass (빈산소 수괴해역 용존산소 환경개선장치 개발과 현장 적용)

  • Oxygen depleted water mass can damage aquatic animals not only in direct way but also in indirect way by generating toxic substances including occurrence of hydrogen sulfide and ammonia which are also highly detrimental to animal life in the water mass. An oxygen dissolution device was developed, which makes turnover of the oxygen rich (over 20 mg/L) surface water down to the bottom where hypoxia is evident and tested the device in terms of oxygen recovery in the oxygen depleted bottom water. the device with turnover rates of $3.6\;m^2$/min at the liquid oxygen injection rate of 48~26.3 L/min could recover dissolved oxygen level to 7~25 mg/L at depth 7 m to lead to the dissolution level of over 90% by the supply of liquid oxygen. The running advantage of the device is that it does not require any auxiliary tank and higher energy for operation. Therefore, it can be highly useful device to relieve damages to the farmed animals in the oxygen depleted waters.

Physiological Changes in Related to Molt Cycle of Macrobrachium nipponense(De Haan) (징거미 새우, Macrobrachium nipponense(De Haan) 유생의 탈피주기와 관련한 생리적 변화)

  • Larvae of the freshwater shrimp, Macrobrachium nipponense(De Haan) were reared in the laboratory under constant conditions ($25^{\circ}C,\;7\%0$ S), and their feeding rate, oxygen consumption, ammonia nitrogen excretion, and growth were measured at regular intervals during development from hatching to post larval stage. Growth was measured as dry weight, carbon, nitrogen, hydrogen, protein and lipid. All these physiological and biochemical traits revealed significant changes from instar to instar. Average feeding rate was high in intermolt stage of the molt cycle and it showed a bell-shaped pattern. Respiration(R) increased from hatching to post larval stage. Excretion(U) increased in intermolt phase of larvae and it showed a bell-shaped variation pattern, in all larval instars with a maximum near the middle of the molt cycle. Regression equations describing rates of feeding, growth, respiration and ammonia excretion as functions of time during individual larval molt cycles were inserted in a simulation model, in order to analyse time-dependent patterns of variation as well as in bioenergetic efficiencies. Carbon was initially increased and nitrogen showed a tendency to increase in premolt phase during individual molt cycles. Protein remained clearly the predominant biochemical constituent in larval biomass.

Molecular Analysis of the Microorganisms in a Thermophilic CSTR used for Continuous Biohydrogen Production (연속수소생성에 사용되는 고온 CSTR 내의 미생물의 분자적 분석)

  • Molecular methods were employed to investigate microorganisms in a thermophilic continuous stirred tank reactor(CSTR) used for continuous $H_2$ production. The reactor was inoculated with heat-treated anaerobic sludge and fed with a glucose-based medium. Denaturing gradient gel electrophoresis showed dynamic changes of bacterial populations in the reactor during 43 days of operation. Gas composition was constant from approximately 14 days but population shift still occurred. Populations affiliated with Fervidobactrium gondwanens and Thermoanaerobacterium thermosaccharolyticum were dominant on 21 and 41 days, respectively. Keeping pH of the medium at 5.0 could suppress methanogenic activity that was detected during initial operation period. $CH_4$ and mcrA detected in the samples obtained from the reactor or inoculum suggested the heat treatment condition employed in this study is not enough to remove methanogens in the inoculum. PCR using primer sets specific to 4 main orders of methanogens suggested that major $H_2$-consuming methanogens in the CSTR belong to the order Methanobacteriales.

Thermophilic Biohydrogen Production from Glucose with a Long-term Operation of CSTR (CSTR의 장기운전을 통한 포도당으로부터의 고온 수소생산)

  • Thermophilic $H_2$ was produced for 1 year using a bench-scale continuous stirred tank reactor(CSTR). The CSTR was inoculated with anaerobically digested sludge after heat treatment and fed with a glucose-based medium. The reactor showed relatively short start-up period(30 days) and high maximal $H_2$ yield(2.4 mol $H_2/mol$ glucose). Keeping pH 5.0 or less suppressed methanogenic activity. Bacteria affiliated with Thermoanaerobacterium thermosaccharolyticum kept being dominant from approximately 40 days as determined by DGGE. Environmental perturbation(pH or temperature) caused the decrease of biomass concentration in the reactor and the instability of reactor performance, $H_2$ production rate and $H_2$ yield. The unstable performance was accompanied with high concentration of lactate in the effluent. Taken together, the poor recovery of CSTR after perturbations could be partly explained by low biomass concentration and/or metabolic shift of the major population in the CSTR.
