A Study on Physicochemical Properties of Achyranthes japonica and Smilax china Extracts (쇠무릎과 청미래덩굴 부위별 추출물의 이화학적 특성에 관한 연구)

  • Physicochemical properties of Achyranthes japonica and Smilax china extracts were investigated for the purpose of functionality research on the natural bio-resources. Extraction contents were order of distilled water>methanol>ethanol solvent, the highest free aminoacids were proline from Achyranthes japonica, phosphoserine and glutamic acid from Smilax china, respectively. BI and TAC by spectrophotometric absorbance were order of methanol>ethanol>water in Smilax china leaf extract, but water>methaol>ethanol in Achyranthes japonica leaf extract. EDA was high in ethanol extract from Smilax china leaf and in methanol extract from Smilax china root, and in water extract from Achyranthes japonica. TBA value of Achyranthes japonica leaf and Smilax china leaf-ethanol extracts on olive oil was 82.1% and 84.0%, respectively, for that of an artificial antioxidant BHT. Antimicrobial effect was observed in Achyranthes japonica stem-methanol extract on Bacillus subtillis, in Smilax china leaf-ethanol extract on Bacillus subtillis, Vibrio vulnificus and Salmonella enterica, respectively. And the adsorption of Pb(II) on Achyranthes japonica was higher than that of Cd(II) on Smilax china under the same metal ion concentration.

Effects of Light and Some Plant Growth Regulators on Ecdysteroids Contents in Polypodium vulgare L. and Achyranthes japonica Nakai (식물의 Ecdysteroids 함량에 미치는 광과 식물생장조절제의 영향)

  • Effects of light, methyl jasmonate(MJ), 6-benzylaminopurine(BA), thidiazouron(TDZ), and 2,4-dichlorophenoxyacetic acid(2,4-D) on the contents of ecdysteroids (${\beta}$-ecdysone+polypodine B) in Polypodium vulgare L. and Achyranthes japonica Nakai were studied. When the plants of P. vulgare were cultured under light control, the ecdysteroids contents in both leaves and rhizomes decreased with increasing light intensity. The ecdysteroids contents in A. japonica were also lower when cultured under light than under dark. Among the tested plant growth regulators, MJ, BA, and TDZ increased the ecdysteroids contents in both P. vulgare and A. japonica.

Identification of Phytotoxic Compounds and Allelopathic Effects of Various Upland Weeds (전작잡초(田作雜草)로부터 생리활성물질(生理活性物質) 탐색(探索))

  • This study was conducted to clarify the identification of phytotoxic compounds from 5 upland weeds which had been collected at their maturity age in the field of Kyungpook province, Korea. 5 weed species including Stellaria aquatica strongly inhibited the lettuce seed germination. These 5 species were Amaranthus lividus, Stellaria aquatica, Achyranthes japonica, Polygonum perfoliata and Galinsoga ciliata. the degree of inhibitory effects according to various solvent extracts such as ethanol, aceton, chloroform and ether was different as species. In total phenol and diphenol content G. ciliata had largest amount of total phenol with 2.75mg/g, P. perfoliata 1.8mg/g, S. aquatica 1.7mg/g, A. lividus 1.28mg/g, A. japonica 1.27mg/g respectively. A. lividus had much amount of p-cresol, hydroquinone, ferulic, caffeic, p-cl-benzoic and p-coumaric in order. S. aquatica, A. japonica, P. perfoliata and G. ciliata had a large quantity of ferulic, caffeic, 7-OH-coumarin and protocatechuic acids in common. The amount of fatty acids was existed 8.49mg/g in S. aquatica, 7.14mg/g in A. japonica, 7.10mg/g in G. ciliata, 5.49mg/g in P. perfoliata and 4.2mg/g in A. lividus respectively. Most of 4 weeds except G. ciliata had much oxalic acid to the extent of 14-22mg/g. however it appeared that G. ciliata had much quantity of feruic and citric acids.

Georeferencing of Primary Species Occurrence Data and Necessity of Data Quality Control - A Case Study of Two Varieties of Ox-Knee, Achyranthes bidentata Blume - (1차종발생자료를 응용한 지리참조연산표준화 및 자료 품질 관리의 필요성 - 쇠무릎과 털쇠무릎의 적용 사례 -)

  • The purpose of this contribution is to develop the framework of a methodology for identifying potential errors in georeferencing and in an application of it using specimens of Ox-Knee, Achyranthes bidentata Blume in Korea. At infraspecific level, uncertainty of identification showed that 41% of A. bidentata var. japonica and 28% of var. bidentata were misidentified, suggesting that the uncertainty level was independent of the reliability of experts' identification. For georeference specimen records, 71 specimens out of total 303 were selected and utilized as occurrence data: Uncertainty was 32.4 km at maximum and was 0.1297 km at minimum (mean = 4,055 m, s.d. = 5,772 m). Var. japonica is common throughout most of the southeastern Korea and west coastal areas, while var. bidentata has been found as far north as Gyeonggi and Gangwon provinces. We modelled the potential distribution of two varieties using Bioclim approach in Korea based on several environmental factors. Our results indicated the most important region for var. japonica lies the west coast ranges and southern area, while for Chungcheongnam-do of potential high diversity occurs for var. bidentata. This study shows that the major factors to determine the distribution patterns of two varieties were thermal factors, rather than precipitation. The Bioclim model using geocode and georeferencing data makes the information increasingly useful and reliable. To improve data quality, it requires full management from data collection to final databases including data cleaning.

Antioxidant Activities of Extracts from Leaves and Stems of Achyranthes japonica (쇠무릎 잎과 줄기 추출물의 항산화 활성 연구)

  • This study was conducted to investigate the physiological characteristics of water and ethanol extracts from leaves and stems of Achyranthes japonica. The highest contents of total polyphenol and flavonoid compounds were 58.27 and 42.22 mg/g in water extract from leaves, respectively. The protein content was the highest at 16.42 mg/g in water extract from leaves. Ethanol extract from stems showed the highest content of reducing sugars at 11.35 mg/g. In the measurement of electron donating ability (EDA), ethanol extract from stems showed the highest EDA at 93.41% at a concentration of 1.0 mg/mL. Superoxide dismutase-like activity of ethanol extract from leaves was the highest at 8.13% at a concentration of 1.0 mg/mL. In the analysis of nitrate scavenging activity, water extract from leaves showed the highest activity at 94.90% at pH 1.2, and the activity increased as concentration increased and pH decreased. In the measurement of xanthine oxidase inhibition, ethanol extract from stems showed the highest inhibitory activity at 66.67% at a concentration of 1.0 mg/mL. Especially, nitrate scavenging activities of water extract from leaves were the highest under all pH conditions. These results verify that extracts from leaves of A. japonica have strong antioxidant activity and can be used as an effective antioxidant source for nutraceutical foods, medicines, and cosmetic stuffs.

Isolation of Genes Involved in Ecdysteroids Biosynthesis from Achyranthes japonica Nakai (쇠무릎(Achyranthes japonica Nakai)으로부터 Ecdysteroid 생합성에 관련된 유전자의 분리)

  • In order to isolate genes involved in ecdysteroids biosynthesis in plants, total RNA was isolated from Achyranthes japonica Nakai, and RT-PCR was performed using degenerate primers selected based on the results of multi-alignment of four cytochrome P450 genes from plants and a putative ecdysone 20-hydroxylase gene from an insect. Fourteen partial cDNA clones showing unique base sequences were obtained, out of which six showed homologies at the levels of nucleotide and amino acid sequences to the other cytochrome P450 genes known to be involved in the ecdysteroid biosynthesis. Of the six clones, four showed relatively high homologies to a putative ecdysone 20-hydroxylase gene isolated from an insect.

Anthracnose of Achyranthes japonica Caused by Glomerella cingulata in Korea (Glomerella cingulata에 의한 쇠무릎 탄저병)

  • Anthracnose symptoms were observed on the community area in Sancheong-gun, Gyeongnam Province in Korea. The symptoms occurred on leaf and stem, and then plants eventually were died. Colony color was whitish gray to dark gray on potato dextrose agar. Conidia were single celled . colorless, cylindrical and was 10.2~20.4$\times$3.6$\times$6.0$\mu$m in size. Appressoria were dark brown, ovate to obovate and 6.2~10.5$\times$4.3$\times$8.6$\mu$m in size. Perithecia were brown to black in color and shaped as globose to obpyriform and 72.6~284.7$\mu$m in size. Asci were clavate to cylindrical in shape and 63.6~98.8$\times$8.3~12.5$\mu$m in size. Ascospores were cylindrical. fusiform, slightly curved at the center and 9.6~21.8$\times$4.0~6.0$\mu$m in size. Optimum temperature fur growth was 3$0^{\circ}C$. On the basis of mycological characteristics and pathogenecity test on host plants, the fungus was identified as Glomerella cingulata . This is the first report on the Anthracnose of achyranthes caused by Glomerella cingulata in Korea.

Screening of Medicinal Plants to Suppress Population of Meloidogyne hapla in Codonopsis lanceolata Trautv (더덕에 발생하는 당근뿌리혹선충의 증식억제 식물 탐색)

  • Total 90 species of medicinal plants were surveyed to see if they have any suppressive effects on the dinsity of M hapla at the exhibition field in the Chinan medicinal herbs experiment station. In 70 species including Achyranthes japonica, root-knot and/or egg sac of M. hapla was not found and these plants were planted in C. lanceolata field to check the degree of M. hapla infection. In 26 species including A. japonica, M. hapla infection was not observed. Simultaneously, 30 species were planted in pots to find out degree of infection by M. hapla. Dianthus chinensis, Rudbeckia bicolor, Sedum kantschaticum, Ricinus communis, Anemarrhena asphodeloides, Malva verticillate, Chelidonium majus, Sesamum indicum, Agrimonia pilosa, Geum aleppicum, Sanguisorba officinalis and Scrophularia buergeriana were free from infection. While the number of galls and density of M. hapla in soil were higher to high innoculation density, and the growth of C. lanceolata was rower.

Micropropagation of Achyranthes japonica Through Axillary Buds Culture (액아배양을 통한 쇠무릎(Achyranthes japonica)의 대량증식)

  • Multiple shoot formation was obtained from excised axillary buds of Achyranthes japonica NAKAI cultured on MS media containing various growth regulators such as auxin and cytokinin. The highest average number of shoots was obtained in 1 mg/L NAA and 2 mg/L BA after 6 weeks (25.8 adventitious shoots per node). Although the regeneration rate was less than the former condition, optimal combination for the production of more shoots with a suitable size was 0.5 mg/L NAA and 1 mg/L BA (19.7 adventitious shoots per node). Roots were induced from regenerated shoots after 3 weeks culture, transferred to 1/2 MS medium supplemented with 0.1 mg/L IBA. Micropropagated plants were successfully transferred to soil.
