  • Sensitivity of the three freshwater fish, Cyprinus carpio, Oryzias latipes (wildtype indigenous to Korea), and O. latipes (Japanese killifish) to 30 pesticide formulations were studied in terms of 48 hr $LC_{50}$ determined with the static method. The correlation between C. carpio and O. latipes (Japanese killifish) was higher than that between C. carpio and O. latipes with correlation coefficients of 0.89 and 0.80, respectively. The sensitivity of O. latipes and O. latipes (Japanese killifish) to pesticides showed very high correlation with a coefficient of 0.93. Therefore, it is suggested that the acute toxicity data concerning O. latipes (Japanese killifish) could represent those C. carpio or O. latipes which are indigenous species in Korea. Also, it is found that the present protocol for the toxicity test with carp in Korea has difficulties in maintaining the proper concentration of dissolved oxygen in the test chamber because of the abrupt decrease of dissolved oxygen to 2mg/l, which is not acceptable according to general guidelines of foreign countries.

The Structures of Feeding Organs in Two Korean Ricefishes (Pisces, Adrianichthyidae), Oryzias latipes and O. sinensis (한국산 송사리속 Oryzias 2종 섭이기관의 구조적 특징)

  • Structure on the feeding organs in two Korean ricefishes, Oryzias latipes and O. sinensis, was examined in the jaws and the teeth. In the jaw, O. latipes has a dent on the dorsal edge of distal arm of premaxilla (vs. straight in O. sinensis) and a alveolar arm of premaxilla running in straight line with distal arm of premaxilla (vs. ventra line in O. sinensis). O. latipes has also a dent on the posterior edge of dentary which is connected to coronoid process of dentary (vs. straight in O. sinensis). Large teeth developing only in the male, which is sexual dimorphism, developed into outside direction of the mandible in O. latipes (vs. dorsal direction in O. sinensis). In the teeth forms, O. latipes has two types (conical type and arrowhead type), whereas O. sinensis has only one type (conical type). Through this study, we confirmed that two Korean ricefishes show distinctive characteristics in the structure of the feeding organs.

  • A ricefish Oryzias latipes sinesis Chen et al. of the family Oryzidae is recorded for the first time from western Korea. The present speciment are similar to O. l. latipes in most morphometric characters, but agree well with the original description of O. l. sinensin based on the diploid chromosome number of 46 with two large metacentric chromosomes and the lesser vertebrae number.

  • A study on the structures of the pharyngeal bones and teeth in two Korean ricefishes, Oryzias latipes and O. sinensis, was carried out to find out interspecfic differences. In the pharyngeal bones, both two species have a pair of upper pharyngeal bone (stilliform shape) and lower pharyngeal bone (rectangular shape). In the lower pharyngeal bones, in particular, the number of the row was 6 to 7 in O. latipes and 5 to 6 in O. sinensis. The pharyngeal teeth also showed different types in two species: O. latipes of a top-hooked type and O. sinensis of a conical type. Through this study, we confirmed that the two Korean ricefishes show distinctive characteristics in the structure of the pharyngeal teeth.

  • The Korean ricefish, Oryzias, consisted of 2 species, O. latipes and O. sinensis. Through investigation on their distributions, it was confirmed that O. sinensis was restricted to western areas, and O. latipes was distributed throughout Korea. Interestingly, Daecheon region near the West Sea was sympatric, existing O. latipes and O. sinensis. In addition to their distributions, O. latipes was well distinguished from O. sinensis as the following morphological characteristics: mean 31 vertebrae (vs. 30), mean 15 gill rakers (vs. 12), lots of dots in both sides of body (vs. absent), and yellowish ventral and anal fins (vs. blackish).

  • 한국산 송사리 (Oryzias)속의 송사리 (O. latipes)와 대륙송사리(O. sinensis)의 집단 내 유전적 변이 및 종간 유연관계를 밝히고자 남한의 16개집단과 일본산 송사리(O. latipes) 1개 집단 등 총 17개 집단에 대한 isozyme 분석을 실시하였다. Isozyme 분석 결과 17개의 효소 및 비효소단백질에서 총 32개의 유전자를 검출한 결과, 전체 유전자중 Est-1과 Est-3 는 종 특유의 유전적 표식인자(genetic marker)였다. 송사리집단 중 진도집단의 유전적 변이 가 가장 낮았으며(Ho=0.011, He=0.043), 대륙송사리 서천집단의 유전적 변이가 가장 높았 다.(Ho=0.114, He=0.124). 한국산 송사리 11개 집단의 평균 유전적 변이 정도는 P=12.7%, Ho=0.029, He=0.042, 대륙송사리 5개 집단의 평균 유전적 변이정도는 P=26.3%, Ho=0.082, He=0.097로 각각 나타나 대륙송사리집단의 유전적 변이 정도가 송사리집단에 비해 약 2.5배 정도 높았고 일본산 송사리의 유전적 변이 정도는 P=15.6%, Ho=0.073, He=0.068로 나타났 다. 한국산 송사리와 일본산 송사리와의 유연관계는 S=0.761(D=0.243)로 나타나 유전적으로 뚜렷한 별개의 분류군으로 생각되며, 대륙송사리와 송사리간의 종간 유전적 근연치는 S=0.648(D=0.389)로 일반적인 척추동물의 종간 유전적 분화 수준을 나타냈다.

Inhibitory Effects of Extracts from Albizzia julibrissin on Gonadal Maturation in a Medaka (Oryzias latipes) (자귀나무 추출물이 송사리(Oryzias latipes) 생식소 성숙에 대한 억제효과)

  • This study was performed to determine whether stem bark extract containing saponin of Albizzia julibrissin in the diet influences gonadal maturation and spawning in medaka (Oryzias latipes). The crude extraction containing saponin (HaBC) was partially purified from n-BuOH extraction of A. julibrissin stem bark by Diaion HP-20, Silica gel and Sephadex LH-20 chromatographies. We fed diets supplemented with HaBC to medaka. We then studied the effects of the HaBC supplement on the suppression of gonadal maturation and spawning in female medaka that were reared in aquaria with recirculation systems. In the experiment with immature female medaka, the periods of initiation of gonadal maturation and spawning were delayed in the fish that were fed diets supplemented with at least HaBC 20 mg/g-feed. In the experiment with mature female medaka, the fish that were fed diets supplemented with at least HaBC 20 mg/g-feed had lower GSIs than the control diet group did. The results showed that the growth of the immature medaka was not correlated with the amount of supplementation of HaBC in the diet. However, the condition factors (CF) in the medaka that were fed diets supplemented with at least HaBC 20 mg/g-feed were higher than in the medaka fed on the control diet. We concluded that extracts containing saponin from the stem bark of A. julibrissin have the potential to inhibit gonadal maturation in female medaka, but they did not act as growth stimulation. Further studies are required to determine the mechanism of the action.

Subchronic Toxicity of Herbicide Butachlor in Fish , the Medaka (Oryzias latipes) (제초제 Butachlor의 송사리에 대한 아급성(亞急性) 독성(毒性))

  • To establish an evaluation system of aquatic toxicity of chemicals at no-effect level, flow through and early life stage toxicity test were performed on a freshwater fish, the medaka (Oryzias latipes). The characteristics of medaka as a bioassay organism for the chronic toxicity test were discussed. Maximum acceptable toxicant concentration(MATC) of butachlor for the madaka in soft water was estimated using survival, growth, and reproduction as indicators of toxic effects. During a 3-month period, the fry of medaka were exposed to butachor concentrations ranging from 0.16 to 0.0l mg/liter and the DO concentration, temperature, and pH in the exposure chamber were measured to check the test condition. The highest concentration showed slight decrease of growth rate in medaka and reduced hatchability of spawning egg. Survival, growth, and reproductive success of adults in butachlor concentration of 0.04 and 0.01 mg/liter were not different from those of the control. The MATC was estimated to be between 0.04 and 0.16 mg/liter for medaka.

Toxicity and Behavioral Changes of Medaka (Oryzias latipes) by Brine Exposure (송사리(Oryzias latipes)를 이용한 고염해수의 생태독성 및 단기적 행동변화에 관한 연구)

  • Acute toxicity test and behavioral change analysis of seawater acclimated Japanese medaka were conducted to identify the brine effects on fish by seawater desalination. 7 day acute toxicity test of brine revealed linear concentration-response relationship from 40.0~80.0 psu treatment groups. There was no significant brine effect for 30-40 psu groups and mass mortality was observed from >50 psu exposure (7-day $LC_{50}$=51.4 psu). Images from the real time camera system were analyzed to observe the changes in behavioral patterns of medaka exposed to various salinity. 40.0 and 50.0 psu exposed groups were stabilized in behavioral patterns after 3.1 and 4.6 hours, respectively and 60.0 psu group showed sharp increase in activity during first 12 hours and 50% mortality thereafter. Similar patterns were observed to 70 and 80 psu groups and both experimental groups showed 100% mortality within 12 hours. Acute toxicity test and behavioral patterns showed very similar toxicity results which revealed the increases in mortality and behavioral activities from 50.0 psu. This critical salinity for fish impacts must be implemented to brine discharge strategy by seawater desalination into the coastal area. Also, we recommend that real time camera monitoring system must be a useful tool for early warning of fish toxicity for other applications.
