Comparison of Loop-Mediated Isothermal Amplification and Real-Time PCR for the Rapid Detection of Salmonella Typhimurium, Listeria monocytogenes and Cronobacter sakazakii Artificially Inoculated in Foods (식품에 인위접종된 Salmonella Typhimurium, Listeria monocytogenes, Cronobacter sakazakii의 신속검출을 위한 Real-time PCR과 Loop-mediated isothermal amplification 비교)

  • The objective of this research was to compare loop-mediated isothermal amplification (LAMP) with real-time polymerase chain reaction (PCR) for the rapid detection of pathogens in foods. In this study, the limits of detection (LODs) for Salmonella Typhimurium, Listeria monocytogenes, and Cronobacter sakazakii were evaluated in various foods. Among 11 samples tested for S. Typhimurium, LAMP and real-time PCR had the same LODs in beef and duck meat whereas real-time PCR was more sensitive than the LAMP in 8 samples. However, S. Typhimurium in chocolate samples was not detected by real-time PCR. The sensitivity of real-time PCR was high in all samples inoculated with L. monocytogenes and C. sakazakii whereas LAMP was more sensitive than real-time PCR in oil-rich foods. Therefore, LAMP can be shown as an easrer, more convenient method, as well as effective analytical method for testing difficult samples when employed in PCR.

Comparison of vitamin K contents in different meats commonly consumed in Korea (국내에서 소비되는 육류의 부위별 비타민 K 함량 분석 및 비교)

  • Vitamin K is a group of fat-soluble vitamins that naturally exist in phylloquinone (vitamin K1) and menaquinone (vitamin K2). In this study, the vitamin K content in different meats commonly consumed in Korea was analyzed using HPLC, and the analytical method was validated. Vitamin K1 was not detected in any of the meat samples. Vitamin K2 contents in different cuts of beef ranged from 0.00 to 5.87 ㎍/100 g, whereas the corresponding value in different parts of chicken ranged from 16.59 to 46.64 ㎍/100 g. In the case of pork, vitamin K2 contents varied from 4.33 to 22.90 ㎍/100 g. Among the different types of meat, the highest vitamin K2 content was found in boiled chicken meat and skin (46.64 ㎍/100 g). The analytical method was found to be reliable and had high accuracy. These results provide accurate nutritional information and contribute a food composition database for meat consumption.

Levels of sulfonamides for animals in food (식품 중 설폰아마이드계 동물용의약품의 잔류실태)

  • To determine levels of 11 sulfonamides for animals in food, simultaneously, a selective method of high performance liquid chromatography with UV detector has been applied. The targets were sulfachlorpyridazine (SCP), sulfadiazine (SDZ), sulfadimethoxine (SDM), sulfisoxazole (SSX), sulfamerazine (SMZ), sulfamethazine (SMT), sulfamethoxazole (SMX), sulfamethoxypyridazine (SMP), sulfamonomethoxine (SMM), sulfaquinoxaline (SQX) and sulfathiazole (STZ). Food samples were beef, pork, chicken, milk and whole egg that were collected at the main 6 cities in Korea as Seoul, Busan, Daejon, Incheon, Mokpo and Gangneung. After homogenizing food samples with sodium phosphate solution and acetonitrile, it was extracted with n-hexane. The mobile phase gradient was a mixture of 5 mM potassium phosphate (pH 3.25) and methanol with a gradient ratio from 100:0 to 30:70. The UV wavelength was 270 nm. The overall recoveries were ranged from 75% to 95% and the limit of detection was minimum 0.004 mg/kg for SMT, and 0.007 mg/kg for STZ at signal/noise > 3, respectively. As results, sulfonamide drugs were not detected in most of the selected food samples, however, sulfamonomethoxine was detected in meat. The determined level of sulfamonomethoxine were 0.03 and 0.06 mg/kg for beef that were below the MRLs.

  • The socio-cultural and economic changes following the Fourth Revolution are driving the growth of the food tech industry. Korea's food tech industry is still focused on delivery apps and the smart farms, robot market including artificial intelligence are in its infancy. In the United States, alternative meat companies are already included in unicorn companies, while Korea, the fourth largest importer of beef, lacks alternative meat development. France, Europe's largest agricultural country, is focusing on Agtech. China has developed the Internet and online e-commerce market with the world's number one population. Korea also needs to change regulations that focus on the past industry and various food tech industries should be developed through political and business-driven research and investment.

  • This study was conducted to evaluate beef using a color machine vision system. The machine vision system has an advantage to measure larger area than a colorimeter and also could measure other quality factors like distribution of fats. However, the machine vision measurement is affected by system components. To measure the beef color with the machine vision system, the effect of color balancing control was tested and calibration model was developed. Neural network for color calibration which learned reference color patches showed a high correlation with colorimeter in L*a*b* coordinates and had an adaptability at various measurement environments. The trained network showed a very high correlation with the colorimeter when measuring beef color.

  • The objective of this study was to reveal the educational meanings of candlelight rallies for the impeachment of President Park Geun-hye. For this, the origin and history of Korean candlelight rallies were divided. The researcher divided Korean candlelight rallies into four stages such as rallies for cherishing Hyosun & Miseon for Stage1, rallies for opposing the import of US beef for Stage2, rallies for the impeachment of President Roh Moo-hyun for Stage3, and rallies for the impeachment of President Park Geun-hye for Stage4. After that, the researcher revealed five educational meanings of the candlelight rallies for the impeachment of President Park Geun-hye. First, it is the change of educational structure. Second, it is the appearance of new subject. Third, it is the place for democratic communication and expression. Fourth, it is the equality and solidarity. Fifth, it is the establishment of an opportunity to have rational growth.

Deep Learning-based Analysis of Meat Freshness Measurement (고기 신선도 측정 데이터의 딥러닝 기반 분석)

  • The measurement of meat freshness at meat markets is important for the health of consumers. Currently a variety of sensors have been studied for the measurement of the meat freshness. Therefore, the analysis of sensor data is needed for the reduction of measurement errors. In this paper, we analyze the freshness measurement data of ten sensors based on deep learning. The measured data are composed of beef, pork and chicken, whose reliability and noise-robustness are examined by a deep neural network. Further, to search for multiple sensors better than a torrymeter, PCA (principle component analysis) is carried. Then, we validated that the performance of the three sensors outperforms the torrymeter in the experiment.

Effect of Antioxidant on Quality of Ground Beef during the Refrigeration Storage (소고기 분쇄육의 냉장 중 품질에 미치는 항산화제의 효과)

  • The objective of this study was to evaluate their effects as the meat antioxidant and on the antioxidant enzymes like superoxide dismutase(SOD) and catalase during the refrigeration storage of ground beef, respectively. Ground beef loin was treated by three natural antioxidants(pycnogenol, catechin, ${\alpha}$-tocopherol) and the synthetic antioxidant(BHT) at the level of 0.01%(w/w) of total fat. Samples were refrigerated at $4{\sim}6^{\circ}C$ for 3, 5, 7 and 10 days to evaluate the color and the pH as the quality parameters, TBA value and fatty acid composition as the parameters of lipid peroxidation, and the activities of SOD and catalase. This study showed that catechin and pycnogenol were excellent in terms of meat color, pH and delaying lipid peroxidation and also maintained the activity of in vivo SOD and catalase better than ${\alpha}$-tocopherol and BHT. These results suggested that the duration of the refrigeration of ground beef may be prolonged up to 10 days in catechin and pycnogenol treated ones in terms of the lipid peroxidation, but 5 days of refrigeration will be more adequate if considering the microbial safety as food, too.

Analysis of synthetic Antimicrobials in Livestock Products by MSPD Method (MSPD법에 의한 축산물 중 합성항균제 동시분석)

  • This study was conducted to evaluate the MSPD and HPLC method about simultaneous determination for residual synthetic antimicrobials of sixteen species such as sulfonamide etc. in livestock products. Elution solvent used in HPLC was ethylacetate:acetonitrile (4:1), and mobile phases for solvent A and B were water:methanol:acetonit rile:phosphric acid (700:250:50:0.2) and 100% acetonitrile respectively. The detector and absorbency used in HPLC was UV 266 nm. This study showed the reduction effect of 99.1% for organic solvents, 94% for experimental steps, 95% for analytical time and manpower and 98.9% for costs compared with korea food standard method. The average recovery rates for chicken, bovine, pork and milk were 67.7% 96.2%, 67.7%~96.6%, 70.0%~96.2%, and 13.8%~97.8%.

The Quality Characteristics of Beef Patty with the Addition of Gynura procumbens (명월초 가루를 첨가한 소고기 패티의 최적화 연구)

  • The purpose of this study was to determine the optimal mixing ratio of Gynura procumbens and milk in the preparation of beef patties. The experiment was designed according to the central composite design for estimating the response surface, demonstrating 10 experimental points including 2 replicates for Gynura procumbens and milk. The physical, mechanical and sensory properties of the test materials were measured, and a canonical form and perturbation plot were used to show the influence of each ingredient on the final product mixture. Results of the measurements showed significant increases or decreases in the following properties: dough moisture (p<0.05); beef patty a (p<0.001) and b (p<0.001); hardness (p<0.01), adhesiveness (p<0.05), chewiness (p<0.05), and gumminess (p<0.05). In addition, the sensory measurements showed significant improvements in color (p<0.001), flavor (p<0.05), greasy (p<0.05) and overall quality (p<0.05). The optimum formulation calculated from the numerical and graphical analysis was 1.75 g Gynura procumbens and 20.44 g of milk.
