Melanogenesis Promotion by 3-Deazaneplanocin A, a Specific Inhibitor of S-Adenosylhomocysteine Hydrolase, in B16/F10 Melanoma Cells (B16/F10 흑색종 세포에서 S-Adenosylhomocysteine Hydrolase 의 선택적 저해제 3-Deazaneplanocin A 에 의한)

  • Skin hypopigmentation, which is observed in albinism or vitiligo, occurs when melanin synthesis is decreased by genetic, epigenetic, and other factors. To identify drug candidates that can promote melanin synthesis in cells, we screened an epigenetic modulator library consisting of 141 cell-permeable, small molecule drugs. B16/F10 murine melanoma cells were treated with each drug at 0.1 𝜇M and melanin synthesis and cell viability were subsequently monitored. As a result, (-)-neplanocin A, 3-deazaneplanocin A (DZNep), and DZNep hydrochloride were found to increase cellular melanin synthesis without causing cytotoxicity. Because these three structurally related drugs exhibited similar dose-dependent effects on melanin synthesis and cell viability, DZNep was selected as a representative drug for additional experiments. DZNep increased intracellular melanin content and tyrosinase (TYR) activity. DZNep also induced the expression of TYR, tyrosinase-related protein 1 (TYRP1), and dopachrome tautomerase (DCT) at the mRNA and protein levels. DZNep also induced the mRNA and protein expression of microphthalmia-associated transcription factor (MITF), a key regulator of melanin synthesis. DZNep is a specific inhibitor of S-adenosylhomocysteine hydrolase and it caused the accumulation of S-adenosylhomocysteine that inhibits histone methyltransferases in cells. This study suggests that melanogenesis can be modulated by targeting S-adenosylhomocysteine hydrolase in certain cellular contexts.

  • We have studied the effects of red ginseng root extract on the decrease of UV-induced genotoxicity in cultured NIH3T3 cells. The increase in survival and the recovery from DNA synthesis inhibition in UV-irradiated cells as a function of normal medium incubation time was potentiated by the presence of the ginseng extract. The extract also increased the UV-induced excision repair as determined by unscheduled DNA synthesis. The amount of UV-induced DNA single strand breaks that are accumulated by polymerase inhibitors was significantly increased by the presence of the extract. These results suggest that the red ginseng extract activates the incision/excision step of UV-induced repair and could be used as a reagent for protecting UV-induced genotoxicity and cytotoxicity.

  • Ten strains of tilapiine species from genus Oreochromis were cytogenetically studied for genetic stock identification. Both the chromosome numbers(2n=44) were identical in all 10 strains. Heteromorphic sex chromosome pair were not found in any strains. Nuclear volumes vary between O. niloticus(21.0 $\mu$ $m^3$) and O. aureus(22.4 $\mu$ $m^3$)

  • At present over 3 dozen mutant genes have been recognized in the Mexican axolotl. These genes, all recessives, are categorized in 5 groups according to the nature of their effects and the developmental stage at which their phenotype is predominately expressed. They are genes affecting the oogenesis (maternal-effect genes), genes affecting the size of the nucleolus, genes affecting the development of specific tissues and organs, genes exert lethal effects on all cells or tissues (autonomous lethals), and genes affecting pigment cells. This report describes briefly the phenotypes and some of the current research applications of those genes.

  • Acridine orange를 이용한 염색법은 OECD guideline 474에 채택되어 널리 사용되고 있는 염색법으로 Giemsa를 이용하였을 때 보다 artifact를 구별하기 쉬우며 숙련되지 않는 관찰자도 쉽게 소핵을 구별할 수 있다는 장점을 가지고 있다. 본 시험에서는 신약의 초기 screening단계에서 유전독성여부를 알아보기 위한 방법으로 마우스 골수세포와 말초혈액을 이용하여 Geimsa와 Acridine orange염색법을 비교하였다. (중략)

Cytoplasmic Inheritance of High Tillering and Earing Characters of a Korean Local Maize Line(MET) (다수.다얼성 옥수수(NET)의 세포질적 유전)

  • A Korean local maize line, MET, which has multi-ears and tillers has been proved as a potential source for silage production. However, no fundamental genetic nature for the line has been investigated. Therefore, this study was done to find genetic information on the multi-earing and -tillering habits of MET line. MET line and a hybrid. (Mo 17 ${\times}$ B68), with monoculm and single ear per plant were used for production of F$_1$(F$\_$1-12/ and F$\_$1-21/), F$\_$2-12/, F$\_$2-21/, BC$\_$1-12/ and BC$\_$1-21/ generations. From the comparison of reciprocal crosses, it was found that the tillering and earing habits of the MET line are controlled by cytoplasmic factors. The tiller and ear numbers, and barren ears were all characters associated with the MET cytoplasm. The cytoplasmic effect of MET on tiller and ear numb en was not evident in F$_1$ generation, probably because of suppressing effect of heterosis on appearance of tillers or ears. Genetic parameters for the gene action for both tiller and ear number also indicated a lack of mono- or digenic-chromosomal gene effects. The heritability (broad) was very low for both characters. Therefore, it is strongly concluded that the tillering and earing characters of MET line are due to cytoplasmic reasons.

  • This study was carried out to investigate the effect of 70% ethanol extract and each fraction from Rhododendron brachycarpum D. Don leaves on cytotoxicity, anticancer, genotoxicity and immunological activity in vitro bioassay. Cytotoxicity for human normal cells (HEL299 and Chang) of the samples was shown below 35% in 0.5 mg/ml concentration of samples except aqueous fraction by SRB assay. DNA damage on the Chang cell of the samples alone in comet assay was observed very weak damage activity even in high concentration (1 mg/ml) of the samples. The anticancer effect of the samples on human cancer cell lines (A549, AGS, Hep3B, MCF7) was indicated that the cancer cells were inhibited gradually in proportion to the increase of the concentration of the samples by MTT assay. The growth of the Raji and Jurkat cells were hastened by adding butanol fraction among the samples. In the genotoxicity on $H_2O_2-induced$ DNA damage in Chang cells using alkaline comet assay, most of samples were shown a strong protective activity from DNA OTM values.

Complete genome sequence of Lactobacillus plantarum SK151 isolated from kimchi (김치로부터 분리한 Lactobacillus plantarum SK151의 유전체 염기서열 해독)

  • Lactobacillus plantarum is a Gram-positive, facultative heterofermentative, nonspore-forming nonmotile bacterium found in a wide range of environmental niches. Here we present the complete genome sequence of L. plantarum SK151 isolated from kimchi, which shows high adhesion to intestinal epithelial cells. The genome is 3,231,249 bp in length and has a GC content of 44.6%. The genome contains genes related to cell adhesion and a complete operon for riboflavin biosynthesis.

Inheritance and Heritability of Telomere Length in Chicken (닭 텔로미어 길이의 유전력 추정과 유전 전이 양상)

  • Telomeres are the ends of the eukaryotic chromosomes and consist of a tandem repetitive DNA sequence and shelterin protein complex. The function of telomere is to protect chromosome. Telomere length in somatic cells tends to decrease with organismal age due to the end replication problem. However, several factors at the genetic, epigenetic and environmental level affect telomere length. In this study, we estimated heritability of telomere length and investigated inheritance of telomeres in a chicken. Telomere length of lymphocytes was analyzed by semi-quantitative polymerase chain reaction using telomere primer and quantitative fluorescence in situ hybridization using telomeric DNA probe. In results, heritability of telomere length was estimated 0.9 at birth by offspring-parent regression analysis and was estimated 0.03 and 0.04 at 10 and 30 weeks old, respectively, by parental variance analysis. There was a significant positive correlation in telomere length between father and their offspring (r=0.348), and mother and their offspring (r=0.380). In inheritance patterns of telomere length, the influence of paternal and maternal effect on their offspring was similar. The influence of inherited telomeres on male and female progeny was also roughly alike. These results implicated that imprinting of parental telomere length was regulated by autosomal genes, not sex linked genes. In addition, telomere length of offspring at birth did not differ along with their maternal age. Thus, maternal age does not affects telomere length in their offspring at birth owing to cellular reprogramming at early embryonic stage.
