Isolation of Myxobacteria Carrying Soraphen Biosynthetic Gene Clusters (Soraphen 생합성 유전자군을 갖는 점액세균의 분리)

  • Polymerase chain reactin (PCR) could be a simple way to screen new microbial strains producing useful secondary metabolites if their biosynthetic genes are known and candidate strains to be screened are available. In this study, we have screened two myxobacterial strains, KYC3047 and KYC3076, carrying genes appeared to be biosynthetic genes of soraphen A, a potent antifungal substance, out of 50 cellulose degrading myxobacteria using PCR. The two strains were identified as Sorangium cellulosum based on morphological, physiological, and molecular biological characteristics. Both of the strains produced substances having strong antifungal activities as expected against Candida albicans, a causative agent of candidiasis, and Colletotrichum acutatum, a causative agent of anthracnose on pepper.

  • 메탄을 탄소원 및 에너지원으로 이용하는 메탄산화균은 물질대사과정 중에 다량의 세포외 고분자물질인 Extracellular polymeric substances(EPS)를 생성하는데, EPS는 카르복실기와 같은 표면흡착 기능을 가지고 있어 생체흡착제로 사용이 가능하다. 따라서 본 연구에서는 메탄산화세균을 이용하여 중금속인 Cd의 흡착성능을 파악하여 활성슬러지의 흡착능과 비교하고, EPS 농도별, pH별 흡착량의 변화를 실험한 후 Freundlich 흡착모델식에 적용하여 흡착공정의 기본적인 설계인자를 도출하고자 하였다. 실험에 사용한 메탄산화세균은 매립지 복토층 상부 토양에서 분리하여 실험실에서 대량으로 배양하였으며, EPS 생성을 위해 메탄을 Head space의 20%를 주입하고 $30^{\circ}C$, 150rpm에서 질소원이 부족한 조건으로 48hr 동안 배양하였다. Cd의 흡착실험은 용액의 pH를 3에서 8까지 변화를 주면서 활성슬러지와 메탄산화세균의 시간별 흡착능을 측정하였다. 또한 중금속의 농도별 흡착능을 측정하여 흡착평형 상수를 파악하였으며, 중금속 흡착 전, 후 미생물의 SEM 촬영, FT-IR 분석, 전자현미분석(EPMA)을 통하여 무기성분 분석 및 표면관찰을 수행하였다. 실험결과 메탄산화세균에 의해 생성된 EPS 물질은 중금속에 대한 강한 결합능력이 있으며, Cd에 대한 최고 흡착능은 26mg Cd(Ⅱ)/g VSS의 값을 보였다. 이러한 미생물의 EPS의 흡착능은 pH와 칼슘이온의 영향을 많이 받았으며, 메탄산화세균의 FT-IR 분석결과 EPS에는 sulfate ester, pyruvate 등과 같은 작용기와 amino sugar, carboxyl 작용기들이 많이 존재하여 활성슬러지에 비해 중금속의 흡착능이 높은 것으로 사료되었다.X>${\mu}_{max,A}$는 최대암모니아 섭취률을 이용하여 구한 결과 $0.65d^{-1}$로 나타났다.EX>$60%{\sim}87%$가 수심 10m 이내에 분포하였고, 녹조강과 남조강이 우점하는 하절기에는 5m 이내에 주로 분포하였다. 취수탑 지점의 수심이 연중 $25{\sim}35m$를 유지하는 H호의 경우 간헐식 폭기장치를 가동하는 기간은 물론 그 외 기간에도 취수구의 심도를 표층 10m 이하로 유지 할 경우 전체 조류 유입량을 60% 이상 저감할 수 있을 것으로 조사되었다.심볼 및 색채 디자인 등의 작업이 수반되어야 하며, 이들을 고려한 인터넷용 GIS기본도를 신규 제작한다. 상습침수지구와 관련된 각종 GIS데이타와 각 기관이 보유하고 있는 공공정보 가운데 공간정보와 연계되어야 하는 자료를 인터넷 GIS를 이용하여 효율적으로 관리하기 위해서는 단계별 구축전략이 필요하다. 따라서 본 논문에서는 인터넷 GIS를 이용하여 상습침수구역관련 정보를 검색, 처리 및 분석할 수 있는 상습침수 구역 종합정보화 시스템을 구축토록 하였다.N, 항목에서 보 상류가 높게 나타났으나, 철거되지 않은 검전보나 안양대교보에 비해 그 차이가 크지 않은 것으로 나타났다.의 기상변화가 자발성 기흉 발생에 영향을 미친다고 추론할 수 있었다. 향후 본 연구에서 추론된 기상변화와 기흉 발생과의 인과관계를 확인하고 좀 더 구체화하기 위한 연구가 필요할 것이다.게 이루어질 수 있을 것으로 기대된다.는 초과수익률이 상승하지만, 이후로는 감소하므로, 반전거래전략을 활용하는 경우 주식투자기간은 24개월이하의 중단기가 적합함을 발견하였다. 이상의 행태적 측면과 투자성과측면의 실증결과를 통하여 한국주식시장에 있어서 시장수익률을 평균적으로 초과할 수 있는 거래전

Antibiotic Production of Pseudomonas otitidis PS and Mode of Action (Pseudomonas otitidis PS 균주의 항생물질 생산과 작용 기작)

  • An isolate capable of inhibiting the growth of gram-positive bacteria was obtained from the soil of Mushim stream, Cheongju. The isolate was identified as Pseudomonas otitidis PS by 16S rRNA gene sequence analysis. P. otitidis PS produced antibiotics as a secondary metabolite when cultured in 1% soybean meal with 0.5% glucose. The maximum yield was about 0.1%. The antibiotic substance of P. otitidis PS extracted using ethyl acetate displayed a minimum inhibitory concentration of $2{\mu}g/ml$ for Staphylococcus aureus KCTC 1261. The antibiotic substance produced an orange halo on chrome azurol S agar due to siderophore activity. Growth inhibition was decreased when the iron was depleted. Since the antibiotic activity was lost upon the addition of the reducing agent ascorbic acid or during anaerobic culture, it was considered that antibiotic of P. otitidis PS strain exerts its bactericidal effect by the generation of reactive oxygen species.

Detection of Conserved Genes in Proteobacteria by using a COG Algorithm (COG 알고리즘을 통한 Proteobacteria의 보존적 유전자 파악)

  • A COG(clusters of orthologous groups of proteins) algorithm was used to detect conserved genes within Proteobacteria and to figure out their relationships. Restricting comparison to the sequences of 42 procaryotes, 33 eubacteria and 16 Proteobacteria, the number of conserved genes was increased. All analyzed procaryotes shared 75 COGs. COG0195, COG0358 and COG0528 were only represented by the 42 procaryotes. Sixtyfour COGs were added as conserved genes in 33 eubacteria. Each Proteobacteria group has a unique repertoire of COGs. Metabolic COGs were more diverse in the beta Proteobacteria group than in the other groups. These results could be used to determine the origins and the evolutionary relationships of Proteobacteria. The possibilities of detecting new biological molecules is high in phylogenetically related organisms, hence the identification of useful proteins by using this algorithm is possible.

  • A monoterpenoid compound, benzylideneacetone (BZA), is a metabolite of an entomopathogenic bacterium, Xenorhabdus nematophila. Its primary biological activity is an inhibitor of phospholipase $A_2$, which catalyzes the committed step of biosynthesis of various eicosanoids that are critically important to mediate insect immune responses. When BZA was applied to two-spotted spider mite, Tetranychus urticae, it exhibited a dose-dependent mortality in leaf-disc assay. Subsequently BZA was tested against T. urticae infesting apples in a field orchard, in which it showed a significant control efficacy, which was not statistically different with that of a commercial acaricide. BZA also had significant antibacterial activities against three species of plant pathogenic bacteria when it was added to the bacterial cultures, in which it showed the highest inhibitory activity against a bacterial wilt-causing pathogen, Ralstonia solanacearum. The bacterial pathogen caused significant disease symptom to young potato plants. However, BZA significantly suppressed the disease occurrence. This study suggests that BZA can be used to develop a novel crop protectant to control mite and bacterial pathogen.

참치정소로부터 핵산복합물질의 추출 및 생리활성 연구

  • 핵산의 생합성은 salvage합성이 우선되며 이 경우 섭취된 음식물유래의 핵산등이 이용되므로 섭취되는 핵산의 영양학적 가치는 매우 높다(송영정사, 1995). Van Buren이나 Kulkarni등은 mouse에의 핵산 경구투여가 면역능을 개선하여 세균감염에 대한 방어에 유효하다고 보고하였고(Van Buren C.T et al, 1983, Kulkarni A.D et a 1986), 이들의 연구가 계기가 되어 섭취 핵산의 유용성에 대한 연구보고가 계속적으로 행하여지고 있는데 그 생리활성을 살펴보면 세포부활작용(Matsunaga et al, 1987) 면역증강작용(Frederic B. Rudolph, 1994) 과산화지질 형성억제작용(Uauy R et al 1985), 장내균총 및 지질대사의 개선작용(Uauy R, 1994, Angel Gil et al, 1986)등이다 본 연구에서는 부산물로 폐기되어지는 참치정소를 이용하여 핵산복합물질을 분리·정제하는 방법 및 그 추출물의 생리활성을 연구하므로써 새로운 기능성 소재로서의 이용가능성을 탐색하였다. Growth promoting effect on Tricholoma matsutake mycelium by bacteria from fairy Ring in Bonghwa-gun, Korea (국내 봉화 송이 자생지 내 균환 유래 토양세균의 송이균사체 생장촉진 효과)

  • As a member of ectomycorrhizal fungi, Tricholoma matsutake has a symbiotic relationship with its host, Pinus densiflora. To cultivate T. matsutake artificially, the co-cultivation of T. matsutake mycelia and bacteria from shiro was introduced. In this study, bacteria were isolated from soil samples in Bonghwa-gun, and seven bacterial isolates (B22_7_B05, B22_7_B06, B22_7_B07, B22_7_B08, B22_7_B10, B22_7_B13, and B22_7_B14) promoted the growth of T. matsutake mycelia (147.48, 232.11, 266.72, 211.43, 175.17, 154.62, and 177.92%, respectively). Sequencing of the 16S rRNA region of the isolated bacteria was performed. B22_7_B05 and B22_7_B10 were identified as Bacillus toyonensis, B22_7_B06 and B22_7_B08 as Paenibacillus taichungensis, B22_7_B07 and B22_7_B14 as P. gorilla, and B22_7_B13 as P. odorifer. These bacterial isolates were associated with the shiro community and are expected to contribute to the cultivation of T. matsutake.

Control Effects of 3-(4-Hydroxyphenyl)-propionic Acid Isolated Xenorhabdus nematophila K1 against Phytophthora Blight and Anthracnose of Red Pepper (Xenorhabdus nematophila K1 대사물질 3-(4-hydroxyphenyl)-propionic acid의 고추 역병과 탄저병에 대한 방제 효과)

  • 3-(4-Hydroxyphenyl)-propionic acid (HPP) is a bacterial metabolite synthesized and released by an entomopathogenic bacterium Xenorhabdus nematophila K1. In this study, the control efficacy of HPP was tested against Phytophthora blight and anthracnose of red pepper plants. HPP suppressed mycelial growth of Phytophthora blight and anthracnose pathogens. Under natural sunlight condition, HPP maintained the antifungal activity on the diseases for at least twenty five days. The antifungal activity was not decreased even in the condition of soil-water. It was proved that HPP was able to penetrate the roots and travel upward of the red pepper plants. When HPP suspension was applied to soil rhizosphere before transplanting the red pepper seedlings or was regularly sprayed to the foliage of the plants with ten days interval, it resulted in significant reduction of the disease occurrences (Phytophthora blight and anthracnose) without any phytotoxicity. These results suggested that HPP can be developed to a systemic agrochemical against Phytophthora blight and anthracnose of red pepper plants.

  • Antibiotics have greatly contributed to the treatment and prevention of bacterial diseases in humans, animals, and fish. However, antibiotic misuse has led to the emergence and spread of multidrug-resistant bacteria. In addition to antibiotic discovery research, efforts are being made to combat such multidrug-resistant bacteria using antimicrobial agents, antioxidants, host immune enhancement, probiotics, and bacteriophages, as well as various symptomatic therapies. To discover novel bioactive compounds, it is crucial to adopt approaches that incorporate fresh ideas, new targets, innovative techniques, and untapped resources. Halophytes are plants that grow in high-salt soils and are known to adapt to salt-induced stress through unique metabolic processes that produce secondary metabolites. This study aimed to investigate the effects of extracts of halophytes native to Korea on oxidative stress and to determine whether they exert inhibitory activity against biofilms, which are major pathogenic factors of infectious bacteria. The Acinetobacter baumannii strain ATCC 17978, a representative drug-resistant bacterium, was used to measure anti-biofilm activity. The results showed that Aster spathulifolius, Carex kobomugi, Rosa rugosa, and Asparagus cochinchiensis exerted strong antioxidant and anti-biofilm effects without affecting bacterial growth itself. The halophytes used in this study are promising candidates for the development of pharmaceutical agents with antioxidant and antimicrobial properties.

Isolation of Antibiotics Effective to Multidrug-Resistant Cancer Cells from Sorangium cellulosum(Myxobacteria). (점액세균 Sorangium cellulosum이 생산하는 약제내성 암세포의 증식억제물질)

  • Drug resistance is one of the most significant impediments to successful chemotherapy of cancer. Multidrug-resistance Is characterized by decreased cellular sensitivity to anticancer agents due to the overexpression of P-glycoprotein. By using adriamycin-resistance CL02 cancer cells, we undertook the screening fur agents which were effective to multidrug-resistant cancer cells from strains of the species Sorangium cellulosum isolated in our laboratory. Sorangium cellulose, cellulose-degrading myxobacteria have recently proved to be a rich source of novel anticancer agents. One of the significant examples is the promising anticancer agent epothilone. JW 1006 is the first strain of Sorangium cellulosum which was selected by us for the isolation of a metabolite by a biological screening because of a high cytotoxic activity against the CL02 cancer cells. Cytotoxicity-guided chromatographic fractionation of the culture broth led to the Isolation of two active principles, disorazole $A_1$ and $A_2$. They showed potent cytotoxicity against CL02 cancer cells with $IC_{50}$ values in the picomolar range, and were as active against drug-resistant cancer cells CL02 and CP70 as against the corresponding sensitive cells.
