Biocontrol Activity of Myxococcus sp. KYC 1126 against Phytophthora Blight on Hot Pepper (점액세균 Myxococcus sp. KYC 1126을 이용한 고추 역병 생물학적 방제 효능)

  • Bacteriolytic myxobacteria have been known to secrete various antifungal metabolites against several soilborne phytopathogens including Phytophthora. Among the three isolates of Myxococcus spp., KYC 1126 and KYC 1136 perfectly inhibited the mycelial growth of Phytophtora capsici in vitro. In order to show the biocontrol activity on Phytophthora blight of hot pepper, we tried to find the best way of application of a myxobacterial isolate. Although KYC 1126 fruiting body was easily grown on the colony of Escherichia coli as a nutrient source, it did not control the disease when it was pre-applied in soil. Before the bioassay of a liquid culture filtrate of KYC 1126 was conducted, its antifungal activity was confirmed on the seedlings applying with the mixture of the pathogen's zoospore suspension and KYC 1126 filtrate. On greenhouse experiments with five and four replications, the control value of KYC 1126 on phyllosphere and rhizosphere was 88% and 36%, respectively. Whereas, the control value of dimetnomorph+propineb on phyllosphere was 100% and that of propamorcarb on rhizosphere was 44%. There was a phytotoxicity of the myxobacterial filtrate when seedlings were washed and soaked for 24 hours. Gummy materials were covered with roots. And stem and petiole were constricted, then a whole seedling was eventually blighted.

  • 트리클로로에틸렌 (trichloroethylene, TCE)는 오랜 시간동안 자연환경에서 잔류할 뿐만 아니라 TCE보다 더욱 더 독성이 강한 중간 생성물들을 만들기 때문에 미국과 대부분의 전세계 국가들로부터 주요 1차 환경오염물질로 분류되었다. 그러한 독성물질들은 혐기성 상태에서는 다이클로로 에틸렌(dichloroethylene, DCE)과 바이닐 클로라이드 (vinyl chloride, VC)와 같은 독성물질들이 생성되고 호기성 상태에서는 TCE epoxide계통의 물질들이 생성된다. 또한 훈증제인 메틸 브로마이드 (methyl bromide)는 대기의 오존층을 파괴하는 것으로 알려져 있고, 2001년경에 미국환경보호청 (USEPA)에 의해 사용이 금지될 것이다. TCE는 혐기성 조건하에서 연속적으로 탈염소화되고, 이어서 호기성 조건하에서 완전 산화될 수 있다. 그리하여 연속적인 혐기성 및 호기성 조건하에서 궁극적으로 TCE의 완전분해를 이루게된다. 메틸브로마이드는 화학적으로 가수분해되어 메틸 알콜 (methyl alcohol)로 되거나 유기물에 강하게 결합 (bound)된다. 또한 그것은 생물학적으로 포름알데하이드 (formaldehyde)로 산화되거나 메틸알콜로 가수분해된다. 수많은 연구자들에 의해 행해진 연구들은 TCE와 MeBr은 메탄 혹은 암모니아 산화 세균에 의한 공동대사과정 (cometabolism)을 통해 분해가 증진될 수 있다는 것을 보여주었다. 두 부류의 세균들이 두 화합물들을 분해시킬 수 있는 monooxygenase를 생산한다는 것은 잘 알려져 있다. 이 연구 논문에서 TCE와 MeBr의 생분해와 관련된 가장 최근의 연구논문들로부터 나온 핵심 연구결과들이 요약 검토된다. TCE와 MeBr로 오염된 현장을 정화하기 위해 이러한 기초연구결과들을 토대로 더욱 더 많은 연구가 필요 할 것으로 사료된다.

Production of Bioactive Substances by a Myxobacterium Myxococcus stipitatus KYC4013 (점액세균 Myxococcus stipitatus KYC4013에 의한 생리활성물질 생산)

  • Myxococcus stipitatus KYC4013 extract exhibited the most potent antifungal activity among the extracts of 207 Myxococcus strains isolated in Korea. High-resolution LC-MS analysis revealed that M. stipitatus KYC4013 produces five antifungal substances and three other secondary metabolites that were predicted to be melithiazol and phenalamide derivatives, respectively. The putative melithiazol derivatives were best produced in CYS medium and the putative phenalamide derivatives were best produced in VY3 medium.

  • The onion moth, Acrolepiopsis sapporensis, was monitored in the farms cultivating the welsh onion, Allium fistulosum, using sex pheromone from transplantation to harvest. Two occurrence peaks were observed at early June and late July after the overwintering population. However, the population sizes were varied among different years and the cultivating environments. To effectively control A. sapporensis with microbial pesticides, different Bacillus thuringiensis strains were screened to select B. thuringiensis kurstaki (BtK). To enhance the insecticidal virulence of BtK, the culture broth of Photorhabdus temperata temperata (Ptt) was added to the BtK. This mixture of two entomopathogenic bacteria was called 'BtPlus', which was superior to BtK alone in the insecticidal virulence. The enhanced virulence was explained by the immunosuppressive activity of the secondary metabolites contained in the Ptt extract. The metabolites inhibited both cellular and humoral immune responses of A. sapporensis, resulting in the enhanced virulence of BtK. These results suggest that A. sapporensis occurs in the welsh onion fields and the resulting economic damage would be effectively prevented by BtPlus application.

  • Subinhibitory concentrations (sub-MICs) refer to concentrations below minimum inhibitory concentrations (MICs). The antimicrobial agents may be present at relatively high concentration, at least higher than bacterial MIC and thereafter be deserted off a surface and function at sub-MICs, perhaps by interfering with bacterial metabolism. Consequently, the aim of this study was to determine the effects of growth, in the presence of sub-MICs of antimicrobial agents, on the cell surface properties and virulence factors of mutans streptococci and to investigate the efficacy of a chemical approach in vitro. Streptococcus mutans Ingbritt and Streptococcus sobrinus 6715-7 were used. Eight antimicrobial agents (Sanguinaria extract;SG, Chlorhexidine digluconate;CHX, Fluoride;F, Propolis;PP, Hydrogen peroxide;HP, Triclosan;TC, Sodium dodecyl sulfate;SDS Cetylpyridinium chloride; CC) were diluted serially in broth to determine MICs and to compare the growth rate, acid production, hydrophobicity, adhesion activity to saliva coated hydroxyapatite, glucan synthesis and cellular aggregation of experiment groups (in the presence of sub-MICs) with those of control (in the absence of antimicrobial agents). Sub-MICs of antimicrobial agents affected the growth of cells, hydrophobicity, and adhesion of bacteria to saliva coated hydroxyapatite and glucan synthesis. They also resulted in a significant reduction in pH after 12 hours (p<0.05). By cells pretreated with proteinase K, either the aggregation induced by antimicrobial agents was completely inhibited or the aggregation titers were markedly increased. According to the results of the present study, each antimicrobial agent at sub-MICs could affect similar as its known action mechanism and could continually inhibit cariogenic bacteria at such concentrations. Thus, the use of these antimicrobial agents would be one of the effective methods to prevent dental caries.

  • Microbial secondary metabolites of marine organisms are regarded as major sources of structurally and biologically novel compounds with numerous potential uses. Sponge-microbe associations are among the most interesting sources for exploring bioactive compounds. In this study, the bacterial strain Microbulbifer sp. (127CP7-12) was isolated from the Asian marine sponge Hymeniacidon sinapium collected at an intertidal zone on the west coast of Korea. Cultured bacteria produced a violet pigment, and optimal culture conditions for violet pigment production were investigated. Maximum production of the violet pigment from the strain culture was observed under the conditions of $25^{\circ}C$, pH 6.0, and 3% NaCl. Acetone provided better extraction of the pigment from fermented broth compared with ethanol and methanol. The proposed structure of the major component in the extracted crude pigment was determined via high-performance liquid chromatography, nuclear magnetic resonance, mass spectrometry, and UV spectra analyses, which showed that the metabolite was the promising bioactive compound violacein. This study describes the examination of marine bioactive materials from microbe-engaged metabolites and the ecological implications of the sponge-microbe association in a changing ocean.

  • The microbiological degradation of 2-(2-methyl-4-chloro-phenoxy) propionic acid (MCPP) was evaluated using mixed cultures of soil bacteria. The mixed cultures comprised Pseudomonas species, Flavobacterium species, and Achromobacter species. The bacteria used MCPP as the sole source of carbon and energy but only a partial degradation of the parent compound occurred MCPP degradation proceeded via the formation of 2-methyl-4-chlorophenol (2, 4-MCP) which was detected by high pressure liquid chromatography (PHLC) and confirmed by gas chromatography-mass sepctrometry. This intermediate occurred only transiently and no evidence was seen for the presence of other intermediates detectable by the reverse-phase HPLC or UV absorbance.

COG 알고리즘으로 파악한 Proteobacteria의 보존적 유전자

  • A COG (clusters of orthologous groups of proteins) algorithm, protein similarities among genomes, was used to detect conserved genes and to figure out their relationships within 42 procaryote, 33 Bacteria and 16 Proteobacteria All analyzed procaryotes shared 75 COGs. COG0195, COG0358 and COG0528 were only represented by the 42 procaryotes. Sixty-four COGs were added as conserved genes in 33 eubacteria. Each Proteobacteria group has a unique repertoire of COGs. Metabolic COGs were more diverse in the beta-Proteobacteria group than in the other groups. The possibilities of detecting new biological molecules is high in phylogenetically related organisms, hence the identification of useful proteins by using this algorithm is possible.

  • Two bithiazole-type antibiotics were isolated from the culture broth of a Myxococcus species which isolated from the marine sediment off the coast of Cheju Island, Korea. The structures of these metabolites were determined as KR025 and melithiazole F, previously reported bithiazoles, using combined spectroscopic methods. Both compounds showed an antifungal activity. In in vivo tests, these compounds exhibited potent controlling activities against tomato late blight, wheat leaf rust, and barley powdery mildew with control values more than 80% at a concentraton of 20 $\mu\textrm{g}$/ml.
