Refinement of Low-grade Clay using Iron-reducing Bacteria [I] : Comparison of Traditional Ripening and Biotechnological Ripening (철환원세균을 이용한 저품위 점토의 개량[I] : 전통적 수비법과 생물공학적 숙성법의 비교)

  • The traditional ripening method of clay was analyzed. An advanced refining method of clay using enrichment cultures of iron r reducing bacteria was developed. After the traditional ripening, the whiteness of the clay was increased due to removal of | iron impurities by inhabitant dissilmaltien with iron reducing bacteria. Other characteristics of the refined clay such as v viscosity, plasticity, and strength were also improved by iron reducing bacteria. An advanced method of clay refinement with a anaerobic enrichment cultivation of iron reducing bacteria supplemented with an extra carbon source such as glucose was s suggested. When the clay was treated by the advanced method. the refinement time could be reduced to 1/6 of that r required by the traditional method. The physical properties of the refined clay by the advanced method were better than t those of the traditionally refined clay.

  • 누룩은 생소맥을 조분쇄하여 물로 혼합시켜 자연적으로 공지중의 여러 종류의 미생물을 배양시켜 만들었다. 그러므로 누룩에서는 많은 종류의 사상균, 호모와 세균이 증식하였다. 누룩의 당화력은 원료 생소맥의 당화력과 대부분의 사상균의 당화력과 알붕의 세균성 당화력에 기인하여, 발효력은 누룩효모와 극소수의 사상균애 의하여 이루어지고 있다. 1945년 이전까지 누룩으로부터 12속 59종의 사상균이, 8속 29종의 효모와 4속 16종의 세균이 분리 되었다. 누룩사상균은 Aspergillus 속이 주종을 이루었고 Rhizopus속, Absidia속, Nucor속의 순으로 분포하고 있었고, Aspergillus속과 Rhizopus속은 중요한 누룩 당화균으로 작용하였다. 누룩의 대표적 효모는 Saccharomyces cerevisiae 이며 , 이균이 알코올 발효에 가장 큰 역할을 담당하며, Saccharomyces cerevisiae도 전통 민속주의 탁주와 약주의 발효역에 깊이관여한다고 사료된다. 누룩 중에 증식하는 세균은 양조학상 특별한 역할을 담당하지않지만 Bacillus 속 과 젖산세균이 많이 분리되었으며, 젖산세균은 담금초기에 pH의 안정화에 기여할 수 있다고 추측할 수 있다.

Effects of Oxygen and Salt on the Growth of Bifidus and Anaerobic Bacteria Isolated from Korean Traditional Fermented Foods (산소와 염농도가 한국전통 발효식품에서 생장하는 혐기성 세균과 Bifidus균의 생육에 미치는 영향)

  • Fourteen samples of kimchies and soy bean pastes were used to isolate strictly anaerobic bacteria on complex BL agar and on a selective BS agar for bifidus bacteria. About $10^7$ ~ $10^8$ colonies per g sample were developed on BL agar under strictly anaerobic conditions, while BS agar supported the growth of $10^3$ ~ $10^6$ colonies per gram sample at the same condition. All colonies developed on BS agar at anaerobic conditions grew in aerobic conditions and did not show fructose6-phosphate phosphoketolase activity. Type cultures of Bifidobacterium did not grow in PYG medium containig more than 3% NaCI. From these results it is conduded that salted fermented food cannot support the growth of strictly anaerobes induding Bifidobactenum.

Biological Control of Stem Rot of Pepper caused by Sclerotium rolfsii using by Bacillus amyloliquefaciens KBC1009 (길항세균 Bacillus amyloliquefaciens KBC1009를 이용한 고추 흰비단병의 생물학적 방제)

  • Sclerotium rolfsii is a well known broad host range soil borne plant pathogenic fungus and caused serious damage to various vegetable crops. To develop an effective biological control agent for S. rolfsii, an isolate which showed strong inhibitory effect on the mycelial growth of S. rolfsii was selected among the antagonistic bacterial isolates collected from vinyl-house soil. The bacterial isolate was identified as Bacillus amyloliquefaciens KBC1009 based on the morphological, physiological characteristics and by 16S rRNA sequence analysis. The growth conditions for B. amyloliquefaciens KBC1009 were optimized in LB media(pH7) by culturing at 30℃ for 72 hrs. Glucose and yeast extract were confirmed as the best carbon and nitrogen sources, respectively. In order to test the inhibitory effect of B. amyloliquefaciens KBC1009 to stem rot of pepper, green house experiment was conducted. Drench of 1/500 diluted bacterial suspension of B. amyloliquefaciens KBC1009(5×108 cfu/ml) to each pepper plant 3 times with 10 days interval showed 66.7% control effectiveness. These results suggest that B. amyloliquefaciens KBC1009 is one of promising biocontrol agent to control stem rot caused by Sclerotium rolfsii.

Effect of Serial Transfer on the Virulence of Xanthomonas oryzae (벼 흰빛잎마름병 병원세균의 계대배양에 병원성에 미치는 효과)

  • Two different isolates of Xanthomonas oryzae, KB 7785 of pathotype I and JN 7721 of pathotype III, that had been the most virulent isolates in the previous inoculation test, were reisolated from cultivar 'Milyang 23' and serially transferred to 10 times. They were inoculated to the 3 cultivars; 'Milyang 23' in Kinmaze group, 'Yushin' in Kogyoku group and 'Tongil' in Rantai-emas group cultivars. It was observed that the virulence of the isolate JN 7721 was more attenuated by the serial transfer on the Wakimoto's agar than the isolate KB 7785. The attenuation of virulence of the isolate JN 7721 was more significant at the cultivar 'Milyang23' than at the other cultivars. This suggests that the host-pathogen interactions and differences of the pathogenicity-maintenance ability among the pathogenic strains may be involved.

  • Various kinds of native herbage grasses like-Miscanthus sinensis, Arundinella hirta, Cymbopogen geirngii, Themeda japonica etc. are widely distributed in every Korean mountain. So we investigated the availability of native grass, Miscanthus sinensis as a substrate for the production of S. C. P. The results were obtained as follows. 1) At the alkali treatment, NaOH was the most effective with the exclusion of lignin, and pretreated Miscanthus with NaOH appeared to be a good substrate for the microbial growth. 2) When Miscanthus was treated with one to 10% NaOH, the microbial growth increased in proportion to the increased alkali concentration. Beyond 4% NaOH, a slight increase was observed. 3) Phosphoric acid, as a neutralizer, was the most effective in cell production after alkali treatment. 4) On the effect of incubation time, the productivity was best found at 60 hours, and the cell weight was 9.23 mg per 1 ml, and the microbial digestibility of substrate was 75.2%.

Antibacterial Effect of Cinnamon (Cinnamomum cassia) Bark Extract against Fish Pathogenic Bacteria (계피 추출물의 어류 질병 세균에 대한 항균 효과)

  • To develop a natural antibacterial agent for fish bacterial diseases, antibacterial activity, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and bactericidal effect of cinnamon bark extract were examined against fish pathogenic bacteria. The antimicrobial effect of the extract to the fish diet was also estimated, Cinnamon bark extract showed the broad spectrum of antibacterial activity against fish pathogenic bacteria, especially, it had strong activity against Streptococcus iniae, Edwardsiella tarda and Listonella anguillarum. Its MIC was $75.8\sim189.6{\mu}g/mL$ against Cram positive bacteria, and $75.8\sim113.8{\mu}g/mL$ against Gram negative bacteria in liquid medium, It was found to show stronger bactericidal action against Gram negative bacteria than Cram positive bacteria. According to increasing concentrations of the extract, it resulted in a proportional reduction of viable cell counts of both S. iniae and L. anguillarum. The former was not detected by addition of $189.6{\mu}g/mL$ after 12 hours incubation and the latter by addition of $151.6{\mu}g/mL$ after 24 hours incubation, respectively. It was reasonable that fish diet was soaked in cinnamon bark extract for ten minutes. The relationship formula between the weight of fish diet and the extract absorbed to fish diet was Y=7.2726X+4.5083 ($R^2=0.9998$). The fish diet soaked in the extract inhibited the growth of all strains used in this study. Its antibacterial activity was stable at the range from $10^{\circ}C\;to\;35^{\circ}C$ during the storage period of 28 days. When the diet soaked in the extract was incubated in liquid medium at $35^{\circ}C$, it inhibited the growth of microorganisms inhabited in the diet.

Comparison of Culture-dependent and DGGE based Method for the Analysis of Marine Bacterial Community (배양법과 DGGE에 의한 해양세균 군집의 비교분석)

  • Seasonal variation of marine bacterial community was analyzed in the surface sea water collected from one of the stations locating at Tongyeoung coastal area, Korea. The results obtained by the culture method through identification with the VITEK Microbe ID system after pure culture in the selective medium were compared with those obtained by the DGGE based 16S rRNA PCR method. The composition of the marine bacterial community in the sea water samples harvested in September, 2004, November, 2004, January, 2005, May, 2005 and August, 2005 determined by the culture method showed 5, 5, 4, 6, and 10 strains respectively. Pseudomonas fluorescens and Acinetobacter lwoffii were detected in all seasons. The other strains were identified to be Pseudomonas stutzeri, Sphingomonas paucimobilis, Burkholderia mallei and Chryseobacterium indologenes. In contrast, the 16S rRNA PCR-DGGE method detected 10, 11, 6, 9 and 13 populations respectively in the same sea water samples and the strains were identified to be Acinetobacter lwoffii, Burkholderia mallei, Pseudomonas fluoresence, Actinobacillus ureae, Burkholderia sp., Pseudomonas stutzeri, Roseobacter sp., Vibrio parahaemolyticue, Sphingomonas paucimobilis and Rugeria algocolus. This results indicated that the DGGE based 16S rRNA PCR method was more efficient than the culture method for the grasp of the characteristics of the marine bacterial community.

A Clinical Study on the Patients with Pyuria (농뇨증이 동반된 환자군의 임상적 고찰)

  • To evaluate the features of pyuria related to the bacteriuria, 140 subjects were studied from Jan. 1987 to Dec. 1987. The pyuria was frequently developed from the age 30 to 60 years old, and male to female ratio was 1:1.41. The most common disease was urethrocystitis that was shown 42.8%. Common precipitating factors were urethral catheterization(25%) and urinary tract obstruction(11.4 %). Through the observation, symptomatic pyuric patients were 66 subjects(47.1%), and the subjects with significant urine culture were 121 subjects(86.4%). In the urine culture, the most common bacteria was E. coli(41.4%), and the next was Pseudomonas(19.3%). A large percentage of E. coli and Pseudomonas was susceptible of amikin. The pyuria due to S. epidermidis and Accinatobacter was well treated. High therapeutic rate was observed in the acute pyelonephritis(71%) and urethrocystitis(67%). In the persistent urinary tract infection, there were relapsing(22 cases) and recurrent urinary tract infection(16 cases).

  • 영양실태조사 및 사람을 대상으로 하는 많은 영양적 연구에서 요중(尿中) 여러 성분(成分)들의 배설량을 측정하기 위해 만 하루의 소변을 완전히 채취하기는 매우 어려우므로 임의시간(任意時間)의 소변을 채취하여 사용한다. 이 경우엔 흔히 요성분(尿成分)의 농도는 요중(尿中) 크레아티닌을 양(量)을 기준으로 하여 표시되는데, 크레아티닌은 요중(尿中) 일일(一日) 배설량이 개인에 따라 알정하고 요양(尿量)에는 상관없이 비교적 일정한 속도로 배설 된다고 간주되기 때문이다. 그러나 비교적 높은 발생율을 갖는 세균재(細菌在) 요도염(尿道炎)에서는 감염균(感染菌)이 크레아티닌을 파괴할 가능성이 있고 따라서 이 경우의 요중(尿中) 크레아티닌의 여러 용도(用途)는 비합리적으로 될 수 있다. 본 연구에서는 이 가설을 규명하려고 한다. 첫 실험에서는 감염균(感染菌)이 요소(尿素)를 암모니아로 파괴함으로써 형성되는 요(尿)의 알칼리성에 대한 크레아티닌의 안정성(安定性)을 알아 보았다. 건강인(健康人)의 요중(尿中) 크레아티닌과 완충액에 용해시킨 순수 크레아티닌을 pH $4.5{\sim}9.0$으로 조정하여 $37^{\circ}C$에서 6일간 배양시켰다. 잔존한 크레아티닌을 측량한 결과, 크레아티닌은 완충용액이나 요(尿)에서서 모두 산성 pH에서 보다 알칼리 pH에서 더욱 안정(安定)함을 보여주었다. 1일간 배양 후엔 거의 변화(變化)가 없었고 6일 후에나 $4.2{\sim}8.0%$의 감소율을 나타냈을 뿐이다. 두번째 실험에서는 감염균이 크레아티닌을 성장(成長)을 위한 질소급원으로 사용하는지를 결정하기 위해 세균성(細菌性) 요도염(尿道炎)에서 자주 발견되는 13종류(種類)의 박테리아를 건강인(健康人)의 요(尿)와 크레아티닌을 질소급원으로 하는 합성배지(合成培地)에 $37^{\circ}C$로 배양하였다. 대부분의 박테리아는 크레아티닌함양(含量)을 감소시키지 않았다. 그러나 Pseudomonas aeruginosa와 Klebsiella pneumoniae는 합성배지중(合成培地中)의 크레아티닌을 상당량 파괴시켰고, 그 파괴율은 그들 성장율과 평행하였다. 배양 6일후에는 크레아티닌이 Pseudomonas aeruginosa에 의해 처음 양(量)(500mg/100m1) 의 12.8%가, Kleobsiella pneumoniae에 의해서는 11.8%가 감소되었다. 감소율은 크레아티닌의 처음 농도가 낮을수록 커져서 50mg/100ml 일 때는 각각 21.1%와 28.2%이었다. 더욱이 Klebsiella pneumoniae는 황산암모늄과 요소(尿素)같은 다른 질소급원이 크레아티닌과 공존(共存)할 때에도 크레아티닌을 어느 정도 파괴함을 보여 주었다. 결론으로, 세균성(細菌性) 요도염(尿道炎)환자의 요(尿)의 알칼리성은 요중(尿中) 크레아티닌양(量)에 중요한 영항을 주지 못한다. 그러나 본(本) 연구(硏究)에서 사용된 Klebsiella pneumoniae와 가능하게는 Pseudomonas aeruginosa 같은 몇 감염균(感染菌)은 크레아티닌을 그들 성장(成長)의 질소급원으로 사용하여 요중(尿中) 크레아티닌양(量)을 저하시킬지도 모른다. 특히 요(尿)에 요소(尿素), 요산(尿酸)같은 다른 질소급원이 크레아티닌에 비해 비교적 낮은 비율로 존재할 때에, 예(例)를 들면 저(低)단백식사(食事)인 경우, 감염균에 의한 크레아티닌의 파괴율이 더 클 것으로 기대된다.
