Biological Traits of the Lesser Strawberry Aphid (Chaetosiphon minus) in Strawberry under Plastic Houses (딸기 재배하우스에 발생하는 애못털진딧물(Chaetosiphon minus)의 생물적 특성)

  • To establish the systematic biological control system for various insect pests in strawberry under plastic houses, we were investigated the biological traits of lesser strawberry aphid, Chaetosiphon minus (Forbes), as a target pest. The lesser strawberry aphid is adapted to low temperatures while examining the availability of and selecting useful natural enemies. The development, survivorship and reproduction of lesser strawberry aphid were evaluated at four constant temperatures (10, 15, 20, and 25℃). The developmental periods of the nymphal stages ranged from 41.7 d at 10℃ to 9.8 d at 25℃. The developmental threshold temperature and degree day of nymphal stages are 5.5℃ and 185 DD. The reproduction rate (R0) was higher at 20℃ (30.16) than at 25℃ (22.38). The 50 % survival rate and maximum longevity of adult females were 31 d and 59 d at 20℃, and 25 d and 36 d at 25℃, respectively. The average progeny per female was 35 at 20℃, and 26 at 25℃. We confirmed that compared to other strawberry aphids under plastic houses, the lesser strawberry aphid is more adapted to lower temperatures. It is, therefore, necessary to commercialize natural enemies such as syrphid flies with high activity at low temperatures.

Developmental and ovipositional charcteristics of Tenodera aridifolia (Mantodea: Mantidae) (왕사마귀(Tenodera aridifolia)의 발육 및 산란 특성)

  • This study was carried out to investigate the development and oviposition characteristics of Tenodera aridifolia. The nymphs of T. aridifolia grew to adults through 6 and 7 nymphal instars. The percentage of 7 nymphal instars (7-instar type) at 25, 28, and $30^{\circ}C$ were 77.3, 76.2, and 73.5%, respectively. In the 7 instars type, total nymphal periods at 25, 28, and $30^{\circ}C$ were 71.9, 53.5, and 54.7 days, respectively. And, there was no difference in deveolpmental periods between 6- and 7-instar types at all temperatures. Egg periods were 41.3 days at $25^{\circ}C$. Adult longevity of females were 82.0 days, deposited average 3.2 oothecae at intervals of 11.7~18.7 days at $28^{\circ}C$ during her life span. The first ootheca was the largest and $40.5mm{\times}22.0mm{\times}17.3mm$ in size. The number of average eggs per oothecae was 178.6 at $28^{\circ}C$.

Evaluating Pesticides for Controlling Apolygus spinolae and A. lucorum Mirid Bug Population Patterns in Jujube Orchards in Boeun, Chungbuk (충북 보은지역 대추나무의 장님노린재 발생 특성 및 활용 가능한 살충제 선발)

  • Jujube tree farms need effective methods of pest control to reduce damage caused by mirid bugs. In this study, we measured the density of mirid bug populations in jujube trees and tested the efficacy of various insecticides. We observed seasonal density patterns, where nymphs were observed from early May to the middle of July; and adults were observed from the end of May to the middle or end of July. Furthermore, we measured the density of two types of mirid bug species, Apolygus spinolae and A. lucorum. Among 45 damaged jujube trees, the dominant species of mirid bugs was A. spinolae (89%). We tested 7 types of insecticides to control mirid bug populations, including pyrifluquinazon, deltamethrin, diazinon, dinotefuran, etofenprox, fenitrothion, and bifenthrin, showing over 80% efficacy. Thus, in this study, we showed that using insecticides was effective for controlling mirid bug populations on jujube trees.

Selection of Entomopathogenic Fungus Isaria javanica FT333 for Dual Control of Thrips and Anthracnose (총채벌레 및 고추탄저병의 동시 방제를 위한 곤충병원성 곰팡이 Isaria javanica FT333 선발)

  • Red pepper is seriously damaged by thrips (Thrips palmi) and anthracnose caused by Colletotrichum acutatum throughout its development. Because of biotic constraints, producers often depend on chemicals that are expensive and have adverse effects on the environment, operator, and beneficial insects. In addition, resistance is developed because of the repeated use of chemicals. In recent decades, the use of microorganisms in crop protection has become a credible alternative because it is eco-friendly. In this study, we aimed to select isolates with insecticidal and fungicidal activities against the pathogens that cause anthracnose and thrips. We treated T. palmi adults and juveniles with 13 strains of entomopathogenic fungi (isolated from the soil by using the insect-bait method), and 6 strains showed excellent insecticidal activity (70-100%) 5 days after the treatment. The selected isolates were cultured with C. acutatum to screen for the strain with excellent anti-fungal activities, among which an isolate FT333 showed more than 95% control efficacy against C. acutatum in vitro. The isolate was identified as Isaria javanica through its morphological characteristics and phylogenetic analysis of the ITS and ${\beta}-tubulin$ nucleotide sequences. The Isaria javanica FT333 isolate could be used effectively for dual bio-control of thrips and anthracnose during red pepper cultivation.

Optimization of Elastic Modulus and Cure Characteristics of Composition for Die Attach Film (다이접착필름용 조성물의 탄성 계수 및 경화 특성 최적화)

  • The demand for smaller, faster, and multi-functional mobile devices in increasing at a rapidly increasing rate. In response to these trends, Stacked Chip Scale Package (SCSP) is used widely in the assembly industry. A film type adhesive called die attach film (DAF) is used widely for bonding chips in SCSP. The DAF requires high flowability at high die attachment temperatures for bonding chips on organic substrates, where the DAF needs to feel the gap depth, or for bonding the same sized dies, where the DAF needs to penetrate bonding wires. In this study, the mixture design of experiment (DOE) was performed for three raw materials to obtain the optimized DAF recipe for low elastic modulus at high temperature. Three components are acrylic polymer (SG-P3) and two solid epoxy resins (YD011 and YDCN500-1P) with different softening points. According to the DOE results, the elastic modulus at high temperature was influenced greatly by SG-P3. The elastic modulus at $100^{\circ}C$ decreased from 1.0 MPa to 0.2 MPa as the amount of SG-P3 was decreased by 20%. In contrast, the elastic modulus at room temperature was dominated by YD011, an epoxy with a higher softening point. The optimized DAF recipe showed approximately 98.4% pickup performance when a UV dicing tape was used. A DAF crack that occurred in curing was effectively suppressed through optimization of the cure accelerator amount and two-step cure schedule. The imizadole type accelerator showed better performance than the amine type accelerator.

Proteomic Study for Low Molecular Weight Peptides in the Mealworm Tenebrio molitor (갈색거저리 유래 저분자단백질체의 분석)

  • In this study, we examined low molecular weight peptides using proteomics in order to identify their original proteins, derive their peptides, and determine the functions of the proteins in Tenebrio molitor, the mealworm (larvae, pupae, or adult) from which the peptides were extracted. Fifty-four proteins were finally identified through an analysis of proteome to derive the analyzed peptides. The proteins that induced low molecular weight peptides were identified to be the most abundant in adults only, and the next highest were derived from a group containing both adults and larva. However, other groups, including pupa, were detected to have a lower frequency of peptides. As a result of orthologous classification of the detected proteins, the general function prediction was only investigated at the highest frequency among the examined proteins. Proteins related to chromatin structure and dynamics were detected by their higher frequency among functional classes. The next highest frequency was shown by proteins related to amino acid transport and metabolism and carbohydrate transport and metabolism. Therefore, it is assumed that proteins correlated with chromatin, amino acid, and carbohydrate metabolisms are easily induced into low molecular weight peptides, and that their peptides could play a role as bioactive substances.

Effects on Control of Pine Wilt Disease (Bursaphelenchus xylophilus) by Thinning Methods in Red Pine(Pinus densiflora) Forest (소나무림 숲가꾸기 종류가 소나무재선충병의 제어에 미치는 영향)

  • This study was conducted to investigate the effect on pine wilt disease by health-thinning, thinning and sapling tending in red pine forest (Pinus densiflora). As a part of developing forestry control methods for pine wilt disease control. In case of putting in pine sawyer (Monochamus alternatus) with pine wood nematode (Bursaphelenchus xylophilus), the specimen trees in health-thinning, thinning and control treatment were withered more than 50%, although there were not statistically significant differences in treatments. In treatment site, thinning slashes had influenced on the spread of pine wilt disease (experiment 1). In thinning stand of sapling pine, site with thinning slashes had highest mortality (> 90%). There were approximately 10% mortality in the site of carried thinning slashes from case and the site of non-thinning with released M. alternatus (experiment 2). The larvae had not appeared in young tree stump with health-thinning and thinning at April, the current emergence year of M. alternatus, but there were larvae in sapling tree stump with thinning at May, the current emergence year of M. alternatus. In case of stands with infected young and sapling tree by pine wilt disease, there is no effects of on pine wilt disease control by health-thinning, thinning, saplings tending at April and May, the current emergence year of M. alternatus, and leaved thinning slashes had influenced on the spread of pine wilt disease as habitation of M. alternatus.

Rapid Detection for Lysinibacillus fusiformis, a Suspicious Pathogen of Bombus terrestris, using Ultra-Rapid PCR (초고속 유전자 증폭법을 이용한 서양뒤영벌 의심병원체 Lysinibacillus fusiformis의 신속 검출법)

  • Lysinibacillus fusiformis has been suspected to be a pathogen of Bombus terrestris in Korea since 2008. In this study, we developed the rapid detection method for the L. fusiformis by utilizing the Ultra-rapid PCR. After optimizing of L. fusiformis-specific Ultra-rapid PCR, it can detect the existence of $1.0{\times}10^8$ L. fusiformis-specific DNA molecules in 4 minute and 22 seconds. Even, only 10 molecules could be detected quantitatively using this method. In addition, for the first time, in our knowledge, L. fusiformis was detected using proposed method from bumblebee produced commercially in Korea. Not only in the laboratory but also in the field, L. fusiformis-specific Ultra-rapid PCR would be applied and might be expected as convenient tools at production of bumblebee or inspection for the import and export of bumblebee.

Spatial Distribution and Host Plants of the Ramulus koreanus (Phasmida; Phasmatidae) in Korean Thistle Cultivation (고려엉겅퀴 재배지에서 발생한 우리대벌레 공간분포 및 기주식물)

  • The Korean thistle (Cirsium setidens) is grown in Gangwon province as a food ingredient called gondre. Based on regular monitoring of Korean thistle cultivation, we detected local outbreaks of a stick insect causing severe damage. Here we report the identification of the stick insect and its ecological characteristics. The survey involved 3 farms located in Jeongeon-gun, Gangwon province and lasted from May 28 to October 1 in 2019. According to morphological and DNA analyses the stick insect was identified as Ramulus koreanus Kwon Ha and Lee. a member of the family Phasmatidae. Its outbreak in the Korean thistle farms was observed from June 11 to August 22 with peak density on July 23. Spatial distribution analyses, using Taylors power law and Greens index showed that R. koreanus was uniformally distributed. Food consumption rate of Korean thistle leaves was estimated as 60.98±4.35 ㎠/day/adult. The main food plants were identified as Juglans mandshurica and Aronia melanocarpa. Further study needs to pursue the mechanism of the outbreak and its possible impact on agricultural and forest ecosystems. Additionally, means to relieve crop damage need to be identified.

Attractiveness of Host Plant Volatiles and Sex Pheromone to the Blueberry Gall Midge (Dasineura oxycoccana) (블루베리혹파리에 대한 기주식물 휘발성 물질과 성페로몬의 유인 효과)

  • The blueberry gall midge, Dasineura oxycoccana (Johnson) (Diptera: Cecidomyiidae), is an emerging pest on cultivated blueberries in Korea. To develop a sensitive tool for monitoring this pest in blueberry orchards, we compared the attractiveness of host plant volatiles and sex pheromone to D. oxycoccana adults. We performed gas chromatography-mass spectrometry (GC-MS) analysis of solid-phase microextraction (SPME)-collected volatiles that were released from blueberry ('Darrow' cultivar). The analysis revealed two major volatiles, cinnamaldehyde and cinnamyl alcohol from flowers; and three major volatiles, β-caryophyllene, germacrene D, and α-farnesene from shoots and young fruits. In field tests conducted in Gunsan, Korea in 2019, commercialized cinnamaldehyde, cinnamyl alcohol, β-caryophyllene, and α-farnesene, used singly or in quaternary combination, were unattractive to the blueberry gall midge. However, traps baited with the known sex pheromone (2R,14R)-2,14-diacetoxyheptadecane attracted significantly more males than the treatments with plant volatiles or the control. No synergistic effect was observed between sex pheromone and plant volatiles. Male D. oxycoccana were captured in the pheromone traps from May to August, with three peaks in mid-May, late June, and late July in Gunsan blueberry fields in 2020.
