Evaluation of Antifungal Activity of Natural Antimicrobials in Fuctional Beverage (기능성 음료의 곰팡이 제어를 위한 천연항균제 활성 평가)

  • Antifungal activity of seven natural antimicrobials, such as $\varepsilon$-polylysine, extract of Yucca shidigera, TLS (vitamin B1 derivative), BMB-FS, chitooligosaccharide, KDSP 001, and KDSP 002 were investigated for their applications in functional beverage. Five fungi including Aspergillus niger, Penicillium citrinum, Rhizopus oryzae, Fusarium moniliforme, and Mucor rouxii were applied as test fungi and mininum inhibitory concentrations (MICs) of antimicrobials were examined. TLS exhibited the strongst antifungal activity among tested antimicrobials and the growth of all fungi was inhibited at 100 ppm. The antifungal activity of BMB-FS appeared different. The growth of Fusarium moniliforme was inhibited by BMB-FS at 100 ppm, but it exhibit antifungal activity on P. citrinum, and M. rouxii at 1000ppm. MICs of TLS appeared to be 60 ppm for four test fungi except A. niger (100 ppm). These results indicate the possible usage of TLS and BMB-FS as natural antimicrobials in functional beverage.

EFFECTS OF SUBINHIBITORY CONCENTRATIONS OF ANTIMICROBIAL AGENTS ON CELL SURFACE PROPERTIES AND VIRULENCE FACTORS OF MUTANS STREPTOCOCCI (아저해농도(亞沮害濃度)의 항균물질이 mutans streptococci의 세포표면성질과 독력인자에 미치는 영향)

  • Subinhibitory concentrations (sub-MICs) refer to concentrations below minimum inhibitory concentrations (MICs). The antimicrobial agents may be present at relatively high concentration, at least higher than bacterial MIC and thereafter be deserted off a surface and function at sub-MICs, perhaps by interfering with bacterial metabolism. Consequently, the aim of this study was to determine the effects of growth, in the presence of sub-MICs of antimicrobial agents, on the cell surface properties and virulence factors of mutans streptococci and to investigate the efficacy of a chemical approach in vitro. Streptococcus mutans Ingbritt and Streptococcus sobrinus 6715-7 were used. Eight antimicrobial agents (Sanguinaria extract;SG, Chlorhexidine digluconate;CHX, Fluoride;F, Propolis;PP, Hydrogen peroxide;HP, Triclosan;TC, Sodium dodecyl sulfate;SDS Cetylpyridinium chloride; CC) were diluted serially in broth to determine MICs and to compare the growth rate, acid production, hydrophobicity, adhesion activity to saliva coated hydroxyapatite, glucan synthesis and cellular aggregation of experiment groups (in the presence of sub-MICs) with those of control (in the absence of antimicrobial agents). Sub-MICs of antimicrobial agents affected the growth of cells, hydrophobicity, and adhesion of bacteria to saliva coated hydroxyapatite and glucan synthesis. They also resulted in a significant reduction in pH after 12 hours (p<0.05). By cells pretreated with proteinase K, either the aggregation induced by antimicrobial agents was completely inhibited or the aggregation titers were markedly increased. According to the results of the present study, each antimicrobial agent at sub-MICs could affect similar as its known action mechanism and could continually inhibit cariogenic bacteria at such concentrations. Thus, the use of these antimicrobial agents would be one of the effective methods to prevent dental caries.

  • Pseudomonas aeruginosa and Staphylococcus aureus are the two most frequently encountered pathogens responsible for chronic wound infections, often coexisting in such cases. These infections exhibit heightened virulence compared to single infections, leading to unfavorable patient outcomes. The interaction among microorganisms within polymicrobial infections has been shown to exacerbate disease progression. Polymicrobial infections, prevalent in various contexts such as the respiratory tract, wounds, and diabetic foot, typically involve diverse microorganisms, with Pseudomonas aeruginosa and Staphylococcus aureus being the most commonly identified pathogens. This study aimed to compare the growth patterns of bacteria under a concentration gradient of toxic chemicals, focusing on a Gram-negative strain of Pseudomonas aeruginosa and a Gram-positive strain of Staphylococcus aureus. The minimum inhibitory concentration (MIC), which signifies the concentration at which bacterial growth is inhibited, was determined by performing broth microdilution and assessing the bacteria's growth curves. The growth curves of both Pseudomonas aeruginosa and Staphylococcus aureus were confirmed, and the exponential growth phases were applied to calculate the doubling times of bacteria. The MIC value for each toxic chemical was determined through broth microdilution. These results allowed for the identification of disparities in growth rates between Gram-positive and Gram-negative bacteria, as well as differences in resistance to individual toxic substances. We expect that this approach has a strong potential for further development towards the innovative treatment of bacteria-associated infections.

Determination of Antibacterial Activity from Tricholoma matsutake Extract and Its Application to Low Salted Jeot-gal (송이버섯 추출액의 항균력 확인과 저염젓갈 생산에 적용 가능성)

  • The antibacterial activity from Tricholoma matsutake extracts were confirmed by minimum inhibitory concentration (MIC) test against various bacteria. Tricholoma matsutake extracts was applied for manufacturing low salted Jeot-gal with sodium DL-malate and sodium nitrite as food preservatives. Due to antibacterial activity from Tricholoma matsutake extracts, MIC of sodium DL-malate and sodium nitrite were significantly reduced to 0.025 g/L and 0.25 g/L, respectively. As a result of this study, a premium low salted Jeot-gal can be developed with low concentration of food preservatives by adding Tricholoma matsutake extracts.

  • This study was performed to examine the cytotoxicity and antimicrobial activity of synthetic or natural preservative. Fibroblast cells L929 were used for cytotoxicity test and Pseudomonas aeruginosa ATCC27853, Staphylococcus aureus ATCC25923, Escherichia coli ATCC25922, Trichoderma reesei ATCC6967 were used for antibacterial and antifungal activities. Benzalkonium chloride(BAC) as a synthetic preservative and chitosan as a natural preservative were used for this study. Minimum inhibitory concentration(MIC) of BAC was 0.1~0.01% for P. aeruginosa and 0.001~0.0001% for S. aureus and 0.1~0.01% for T. reesei, MIC of chitosan was 2% for P. aeruginosa and 1I % for S. aureus. This study suggest that chitosan might be useful as an eyedrop.

  • A sediment exposure experiment was conducted to investigate the influence of acid volatile sulfides (AVS) and simultaneously extracted metals (SEM) in sediments on the bioavailability and toxicity of Cd, Ni and Zn to a marine polychaetes Neanthes arenaceodentata. The test animals were exposed to contaminated sediments spiked by metal mixtures of Cd, Ni, Zn (0.5~15 $\mu$mol/g of total SEM) in low (~1 $\mu$mol/g), medium (~5 $\mu$mol/g) and high AVS series (~10 $\mu$mol/g) to determine bioaccumulation, mortality and individual growth rate in each treatment after 20 days. Cd and Zn bioaccumulation in test animals increased with increasing of overlying water (OW) concentration controlled by AVS. In contrast, Ni bioaccumulation increased with increase of SEM concentration. Mortalities and growth inhibitions of N. arenaceodentata observed in only treatments with [SEM-AVS]>0, due to a high level of OW-Zn. With regard to the mortality, the 20-d LC5O value fur OW-Zn was 9.3(8.0$\pm$11.0) $\mu$M. The LOEC (Lowest Observed Effect Concentration) for Tissue-Zn was 7.8 $\mu$mol/g and the NOEC (No Observed Effect Concentration) was 6.2 $\mu$mol/g. Regarding the inhibition of individual growth rate, the LOEC fer Tissue-Zn was 5.9 $\mu$mol/g, and NOEC was 5.1 $\mu$mol/g. In this study, the toxicity of dissolved metals, especially for Zn, was overemphasized due to the reduced distribution coefficients (K$\_$d/s) of metals in the experimental sediments.

  • The effect of combinations of 8 antimicrobial agents on the growth of Streptococcus mutans and Streptococcus sobrinus was investigated with the minimum inhibitory concentration and fractional inhibitory concentration(FIC) and ${\Sigma}FIC$ index. According to the ${\Sigma}FIC$ values by. The American Society for Microbiology and Berenbaum, Approximately 34% and 82% of the combinations were synergistic respectively. Partial synergy described by Isenberg was also observed in the half of the combinations. There was a tendency for additive antimicrobial effect against cariogenic bacteria though the test results showed difference according to the applied values. It may be beneficial to use combined antimicrobial agents that have various activities against an ecosystem and metabolism of bacteria than using individual agents.

Study on the Effect of Blending Ratios on the Antibacterial Activities of Chitosan/Gelatin Blend Solutions (혼합비율에 따른 키토산/젤라틴 혼합용액의 항균활성에 관한 연구)

  • Chitosan, second largest biomass after cellulose on earth, has potential for use as functional food package due to its antibacterial activity. However, due to high melting temperature of chitosan, chitosan films have been made by casting method. Because gelatin has relatively low molting temperature depending upon amount of plasticizer added, it was added to chitosan to produce commercially feasible film. The objective of the current study was to determine optimum blend ratio and amount of chitosan/gelatin blend solutions against antibacterial activities for extruder resin. Gram-positive bacteria (Bacillus cereus ATCC 14579 and Listeria monocytogenes ATCC 15313) and -negative bacteria (Escherichia coli ATCC 25922 and Salmonella enteritidis IFO 3313) were used. Paper (8 mm) diffusion and optical density methods were used to evaluate effect of different blending ratio solutions on the inhibition of bacterial growth. Measured clear none size ranged from 8 mm to 18.07 mm in paper diffusion test. For B. cereus, E. coli, and S. enteritidis, addition of $50\;{\mu}L$ blend solution (chitosan/gelatin = 2/8: 0.3 mg) resulted in clear zone on paper disc. In L. monocytogenes, inhibition effect was observed with 0.6 mg chitosan (chitosan/gelatin=4/6). Minimum inhibitory concentration (MIC) values of B. cerues, L. monocytogenes, E. coli, and S. enteritidis with addition of chitosan were 0.1461, 0.2419, 0.0980, and 0.0490 mg/mL, respectively, These results indicate possibility of producing commercially feasible film with addition of optimum chitosan/gelatin amount.

  • 본 연구는 Se과 Vit. E 투여수준이 한우 종빈우 번식효율에 미치는 영향을 규명하기 위하여, 한우 종빈우의 임신말기에 적정량의 Se와 Vit .E를 투여하여, 종빈우의 혈액화학치 변화 및 혈중 Vit. E 농도변화를 규명하였다. 1. 분만 1개월 전 Se 과 Vit. E 투여가 한우 종빈우의 혈액성분에 미치는 영향을 검토한 결과, 평균 혈중 albumin 과 calcium 함량은 각각 투여구가 대조구보다 유의적으로 높았으며 (P<0.05), 평균 혈중 IP 함량은 각각 Se 과 Vit. E 혼합투여구가 여타구보다 높은 성적을 나타냈다 (P<0.05). 그러나 평균 혈중 BUN, creatinine, glucose, cholesterol 및 triglycerides 의 함량은 처리구간 유의적인 차이가 인정되지 않았다 (P>0.05). 2. 분만 2개월 전 Se과 Vit. E 투여가 한우 종빈우의 혈액성분에 미치는 영향을 검토한 결과, 혈중 평균 albumin, BUN, cholesterol, creatinine, glucose, IP, TP, triglycerides 및 Ca 함량은 처리구간 유의적인 차이는 인정되지 않았다 (P>0.05). 3. 한우 종빈우의 분만 l 개월전 Se 및 Vit. E 투여가 한우 종빈우의 혈중 Vit. E 농도에 미치는 영향을 검토한 결과, 평균 혈중 Vit. E 농도는 투여구가 대조구보다 다소 높은 농도를 나타냈으나, 처리구간 유의적인 차이는 인정되지 않았다 (P>0.05). 4. 한우 종빈우에서 분만 2 개월전 Se 및 Vit. E 투여가 혈중 Vit. E 농도에 미치는 영향을 검토한 결과, 평균 혈중 Vit. Antimicrobial Activities of Medicinal Herb Extracts (한약재추출물의 항균활성)

  • In this study, 18 kinds of Korean medicinal herb extracts were examined for anti-microbial activities against pathogenic microorganisms. The methanol (MeOH) extracts from Schizandra chinensis, Rhus javanica and Caesalpinia sappan exhibited antimicrobial activities against most pathogenic microorganisms at concentrations of 5 mg/mL, whereas the other 15 extracts exhibited anti-microbial activities at concentrations of 30 mg/mL. The minimum concentration at which Schizandra chinensis extracts inhibited for S. epidermidis and Bor. bronchiseptica was 0.6 mg/mL. The MeOH extracts from Schizandra chinensis, Caesalpinia sappan, Rhus javanica and Seutellaria baicalensis which had higher anti-microbial activities were subsequently fractionated using 5 different solvents, and further screened for anti-microbial activities. The inhibitory effects of ethyl acetate (EtOAc) extracts on microbial growth were greater compared to any other solvent extracts. In order to investigate the inhibitory effect of Korean medicinal herbs with high anti-microbial activities on microbial proliferation, the MeOH extracts at concentrations of 0, 100, 300 and 500 ppm were added to the media. No addition of extracts caused rapid growth of microbes after 12 hours incubation. As the concentration of extracts from Rhus javanica and Caesalpinia sappan increased, the growth-inhibiting effect on gram-positive bacteria including S. aureus, S. epidermidis, and L. monocytogenes was prominent. Rhus javanica extracts exhibited growth-inhibiting activity for gram-negative bacteria including Sal. Pullorum and Sal. Choleraesuis. The low concentration of extracts from Rhus javanica and Caesalpinia sappan exhibited the growth of Bor. bronchiseptica and E. coli serotype $O_8$. However, the higher concentration of extracts from Rhus javanica and Caesalpinia sappan exhibited a strong inhibitory effect on microbial proliferation.
