Essential Oils of Thymus quinquecostatus Celakov. and Thymus magnus Nakai (백리향(百里香)과 섬백리향(百里香)의 정유성분(精油成分) 조성(組成))

  • The essential oils of Thymus quinquecostatus Celakov. and T. magnus Nakai, respectively, were isolated by using a modified Likens-Nickerson type steam distillation and extraction apparatus and analyzed by gas chromatography and gas chromatography-mass spectrometry. The oil content of T. quinquecostatus was 1.94%, and that of T. magnus was 1.91% in mixed leaves and stems and 0.68% in flowers. Among 38 components identified in either mixed leaves and stems or flowers the major components in essential oil isolated from T. quinquecostatus were thymol(39.8%), ${\gamma}-terpinene(10.0%)$ ${\rho}cymene(9.2%)$ and camphor(5.9%) while those from mixed leaves and stems of T. magnus were thymoI(54.7%), ${\gamma}-terpinene(15.8%)$, ${\rho}cymene(6.7%)$ and carvacroI(3.2%). The contents of ${\alpha}-pinene$, camphene, camphor, bornyl acetate and ${\alpha}-terpinene+borneol$ were higher in T. quinquecostatus than in T. maglnus but ${\gamma}-terpinene$ and thymol were higher in T. magnus than in T. quinquecostatus. Comparing leaves and stems with flowers in T. magnus, peak area percentage(%) of ${\gamma}-terpinene$, ${\alpha}-terpinene$ were higher in mixed leaves and stems than in flowers, whereas ${\rho}cymene$ was predominantly higher in flowers than in leaves and stems.

Lectotypification and identity of Thymus quinquecostatus var. magnus(Nakai) Kitam. (Labiatae) (섬백리향(꿀풀과)에 대한 선정기준표본 설정과 분류학적 검토)

  • Thymus quinquecostatus var. magnus (Nakai) Kitam., an endemic taxon in Ulleung Island, is lectotypified. The lectotype is designated based on careful examination of the original materials (deposited in TI) cited in the protologue of the variety. In addition, taxonomic identity of T. quinquecostatus var. japonicus H. Hara, a problematic name often mistakenly referred to the above taxon, was throughly reexamined.

Hyaluronidase Inhibitor 개발에 관한 연구

  • 총 40종 생약의 hyaluronidase에 대한 저해 효과를 검색한 결과, 백지, 오수유, 향부자, 섬백리향, 배풍등, 황금, 황련, 진피, 소리쟁이, 반디나물 등 10종 생약에서 저해 활성을 관찰할 수 있었다. 또한 권백의 n-BuOH 분획에서 2개의 biflavonoid 성분인 amentoflavone, isocryptomerin을 분리하였으며, chloroform 분획에서 cryptomerin B를 분리하였다. 이들 화합물의 화학구조는 각종 기기분석 data를 이용하여 결정하였으며, 이들 화합물중 cryptomerin B는 Selaginellaceae에서 처음으로 분리 보고되는 화합물이다. 현재 이들 화합물의 생리 활성에 대하여 검사중이다.

Anti-nociceptive, Anti-inflammatory, Mental Effects of Essential Oil from Thymus magnus (섬백리향 정유의 진통, 항염증, 정신적인 작용)

  • Thymus magnus is an endemic (Ulleung Island) species in Korea. This plant is used as diaphoretics and carminatives in traditional medicine. In the literature, few scientific assays were realized on this species, such as antibiotic (Streptococcus pneumoniae, Staphylococcus aureus, Salmonella enteritidis, and S. typhimurium) and antifungal activities. In order to clarify whether essential oil of T. magnus have pharmacological effects, anti-inflammatory, sedative, anti-depressant, analgesic, and sleep-prolonged effects were investigated using animal models. From this study, the following conclusions were attained; 1) Essential oil of T. magnus did not show any acute toxicity on mice when orally administered at the dose of 2-3 g/kg body weight. 2) Essential oil of T. magnus possessed strong anti-inflammatory activity, similar to that of a positive control prednisolone. 3) Essential oil of T. magnus had excellent analgesic activity, comparable to that of aspirin. 4) The essential oil of T. magnus possessed strong sleep-prolonged effect on pentobarbital induced-sleep test in mice model. 5) In the hot plate test, the essential oil of T. magnus had moderate effect. 6) And the essential oil of T. magnus had no significant effects in forced-swimming test and open-field test.

Anti-Inflammatory Effect of the Extracts from Leaves and Stems of Thymus quinquecistatus var. japonica (H.Hara) (섬백리향 잎과 줄기 추출물의 항염 활성에 관한 세포생물학적 연구)

  • Objectives : Thymus quinquecistatus var. japonica (H.Hara) is a member of the genus Thymus of perennial aromatic herb, and it's designated as a natural monument of South Korea. It has traditionally been known to have protective or therapeutic effects on various human disease including cerebrovascular and neurological disease. Recently it was suggested that essential oil extracted from thyme has anti-fungal and anti-bacterial effect. The aim of this study is to investigate anti-inflammatory effect of Thymus quinquecistatus var. japonica in Raw 264.7 macrophage cell line. Methods : The cytotoxic effects of water and 70% ethanol extracts from Thymus quinquecistatus var. japonica, was tested using MTT assay. Inhibitory effects of the extracts to nitric oxide production and mRNA expression of inflammatory cytokines were examined by RT-PCR. Also, MitoSOX-red assay and JC-1 assay were performed to determine if the extracts can inhibit mitochondrial ROS accumulation and maintain mitochondrial membrane potential. Results : In LPS-induced inflammatory response, the extracts efficiently reduced nitric oxide NO production through inhibiting mRNA expression of iNOS enzyme. In addition, expression of the proinflammatory cytokines, IL-1𝛽 and IL-6, was also down-regulated by the extract treatments. Excessive accumulation of mitochondrial ROS induced by LPS was inhibited in the extract treated cells, which finally protected mitochondrial membrane potential. Conclusions : These results showed that water and 70% ethanol extracts from Thymus quinquecistatus var. japonica have anti-inflammatory effect through down regulation of IL-1𝛽, IL-6, and iNOS, and also have antioxidative effect against mitochondrial ROS accumulation that promote inflammatory response.

Comparison of Volatile Compounds from Thymus Magnus Nakai by Three Different Extraction Methods (추출방법에 따른 섬백리향의 휘발성 향기성분 비교)

  • The purpose of this study was to analyse the volatile components of Thymus magnus Nakai extracted by different extraction methods and reproduce scent close to original plant based on the results. For this purpose, the essential oil of T. magnus was extracted by supercritical fluid extraction (SFE), water and steam distillation (WSD) and simultaneous steam distillation and extraction (SDE) methods. The compositions of the essential oil were analyzed by gas chromatography-mass spectrometry (GC-MS). Consequently, linalool (0.1%) and trans-sabinene hydrate (0.9%) contents in the essential oil extracted by SFE method of $40^{\circ}C$ - 400 bar condition were relatively higher than compositions of the essential oil extracted by different conditions. The contents of borneol (3.82%), terpinen-4-ol (0.3%) and caryophyllene oxide (2.2%) were relatively higher at $50^{\circ}C$ - 400 bar and the contents of ${\beta}$-bisabolene (5.88%), 1-octen-3-ol (0.31%), caryophyllene (2.91%), p-cymene (2.04%) and ${\gamma}$-terpinene (0.52%) were extracted relatively higher at $50^{\circ}C$ - 300 bar. The compositions of the essential oil extracted by SFE method of $50^{\circ}C$ - 200 bar condition contained relatively higher contents of thymol (77.63%) and carvacrol (5.65%). The contents of ${\alpha}$-bisabolol (0.17%), caryophyllene (6.46%), cis-${\alpha}$-bisabolene (1.52%) and ${\beta}$-bisabolene (20.65%) in the essential oil extracted by WSD method were relatively higher than compositions of the essential oil extracted by SFE method, and by SDE method we couldn't obtained essential oil. The results of this study could be utilized to reproduce scent close to original scent of T. magnus.

Covering Types and Covering Ratio Changes of Planted Species on an Extensive Green Roof (관리조방형 옥상녹화 식재식물의 피복률 변화 및 피복유형)

  • The purpose of this study was to analyze the characteristics of plant covering in the extensive green roof site. Eighteen herbaceous perennial grass such as Caryopteris incara and sedum species such as Sedum Kamtschaticum were planted on an experimental green roof with 10cm substrate depth in 2007. This study investigated vegetation change over 3 growing seasons 2007-2009. The covering rates of planted species mostly increased in 2008, but declined in 2009 except 6 species such as sedum specis and Thymus quinquecostatus var. japonica etc. There were four categories of covering characteristics generated from the results of this study. These include the type of sustain planting site, the type of encroach adjacent site, the type of creeps and spreads, and the type of scatters and spreads. The covering models of eight planted species were drawn by simple regression analysis. However more monitoring of various plants will be needed to establish the information for sustainable roof planting plan.

Use of Auxins for Rhizome Propagation and Shading Effect on Growth of Asarum sieboldii Mio. (세신(細辛)의 근경삽(根莖揷)번식에 있어서 오옥신의 이용과 차광재배(遮光栽培))

  • This study was carried out to investigate the auxin(IAA, IBA, NAA) treatment and the effect of shading rate in Asarum sieboldii. The results obtained were summerized as follows: By the soaking treatment of auxins to the cutted rhizome enhanced root growth and plant weight. By the increment of shading rate, plant growth was much better compare to the control. Leaf fallen times appeared about 20 days more earlier at plain area then the alpain area. Root yield was much higher by the treatment of shading then the conventional cultivation so it seem to be the useful for large scale cultivations of A. sieboldii.
