Exploration and functional expression of homologous lipases of Candida antarctica lipase B (Candida antarctica lipase B의 상동체 효소 탐색과 발현)

  • Candida (also known as Pseudozyma) antarctica lipase B (CAL-B) has been intensely studied in academic and industrial fields. However, the research related to its homologous enzymes has been rarely reported. In the current investigation, protein sequence similarity search of CAL-B has been conducted and six homologous protein sequences were identified. After the syntheses of their codon-optimized genes, the synthetic genes have been cloned into a periplasmic expression vector to express in Escherichia coli. Among six homologous sequences, four sequences were successfully expressed in E. coli. The hydrolytic activities of the expressed proteins towards 4-nitrophenyl acetate and 4-nitrophenyl butyrate were measured and compared with those of CAL-B to identify whether the expressed proteins work as a hydrolase. It has been revealed that the expressed proteins can hydrolyze the substrates and the specific activities were determined as $(1.3-30){\times}10^{-2}{\mu}mol/min/mg$, which are lower than those of CAL-B. Among these homologous enzymes, Pseudozyma hubeiensis SY62 exhibits the comparable enantioselectivity to that of CAL-B towards the hydrolysis of (${\pm}$)-1-phenylethyl acetate.

Cloning and Molecular Characterization of Epoxide Hydrolase from Aspergillus niger LK (Apergillus niger LK 유래의 Epoxide Hydrolase 클로닝 및 특성 분석)

  • Aspergillus niger LK harboring the enantioselective epoxide hydrolase (EHase) activity was isolated, and enantioselectivity of EHase was tested for various racemic aromatic epoxides. The gene encoding epoxide hydrolase was cloned from cDNA library generated by reverse transcriptase-polymerase chain reaction of the isolated total mRNA. Sequence analysis showed that the cloned gene encodes 398 amino acids with a deduced molecular mass of 44.5 kDa. Database comparison of the amino acid sequence reveals that it is similar to fungal EHase, whereas the sequence identity with bacterial EHase is very low. Recombinant expression of the cloned EHase in Escherichia coli BL21 yielded an active EHases, which can offer a potential biocatalyst for the production of chiral epoxides.

  • Research, such as developing alternative energy in the transportation field, including aviation, is being actively conducted to solve the issue of current climate change. Interest in ammonia fuel as a carbon free energy (CFE) source is increasing due to the ease of liquefaction and transportation and similarity in energy density to that of methanol. However, explosiveness and toxicity of ammonia make it difficult to handle. Therefore, in this study, stable ammonia production was attempted using relatively easy-to-handle urea water solution (UWS). High temperature steam was used to promote the hydrolysis of ammonia. In order to determine the causes for ammonia production below the theoretical equivalent ratio, it was suggested that there were not enough collisions to promote the hydrolysis based on the kinetic theory of gases. The hydrolysis of unreacted isocyanic acid (HNCO) was tested according to the change in water supply. As a result, an increased amount of ammonia produced was confirmed. The increased amount of ammonia produced in a certain section was dependent on the steam temperature and the flow rate of water supplied.

Oxalate Chelating Activity of Egg White Proteins and Their Hydrolysates

  • Major egg white proteins and their hydrolysates serve as functional food ingredients that have certain metal-chelating properties. Employing egg white proteins and their hydrolysates to scavenge dietary oxalates is anticipated to have beneficial effect in the prevention of kidney stones. The objective of this study was to determine the biogenic oxalate-chelating activity of ovalbumin, ovomucin, and ovotransferrin and their hydrolysates. To prepare oxalate extracts, 30 mL of 0.25 N HCl was added to separately to 0.5 g of dried spinach and starfruit powders followed by boiling for 15 min, and after cooling, the addition of a further 20 mL of 0.25 N HCl. Having prepared these extracts, ovalbumin, ovomucin, and ovotransferrin and their hydrolysates were separately mixed with oxalate extracts and incubated at 3℃ for 24 h. Following centrifugation, supernatants were analyzed by HPLC using a reverse-phase C18 column coupled with a diode array detector. We found that all assessed proteins and their hydrolysates showed biogenic oxalate-chelating activity against the oxalates of spinach. In contrast, however, only ovalbumin, ovalbumin-hydrolysate, and ovomucin showed chelating activity (57.10%±8.84%, 85.44%±5.30%, 73.20%±4.13%, respectively) against the oxalates of starfruit (P<0.05). Overall, hydrolyzed ovalbumin was identified as the most effective chelator of the oxalates both spinach and starfruit. In this study, we thus established that the assessed egg white proteins and their hydrolysates have oxalate-chelating activity in vitro, thereby indicating that these compounds have potential utility as nutraceuticals for the chelation of dietary oxalate. However, further research will be necessary to verify their oxalate-chelating activities against different fruits and vegetables and under specific in vivo conditions and against purified oxalate.

  • The epoxide hydrolase (EH) of Aspergillus niger LK was expressed to high levels in Escherichia coli based on codon usage. E. coli, Rosetta (DE3)PLysS, containing a large number of tRNAs for rare-codons was employed as a host strain. The recombinant E. coli expressing A. niger EH showed an enhanced enantioselective hydrolysis activity toward racemic styrene oxide. Enantiopure (S)-styrene oxide with a high enantiopurity of 99% ee was obtained from racemic substrates.

Hydrolysis of Isolate Soybean Protein Using Subcritical Water (아임계수를 이용한 분리대두단백질의 가수분해)

  • Hydrolysis of isolate soybean protein (ISP) using subcritical water (SCW) was conducted to study the feasibility for producing protein hydrolyzate. SCW hydrolysis of SPI suspension (5-15%) was conducted in an electrically heated batch reactor (2 L). The effects of temperature (230 to $270^{\circ}C$) and holding time (10 to 50 min) on the degree of hydrolysis (DH) and the production of amino acids were studied by surface response method. The DH was determined by derivatizing the hydrolyzates with ortho-phthalaldehyde (OPA) solution. It was confirmed that reaction temperature and holding time affected the hydrothermolysis of soybean protein. However, the holding time was less effective on amino acid yield when the temperature was higher than $230^{\circ}C$. In order to achieve optimal yields of amino acids exceeding 43%, the temperature should be within the range between 256 and $268^{\circ}C$ with holding time from 29 to 41 min, respectively. A maximum estimated amino acid yield of 43.5% was obtained at $268^{\circ}C$ for 35 min.

  • Epoxide hydrolase (EH) gene of Caulobacter crescentus was cloned by PCR and expressed in Escherichia coli. The C. crescentus EH (CcEH) primarily attacked at the benzylic carbon of (S)-styrene oxide, while the CcEH preferentially attacked at the terminal carbon of (R)-styrene oxide, thus leading to the formation of (R)-phenyl-1,2-ethanediol as the main product. (R)-phenyl-1,2-ethanediol was obtained with 85% enantiomeric excess and yield of 69% from racemic styrene oxide via enantioconvergent hydrolysis by using recombinant CcEH as the single biocatalyst.

  • The selective removal and recovery of fermentation inhibitors during purification of sugars from biomass hydrolysates can increase the economic efficiency of the entire process to produce bioalcohol from lignocellulosic biomass. This study investigated the effect of furans in phenols-free biomass hydrolysate on acetic acid extraction in an emulsion liquid membrane system. Under specific operating conditions, more than 99% of acetic acid could be extracted within 5 minutes, and the degrees of extraction of furfural and 5-hydroxymethylfurfural were about 10% and 4%, respectively. The extraction rate of acetic acid was also lower at a higher initial concentration of furfural in the feed phase, which was greater for furfural than 5-hydroxymethylfurfural. Thus, if furfural is first removed from the hydrolysate prior to acetic acid extraction, emulsion liquid membrane would be a more economically efficient way of removing acetic acid.

  • The bacterium Serratia marcescens QM Bl466 produces selectively large amount of chitinolytic enzymes(about 1mg/L medium). Enzymatic hydrolysis of chitin to N-acelyl-${\beta}$-D-glucosamine(NAG) is performed by a system consisting of two hydrolases : chitinase and chilobiase. Objectives of this study included optimization of a microbial host by using chitin particles for chitinase/chitobiase production and secretion and also development of batch fermentation system for high cell density cultivalion of S. marcescens QM B1466. Also, the influence of chitin source and carboxymethyl(CM) chitin on chitinase/chitobiase production and NAG production was investigated. When carboxymethyl chitin was substituted for colloidal and practical grade chitin, the chitinase activity was increased about 7∼10U/mL. In this case, the ratio of chitinase/chitobiase was 30.03U/3.44U(9:1). The highest amounts of NAG(3.0g/L) was obtained.

Preparation of CuO-CeO2 mixed oxide catalyst by sol-gel method and its application to preferential oxidation of CO (졸-겔법에 의한 CuO-CeO2 복합 산화물 촉매의 제조 및 CO의 선택적 산화반응에 응용)

  • For the preferential oxidation of CO contained in the fuel of polymer electrolyte membrane fuel cell (PEMFC), CuO-$CeO_2$ mixed oxide catalysts were prepared by the sol-gel and co-precipitation methods to replace noble metal catalysts. In the catalyst preparation by the sol-gel method, Cu/Ce ratio and hydrolysis ratio were changed. The catalytic activity of the prepared catalysts was compared with the catalytic activity of the noble metal catalyst($Pt/{\gamma}-Al_2O_3$). Among the catalysts prepared with different Cu/Ce ratios, the catalyst whose Cu/Ce ratio was 4:16 showed the highest CO conversion (90%) and selectivity (60%) at $150^{\circ}C$. As the hydrolysis ratio was increased in the catalyst preparation, surface area increased, and catalytic activity also increased. The highest CO conversions with the CuO-$CeO_2$ mixed oxide catalyst prepared by the co-precipitation method and the noble metal catalyst (1wt% $Pt/{\gamma}-Al_2O_3$) were 82 and 81% at $150^{\circ}C$, respectively, whereas the highest CO conversion with the CuO-$CeO_2$ mixed oxide catalyst prepared by the sol-gel method was 90% at the same temperature. This indicates that the catalyst prepared by the sol-gel method shows higher catalytic activity than the catalysts prepared by the co-precipitation method and the noble metal catalyst. From the CO-TPD experiment, it was found that the catalyst having CO desorption peak at a lower temperature ($140^{\circ}C$) revealed higher catalytic activity.
