생약재 추출물의 수율 및 항고혈압 활성

  • 본 실험에 사용한 생약재는 금산 약초시장에서 2003년 6월에 구입하여 추출수율을 조사하였다. 그 결과, 생약재의 물 추출물의 동결건조 중량(%)이 30~40%에 달하는 생약재는 가자, 갈근, 맥문동, 산수유가 있고, 20~30%에 달하는 생약재는 감초, 구기자, 당귀, 오미자, 천궁, 황기가 이에 속했으며, 중량(%)이 10~20%에 달하는 생약재는 복분자, 오수유, 애엽, 작약, 진피, 지유, 파고지, 황련이 이에 속했다. 마지막으로 10%이하의 추출 수율을 나타낸 생약재들은 가시오가피, 강황, 계피, 석곡, 소목, 영지, 오약, 육두구, 정향, 죽엽, 필발, 초두구, 호장근이 이에 속했다. 생약재의 70% Ethanol 추출물의 동결건조중량을 살펴본 결과, 동결건조 중량(%)이 30~40%에 달하는 생약재는 가자, 당귀, 산수유가 이에 속하였고, 20~30%에 달하는 생약재는 감초, 맥문동, 구기자, 오미자, 오수유, 천궁, 황기가 속하였으며, 동결건조 중량(%)이 10~20%에 달하는 생약재는 복분자, 육두구, 애엽, 작약, 정향, 지유, 파고지, 호장근, 황련이 이에 속했다. 또한 10% 이하의 수율을 나타내는 생약재는 가시오가피, 강황, 갈근, 계피, 석곡, 소목, 영지, 오약, 익지인, 죽엽, 필발, 초두구가 이에 속하였다. 생약재의 추출 시간 및 온도에 따른 수율을 조사한 결과, 추출 시간이 경과함에 따라서 수율이 점차 증가하는 경향을 보였으며, 가자, 갈근, 황련, 복분자, 지유, 정향, 계피, 소목 순으로 높은 수율을 얻을 수 있었다. 생약재에 여러가지 효소를 이용한 가수분해 결과, 대부분의 생약재에 대해 95$^{\circ}C$에서 2시간동안 추출한 수율과 그다지 큰 차이는 보이지 않았지만 대부분의 생약재가 Termamyl 효소를 사용하여 가수분해하였을 때 가장 높은 수율을 나타내었다. 또한 지유의 경우는 Viscozyme 효소를 처리하였을 경우 가장 좋은 수율을 나타내었다. 또한, 효소의 최적 온도로 조정한 Water bath에서 0, 30, 60, 120, 210분간 효소 반응 시켜, 420nm에서 갈변도를 시간별로 측정해본 결과, Viscozyme 효소를 처리한 생약재에서 시간이 흐름에 따라 흡광도 수치의 변화가 크게 나타났다. 32종의 생약재 물 추출물의 항고혈압 활성을 측정한 결과, 파고지, 소목, 죽엽의 항고혈압 활성이 높은 것으로 나타났으며, 파고지의 효과가 가장 우수한 것으로 나타났다. 10%에탄을 추출물의 경우에는 오미자, 오수유, 소목에서 항고혈압 활성이 높은 것으로 나타났으며, 오미자의 효과가 가장 우수한 것으로 나타났다. 파고지 물 추출물을 칼럼(Sephacryl S-300, High Resolution)으로 분획하여 3개의 Peak를 얻었으며, 2번째 Peak의 항고혈압 활성이 매우 높은 것으로 나타났다.

Restoration of endangered orchid species, Dendrobium moniliforme (L.) Sw. (Orchidaceae) in Korea (멸종위기 난과 식물 석곡의 복원)

  • A total of 13,000 individuals of Dendrobium moniliforme (L.) Sw. artificially propagated in laboratories and greenhouses were restored in their natural habitat of Bogildo Island, Wandogun, in the southern part of Korea in June of 2013. The growing conditions of the individuals were monitored for two years. The parental individuals for the restoration were obtained from a wild population in southern Korea, from which seeds were produced via artificial crossings. These seeds were germinated and cultivated in growing media and two-year-old plants were then grown in greenhouse beds. The genetic diversity among the propagated individuals was confirmed by examining DNA sequences of five regions of the chloroplast genome and the nuclear ITS region. The diversity values were as high as the average values of natural populations. All propagated individuals were transplanted into two different sites on Bogildo by research teams with local residents and national park rangers. After restoration, we counted and measured the surviving individuals, vegetative propagated stems, and growth rates in June of both 2014 and 2015. There was no human interference, and 97% of the individuals survived. The number of propagules increased by 227% in two years. In contrast, the average length of the stems decreased during the period. In addition, different survival and propagation rates were recorded depending on the host plants and the restored sites. The shaded sides of rock cliffs and the bark of Quercus salicina showed the best propagation rates, followed by the bark of Camellia japonica. A few individuals of D. moniliforme successfully flowered, pollinated, and fruited after restoration. Overall, our monitoring data over two years indicate that the restored individuals were well adapted and vigorously propagated at the restored sites. In order to prevent human disturbance of the restored sites, a CCTV monitoring system powered by a solar panel was installed after the restoration. In addition, a human surveillance system is operated by national park rangers with local residents.

Anti-oxidative Effects of Dendrobii Herba on Toxic Agent Induced Kidney Cell Injury (석곡(石斛)의 항산화 효과)

  • Objectives : This study was carried out to determine if Dendrobii Herba have protective effect against cell injury induced by various toxic agents in rat kidney slices. Water(DWe) and methanol(DMe) extracts were prepared for this experiment. Methods : Cell injury was estimated by measuring lactate dehydrogenase(LDH). Lipid peroxidation was examined by measuring malondialdehyde, a product of lipid peroxidation. Results : DMe prevented the LDH release by $CCl_4$, menadione, tert-butyl hydroperoxide and mercury treatment in vitro in kidney slices, but DWe prevented the LDH release by $CCl_4$ and mercury. DMe also prevented reduction in GSH and lipid peroxidation induced by $CCl_4$ and mercury. Conclusion : Thus, DMe may have more powerful efficacy on anti-oxidative effects when compared with DWe. And further studies have to be followed concerned with extraction of Dendrobii Herba and its change of effects.

Effects of Dendrobii herba against Intracerebral Hemorrhage in Rats (석곡(石斛)이 흰쥐의 뇌조직출혈에 미치는 영향)

  • Objects : This study was performed in order to observe the effects of water extract of Dendrobii herba on intracerebral hemorrhage(ICH), Method : ICH was induced by the stereotaxic intrastriatal injection of bacterial collagenase type IV in Sprague-Dawley rats. After the water extracts of Dendrobii herba were administrated orally once a day for 3 days, hematoma volume, percentage of brain edema, expression of iNOS and MPO were observed using immunohistochemistry. Results : Rats fed with water extracts of Dendrobii herba showed reduction of hematoma volume and percentage of brain edema compared with controls. In addition, Infiltration of myeloperoxidase (MPO) expressing neutrophil and expression of inducible nitric oxide synthatase(iNOS) were significantly reduced in rats fed with water extracts of Dendrobii herba. Conclusion : These results demonstrated that water extracts of Dendrobii herba reduced brain damage of intracerebral hemorrhage(ICH) and subsequent ICH-induced cerebral edema, and inhibited neutrophil infiltration.

Review and Reflection of Studies on Gyoojoon Lee (석곡 이규준 연구의 성찰과 모색)

  • Gyoojoon Lee is a prominent figure of Confucianism and traditional medicine representing southern region of Korea during late $19^{th}$ and early $20^{th}$ century, but studies on his achievements were just started merely a decade ago. We would provide foundation for the further multidisciplinary study on his legacy. We reviewed his chronology, written works, interactions with intellectual figures, and current studies on his works to provide foundations for more thorough research on his legacy. We found he has wide spectrum of studies in medicine, confucianism, literature, humanism and philosophy during the late Chosun dynasty, Korean empire and Japanese colonization period. He wrote twenty five books in diverse fields and was found to have interactions with fifty seven figures with idea during his lifetime which need more detailed examination. Multidisciplinary studies on Gyoojoon Lee is guaranteed for the future in depth study on his works including Buyunglon (theory of supporting Yang).

  • This study is on the philosophy and medical science of Lee Gyu Jun, who is the author of SoMunDaeYo and UiGamJungMa. Lee Gyu Jun proposed that Providence is made of Ki, and the Pure Good of Ki is the composing principle of the universe. The Ki could be Pure Good because it embraces Reason of Law inside itself, as a part of itself. This developed as a result of absorbing the ideas from Han and Tang Dynasty's' Ki-Philosophy trend, the latter half of the Joseon Dynasty's Heart Nature Doctrine centered Ri(理:reason)-Monism trend, and Sim (心:heart) centered teachings of Wang Yang Ming, etc. Later, his philosophical Ki-Monism(氣一元 GiIlWon) and SimAngKiReon(心陽氣論) appears exactly in medical science and continues on to Ki-Monism(氣一元 GiIlWon) and SimHwaReon(心火論).

Study on Suk Gock's Fortify Yang Theory (석곡 이규준의 부양이론에 관한 연구)

  • With regard of YinYang and Five Elements Theory, predominance of Yin or Yang is a key point to discuss Nourishin Ying or Reinforcing Yang. There are two major branches about studying predominance of Yin or Yang. Zhu Danxi advocated the theory that Yang was always in excess while Yin was often deficient, and thus belonged to the School for Nourishing the Yin. Zhang Jie-bin advocated that Yang is easily deficient while Yin is easily sufficient. It seems that they studied Wang Bing's 'Ministerial fire rule on all thing instead of Imperial Fire' and right Kidney Mingmen theory in Nan Jing, which are both related to the First Yang in the Book of Changes and say Mingmen fire is the host of the body. Zhu Danxi insisted that Mingmen fire was easily in excess and likely to become pathogenic fire while Water phase was easily in deficiency, so the doctors should nourish Yin and suppress Yang. Zhang Jie-bin said that Mingmen fire is vulnerable so the doctors should fortify yang and reinforce Mingmen fire. Suk Gock Master Lee thought that the Book of Changes and Nei Jing had a viewpoint of Fortify Yang as regards of some distinguished Physicians' articles about predominance of YinYang. The author's going to discribe Suk Gock's Fortify Yang Theory with viewpoints of Nei Jing and the Book of Changes.

In vitro Propagation and Protocorm-like Body Formation of Endangered Species, Dendrobium moniliforme (멸종위기 수종 석곡(Dendrobium moniliforme)의 Protocorm-like Body 형성 및 기내증식)

  • This experiment was conducted to establish the in vitro propagation of Dendrobium moniliforme through protocorm like body (PLB) induction from the culture of leaf, stem and root explants. Frequency of PLB formation from root explants was higher than those of leaf and stem explants. PLB's proliferation in MS medium including 1.0 mg/L NAA was better than 1.0 mg/L IBA. Frequency of PLB's proliferation on medium with various concentrations of BA, Kinetin, Tidiazuron (0. 1.0, 3.0 mg/L) and NAA (0, 1.0 mg/L) was tested. The maximun proliferation of PLB's was obtained on MS medium supplemented with 3.0 mg/L BA and 1.0 mg/L NAA after 6 weeks of culture. Addition of 200 mg/L activated charcoal (AC) and 30 g/L sucrose was effective on PLB proliferation for D. moniliforme.

Mass production of the seedlings of Dendrobium moniliforme using bioreactor culture (바이오리액터 배양기에 의한 석곡 유식물체 대량 증식)

  • Protocorms were newly formed from the culture of axillary buds, obtained in the seedlings of Dendrobium moniliforme in vitro. Its formation ratio was calculated to 43.7% on MS medium containing 1.0 mg/L BA. To test their survival ratio, we gradually increased the inoculation of transplant populations from single to more than three, and then found that the ratio in three populations went up as high as 95.2% rather than those of one or two. In bioreactor, explant obtained from the axillary bud grew well in lower concentration as 1/4 MS medium, while clearly grew slow in a little bit high concentration as 1/2 MS medium. We found that the explant of axillary bud, obtained from the Dendrobium moniliforme seedlings, would grow five times after culturing in a bioreactor for six weeks in 1/4 MS medium.

Phytochemical Identification of Bioactive Constituents of Dendrobii herba (생약석곡(Dendrobii herba)의 생리활성 성분 규명)

  • Two new phenanthrenes, namely, (1R,2R)-1,7-hydroxy-2,8-methoxy-2,3-dihydrophenanthrene-4(1H)-one (1) and 2,7-dihydroxyphenanthrene-1,4-dione (2), were isolated from the ethyl acetate-soluble fraction of Dendrobii Herba, together with the eleven known compounds, densiflorol B (3), 6,7-dimethoxyphenanthrene2,5-diol (4), dehydroorchinol (5), 1,5,7-trimethoxy-2-phenanthrenol (6), denthyrsinin (7), ephemerantol A (8), lusianthridin (9), moscatilin (10), gigantol (11), 3-[(1E)-2-(3-hydroxyphenyl)ethenyl]-5-methoxyphenol (12) and (-)-syringaresinol (13). Structures of 1 and 2 were elucidated by analyzing 1D and 2D NMR as well as HR-ESI-MS data. The absolute configuration of compound 1 was confirmed by ECD spectroscopic method. In cytotoxicity evaluation using FaDu human hypopharyngeal squamous carcinoma cell line, compounds 3-6, 8, 10 and 12 showed activities, with IC50 values ranging from 2.55 to 17.70 μM. Among them, compound 10 exhibited remarkable cytotoxic activity with IC50 value of 2.55 μM. This is the first report on the anticancer mechanistic study of compound 10 in HNSCC including FaDu cells.
