Antioxidant Activity and DNA Damage Protective Effect of a Robinia pseudoacacia L. Flower Extract (아까시 꽃 추출물의 항산화 활성 및 DNA 손상 억제 효과)

  • The antioxidant properties of Robinia pseudoacacia L. water and 70% ethanol extracts were evaluated by determining total phenolic content (TPC), DPPH radical scavenging activity (RSA), and reducing power (RP). The water extract showed higher TPC (9.07 mg/g gallic acid equivalents) and RP than those of ethanol extract, whereas ethanol extract had greater DPPH RSA. The R. pseudoacacia L. extracts also showed antigenotoxic effects for 200 ${\mu}M$ $H_2O_2$-induced DNA damage in human leukocytes. The 200 ${\mu}M$ $H_2O_2$-induced DNA damage decreased following treatment with the water extract. Reductions in DNA damage with 50 ${\mu}g/ml$ of the water and ethanol extracts were 46.5 and 32.4%, respectively.

Physicochemical and Sensory Evaluation of Cured and Short-Ripened Raw Hams During Storage at 10 and 25°C (단기 숙성 생햄의 저온 및 실온저장 과정 중 물리화학 및 관능학적 품질 특성 변화)

  • Twenty pork loins were processed for manufacturing raw hams according to a short-ripening procedure including dry-and wet-curing for 1 week each, followed by ripening for 2 weeks. Raw ham cuts were vacuum-packaged and stored in darkness at 10 and $25^{\circ}C$ for 90 days, and their physicochemical and sensory quality characteristics were investigated. The sodium chloride content of raw hams stored at 10 and $25^{\circ}C$ was maintained at approximately 5.1% throughout storage at either temperature. No significant changes in water, crude protein, crude fat and ash contents were observed in all samples regardless of storage temperature and storage length. Thiobarbituric acid and volatile basic nitrogen values increased continuously during the storage period. The changes in physicochemical characteristics including pH, water activity texture lipid oxidation and protein degradation, and sensory attributes appeared to be more pronounced at $25^{\circ}C$ than at $10^{\circ}C$ over the storage period. At prolonged storage periods, a significant quality loss in the aspect of texture changes including hardness, brittleness, elasticity, cohesiveness, gumminess, and adhesiveness was observed (p<0.05). Based on sensory evaluation scores, It appeared that vacuum-packaged raw ham cuts stored at 10 and $25^{\circ}C$ were not acceptable after 75 and 45 days, respectively.

  • In order to investigate the changes and characteristics of biochemical metabolic substances of soybean tissue culture during the cultural period, immature cotyledons were detached form the plant on 15th days after flowering and cultured in vitro for 3 weeks. The cultures were classified into embryogenic(EC) and non-embryogenic callus(NEC). A part of the EC lines were subcultured for another 3 weeks and classified into root forming(RFC), and shoot forming cultures(SFC). Another part of the EC lines were used for isolation of protoplasts, which were subsequently cultured in vitro for 4 weeks. The cultures were classified into embryogenic(PEC) and non-embryogenic callus(PNEC) derived from the protoplasts. The cultures of EC and PEC lines showed higher phenylalanine content and lower methionine content than those of NEC and PNEC. At organ differentiation stage, both cultures showed the content of aspartic acid decreased, while the other amino acids increased as a whole. The protein pattern analysis of the cultures revealed that EC and NEC lines contained distinctive polypeptides, with mass of ca. 18KD for EC and ca. 22KD for NEC respectively. The EC and PEC lines also showed high activity of peroxidase isozyme A(piA), while the RFC and SFC lines showed that of peroxidase isozyme B(piB).

Mechanical and Oxygen Permeation Properties of Layered Double Hydroxide/Ethylene Vinyl Acetate Nanocomposite Membranes (Mg-Al Layered Double Hydroxide/Ethylene Vinyl Acetate 나노복합막의 기계적 특성과 기체투과 특성에 관한 연구)

  • The effect of layered double hydroxides (LDH) on the gas separation properties of ethylene vinyl acetate copolymer was investigated. Mg-Al LDH/EVA nanocomposite membranes were prepared from solution intercalation using organically modified LDH (DS-LDH). Dodecyl sulfate (DS)-LDH was obtained by the intercalation of DS anion in the interlayer. The nanocomposite structure has been elucidated by X-ray diffraction (XRD), Fourier transform infrared (FT-IR) spectroscopy and scanning electron microscopy (SEM). XRD pattern clearly shows that the DS-LDH layers are disorderly well dispersed in the EVA matrix. The maximum tensile strength and elongation of the LDH/EVA nanocomposite membrane were found with the LDH content 3 wt%. The thermal properties of nanocompostie membrane were enhanced by the incorporation of LDH in EVA matrix. Gas permeation of LDH/EVA nanocomposite membranes with LDH contents of 1, 3, 5 wt% was studied for $O_2$ and $CO_2$ single gases. The presence of 3 wt% LDH decreased $O_2$ permeability by up to 53% compared to the EVA membrane. In spite of barrier property of nanocomposite membrane, however, the gas permeability for $CO_2$ was increased due to its strong affinity with the residual OH groups on the LDH.

  • 1. Chemical components of fresh tobacco leaves at topping stages were affected variously by fertilizer application level. The more fertilizers were applicated, the higher nitrogen content of leaves was shown regardless of the soil fertility, but phosphorus content was not affected either by phosphorus rate or soil fertility. Potassium content was higher in the leaves grown in fertile soil than infertile at the same application rate. 2. Maturation of tobacco leaves was delayed by applying high level of nitrogen fertilizer, especially in fertile soil. The excessive accumulation of nitrogen in tobacco leaves at later stage of growth resulted in poor quality index for the high content of nicotine and low content of reducing sugar in cured leaves. 3. Nicotine content of cured leaf was increased significantly as nitrogen content increased, regardless of soil fertility, but reducing sugar content was reduced. Nicotine and reducing sugar content of cured leaf were higher in fertile than in infertile soil. 4. Resulting from the facts that nicotine contents were negatively correlated and reducing sugar contents were positively correlated with grading value (Won/Kg), authors suggested that grading index (Won/Kg) of the Office of Monopoly be based on quality index from chemical components of cured leaves.

  • 유역의 자연 유출량이 줄어드는 갈수기 동안 낙동강 본류의 수질은 상류지역의 댐 방류량에 큰 영향을 받는다. 따라서 수질이 취약한 중 하류부지역의 수량과 수질을 종합적으로 고려한 낙동강 수계의 물 관리를 위해서는 댐 방류량과 하류 수질의 정량적인 관계에 근거한 다목적댐군의 연계운영이 필요하다. 본 연구의 목적은 정상상태 하천수질모델을 이용하여 낙동강수계 다목적댐군의 방류량과 하류 하천의 수질 관계를 정량적으로 분석하고, 갈수기 동안 낙동강 수질개선을 위해 필요한 적정 댐 용수공급 시나리오를 제시하는데 있다. 낙동강수계 댐 군의 하류 하천 수질을 고려한 저수지운영 계획 수립을 지원하기 위하여 1차원 정상상태 수질예측모델인 QUAL2E를 구축하고 실측 유량과 수질자료를 이용하여 모델을 보정하고 검증하였다. 검증된 모델은 낙동강수계 상류에 위치한 안동댐, 임하댐, 그리고 중 하류 지류에 위치한 합천댐과 남강댐의 용수공급 시나리오별 하천 수질개선 효과를 분석하는데 적용하였다. 연구결과 낙동강 수질이 악화되는 갈수기 동안 중 하류부에 위치한 상수원의 수질을 개선하기 위해서는 합천댐과 남강댐에서보다는 상류에 위치한 안동댐과 임하댐에서 용수를 추가 공급하는 것이 더욱 효과적인 것으로 평가되었다. 댐 방류량 증감에 따라 수질영향을 가장 크게 받는 하천구간은 수질오염도가 가장 크게 나타나는 금호강합류점${\sim}$고령교구간인 것으로 분석되었다. 본 연구결과는 낙동강과 같이 수량이 부족한 하천은 오염부하량 삭감 대책과 함께 갈수기 유량조절이 매우 중요한 요소임을 보여주며, 주기적인 수질악화를 겪는 낙동강 중 하류 지역의 수질개선을 위한 상류 다목적댐군의 연계운영 계획 수립에 활용될 수 있을 것으로 사료된다.치모의 결과와 현장에서 관측한 유속장을 비교함으로써 본 연구에서 제시한 실제 탁수배제능력을 검증하였다.를 구축하였다는데 의의가 있다.로와 접하는 건물의 경우 모서리부 광고 효과가 지배적이며 대부분 곡선돌출형이 사용되고 있었다. 그러므로 모서리 저층부를 필로티로 계획하여 보행흐름을 원활하게 하고 대신 입면을 투명하게 하여 간접광고(내부전시) 효과를 유도하는 것이 좋다. 특히 원형모서리는 건물 특화 성격이 강하므로 불가피할 경우 소형 액센트 광고 위치를 미리 벽면으로 할애하는 것이 경관 및 입면계획에 유리한 것으로 분석되었다. 불확실도 해석모형 등의 새로운 기능을 추가하여 제시하였다. 모든 입출력자료는 프로젝트 단위별로 운영되어 data의 관리가 손쉽도록 하였으며 결과를 DB에 저장하여 다른 모형에서도 적용할 수 있도록 하였다. 그리고 HyGIS-HMS 및 HyGIS-RAS 모형에서 강우-유출-하도 수리해석-범람해석 등이 일괄되게 하나의 시스템 내에서 구현될 수 있도록 하였다. 따라서 HyGIS와 통합된 수리, 수문모형은 국내 하천 및 유역에 적합한 시스템으로서 향후 HydroInformatics 구현을 염두에 둔 특화된 국내 수자원 분야 소프트웨어의 개발에 기본 토대를 제공할 것으로 판단된다.았다. 또한 저자들의 임상병리학적 연구결과가 다른 문헌에서 보고된 소아 신증후군의 연구결과와 큰 차이를 보이지 않음을 알 수 있었다. 자극에 차이가 있지 않나 추측되며 이에 관한 추후 연구가 요망된다. 총대장통과시간의 단축은 결장 분절 모두에서 줄어들어 나타났으나 좌측결장 통과시간의 감소 및 이로 인한 이 부위의 통과시간 비율의 저하가 가장 주요하였다. 이러한 결과는 차가운 생수 섭취가 주로 결장 근위부를 자극하는 효과를 발휘하는 것이

Distribution and Characteristics of Acidotolerant Heterotrophic and Naphthalene­Degrading Bacteria in Acidic Soils (산성토양에서 내산성 종속영양세균과 나프탈렌분해세균의 분포 및 특성)

  • The distribution and characteristics of acidotolerant heterotrophic and naphthalene-degrading bacteria were investigated in two forest areas, one near Ulsan petrochemical industrial complex (Sunam) and the other in countryside (Daeam). Average values of soil pH at Sunam and Daeam were 3.8 and 4.6, respectively. When het­erotrophic and naphthalene-degrading bacteria were enumerated by most probable number (MPN) procedures at Sunam, the median values of heterotrophs growing at pH 7.0 and pH 4.0 were $5.3{\times}10^7\;and\;3.3{times}10^7$ MPN/g, whereas those of naphthalene-degraders were $5.6{\times}10^4\;and\;4.0{times}10^5$ MPN/g, respectively. While the medians of heterotrophs at Daeam were larger than those at Sunam, the concentrations of naphthalene-degraders were higher at Sunam compared to those at Daeam. From the MPN tubes and enrichment cultures, we obtained 17 isolates of naphthalene-degraders which were identified as Sphingomonas paucimobilis, Brevundimonas vesic­ularis, Burkholderia cepacia, Ralstonia pickettii, Pseudomanas fluorescens, and Chryseomonas luteola. Among them, 6 isolates showed higher naphthalene-degrading activity on minimal media of pH 4 compared to pH 7, whereas the extent of growth was not greater at pH 4 than at pH 7 when they were inoculated on nutrient-rich media. It is plausible that the pH may affect naphthalene-degrading activity of the isolates by changing fatty acid composition of bacterial membrane.

Quantitative Assessment of the Quality of Regional Adaptation Trial Data for Crop Model Improvement (작물 모형 개선을 위한 지역적응시험 자료의 정량적 품질 평가)

  • Cultivar parameters, which are key inputs to a crop growth model, have been estimated using observation data in good quality. Observation data with high quality often require considerable labor and cost, which makes it challenging to gather a large quantity of data for calibration of cultivar parameters. Alternatively, data in sufficient quantity can be collected from the reports on the evaluation of cultivars by region although these data are of questionable quality. The objective of our study was to assess the quality of crop and management data available from the reports on the regional adaptation trials for rice cultivars. We also aimed to propose the measures for improvement of the data quality, which would aid reliable estimation of cultivar parameters. DatasetRanker, which is the tool designed for quantitative assessment of the data for parameter calibration, was used to evaluate the quality of the data available from the regional adaptation trials. It was found that these data for rice cultivars were classified into the Silver class, which could be used for validation or calibration of key cultivar parameters. However, those regional adaptation trial data would fall short of the quality for model improvement. Additional information on management, e.g., harvest and irrigation management, can increase the quantitative quality by 10% with the minimum effort and cost. The quality of the data can also be improved through measurements of initial conditions for crop growth simulations such as soil moisture and nutrients. In addition, crop model improvement can be facilitated using crop growth data in time series, which merits further studies on development of approaches for non-destructive methods to monitor the crop growth.

Enhanced Nasal Absorption of Ketoconazole by Inclusion with Cyclodextrin (시클로덱스트린과의 포접에 의한 케토코나졸의 비점막 흡수증가)

  • Inclusion complexes of ketoconazole(KT) with ${\alpha}^_$, ${\beta}^_$cyclodextrin(CD) and $dimethy1-{\beta}-cyclodextrin$ (CD) and $dimethy1-{\beta}-cyclodextrin(DM{\beta}CD)$ as nasal absorption enhancer were prepared in 1: 2 molar ratios by freeze-drying and solvent evaporation methods. In order to compare with the intrinsic absorptivity of KT in the jejunum(J) and the nasal cavity(N), the in situ simultaneous perfusion method was employed. The in situ recirculation study revealed that KT-CD inclusion complexes with the greater stability constant and the faster dissolution rate proportionally increased the absorption of KT in the J and N of rats. The rank order of apparent KT permeability$(P_{app}\;:\;cm/sec\;{\time}\;1O^{-5}{\pm}S.E.)$, corrected by surface area of absorption, was $5.10{\pm}0.3(N,\; KT-DM{\beta}CD)$ )> $4.13{\pm}0.4(N,\;KT-{\beta}-CD)$ )> $3.52{\pm}0.2(N,\;KT-{\alpha}-CD)$ )> $2.76{\pm}0.3(J,\; KT-DM{\beta}CD)$ )> $2.61{\pm}0.5(J,\;KT-{\beta}-CD)$ )> $2.42{\pm}0.4(J,\;KT-{\alpha}-CD)$ at pH 4.0. The in crease in permeability of $KT-DM{\beta}CD$ inclusion complex was 2.6 folds in the J and 4.5 folds in the N when the perfusing solution was changed from the buffer(pH 4.0) to saline. The absorption rate of $KT-DM{\beta}CD$ inclusion complex after nasal administration was more rapid than those of ketoconazole alone and $KT-DM{\beta}CD$ inclusion complex after oral administration to rats. In comparision with an oral administration of ketoconazole suspension in corn oil, the relative bioavailability was calculated 137.3% for the oral and 195.0% for nasal $KT-DM{\beta}CD$ inclusion complex in rats. The present results suggest that $KT-DM{\beta}CD$ inclusion complex may serve as a potential nasal absorption enhancer for the nasal delivery of ketoconazole.

  • In the case of the direct measurement of $^{226}Ra$ using a HPGe gamma-ray spectrometer, the interference between gammarays with 186.21 keV of $^{226}Ra$ and 185.7 keV of $^{235}U$ should be corrected to calculate the net peak area in the energy spectrum. In general, it is very difficult to conduct peaks stripping with difference of about 0.5 keV, although a HPGe with the superior resolution is applied and the maximum channels is applied to the spectrometer. In this study, several interference correction techniques in the direct measurement were surveyed to evaluate the feasibility for the measurement of $^{226}Ra$ using the gamma-ray spectrometery. Applying the interference corrections to the analysis of raw materials and by-products, the method validation for the direct measurement of $^{226}Ra$ was conducted by evaluating the measurement uncertainty, linearity, and range. As a result, the optimum method of the interference correction was selected by comparing with the indirect measurement of which progenies of $^{226}Ra$, such as $^{214}Pb$ and $^{214}Bi$, were analyzed in the secular equilibrium state.
