형상기억합금의 특성 및 응용

  • 형상기억합금(SMA : Shape Memory Alloy)은 일반적인 금속이나 합금에서는 찾아볼 수 없는 형상기억효과(shape memory effect)와 초탄성 (superelasticity) 거동을 보이고 있다. 이러한 특성은 1951년에 금-카드뮴(Au-Cd) 합금에서 처음으로 발견되었으며, 1963년에 미국 해군병기연구소(Naval Ordnance Laboratory)에서 니켈-티타늄 (Ni-Ti) 합금에서 형상기억효과를 발견한 후로 널리 상용화되었다. 니티놀(nitinol)이라고 불려지는 니켈-티타늄 계열의 형상기억합금은 단위 부피당 많은 에너지를 낼 수 있고, 내 부식성(corrosion resistance)과 생화학적 적합성(bio-compatibility)이 뛰어나다. 또한 100,000사이클 이상의 긴 사용수명을 갖기 때문에 작동기(actuator)로서 우수한 특징을 갖는다.   • 잠재적인 생체의료용 소재로서 홍합접착단백질은 그 동안 커다란 관심을 받아왔었다. 홍합접착단백질은 환경친화적인 수중접착제로써 강하면서도 유연하게 다양한 물질들의 표면에 접착하며 수분에 강하고 인체에 무해하며 생분해되는 특성을 가지고 있다. 여러 가지의 홍합접착단백질들이 홍합으로부터 발견되어 그 특성들이 연구되었으며 홍합의 접착기작에 대한 생화학적 지식들이 축적되어 왔다. 또한 이렇게 가능성이 높은 해양홍합유래의 바이오-접착소재를 현실화하려는 많은 노력들이 시도되어 왔다. 본 고에서는 홍합접착단백질의 기능적 생산에 초점을 맞추어 다양한 개발 접근방법들의 진행들의 추이를 정리하였다.

Screening and Identification of Soy Curd-Producing Lactic Acid Bacteria (두유 커드를 생산하는 김치 유래 젖산균의 동정)

  • The purpose of this study was to isolate soy curd forming bacterial strains. Soy curd forming bacteria were isolated from Kimchi, a traditional Korean vegetable food that is fermented using lactic acid bacteria. Among 196 bacterial strains, ten isolates (strain No. 2-2-2, 2-15-2, 2-18-1, 2-19-2, 3-4-1, 3-4-2, 3-8-1, 3-8-3, 3-17-1, 4-39-5) formed firm soy curd. The isolated bacterial strains were identified by molecular biological and biochemical analyses. The genomic DNAs extracted from the isolated bacterial strains were used as a template for PCR amplification of 16S rDNA region. By comparing the results of the 16s rDNA sequences with GenBank data, the isolated strains were identified as Leuconostoc mesenteroides group and Lactobacillus sakei group. The phylogenetic position of soy curd forming strains and their related taxa were investigated using neighbor-joining method. L. mesenteroides group was further identified as L. mesenteroides subsp. dextranicum based on biochemical properties. L. sakei group was named Lactobacillus sp., because it showed a variety of biochemical properties.

  • 1982년도 1월부터 12월 1년간 서울을 비롯한 전국 보건연구소, 검역소, 서울 시내 종합병원 임상병리실험실에서 수거한 가검물 및 분리한 세균 등 총 1,518건을 시험한 결과 이중 살모넬라균속이 687건이 동정되었다. 이중 441건이 장티푸스였으며 그외 살모넬라균이 246건이었다. 이에 대한 생화학적 특성, 혈청학적 특성, 지리적, 계절적, 성별에 대한 발생특성, 앰피실린 등 12가지 항생제에 대한 감수성 시험을 하였다.

  • 굴을 포함한 대부분의 해양 무척추동물들은 동일종이라도 먹이조건과 온도조건에 의해 성장과 발육, 번식초기연령이나 최소번식 크기 등에 큰 차이를 보인다. 굴의 번식수단 및 생활사 를 이해하기 위해서는 번식이나 생활사에 관한 속성을 보다 수량적으로 확인하고 동시에 개체군 생태학적인 지견을 가질 필요가 있다 (윤, 1996). 대서양산 굴, Crassostre virginica에 대하여 Lee and Heffernan (1991)은 알의 지질과 단백질 조성을 밝히고 생화학적 특성을 보고하였으며, Choi et al. (1993)는 면역학적 방법을 이용하여 번식량에 대한 정량적인 연구를 보고한 바 있으나, 이와는 대조적으로 참굴 C. gigas의 알 및 번식량에 대한 연구는 미비한 실정이다. (중략)

The study on the causal agent of Streptococcicosis (Lactococcus garvieae), isolated from cultured marine fishes (해산 양식어류로부터 분리된 연쇄구균증의 원인균, Lactococcus garvieae에 대한 연구)

  • The purpose of this study was to investigate the microbiological characteristics and the distributions of the bacteria causing streptococcicosis occurred in marine fish farm, Korea. Many kinds of cultures fishes suffered from the disease accompanied with typical symptoms, including darkening of the skin, exophthalmia, petechiae inside of the opercula and distended abdomen. The isolates from the diseased fishes were compared with Lactococcus garvieae by biochmical, biophysical and serological methods and polymerase chain reaction(PCR) assay. We isolated 35 strains of the geuns Streptococcus from the diseased olive flounder, Paralichthys olivaceus, yellow tail, Seriola quinqueradiata and Korean rockfish, Sebastes schlegeli. 15 strains out of the isolates were identified to L. garvieae and the others were not because of their different biochemical and biophysical charateristics. Seven strains of the isolates were agglutinated by rabbit serum raised against L. garvieae $KG^+$ phenotypic cells(ATCC49156)as a reference strain. Twenty-one strains of the isolates identified to L. garvieae since they were formed the expected band through performing PCR assay using specific primers, pLG-1(5'-CATAACAATGAGATCGC-3') and pLG-2(5'-GCACCCCGCGGTTG-3'). In the present study, it showed that L. garvieae was a dominant strain causing streptococcicosis in the tested area due to occurrence of 21 strains as L. garvieae out of all the isolates, 9 atrains as Streptococcus sp. and 5 strains as Enterococcus sp.

  • From 2002 to 2004, various vibrios were isolated from the olive flounder, Paralichthys olivaceus of culturing size with disease signs. During this survey, it was known that the high proportion of Vibrio ichthyoenteri was occupied among the isolated vibrios. Generally, V. ichthyoenteri is well known as the pathogen of bacterial enteritis of olive flounder larvae. The aim of the present study was the compare the characteristics of two groups of V. ichthyoenteri, culturing sized olive flounder, and larvae of olive flounder showing the intestinal necrosis. The research was focused on the physiology, biochemistry, genetics in the two bacterial groups. The physiological and biochemical characteristics of the tested strains were very similar. The intergenic spacer (IGS) region between the 16S and 23S rRNA genes of 21 isolated strains and 3 reference strains, V. ichthyoenteri, were investigated by PCR fragment length typing and DNA sequencing. After the isolated strains were identified as V. ichthyoenteri, not only phenotypic characteristics of the isolated and reference strains but also homology of 16S-23S IGS of all isolated strains and reference strains as 99.1~100%. The V. ichthyoenteri showed 4 specific 16S-23S patterns and contained no-tRNA, tRNAGlu(TTC) , tRNAIle(GAT) tRNAAla(TGC) type .

  • The diseased larvae of the flounder, Paralichthys olivaceus, were sampled several times from a fish hatchery located in Cheju Island between December, 1991 and April, 1992. They turned out to be infected with Vibrio sp. and diagnosed as intestinal necrosis of flounder larvae(INFL) based on morphological, biological, and biochemical examinations. INFL was known to be caused by the live food organism infected with Vibrio sp. The optimal growth conditions of the isolates for temperature, pH, and NaCl concentration were $20\sim30^{\circ}C$, 6~8, and 2~4%, respectively. In the drug sensitivity test, the isolates were sensitive to oxytetracycline, nalidixic acid, kanamycin, and novobiocin, but resistant to ampicillin, erythromycin, spiramycin and sulfa-drug.

Characteristics Comparisons of Edwardsiella tarda Isolated from Cultured Olive Flounder and Eel (양식넙치와 뱀장어에서 분리된 Edwardsiella tarda의 특성 비교)

  • The objective of this study was to determine comparative biochemical characteristics and RAPD (random amplified polymorphic DNA) profiles of 18 strains of Edwardsiella tarda isolated from cultured olive flounder (Paralichthys olivaceus) and eel (Anguilla spp) that showed diseases between 1996 and 2010 in Korea. In terms of biochemical properties, they showed four patterns with differences in citrate degradation and production of H2S and indole. All strains were identified as E. tarda. Characteristics of isolates were not classified according to their host. As a result of PCR with E. tarda-specific primer, EDtT, the same band of about 270 bp was detected in all 18 isolates. However, no specific band was detected in type strains of E. tarda or Edwardsiella ictaluri. As a result of RAPD PCR performed with primer No. 5 and No. 6 of a Ready-To-Go-RAPD kit, the band profile showed clear differences among isolates of olive flounder, isolates of eel, and E. tarda and E. ictaluri type strains. It was possible to organize their characteristics according to the origin of host with possibility to develop tools for pathogen monitoring.
