Biochemical Characterization of α-Galactosidase-Producing Thermophilic Bacillus coagulans KM-1 (α-Galactosidase를 생산하는 고온성 Bacillus coagulans KM-1 균주의 생화학적 특성)

  • A bacterium producing ${\alpha}$-galactosidase (${\alpha}$-$\small{D}$-galactoside galactohydrolase, EC 3.2.1.22) was isolated. The isolate, KM-1 was identified as Bacillus coagulans based on its 16S rRNA sequence, morphology, and biochemical properties. ${\alpha}$-Galactosidase activity was detected the culture supernatant of B. coagulans KM-1. The bacterium showed the maximum activity for hydrolyzing para-nitrophenyl-${\alpha}$-$\small{D}$-galactopyranoside (pNP-${\alpha}Gal$) at pH 6.0 and $50^{\circ}C$. It hydrolyzed oligomeric substrates such as melibiose, raffinose, and stachyose liberating a galactose residue, indicating that the B. coagulans KM-1 ${\alpha}$-galactosidase hydrolyzed ${\alpha}$-1,6 linkage. The results suggest that the decreased stachyose and raffinose contents in fermented soybean meal are due to the ${\alpha}$-galactosidase activity.

  • Nine biochemical characteristics of female Oncorhynchus keta subpopulations that migrate to three streams (Namdaechun, Myeongpachun, Oshipchun) in Gangwon, Korea, were evaluated during November 2006: aspartate aminotransferase, alanine aminotransferase, creatine kinase, triglyceride, cholesterol, glucose, hemoglobin, and urea in the blood and RNA/DNA in muscle. The three subpopulations were identical morphologically, but differed in their biochemical characteristics. The RNA/DNA ratio could be useful for distinguishing the subpopulations. RNA/DNA ratios of the Namdaechun and the Myeongpachun and Oshipchun subpopulations were $3.52{\pm}0.7$ and $2.9{\pm}0.7$ and $2.4{\pm}0.8$, respectively. The difference was statistically significant (P<0.05). By contrast, the blood parameters were somewhat ambiguous.

  • The isolation and characterization of type II restriction endonuclease from Clostridium thermocellum ATCC 27405 were described. This enzyme (Cth I endonuclease) is an isoschizomer of Bcl I endonuclease recognizing 5'-TGATCA-3'. Cth I endonuclease requires MG$^{2+}$ ion for its activity and is maximally active at PH 1.5 to 10.5 in the Presence of 0 to 10mM NaCl. Cth I endonuclease is heat stable and has an optimum temperature of 6$0^{\circ}C$. The activity of Cth I enzyme is sensitive to dam methylation.

Purification and Characteristics of ${\beta}$-Galactosidase from Strawberry (딸기 ${\beta}$-Galactosidase의 정제 및 생화학적 특성)

  • ${\beta}$-Galactosidase was extracted and purified from strawberry. The purified ${\beta}$-Galactosidase from strawberry was investigated their physicochemical characteristics. ${\beta}$-Galactosidase was purified 25.74 fold from strawberry. The purification procedure include ammonium sulfate fraction, acetone powder treatment and gel and ion exchange chromatography. Yield of the enzyme purification was 18.11%. The purified enzyme has native molecular weight of 116,000 dalton. Vmax value and Km value of ${\beta}$-Galactosidase were 0.077 mM ONPG/ml/15mim and 1.75x10-2mM, respectively. The optimum temperature and pH of ${\beta}$-Galactosidase were 43$^{\circ}$C and pH 4.0, respectively. The ${\beta}$-Galactosidase activity was stable below 50$^{\circ}$C and at pH 4.0 to pH 6.0. Among the metal ions Ca and Mg were did not affect, whereas K, Cu and Zn show a little effect on the enzyme activity. The ${\beta}$-Galactosidase activities were inhibited by treatment with EDTA and SDS.

Biochemical Characteristics of the Granulosis Viruses DNA of Common Cabbage Worm, Pieris rapae and Pieris brassicae (배추흰나비 과립병바이러스 DNA의 생화학적 특성)

  • This study was carried out to acquire some basic biochemical informations on the granulosis virus (GV) DNA of Pieris rapae and Pieris brassicae. The thermal denaturation temperature (Tm) and G+C content of the DNA of the viruses were $83.7^{\circ}C$ and 35.5% for P. rapae GV, $84.0^{\circ}C$ and 35.9% for P. brassicae GV, respectively. There were some differences in the DNA fragmentation patterns of the two GV's produced by digestion with restriction endonucleases such as EcoR I , BamH I and Hind m . The homololgy between the two DNAs was caculated to be 97.0%. The size of the genome was estimated to be 103 kbp for P. rapae GV and 108 kbp for P. brassicae GV.

  • An agar degrading bacterium, This strain was isolated from sea water in Jeju. The strain is Gram-negative, rod and strictly aerobe for growth. The identification bases on the 16S rRNA gene sequencing showed that the strain was closely related to the genus Agariovarans sp. and named Agariovorans sp. JA1. The strain grew on agar as a sole carbon and energy source and produced an extra cellular agarase. For the increase of agarase productivity, 0.5% agar, yeast extract and $NaNO_3$ were used as carbon, organic and inorganic nitrogen source, respectively. For effective production of agarase and growth, the pH, temperature and NaCl concentration were was pH 7, $25^{\circ}C$ $^{\sim}$ $30^{\circ}C$ and 2%, respectively.

  • The present study was conducted to investigate biochemical properties and antimicrobial drug susceptibilities of 57 strains of Pasteurella haemolytica (P haemolytica) isolated from pneumonic calves and goats in Youngnam province during the period from September 1989 to February 1991. P haemolytica was isolated from 95 of 142(31.7%) pneumonic calves of 1 to 6 months of age and from 9 of 24(37.5%) pneumonic goats of 2 to 10 months of age. Seasonal isolation frequency of P haemolytica in calves and goats varied from 16.6 to 35.7%, and it was higher in winter. The majority of biochemical and cultural properties of P haemolytica isolated from calves and goats were identical to those of the reference strains employed. All isolates were susceptible to ceftiofur, erythromycin, kanamycin, gentamicin, and chloramphenicol (MIC: <$25{\mu}g/ml$), some of them were resistant to amikacin, sulfadimethoxin, and streptomycin (MIC: >200IU or ${\mu}g/ml$).

Incidence of Pasteurella multocida infection in Youngnam swine herds and the biochemical properties of the organisms recovered from pigs with atrophic rhinitis and pneumonic lungs (영남지방 돼지의 Pasteurella multocida 감염상태 및 분리균의 생화학적 특성)

  • The present study was conducted to investigate the incidence of Pasteurella multocida infection in Youngnam swine herds during the period from March 1988 to February 1989 and some properties of the isolated organisms. P multocida was isolated from 22(43.1%) of 51 growing pigs of 4 to 12 weeks of age and from 8(80.0%) of 10 herds. From nasal turbinates of 102 slaughtered pigs, 47(46.1%) pigs were culture positive and pigs from 8(88.9%) of 9 herds were found to be infected with P multocida. From lungs of 101 slaughtered pigs, 42(41.6%) pigs were culture positive and the pigs from 11(91.7%) of 12 herds were found to be infected with P multocida. The majority of biochemical and cultural properties of the P multocida isolates were identical to those of the standard strains. The isolation frequencies of P multocida in relation to pig snout lesion grades of 0 to 5 were 28.6%, 41.6%, 48.0%, 50.0%, 85.7%, and 100%, respectively.

  • Protease producing bacterium, PB16 was isolated from food processing wastewater sludge and paddy field soil samples and selected by the clear zone and enzyme activity test. The isolate was gram negative, rod type and its protease productivity was 6.49 U/ml. As a result of API20NE kit test and 16S rDNA sequencying, the isolated PB16 was identified as Aeromonas hydrophila (99%). The growth rate ($h^{-1}$) was 0.21 in synthetic waste water only and 0.26 in synthetic waste water containing vitamin and mineral using a bioscreen C. Synthetic wastewater removal rate was 59 and 87%, respectively after 1 and 3 day reaction (intial CODcr was 2,472 mg/l).

  • The present study was conducted to investigate biochemical properties and antimicrobial drug susceptibilities of 47 strains of E rhusiopathiae isolated from the cases of acute septicemic swine erysipelas in Youngnam and Kyunggi provinces during the period from June 1988 to December 1989. The isolants were identified as E rhusiopathiae on the basis of cellular and colonial morphology, and characteristic reactions in some biochemical tests. All the organisms produced hydrogen sulfide in triple sugar iron agar and showed the characteristic "pipe cleaner" type of growth in gelatin stab cultures. The majority of biochemical and cultural properties of E rhusiopathiae isolated from pigs affected with acute erysipelas were identical to those of the reference strains employed. All the isolates were highly susceptible to penicillin G, ampicillin, erythromycin (MIC:0.025~0.39IU or ${\mu}g/ml$), and moderately susceptible to oleandomycin, oxytetracycline, chloramphenicol(MIC:$0.78{\sim}25{\mu}g/ml$). Kanamycin and sulfadimethoxine showed no activity against the isolates(MIC:>$400{\mu}g/ml$). The MICs of dihydrostreptomycin presented two distribution peaks; of 47 strains, 5(10.6%) were resistant to dihydrostreptomycin (MIC:$400{\mu}g/ml$), while the majority of them were susceptible to the drug.
