Studies on the Survival and In Vitro Fertilization Ratesof Frozen-Thawed Bovine Immature Oocytes (소 동결 미성숙 난자의 생존율과 체외수정율에 관한 연구)

  • 본 연구는 소 미성숙 난자의 동결에 있어서 미성숙 난자의 1, 2, 4, 8 시간 성숙배양한 다음 동결 융해 후 체외배양하였을 때 체외수정율과 생존율을 조사하고저 수행하였다. 미성숙 난자들 1∼8시간 성숙배양후 동결융해 및 체외수정시켰을 때 수정율은 12.8∼30.9%로서 단시간의 체외성숙 배양이 높게 나타났으며(23.8∼28.8%), 또한 체외배양시 생존율은 11.8∼29.9%를 나타냈다.

  • 모바일 로봇 센서가 개입하여 네트워크의 단절을 억제하여 모바일 로봇 센서를 통해 센서 네트워크의 생존시간을 향상시키는 것을 목표로 한다. 센서노드가 비교적 적은 희소한 환경에서는 베터리 소모 등의 다양한 원인으로 인하여 매개체 역할을 수행하는 브릿지 노드가 제거 될 경우 해당 브릿지 하부의 네트워크는 단절되어 더 이상 쓸모없게 되는 심각한 상황을 초래하기 쉽다. 본 논문에서는, 모바일 로봇 센서가 개입하여 단절된 브릿지를 효율적으로 찾아가 그 역할을 대행함으로써 네트워크의 생존시간을 연장 시키는 것을 목적으로 하는 센서 네트워크를 설계하고 구현했다.

Cryopreservation of the Entomopathogenic Namatode, Steinernema carpocapsae Weiser (곤충병원선충(Steinernema carpocapsae Weiser)의 냉동저장법)

  • Cryopreservation of infective juveniles of entomopathogenic nematode, Steinernema carpocapsae Weiser, was conducted at $-190^{\circ}C$ liquid nitrogen and its, efficacy was analysed on nematode survival and pathogenicity with glycerol pretreatments and storage periods. Infective juveniles were pre-treated before being frozen by incubating the nematodes in 22% glycerol for each of 6, 12, and 24 h, followed by 70% methanol at $0^{\circ}C$ for 10 minutes. Just after glycerol and methanol incubations, subsamp1es of the nematodes were resuspended in 0.85% saline and maintained during 24h for viability determination. Different glycerol incubation periods significantly affected the nematode susceptibility to methanol infiltration. Six hour incubation in glycerol resulted in much less nematode survival than did 12 h or 24 h incubation. About 70% of the infective juveniles frozen at $-190^{\circ}C$ for 5 months, preincubat-ed in glycerol at least for 12h, were able to survive after being resuspended in 30°C saline. They did not also show any change in their pathogenicity during cryopreservation. These results suggest an improved technique for long-term storage of the entomopathogenic nematodes.

An extension of multifactor dimensionality reduction method for detecting gene-gene interactions with the survival time (생존시간과 연관된 유전자 간의 교호작용에 관한 다중차원축소방법의 확장)

  • Many genetic variants have been identified to be associated with complex diseases such as hypertension, diabetes and cancers throughout genome-wide association studies (GWAS). However, there still exist a serious missing heritability problem since the proportion explained by genetic variants from GWAS is very weak less than 10~15%. Gene-gene interaction study may be helpful to explain the missing heritability because most of complex disease mechanisms are involved with more than one single SNP, which include multiple SNPs or gene-gene interactions. This paper focuses on gene-gene interactions with the survival phenotype by extending the multifactor dimensionality reduction (MDR) method to the accelerated failure time (AFT) model. The standardized residual from AFT model is used as a residual score for classifying multiple geno-types into high and low risk groups and algorithm of MDR is implemented. We call this method AFT-MDR and compares the power of AFT-MDR with those of Surv-MDR and Cox-MDR in simulation studies. Also a real data for leukemia Korean patients is analyzed. It was found that the power of AFT-MDR is greater than that of Surv-MDR and is comparable with that of Cox-MDR, but is very sensitive to the censoring fraction.

  • Crossover trials of new drugs in the treatment of angina pectoris, which frequently use treadmill exercise test for the assessment of its efficacy, produce censored survival times. In this paper we consider analysis approaches for censored survival times from crossover trials. Previously, a stratified Cox model for paired observation and nonparametric methods have been presented as possible analysis methods. On the other hand, the differences of two survival times would produce interval-censored survival times and we propose a Cox model for interval-censored data as n alternative analysis method. Example data is analyzed in order to compare these different methods.

비단가리비 인공종묘 생산

  • 비단가리비의 인공 종묘 생산을 목적으로 전라남도 신안군 대흑산도 주변에 서식하는 비단가리비를 대상으로 인공 종묘 생산을 위한 산란 유발, 수정란의 발생 과정, 유생 사육, 채묘 및 중간 육성 등 양식 생물학적 연구를 실시하였다. 어미의 각종 산란 유발 자극에 대한 반응은 Serotonin 주사, 온도 자극, 혼합 자극에서 반응율이 가장 높았으며, 자외선 조사 해수 자극은 수컷만 반응을 하였고 간출 자극은 반응이 없어 실효성이 없는 것으로 나타났다. 수정란의 크기는 $69.5{\mu}m$이었고, 수정 후 약 2시간에 2 세포기, 8시간 후에 8 세포기, 20시간 후에는 담륜자 유생으로 부화하였으며 40시간 후에는 D상 유생으로 되었다. 수온별 비단가리비 유생의 성장은 수온 $20^{\circ}C$에서 각장 $178.9{\mu}m$로 가장 좋았으며, 이 때의 생존율은 15.5%이었다. 그러나 수온 $15^{\circ}C$에서는 낮은 성장을 보여 각장 $135.9{\mu}m$로 성장하였으며, 생존율도 9.8%로 저조하였다. 수용 밀도별 사육 시험에서 $1m{\ell}$당 1개체와 5개체에서 성장 및 생존율이 양호하여 성장 및 생존율을 볼 때 적정 사육 밀도는 $1m{\ell}$당 5개체 이하로 나타났다. 먹이 생물 종류에 따른 유생의 성장을 알기 위하여 I, gal-baba, C, calcitrans, N, oculata를 단독 또는 혼합으로 공급하였을 때 실험 종료후 각장의 성장은 I, galbana+C. calcitrans+N. oculata구가 $194.2{\mu}$로 가장 좋았고 N. oculata구는 $162.2{\mu}m$로 가장 낮은 성장을 보였다. 생존율에서는 I. galbana+C. calcitransoculata구는 9.4%로 가장 낮은 생존율을 보였다. 채묘기별 유생의 부착률은 염화비닐판을 수평으로 놓은 것과 패각이 각각 3.43%와 3.17%로 가장 양호하였으나, 양파망과 염화비닐판을 수직으로 놓은 것은 각각 1.52%와 1.61%로 비교적 저조하였다. 수정 후 40일째부터 90일째까지 측정한 부착치패의 경과 일수에 따른 각장의 성장은 $SL=184.44e^{0.0335X}(r^2=0.9861)$의 회귀직선식으로서 나타났다. 중간육성 시험에서 수심별 성장을 분석한 결과, 비단가리비 치패는 저층보다 표층이 각장 5.92mm, 전중량 6.07g 정도 더 빨리 성장하였다.

Survival on Water Temperature, Salinity and Air Exposed Time and Sand Immersion Ability of the Surf Clam, Spisula sachalinensis Spat (북방대합 치패의 수온, 염분 및 공기노출에 따른 생존율과 잠사능력)

  • Survival with various conditions (water temperature, salinity and air exposure) and sand immersion ability for release of surf clam, Spisula sachalinensis spats, which were artificially seed produced, were investigated. Survival with water temperature conditions of spats showed no difference within the range of 4∼28 $^{\circ}C$ for 10 days. In salinity conditions, survival (%) of spats showed more than 50% in over 22 psu. In air exposure conditions, survival (%) of spats air exposed for 12 hours was no difference within below 2$0^{\circ}C$, however, that of spats air exposed for 16 hours showed below 50% at $25^{\circ}C$. In sand immersion ability, the immersion time was increased with the increase of shell length, and the immersion rate was decreased. Spats of below 15 mm in shell length were emersed more than 90% within 10 min. There was no difference in the immersion time and the immersion rate with the grain sizes of the sand bottom.

Attachment of Hard Shelled Mussel, Mytilus coruscus and Blue Mussel, Mytilus edulis (홍합 Mytilus coruscus과 지중해담치 Mytilus edulis의 부착 비교)

  • In this study, the reattachment processes of small and medium size of spot of Mytilus coruscus and M. edulis were observed. The small spats (Shell length, SH, $3{\sim}7mm$) of hard shelled mussel, M. coruscus showed 100% reattachment rate within 7 minutes after the detachment of byssus thread and 100% survival rate. While large spats (SH 25 mm) showed 85% reattachment rate within 5 hours and 100% survival rate for 24 hours. The reattachment rate of M. edulis (SH 30 mm) was higher than that of M. coruscus (SH, 28 mm). The thread consisted of three identifiable structures; adhesive disc, adhesive thread and adhesive root. The adhesive disk and adhesive thread of M. coruscus were larger and thicker than those of M. edulis, whereas the thickness of adhesive root was the other way round. Further studies are required to identify the relationship between the structure of byssus thread and attachment ability. This study suggests that the sizes of spats could be an important factor in determining the timing of removing and reattaching mussel seedling for aquaculture or releasing to the sea.

Effect of Suboptimal Temperature Incubation on the Resistance of Lactobacillus acidophilus CT 01 to Storage and Drying (저온배양에 따른 Lactobacillus acidophilus CT 01의 저장 및 건조에 대한 저항성)

  • This study was carried out to determine the storage, cryotolerance, heat and drying resistance, when Lactobacillus acidophilus CT 01 isolated from preweaned piglet feces growing at suboptimal temperature. L. acidophilus CT 01 suboptimal temperature incubated for 48 hours had the slowest growth rate at 22℃ but the highest viable cell number after 36 hours at 22℃, with 1.3×10/sup 9/ CFU/mL. In case of 4 and 20℃ storage, the suboptimal temperature incubated groups had a viability higher than the control (p<0.01). The cryotolerance of suboptimal temperature incubated L. acidophilus CT 01 was a higher than the control (p<0.01). When L. acidophilus CT 01 was heat treated at 60℃ for 15 minutes and 30 minutes, the suboptimal temperature incubated L. acidophilus CT 01 at 22℃ had a viability higher more than the control (p<0.01). L. acidophilus CT 01 incubated suboptimal temperature was inoculated by 30% to the carrier, and dried at 50℃ for 12 hours had the highest viability in the suboptimal temperature incubated L. acidophilus CT 01 at 28℃.

Yolk resorption, onset of feeding and survival potential of larvae of red spotted grouper, Epinephelus akaara (붉바리 자어의 난황흡수 및 첫 먹이 섭취시기와 관련한 생존 특성)

  • Yolk resorotion and commencement of external feeding in larvae of red spotted grouper, Epinepheus akaara were studied to understand the high mortality of larvae. Consumption of yolk was higher than that of oil globule for the first 24hs after hatching. The resorption rates of yolk and oil globule increased with the increase of temperature. The time yolk was comsumed up to 99% was 84hs after hatching in 23~$25^{\circ}C$,72hs at ~$27^{\circ}C$, and 60hs in 29~~$31^{\circ}C$, while the time for oil globule was 96hs in 23~$25^{\circ}C$, 84hs at $27^{\circ}C$, and 60hs in 29~$31^{\circ}C$. The relationship between mouth openning time (Y) and water temperature (X) was repersented as a linear regression, Y=-2.53X+118.4, (r2=0.89). In 72~96hs after hatching, upper jaw sizes of the larvae reached around 0.114 mm. By the time the larvae comsumed most of the yolk and oil globule, and started external feeding. The relationship between upper jaw length (Y) and total length (X) was represented as a linear regression, Y=0.139X-0.165, (r2=0.88). Rotifer was not found in the guts of larvae before 72hs after hatching at $28^{\circ}C$, when larvae resorbed their yolk and oil globule over 99%. But, at 96hs after hatching, about 33% of the larvae had the rotifer in their guts. Delayed feeding experiments revealed that the earlier food supply within 84hs after hatching showed the better survival rates for the larvae. The larvae fed at 96hs after hatching died all within 120hs.
