Increase of Ginsenosides Production by the Treatment of Chitosan and Jasmonic Acid in the Adventitious Roots of Korean Ginseng (Panax ginseng C.A. Meyer) (Chitosan과 Jasmonic acid 처리에 의한 인삼 부정근의 Ginsenosides의 생산성 증대)

  • In order to investigate the effects of elicitors on the growth and ginsenosides biosynthesis of ginseng adventitious roots, chitosan and jasmonic acid were treated with various concentrations. The growth rate of adventitious roots was increased with the addition of chitosan at higher concentrations (10 mg/L), but the best accumulation of ginsenosides was observed at the lower concentration (5 mg/L). Jasmonic acid was an effective elicitor for ginsenosides biosynthesis in ginseng adventitious roots. The maximum accumulation of ginsenosides was observed at the treatment of 10 uM jasmonic acid. But the jasmonic acid was found to decrease the growth rate of adventitious roots.

Effect of Seeding Rates and Growth Regulator Application on Lodging Resistance and Yield in Direct Seeding on Flooded Paddy Surface (벼 담수표면직파재배에서 파종량과 생장조절제 처리에 따른 내도복성과 수량성)

  • Experiments were conducted to effect of seeding rates and growth regulator (KIM-112) application on lodging resistance and yield in direct seeding on flooded paddy surface. The number of seedlings per $m^2$ were ranged from 61 to 143, and seedling ratios were from 71.7 to 76.1%. The culm length was increased with the higher seeding rates. The culm length was significantly shortened by KIM-112 application, and the shortened effect of internodes were various with application times. The leaf angles with the culm and top leaves were errected and position of light reception by KIM-112 application, also the weight of culm base was increased. The bending moment at breaking of culm and culm diameter were decreased according to the higher seeding rates. The number of panicles per $m^2$ were increased according to the higher seeding rates. The ripened grain rate and 1, 000 grains weight became higher than the untreated control.

Culture of Tricholoma matsutake Mycelium using Solid Matrix (고체 매질을 이용한 송이(松栮)(Tricholoma matsutake)균 배양)

  • This study was conducted to develop optimal solid culture medium for Tricholoma matsutake. As the solid matrix, granitic soil, perlite, vermiculate, pine sawdust and peat moss were compared regarding their effected on mycelial growth. Ergosterol content which is a fungal wall component was used as the growth index of the mycelia. Among the various solid matrixes, the granitic soil, perlite and mixture of the two supported the growth most. Barely flour appeared to be very effective on the stimulating of the mycelial growth when added to the solid matrix. An mixture of the matrix contained an even (1:1:1:1, v/v/v/v) mixture of granitic soil, perlite, vermiculate and pine sawdust. T. matsutake started growth 2 weeks after inoculation and reached stationary growth phase after 8th weeks in the solid matrix mixture. The mycelial density in the solid matrix was 7 times higher than that in fairy-ring soil. In addition, 30~70% water content and 10% humus soil in the solid matrix also supported good growth suggesting that T. matsutake needs humus soil for a nutrient sources. The solid matrix developed in the present study could be used to study physiological characteristics of T. matsutake as well.

Effects of Whey Protein Hydrolyzates Fractionated by Molecular Weight on the Growth of Bifidobacterium bifidum Bb-11 (분자량에 따라 분획된 유청단백분해물이 Bifidobacterium bifidum Bb-11의 생장에 미치는 영향)

  • This study was carried out to evaluate the effect of whey protein concentrate-80%(WPC-80) and whey protein isolate(WPI) on the growth of B. bifidum Bb-11. Whey proteins($\alpha$-lactalbumin, $\beta$-lactoglobulin) were digested with trypsin, then their hydrolyzates were separated into three fractions (>10,000Da, 3,000∼10,000Da, <3,000Da) by two-step ultrafiltration process with Centriprep 10 and Centricon-30. These three fractions by molecular weight were evaluate growth-promoting effects for the B. bifidum Bb-11. The results obtained were summarized as follows; The growth rate of B. bifidum Bb-11 tended to increase by supplementation of WPC-80 to basal medium, but decreased by supplementation of WPI. Two whey proteins were hydrolyzed by trypsin at 40$\^{C}$ for 6 hrs, and three fractions were collected by UF treatment and concentrated by Centricon-30. Collected concentrations of protein of F-I and F-II and F-III from $\alpha$-lactalbumin were 11.53mg, 7.79mg, and 5.21 mg and those of protein from $\beta$-lactoglobulin were 4.13mg, 5.30mg, and 9.351mg, respectively. Three fractions of $\alpha$-lactalbumin hydrolyzates promoted the growth rate of B. dbifidum Bb-11. Growth promoting activities of hydrolyzates(F-I and F-II) with molecular weight below 10,000Da were stronger than that of hydrolyzate(F-III) above 10,000Da. However, there was no significant difference between the hydrolyzate F-I and F-II. Three fractions of $\beta$-lactoglobulin hydrolyzates improves the growth rate of B.bifidum Bb-ll. The growth of B.bifidum Bb-ll was decreased after 24 hr incubation by supplementation of either F-II or F-III fraction compared to basal Whey medium, but maintained the enhancement by supplementation of F-I.

Correlation of Growth Performance with Total Nitrogen, Carbon and Nitrogen Isotope Compositions in the Xylem of Pinus koraiensis (잣나무 생장과 목질부내의 전질소, 탄소 및 질소 동위원소 조성과의 상관관계)

  • In this study, we investigated the relationship of tree-ring growth with total nitrogen content, and stable carbon and nitrogen isotopes from the core samples of Pinus koraiensis (35-year-old). Annual ring width showed significant positive correlations with the ${\delta}^{13}C$ (P=0.003). The total nitrogen content (P=0.024), and the ${\delta}^{13}C$ content was also correlated with total nitrogen content (P=0.038), indicating that the growth of P. koraiensis was stimulated as the contents of both ${\delta}^{13}C$ and total nitrogen were increased. On the other hand, the less the ${\delta}^{15}N$ content and the C/N ratio were, the larger the annual ring width was. Moreover the families with relatively better growth performance contained the higher levels of ${\delta}^{13}C$ in the xylem compared to other families. These results suggest that the ${\delta}^{13}C$ and total nitrogen contents are the important determinants in the growth performance of P. koraiensis.

Ergosterol Contents and Enzymatic Characteristics of Lentinula edodes During Culture and Fruiting Periods (표고 균주의 배양 기간과 자실체 발생 기간에 따른 에르고스테롤 변화와 효소적 특성)

  • Three different strains of Lentinula edodes, Sanlim 5-Ho, Sanlim 6-Ho and Nongki 3-Ho, were cultured in the sawdust media of Mongolian oak(Quercu mongolica Fisch) for 90 days under dark and light conditions(each 30 days) and fruiting period(30 days). Weight loss of sawdust media was determined after fungal cultures and the contents of ergosterol in fungal mycelia were quantified by HPLC analysis followed by solvent extraction. Compared with the two other fungal strains$(8\%)$, weight loss of Sanlim 5-Ho was slightly lowered to $7\%$. The level of ergosterol content, a parameter for fungal growth, was continuously enhanced in Sanlim 5-Ho for dark and light incubation periods. However, Sanlim 6-Ho and Nongki 3-Ho recorded the maximized fungal growth under light condition. In fruiting periods the ergosterol contents were lowered in the three strains. Intra- and extracellular enzymes during cultural and fruiting periods were also characterized. The activity of Mn-peroxidase and laccase, which are characteristics enzymes for white rot fungi as lignin degrading enzymes, were determined as a high level overall the periods. As cellulose degrading indicators, the activity of CMCase, avicelase, xylanase and glucanase were detectable in initial incubation period.

Development of Ingrowth Estimation Equations for Pinus densiflora in Korea Derived from National Forest Inventory Data (국가산림자원조사 자료를 이용한 소나무의 진계생장 추정식 개발)

  • This study was conducted to develop ingrowth estimation equations on Pinus densiflora found in Gangwon Province and in the center of Korean Peninsula, based on the National Forest Inventory (NFI)'s permanent sampling plot data. For this study, identical sampling plots in $5^{th}$ and $6^{th}$ NFI data were collected in order to identify ingrowth amounts for the last 5 years. Following two-stage approaches in developing the ingrowth estimation equations, the logistic regression model was used in the first stage to estimate the ingrowth probability. In the second stage, regression analysis on sampling plots with ingrowth occurrence was used to estimate the ingrowth amount. A candidate model was finally selected as an optimal model after a verification based on three evaluation statistics which include mean difference (MD), standard deviation of difference (SDD) and standard error of difference (SED). In results, a logistic regression model based on the number of sampling plot which did not result in ingrowth (model VI), was selected for an ingrowth probability estimation equation and exponential function including the species composition (SC) variable was optimal for an ingrowth estimation equation (model VII). The ingrowth estimation equations developed in this study also evaluated the estimation ability in various forest stand conditions, and no particular issue in fitness or applicability was observed.

Analysis of Growth Characteristics and Aboveground Carbon Storage for Zelkova serrata Artificial Forests in Gwangneung Experimental Forest (광릉시험림 내 느티나무(Zelkova serrata) 인공림의 생장특성 및 지상부 탄소저장량 분석)

  • This study was conducted to analyze the growth characteristics and aboveground carbon storage for old growth Zelkova serrata artificial forests (site1: age class IX, site2: age class VIII) in Gwangneung Experimental Forest. The trees were classified by crown classes for analyzing forest stand structure. The growth characteristics were analized through ringwidth increment by crown classes and stem analysis of dominant trees. There were a wide range of DBH (site1: 8~62 cm, site2: 14~40 cm) and height (site1: 8~26 m, site2: 12~26 m) distributions and revealed different growth characteristics by crown classes in both sites. The mean annual increment (MAI) of ringwidth for the last 5 years of dominant trees for site1 (3.3 mm) was higher than MAI of ringwidth of total growth period (2.3 mm) and MAI of ringwidth for the last 5 years of dominant trees for site2 (2.2 mm) was equal to MAI of ringwidth of total growth period (2.2 mm). Also, the growth increment of ringwidth by crown classes had significant differences between dominant tree and the others crown classes (p<0.01) in both sites. As a results of stem analysis of dominant trees in both sites, there were similar to their volume between site1 (1.106 $m^3$) and site2 (1.035 $m^3$). In spite of old age, the annual increment of volume has been increasing steadily until recent year. Meanwhile, total aboveground carbon storage of site1 (65.6 Mg C $ha^{-1}$) was higher than that of site2 (56.1 Mg C $ha^{-1}$). The proportion of dominant and co-dominant trees to total aboveground carbon storage was more than 90% and the greatest individual aboveground carbon storage by crown classes was dominant tree in all both sites. However, individual aboveground carbon storage of dominant tree in site1 had 0.054 Mg C $tree^{-1}$ more than site2 owing to the differences from average DBH of dominant trees by sites. We think that these results will contribute to the forest practice for Zelkova serrata artificial forests as a basic information.

Daily Shoot Growth Measurement of Zoysiagrass (Zoysia japonica) to Determine Mowing Interval (한국잔디(Zoysia japonica)의 깎기주기 결정을 위한 지상부 생육 조사)

  • Mowing is an important practice in golf course management. Mowing interval and height can affect net photosynthesis of turfgrass and finally shoot density, tillering, rooting and regrowth of turfgrass. The objectives of this study were to investigate the regrowth rate of zoysiagrass shoot under different mowing height for determination of proper mowing frequency. Recommended mowing interval were calculated by a rule that no more than 33% of the above ground height be removed in a single mowing. Daily shoot growth of zoysiagrass at 10 mm mowing height were 2.1~4.7 mm (July), 1.9~2.9 mm (August), 0.9~1.5 mm (September), and 0.6 mm (October). These results indicate that recommended mowing interval were 1.1~2.3 day for July, 1.7~2.4 day for August, 3.5~5.4 day for September, and 8.5 day for October, respectively. Daily shoot growth of zoysiagrass at 15 to 17 mm mowing height were 4.0~5.3 mm (July), 2.9~4.5 mm (August), 1.4~3.7 mm (September), and 1.3 mm (October). These results indicate that recommended mowing interval were 1.4~1.9 day for July, 1.7~2.5 day for August, 2.3~6.3 day for September, and 6.8 day for October, respectively. Daily shoot growth of zoysiagrass at 18 to 21 mm mowing height were 3.5~4.7 mm (July), 2.9~4.9 mm (August), and 1.5~1.8 mm (September). These results indicate that recommended mowing interval were 1.9~2.6 day for July, 1.8~3.1 day for August, and 5.9~7.0 day for September, respectively. Daily shoot growth of zoysiagrass at 50 mm mowing height were 4.6~4.9 mm (July), 5.0~6.5 mm (August), and 2.5~4.7 mm (September). These results indicate that recommended mowing interval were 5.1~5.4 day for July, 3.9~5.0 day for August, and 5.3~9.8 day for September, respectively.
