Genetic Transformation of Intact Potato Microtuber by Particle Bombardment (Particle Bombardment 방법을 이용한 인공 씨감자의 형질전환)

  • In vitro grown microtubers of potato (cv Jaju) were used for introduction of herbicide resistance gene using bombardment with DNA-coated particles. The apical shoot-tip area of newly sprouted microtubers were intensively bombarded. After bombardment, microtubers were germinated and transplanted in a greenhouse. Northern blot analysis indicated that bar gene was expressed in two plantlets. After 5 weeks of growing, commercial herbicide Basta was sprayed to screen the resistant plants. All untransformed potato plants died after 7 days while two transgenic plants survived.

Taxonomic identities of Artemisia capillaris Thunb. and A. scoparia Waldst. & Kit. (사철쑥과 비쑥의 분류학적 실체)

  • Artemisia capillaris Thunb. and A. scoparia Waldst. & Kit. are classified by their root shape, leaf size and capitulum size. However, these characteristics are not nearly as useful when attempting to identify the two species owing to the continuous variation in them. Another important taxonomic characteristics is the habit, whether it is the subshrubs or perennial herbs of A. capillaris Thunb. or the annual or biennial herbs of A. scoparia Waldst. & Kit.. All observed specimens and living individuals in Korea are subshrubs or perennial herbs, and annual or biennial examples are not found. Therefore, it is considered that A. scoparia Waldst. & Kit. is not distributed in Korea.

Development of Bactericidal Yeast Strain by Expressing the Leucocin A Gene on the Cell Surface of Saccharomyces cerevisiae (Saccharomyces cerevisiae세포 표면에 leucocin A유전자의 발현에 의한 항균활성 효모의 개발)

  • ln order to develop yeast cells that produce a bacteriocin on their cell surfaces, the 114 bp Leucocin A gene with stop codon was ligated into pYDl, an yeast vector. The recombinant DNA, pYDl-LeucoA was used to transform yeast (Saccharomyces cerevisiae) cells. Yeast cells harboring pYDl-LeucoA showed antibacterial activity against Bacillus subtilis. To confirm these bacteriocidal yeast cells possess the Leucocin A gone, PCR was performed with plasmid prepared from transformed yeast cells as a template and two Leucocin A-specific primers. In this study, bacteriocidal yeast cells that can be used as an antibiotic or a food preservative were developed.

The Selection Alisma plantago Varieties Suitable for the Southern Part of Korea (남부 지역에 적응한 택사의 품종선발)

  • In order to obtain basic information for breeding varieties of Alisma plantago which is suitable for the southern part of Korea, nine local varieties were grown and yield components of plant were observed from Jun. 1999 to Dec. 1999 at farm field of Chonnam Sunchon Korea. Alisma plantago cv. Sunwol showed higher dry root yield than the other varieties used in the experiment. It showed relatively higher length of plant height and lower numbers of floral axis. Therefore, it was concluded that Sunwol was the most suitable variety with high yield and low floral axis at the southern part of Korea. The heritabilities of all observed characters were estimated to be large. Dry root yield was shown highly significant positive correlations with plant height and numbers of leaves. Dry root yield, plant height and numbers of leaves were shown negative correlation with number of floral axis and nursery leaves.

Characteristic of Progeny in Pepper Transformants (고추 유전자변형체 후대 생육특성 검정)

  • For the resistance test for Phytophthora blight of $T_1$ and $T_2$ transformants in pepper, Phytophthora blight fungus was inoculated to seedlings of the $T_1$ and $T_2$ transformants by concentration (density: zoospore $10^3/ml$). Occurrence rate of blight at 5days after inoculation was 4.0 % in T1-1 line and 10.0% in $T_1-2$ line, and its rate for 12 days after inoculation was 52.0% in $T_1-1$ line, 64.0% in $T_1-2$ line, respectively. Therefore, the lower occurrence rate to blight was enable to select resistant transformants in the some inoculation density (zoospore $10^3/ml$), meanwhile 'Kumtap' and 'Subicho' were 100% in highest occurrence rate to blight. For field test, in which blight was commonly occurred, of the Youngyang Pepper Experiment Station, the acquired transformant resisting to blight was similar to characteristics of domestic varieties, 'Subic ho' for fruit shape, but there are some differences in growth, days to flowering, fruit characteristics. Occurrence of blight in $T_2-1-6$, and $T_2-4-9$ lines was smaller approxmately 30% than commercial varieties, 'Kumtap', although occurrence of blight in field was showed higher difference among tested lines. In this study, we concluded that the transformants showing blight resistance selected from habitual field could be fixed at every generation, and the developed transformation system was also considered to develop transformants in pepper.

Combining Ability Test of F1 Generation by Diallel Cross in Kidney Bean (이면교잡에 의한 강낭콩 F1 세대의 조합능력 검정)

  • The studies were conducted to obtain basic informations on inheritance of some quantitative characters in kidney bean (Phaseolus vulgaris L.). Seven parents (Felibon, Renka, Processor, Kaboom, $BO_{22}$, Local variety $\#1$ and Local variety $\#2)$ and $F_1$ hybrids of 21 crosses from a set of diallel cross among varieties were used to estimate combining ability for eight agronomic characters such as days to flowering, days to maturity, stem length, pod numbers per plant, pod length, grain numbers per plant, 100-grain weight and grain weight per plant. General combining ability (GCA) and specific combining ability (SCA) were significantly different among all characters, and values of GCA were greater than those of SCA in all characters except number of grains per plant. In effect of GCA, Felibon expressed high GCA effect for days to flowering and number of grains per plant. Local variety $\#1$ showed high GCA effect for 100-grain weight and stem length. Local variety $\#2$appeared to high GCA effect for 100-grain weight and grain weight per plant. Processor expressed high GCA effect for days to maturity and grain weight per plant. Kaboom showed high GCA effect days to flowering and days to maturity. $BO_{22}$ appeared high GCA effect for all characters except for days to flowering, days to maturity and stem length. In the SCA effect, crosses of Felibonx Renka and Local variety $\#2{\times}Processor$ exhibited in high negative effect for days to flowering. Thus, these crosses were evaluated to useful for breeding early maturing variety. Crosses of breeding high yield variety were considered of $Felibon{\times}Local$ variety $\#1$, $Felibon{\times}Renka$ and Local variety $\#2{\times}BO_{22}$.

Classification of Garlic Germplasms Based on Agronomic Characteristics and Multivariative Analysis (마늘 유전자원의 작물학적 특성과 다변량 분석에 의한 품종군 분류)

  • This research was conducted to investigate the genetic diversity and select useful accession with agronomic characteristics of garlic (Allium sativum L.). germplasms at Garlic Research Institute in Chungbuk Agricultural Research and Extension Service. Morphological diversity and relationships among 160 germplasms collected from 26 countries were assessed by methods of clustering and principal component analysis. Among 11 types of leaves and bulbs characteristics, emergence days of leaf showed the highest variation with coefficient of variation of 84.8%, and the bulb weight and the number of scales showed higher variability with 24.3%. Correlation analysis based on 11 quantitative traits showed that bulb weight and bulb length have very high positive correlation with bulb quantity. Plant height, leaf length, and number of leaves showed positive correlation with bulb weight as collections with better performance in growth produced large bulb with higher quality. The cluster analysis based on 5 principal components generated 6 clusters with an average distance of 1.6 among clusters. Domestic genetic resources were the largest with 36 species (22.5%) in group II.

Effects of Planting Density and Fertilizer Level on Agronomic Characteristics and Yield of Zingiber mioga ROSC (재식밀도와 시비량이 양하의 주요형질과 수량에 미치는 영향)

  • The experiment was carried out to study the influence of planting densities and fertilizer application levels on some agronomic characters and yield of flower-head in Zingiber mioga ROSC. The results obtained are summarized as follows ; Plant hight was long in dense planting, and short in spacious planting. Stem diameter was thick in spacious planting, and was thin in dense planting. Flower-head weight per plant was decreased in dense planting and was increased in spacious planting. Yield was higher in dense planting densities (10$\times$20 : 50plants /m$^2$, 20$\times$20 : 25plant /m$^2$). Fertilizing (N : P$_2$O$_{5}$ : $K_2$O=15 : 15 : 15kg /10a) was increased than without fertilizing because plant height and number of stem was good.d.

Effects of Mulching Materials on Growth and Yield of Atractylodes macrocephala Koidz (피복재료(被覆材料)가 백출(白朮)의 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • This study was conducted to investigate the soil mulching effect on growth and yield of Atractylodes macrocephala Koidz. The number of stem, stem diameter, and fresh weight of aerial part in black poly-ethylene film and transparent poly-ethylene film mulched condition were higher than those of non-mulching and rice-straw mulching. Number of flowers were high in the order of non-mulching, rice-straw, trans-P. E. film, and black P. E. film. Yield of underground part was higher in mulching condition than that of non-mulching. Dry rhizome yield in black P. E. film mulching was significantly high to compare with the other treatments. There was lower disease incidence of Phytophthora blight in trans-P. E and black P. E. film compare with the other treatments but any difference was not observed in essential oil content of rhizome. Significant positive correlation was found between rhizome weight and fresh weight of aerial part. It was also shown that the fresh weight of aerial part was greatly dependent on the number of stems and stem diameter.
