Species Identification and Developmental Biology of a Red Bean Pest in Ostrinia sp. (Lepidoptera: Crambidae) (Ostrinia속(나비목: 포충나방과) 팥 해충의 종 동정과 발육 특성)

  • Ostrinia larvae feed the pods and stem of red bean and seriously damage the bean production from farmers. In this study we investigated biological and developmental characteristics including field collection, host feeding preference, artificial rearing diet, morphological and molecular taxonomical identification, and pheromone analysis for an Ostrinia sp. in Korea. The male adults have massive tibia in the middle legs and 3-lobed uncus in the genitalia. The partial nucleotide sequences of mitochondrial cytochrome oxidase I (COI) and II (COII) were not corresponded to those DNA sequences from other Ostrinia species reported previously in Japan and China. Host plants for this species are also different from the previous species reported. In the gas chromatography (GC) analyses, (Z)-9-tetradecenyl acetate was not detected from the pheromone gland of our species while the component as a sex pheromone was found in O. zaguliaevi and O. zealis, With taken results, we conclude this Ostrinia species in Korea is Ostrinia scapulalis or closely related species. When larvae collected in a fall were incubated in the outdoor condition, they emerged to adult between June and July in the next year. The result indicates that the winter diapause could be started in late larval stage. In addition, we developed a semi-synthetic artificial diet adopted for mass rearing of the O. scapulalis in laboratory.

  • 본 연구는 수도작에서 토양과 식물체의 분광반사특성과 영상처리를 이용한 기계시각 잡초검출 센서를 개발하기 위한 기초연구로서 분류하고자 하는 대상체들의 분광반사율을 조사하여 주요한 파장을 선정하고 선정된 파장을 이용한 판별분석을 통해 각 대상체에 대한 분류 정확도를 중심으로 잡초검출 가능성을 조사하기 위하여 수행하였으며, 실험으로부터 얻은 결론은 다음과 같다. 1. 토양과 식물체를 구분하는데 효과적인 파장은 마른 토양의 경우 680 nm, 배수 토양에 있어서는 810 nm로 선정하였고, 토양을 배제한 후 벼와 잡초를 구분하기에 효과적인 파장은 580, 680 nm로 선정하였다. 2. 토양과 식물체를 구분하기 위한 판별분석 결과 2가지 토양상태 모두 식물체와 완전히 구분 가능한 것으로 나타났다. 벼와 잡초를 구분하기 위한 실험에서, 벼는 98%의 분류정확도로 구분이 가능하였고, 잡초는 83%의 분류정확도로 구분이 되는 것으로 나타났다. 따라서 차후 분광학적 원리를 이용한 센서를 제작할 때 본 연구에서 선택한 주요 파장과 판별함수를 이용하여 장치를 구성하고 알고리즘을 제작한다면 벼, 잡초, 토양을 효과적으로 구분이 가능할 것으로 판단되었다. 3. 컬러 CCD 카메라를 사용하는 경우에 있어 식물체와 토양을 구분하기 위해 3 종의 파장 중 630 nm 파장만의 이용을 고려하여 그 분류성능을 분석한 결과, 식물체와 토양은 소수의 관측치를 제외하고 완전히 구분이 가능했고, 벼와 잡초를 구분한 결과에서는 비교적 높은 분류능력을 가진 것으로 나타나 차후 컬러 CCD 카메라를 이용하여 장치를 구성하는데 좋은 기초가 될 것으로 판단된다. 배양체의 접종작업은 모든 배양실이 인력에 의존하였으며, 배양체를 배지와 분리하여 불필요한 부분을 제거하고 배양작물에 따라 생육정도를 2~3등급으로 구분하여 배양용기의 배지 위에 치상하는 과정으로 수행되었으며, 작업능률은 호접란의 경우 배양병에 25본을 접종하는데 시간당 6병, 심비디움은 원형 플라스크에 25본을 접종하는데 시간당 10병 정도였다. 바. 식물체의 대량증식에 사용되는 플라스크, 배양병, PE용기 등 배양용기의 세척작업은 농원의 1개배양실에서 간이식 세척기, 이 외의 9개배양실은 모두 물에 담겨 두었다가 세제와 브러쉬 등을 사용하여 인력으로 세척하고 있어 생력화 기술개발이 요구되었다.도가 빠를수록 건조속도가 빨라졌으며, 건조에너지도 1,334kcal/kg.water로 비슷하게 소요되었다. 마. 시험구와 대비구의 건감률은 시험구에서 1.08~1.36w.b./h로 나타나 대비구보다 약 9.9~18.3%가 높게 나타났고, 건조에너지는 10.2~14.6%가 절감되었다. 발아율은 열풍온도가 낮을수록 높게 나타났고 시험구가 대비구보다 발아율이 낮게 나타났으며, 동할률 증가량도 원적외선.열풍 복합건조방법이 높게 나타나 이것은 곡물 표면에 원적외선 방사에의한 복사열이 전달되어 열장해를 받았기 때문으로 판단되며, 금후 더 연구하여 적정 열풍온도 및 방사체 크기를 구명해야 할 것이다.으로 보여진다 따라서 옻나무 유래 F는 포유동물의 생식기능에 중요하게 작용하는 것으로 사료된다.된다.정량 분석한 결과이다. 시편의 조성은 33.6 at% U, 66.4 at% O의 결과를 얻었다. 산화물 핵연료의 표면 관찰 및 정량 분석 시험시 시편 표면을

  • Anther is the major organ of flower in responsible to reproduction and outward appearance. From anther-specific cDNA library of Lilium Oriental Hybrid 'Acapulco', 2000 expressed sequence tags were selected randomly. Differential slot blot analysis with cDNA probes from the anther and leaf was used to get anther-expressed clone and 570 non-redundant ESTs were obtained and sequenced. Compared to the GenBank database using BLASTX algorithm, 191 clones showed significant similarity but others (66.5%) did not measured to known sequence. Functional categories according to gene ontology (GO) annotation included sequence representing a significant portion of protein in cell and cell part respectively. A transcriptional analysis at 7 different organs and developmental stage was performed using northern blot with thirty ESTs as putative anther specific gene. This report suggest that selection of anther expressed clone using differential slot blot was considered as very effective tool and our current study can provide fundamental information on the lily anther including pollen furthermore.

Effects of Changes of Nutrient Solution Concentration According to Growth Stage on Growth and Flowering of Cut Chrysanthemum Grown Hydroponically in Perlite (국화의 펄라이트 양액재배시 생육단계에 따른 양액농도의 변화가 생육과 개화에 미치는 영향)

  • This study was carried out to investigate the effects of changes of ionic strength according to growth stage on growth and flowering of Dendranthema grandiflorum (Ramat.) Kitamura 'Seiun' grown hydroponically in perlite. The stage I, II, and III covered early vegetative growth (27-40 days after planting), latter vegetative growth (41-54 days), and reproductive growth (55-80 days), respectively. The 2 strength (1S and 2S) of nutrient solution were treated in stage I, whereas 3 strengths (1S, 1-2S, and 2S) were treated in stage II. Then, total 9 treatments in stage III were designated by 3 treatments (tap water, 1S, and 2S) for each 3 strengths in stage II. Each nutrient solution was applied 8 times per day. At vegetative growth stage (54 days after planting), stem length was highest when irrigated 8 times a day with 1S nutrient solution. Both photosynthesis and transpiration rate were higher in 1S than those in other treatments (1-2S, 2S), whereas leaf chlorophyll content was highest in 2S treatment. Ion content of plant treated with 2S was higher than other treatments. Growth (plant height, leaf area, stem length), fresh weight, and dry weight of each plant organ after flower bud formation were better in tap water treatment (1-1-0) than other 1S treatments (1-1-1, 1-1-2). Regarding the number of days to flowering, tap water treatment was the most effective. Thus, after flower bud formation supplying tap water or lower concentration of nutrient solution than those used during the vegetative growth stage was economical in saving chemical fertilizers, shortening the number of days to flowering, reducing salt accumulation in media, saving efforts of leaching, and reducing ground water contamination.

Time-based Expression Networks of Genes Related to Cold Stress in Brassica rapa ssp. pekinensis (배추의 저온 스트레스 처리 시간대별 발현 유전자 네트워크 분석)

  • Plants can respond and adapt to cold stress through regulation of gene expression in various biochemical and physiological processes. Cold stress triggers decreased rates of metabolism, modification of cell walls, and loss of membrane function. Hence, this study was conducted to construct coexpression networks for time-based expression pattern analysis of genes related to cold stress in Chinese cabbage (Brassica rapa ssp. pekinensis). B. rapa cold stress networks were constructed with 2,030 nodes, 20,235 edges, and 34 connected components. The analysis suggests that similar genes responding to cold stress may also regulate development of Chinese cabbage. Using this network model, it is surmised that cold tolerance is strongly related to activation of chitinase antifreeze proteins by WRKY transcription factors and salicylic acid signaling, and to regulation of stomatal movement and starch metabolic processes for systemic acquired resistance in Chinese cabbage. Moreover, within 48 h, cold stress triggered transition from vegetative to reproductive phase and meristematic phase transition. In this study, we demonstrated that this network model could be used to precisely predict the functions of cold resistance genes in Chinese cabbage.

Development of PCR-based markers specific to Solanum brevicaule by using the complete chloroplast genome sequences of Solanum species (엽록체 전장유전체 비교를 통한 PCR 기반의 Solanum brevicaule 특이적 분자마커 개발)

  • Solanum brevicaule is one of the tuber-bearing wild Solanum species. Because of its resistance to several important pathogens infecting potatoes during cultivation, it can be used for potato breeding. However, the fact that S. brevicaule used in this study has an EBN value of two causes the sexual reproduction barriers between the species and cultivated potatoes. In this study, specific markers for discriminating S. brevicaule from other Solanum species were developed on the basis of the results of sequence alignments with the whole chloroplast genomes of S. brevicaule and seven other Solanum species. The chloroplast genome of S. brevicaule was completed by next-generation sequencing technology described in other recent studies. The total sequence length of the chloroplast genome of S. brevicaule is 155,531 bp. Its structure and gene composition are similar to those of other Solanum species. Phylogenetic analysis revealed that S. brevicaule was closely grouped with other Solanum species. BLASTN search showed that its genome sequence had 99.99% and 99.89% identity with those of S. spegazzinii (MH021562) and S. kurtzianum (MH021495), respectively. Sequence alignment identified 27 SNPs that were specific to S. brevicaule. Thus, three PCR-based CAPS markers specific to S. brevicaule were developed on the basis of these SNPs. This study will facilitate in further studies on evolutionary and breeding aspects in Solanum species.

  • The Pseudogobio esocinus were caught at Wyuleo-ri, Gyeombaek-myeon, Boseong-gun, Jeollanamdo from April to May 2003. The fishes were incubated in transparent aquarium located at the laboratory of Chonnam National University, and their embryonic and larval development were observed. The fertilized eggs were spherical, semitransparent, and adhesive, and were $1.98{\pm}0.19mm$ (n=50) in diameter. The embryo, including 31$\sim$32 myotomes, hatched through egg membrane at 164 hrs after fertilization. The newly-hatched larvae were $4.61{\pm}0.83mm$ (n=10) in total length (TL). At that moment, yolk was not absorbed, and mouth and anus were not open. Star and spot shaped melanophores were distributed on the lens, and dorsal, ventral, and caudal parts. At 42 days after hatching, larva was $16.22{\pm}0.65mm$ (n=10) in TL. Melanophores were scaterred at head, back, and side parts. Morphological features of the embryo were transferred to juvenile stage showing similar features with those of the adult fish.

Occurrence of Regalecus russellii off the Coast of Gangwon-do, Korea and Coastal Environment (강원도 속초 연안에서 산갈치(Regalecus russellii) 출현과 연안환경)

  • Regalecus russellii, which spends most of its life in the deep sea, occasionally appears in coastal waters. However, the reasons for its appearance remain unclear. In Korea, R. russellii mainly appears along the eastern coastal waters, and most of them are caught in fishing gear, such as gill nets, or are stranded on the shore; nevertheless, the frequency of appearance is extremely low. Even if found, this species is often identified to be morphologically similar to Trachipterus ishikawae, and comprehensive analysis to identify the species through sample collection is limited. Consequently, information on the biological characteristics of R. russellii appearing in the coastal waters of Korea is scarce. Herein, the anatomical characteristics of R. russellii caught in a gill net off the Gangwon-do coast on March 14, 2023, were analyzed, and coastal water temperature was measured using an ocean buoy. Our results showed that the individual was male, its total length was 320 cm, body weight was 27.52 kg, body length was 26.62 cm, gonad weight was 619.45 g, and liver weight was 218.71 g. The stomach was full of euphausiids. The water temperature changed drastically at 15-30 m roughly a week before the R. russellii individual was caught, and the subsurface water temperature was lower than 10 ℃. Our findings provide baseline data to understand the ecological characteristics of R. russellii appearing along the eastern coast of the Korea.

Effects of Progesterone (P4), 17β-estradiol (E2), Melatonin and Serotonin (5-HT) on the mRNA Expression of Reproduction-related Genes in the Pituitary Cells of Eels (Anguilla japonica) (뱀장어(Anguilla japonica) 뇌하수체 세포의 번식 관련 유전자 mRNA 발현에 미치는 Progesterone (P4), 17β-estradiol (E2), Melatonin 및 Serotonin (5-HT)의 영향)

  • Fish reproduction is regulated by various neurohormones secreted from the brain and gonadotropic hormones secreted from the pituitary. Reproduction of eel (Anguilla japonica) is also regulated by these hormones. However, how the neurohormones regulate the secretion of pituitary hormones during sexual maturation is not completely understood. Previous studies have shown that neurohormones such as progesterone (P4), melatonin and serotonin (5-HT) are involved in the regulation of reproductive processes in some fish. In this study, the eel pituitary was primary cultured, and stabilized pituitary cells were treated with P4, 17β-estradiol (E2), melatonin, or 5-HT. The effect of these treatments on the expression of FSHβ, LHβ, GH and SL mRNA was, then, investigated. P4 increased the expression of FSHβ and LHβ in pituitary cells, and melatonin increased the expression of GH and SL as well as FSHβ and LHβ. However, 5-HT did not significantly affect the expression of these mRNA. These results suggest that P4 and melatonin may play some important roles in the early sexual maturation of eels.

Production of Transgenic Murine Embryos using Haploid Spermatids Transfected with EGFP Gene (EGFP 유전자가 도입된 반수체 정자세포에 의한 형질전환 설치류 난자의 생산)

  • In this study, the production of transgenic embryo was attempted by microinjection or round spermatid cultured with foreign DNA. At first, the expression of haploid spermatids specific gene, mTP1 in mouse and hPrm2 in hamster spermatids were investigated by RT-PCR method in testes of young mice and hamster testis. The specific gene expression first appeared at 18 days post partum (dpp) in mice spermatid and 20 dpp in hamster spermatid. Therefore, the round spermatids isolated from 17 dpp mice and 19 dpp hamster were used for the introduction of foreign EGFP gene into haploid round spermatids. For the introduction of EGFP gene haploid round spermatids suspended in medium including EGFP gene were treated with a different electric field strength at 0.11, 0.18 and 0.44 ㎸/cm. After electrical stimulation, viability of testicular sperm cells and 67.6%, 66.4% and 49.9%, in mice and 62.6%, 57.9% and 27% in hamster, respectively. These values were significantly lower than those of non-treated control groups 80.5% in mouse and 69.1% in hamster After 72 hrs culture, the highest expression rate of EGFP gene, 28.5% in mice and 32.1% in hamster were obtained from tile spermatogenic cells electroporated by the field strength or 0.18 ㎸/cm. Then, the ability of fertilization and embryonic development of haploid spermatids transfected with foreign EGFP gene were estimated by the microinjection of spermatids into hamster oocytes. The Irate pronuclear formation rate (77.5%) was lower than non-treated control (80%), and the cleavage rate of the treated group (58.8%) was lower than control (65%). To prove the foreign EGFP integration in hamster embryos, 2-cell stage hamster embryos were subjected to the observation under the fluorescence microscope, and the PCR analysis. As a result, about 44% of 2-cell embryos were showed the integration of EFGP gene into their genome. Therefore, These results suggest the possibility to produce transgenic hamsters by microinjection of haploid spermatid transfected with foreign DNA.
