In Vivo Embryo Production and Embryo Transfer in Hanwoo and Jeju Black Cattle Using CIDR (CIDR를 이용한 제주 한우 및 흑우의 체내 수정란 생산과 이식)

  • The objectives of this study was to produce valuable offsprings of Hanwoo and Jeju black cattle using in vivo embryo production and embryo transfer techniques during 5 years ($2001{\sim}2005$) in Jeju. Two hundred and eighty six Hanwoo and sixty nine Jeju black cattles, at random stages of the estrous cycle, received a CIDR. Seven days later, the animals were superovulated with a total of 400 mg pFSH ($Folltropin^{(R)}-V$) administered twice daily in constant doses (each 50 mg) over 4 days. On the 6th administration of FSH, CIDR was withdrawn and 25 mg $PGF_{2{\alpha}}$ was administered. Cows were artificially inseminated thrice after estrous detection at 12 hr intervals. The cows received $250{\mu}g$ GnRH at the time of 2nd insemination. Embryos were recovered 7 or 8 days after the 1st insemination. Recipients were synchronized with donors by insertion of a CIDR for 7 days and administration of 25 mg $PGF_{2{\alpha}}$ at the time of CIDR removal. The collected embryos were transferred to 1,219 recipients by 6 transfer persons. The mean numbers of total ova and transferable embryos from Hanwoo and Jeju black cattle donors were 7.4 and 4.7, respectively The number of transferable embryos differed between Hanwoo (5.0) and Jeju black cattle (3.5, p<0.05), while that of total ova did not differ. Repeated superovulation treatments decreased (p<0.05) the ratio of numbers of the flushed animals vs. superovulated animals in Jeju black cattle, and the numbers of total ova and transferable embryos in Hanwoo. More transferable embryos were collected at summer (5.6) than winter (2.9, p<0.01). The mean pregnancy rate was 40%. The pregnancy.ate was affected by transfer year (2001<2004, p<0.05) and transfer person ($33.0{\sim}41.9%$, p<0.01), while not by donor (embryo) breed. These results showed that in vivo embryo preduction and embryo transfer techniques using CIDR for Hanwoo and Jeju black cattle donors as well as recipient, regardless of their estrous cycle, may enable a stable embryo production and recipient preparation.

Production of Transgenic Porcine haboring the Human Erythropoietin(EPO) Gene (사람 조혈인자 유전자(Human Erythropoietin Gene)를 도입한 형질전환돼지 생산)

  • This study was performed during the four seasons for the production of transgenic pigs containing the human erythropoietin(hEPO) transgene. Purebred Landrace gilts and sows approximately 8∼15 months of age (n=42) were used fur the collection of 1-cell zygotes for DNA microinjection and transfer. Retrospectively, estrus synchronization and superovulation schemes were evaluated to assess practicality for zygote collection. Synchronization and superovulation procedures were used that cyclic gilts were synchronized with 20mg altrenogest (ALT) per day for 9days after PG600 administration followed by superovulation with 1500IU pregnant mares serum gonadotropin (PMSG) and 500IU human chorionic gonadotrophin (hCG). Preparation of recombinant gene for microinjection is mice whey acidic protein promoter (mWAP) linked to human erythropoietin (hEPO) gene. After hormone treatment, 650 embryos were collected from 23 donors and 83.1% (540/650) embryos were in 1-cell stage which can be visualized the pronuclei for DNA microinjection. A total of 543 DNA microinjected embryos fiom donors were transferred to 19 synchronized recipients, seven of them maintained pregnancy and delivered 47 piglets. One of the 47 offsprings were determined to have transgene by PCR analysis. The overall rate of transgenic production was 2.13% (tansgenic/offspring). This study provides the success and useful information regarding production of transgenic pig for bioreactor research.

  • In the era of the 4th Industrial Revolution, manufacturing is the implementation of smart factories through digital transformation, and refers to consumer-centered intelligent factories that combine next-generation digital new technologies and manufacturing technologies beyond the existing factory automation level. In order to successfully settle such a smart factory, it is necessary to train professionals. However, education for smart factories is difficult to have actual field mechanical facilities or overall production processes. Therefore, there is a need for a system that can visualize and control the flow and process of logistics at the actual production site. In this paper, the logistics flow of the actual site was implemented as a small FMS, a physical system, and the production process was implemented as a digital system. In real-time synchronization of the physical system and the digital system, the location of AGV and materials, and the process state can be monitored to see the flow of logistics and process processes at the actual manufacturing site. The developed digital twin system can be used as an effective educational system for training manpower in smart factories.

Variation of Rice Production for Two Decades before and after Breeding Tongil Variety in Korea (수도 통일품종 육성보급 전후 20년간의 생산성 변이)

  • The variability of rice productivity during last 2 decades (1961-1980) of ten years before and after the introduction of"Tongil" was reviewed from the epochal, regional and varietal points of view. During that period the cultivated area of paddy rice have remained almost unchanged, while the total rice production have got elevated from 3, 463 million metric tons in 1961 to 6.006 million metric tons in 1977, recording 73.4% increase. This remarkable increase in rice production is considered to be attributable much to the development and release of new high yielding variety, "Tongil", coupled with the amelioration of cultural techniques. However, in 1978 Tongil type varieties experienced the epidemic outbreak of blast disease due to the shifted race population of blast fungus and in 1980 recorded poor rice production as low as in 1960's due to the unfavorable weather stress throughout the rice growing season, giving rise to many problems awaiting solutions for securing the stabilized high production of rice. The rice yield has continued the gradual increase during last two decades but its difference between farmer and research organization have got wider from 79kg/10a during 1960 to 1971 to 101kg/l0a during 1972 to 1980, and also the inter-regional differences have been increased from 50-60kg/10a to 80kg/10a during those periods. Therefore, this proves that we have raised the upper boundary of rice yield by increasing the yield potential of rice variety but have not changed those absolute deviations. Estimates indicate that the increased rice production during that period was indebted 40 percent to the varietal improvement and 13 percent to the ameliorated agro-technologies, and the rest, 47 percent, could be ascribed to the other factors besides varieties and cultural technologies such as the improved agricultural environments, etc. Of course, even though it cannot be expected to unify the cultural environments and the cultural technologies, provided that much efforts are to be endeavored to minimize the yield difference of 20 percent between farmer and research organizations and the inter-regional yield difference of 20 percent, much increased rice production can be expected to be achieved with the current level of cultural technology and the yielding potential of the present rice varieties. In order to expedite the above effects on rice production the followings are to be put into practices consitently and steadfastly. 1. Reinforcement of breeding for varieties with high yielding potential and less susceptible to climatic-stress and pests, and of basic physicoecological studies of rice plant for improving the cultural technologies. 2. Continuous endeavor to secure the stabilized cultural environments by improving the soil fertility and increasing the drainage and irrigation facilities. 3. Political back-up to encourage the farmers' incentives for production 4. Precise surveys for agricultural statistics to facilitate the long-term planninge long-term planning.

섬유소 분해효소 유전자 도입 형질전환 돼지 생산

  • 본 연구는 섬유소 분해효소 유전자 Cel D(Cellulase Digestion)가 도입된 형질전환 돼지 생산을 통하여 섬유소 함유 사료의 이용 효율을 증대시키고, 나아가 장기이식동물 및 고가의 의료용 단백질 생산가축을 개발하기 위한 원천기술 확보에 있다. 섬유소분해 유전자의 크로닝 및 조직 특이적 발현벡터를 개발하기 위하여, 우선 소의 위중 제4위 내의 미생물로부터 전체 유전자를 분리하였고, 이렇게 작성된 DNA library에서 섬유소분해 관련 유전자인 약 2.0 kb의 Cel D유전자를 크로닝하였으며, 췌장 특이적 발현 프로모터(rat elastase I: 약 200bp)를 크로닝한 후, 미세주입용 형질전환 재조합 벡터를 구축하기 위하여 rElastase I 프로모터 하류에 섬유소 분해 유전자(Cel D)를 연결하여 약 3.0 kb 크기의 재조합 벡터를 준비하였으며, 재조합 유전자를 1세포기 수정란 전핵내에 미세주입 하기 위해 Sal I과 BglII를 이용 유전자 단편을 만들었다. 구축된 유전자를 미세주입하기 위한 수정란을 회수하기위해 총68두의 돼지를 4-5두씩 분리사육하면서 발정동기화 및 과배란 유기를 위해 PG600, Altrenogest, FSH, hCG를 투여하였으며 hCG투여후 약54시간에 외과적 방법에 의해 총 1,359개의 수정란을 회수하였고, 이중 미세주입가능한 1세포기 수정란은 1,296개로 두당 평균 15.9개 였다. 1,296개의 1세포기 수정란 중에서 재조합 유전자(rE I-CelD)가 미세주입된 660개의 수정란을 32두의 수란돈에 외과적 방법에 의해 이식하였으며, 이식되어진 모돈 13두가 분만하여 40.6%의 임신율을 나타내었다. 이렇게 분만된 13두에서 총 65두(암:33두, 수:32두)의 자돈이 생산되었으며, 형질전환 여부를 판명하기 위해 자돈의 꼬리조직으로 부터 genomic DNA를 추출하고 PCR 검정을 실시하였다. PCR 검정 결과, 섬유소 분해 유전자가 도입된 자돈은 5두 이었으며, 그 결과를 Table에 나타내었다.(Table Omitted) Table 1 에서와 같이 섬유소 분해효소유전자가 형질전환된 자돈은 65마리 중 5마리로 7.69%의 형질전환율을 나타내었으며, 5마리의 자돈중 2두(암:1두, 수:1두)는 분만 후 즉시 폐사되었으며 2두(암:1두, 수:1두)는 86일령 그리고 14일령에 폐사하여 현재 1두(암)가 생존하여 섬유소 분해 사양 실험 중에 있다.

Retrospectively, estrus synchronization and superovulation schemes were evaluated to assess practicality fur zygote collection. Synchronization and superovulation procedures were used that cyclic gilts were synchronized with 20mg altrenogest (ALT) per day for 9 days after PG600 administration followed by superovulation with 1000 IU pregnant mares serum gonadotropin (PMSG) and 750IU human chorionic gonadotrophin (hCG). The cellulase digestion gene for microinjection is rat elasterase promoter (rEl) linked to CelD gene. After hormone treatment, 1,422 embryos were collected from 91 donors and 95.6% (1,359/1,422) embryos were in 1-cell stage which can be visualized the pronuclei for DNA microinjection. A total of 725 DNA microinjected embryos transferred into 35 recipients and produced 65 piglets from 13 litters. Pregnancy rate according to the number of transferred embryos to recipients was higher the group which received 21 to 24 embryos (50.0%) than other groups 20.0% in less and 33.3% in more. A tail tissue was collected from 65 piglets for biopsy. PCR screening was performed on each DNA sample using two separate sets of primers specific for the 5'- and 3'-flanking region of the rEl-CelD gene. Five of the 65 piglets (7.69%) were positive for the transgene. This study provide useful information regarding production of transgenic pig for bioreactor research.

Object Recognition and Target Tracking Using Motion Synchronization between Virtual and Real Robots (가상로봇과 실제로봇 사이의 운동 동기화를 통한 물체 인식 및 목표물 추적방안)

  • Motion synchronization between developed real and virtual robots for object recognition and target tracking is introduced. ASUS's XTION PRO Live is implemented as a sensor and configured to recognize walls and obstacles, and perceive objects. In order to create virtual reality, Unity 3D is adopted to be associated with the real robot, and the virtual object is controlled by using an input device. A Bluetooth serial communication module is used for wireless communication between the PC and the real robot. The motion information of a virtual object controlled by the user is sent to the robot. Then, the robot moves in the same way as the virtual object according to the motion information. Through motion synchronization, two scenarios, which map the real space and current object information with virtual objects and space, were demonstrated, yielding good agreement between the two spaces.

  • This paper introduces the secondary cultural productions of game fandom that have been underestimated. The participatory cultural activities of game users are broadly expanding owing to machinima technology based on game engine. It allows gamers to make the three dimensional digital animations simple and easy way. This paper studies that how simple game consumers change their role as producers through machinima. This study is focused on the relationship between polysemy of participatory Machinima and former fandom's the secondary culture studies using ethnographic research method. It is to clarify relationship between the game users' psychological motivation for culture practice and the machinimas. We verify practically the machinima's effects on fandom culture through analysing viewers' reaction using replies on the machinimas and then find out machinima is a very important tool for expanding the game culture realm as the main body.

  • These days, the number of automated container terminals is increasing to encourage the productivity of the container terminal inside and outside of the country. So far, there have been a lot of researches on the operating one type of equipments in a container terminal. However, there is still room for further improvement as none of equipment works itself but cooperate each other to finish a job, which means synchronization among the equipments is necessary. Among lots of equipments in a terminal, this paper concerns with the operation of ATCs (Automated Transfer Crane) and YTs (Yard Truck). The purpose of this paper is to find the efficient heuristic methods for operating ATCs and YTs that can set up a schedule in a real time. Moreover, using simulation this paper shows the efficient stacking strategy to decide the location of containers to be put and the proper selection range of YTs.

3D Object Location Identification Using Finger Pointing and a Robot System for Tracking an Identified Object (손가락 Pointing에 의한 물체의 3차원 위치정보 인식 및 인식된 물체 추적 로봇 시스템)

  • In this work, a robot aimed at grapping and delivering an object by using a simple finger-pointing command from a hand- or arm-handicapped person is introduced. In this robot system, a Leap Motion sensor is utilized to obtain the finger-motion data of the user. In addition, a Kinect sensor is also used to measure the 3D (Three Dimensional)-position information of the desired object. Once the object is pointed at through the finger pointing of the handicapped user, the exact 3D information of the object is determined using an image processing technique and a coordinate transformation between the Leap Motion and Kinect sensors. It was found that the information obtained is transmitted to the robot controller, and that the robot eventually grabs the target and delivers it to the handicapped person successfully.
