New series of Mn(II), Ni(II), Co(II), Cu(II) and Zn(II) of the 2-(benzothiazol-2-yl)-N'-(2,5-dihydroxybenzylidene) acetohydrazide have been synthesized and characterized by elemental analysis, IR, UV-vis, $^1H$-NMR, mass and ESR spectra, magnetic susceptibility and molar conductivity measurements. The spectral data and magnetic measurements of the complexes indicate that, the geometries are either square planar or octahedral. The biological activity of the ligand and its complexes against fungi (Aspergillus nigar and Fusarium oxysporium) were investigated. The metal complexes exhibited higher activity than both the parent ligand and the corresponding metal ion.
The design procedures for a biological reactor and a secondary settling tank (SST) of an activated sludge system are based on the steady state design method (Ekama et al., 1986; WRC, 1984) and the 1-D flux theory design method (Ekama et al., 1997), respectively. This study combined both of the design procedures, to determine the optimum sludge concentration in the reactor and the best design with the lowest cost. The best design of the reactor volume and the SST diameter at the optimum sludge concentration were specified with varying wastewater and sludge characteristics, temperature, sludge retention time (SRT) and peak flow rate. The effects of the influent wastewater characteristics, such as substrate concentration and unbiodegradable particulate fraction, were found to be considerable, but the effect of unbiodegradable soluble fraction was to be negligible. The effects of sludge settling characteristics, were also significant. SRT, as an operating parameter, was found to be an important factor for determining the optimum sludge concentration. However, the effect of temperature was found to be small. Furthermore, for designing a large scale wastewater treatment plant, the number of reactors or SSTs could be estimated, by dividing the total reactor volume or SST area. The new combined design procedure, proposed in this research, will be able to allow engineers to provide the best design of an activated sludge system with the lowest cost.
A streptomycete strain was isolated from the soil samples from Korea. The chemotaxonomy and 16S rDNA sequencing confirmed that the strain belonged to the genus Streptomyces and we named it Streptomyces sp. AO-0511. Two antibiotics, herbimycin A and dihydroherbimycin A produced by this strain were tested for their physico-chemical and biological characteristics. Both compounds were stable under acidic pH. Dihydroherbimycin A was more heat-stable and polar compared with herbimycin A. Only weak antibacterial activities were detected against Bacillus subtilus ATCC 6633 and Micrococcus luteus ATCC 9341. However, herbimycin A and dihydroherbimycin A showed strong inhibitory activities on lung cancer cells (A549 cells) and leukemia cells (HL-60). The cytotoxicity was determined using L5178Y and P388 cell lines. The results showed that herbimycin A and dihydroherbimycin A had lower toxic effects on the cells compared with the standard compounds, comptothecin and cyclosporin A. Therefore, both compounds could be good candidates for the development of new anticancer drugs.
The removal yield of dissolved organic matter in drinking water by biological activated carbon (BAC) process was investigated. The tested processes wer raw water-AC process (BAC1), raw water-ozonation-BAC process (BAC2), and raw water-ozonation-coagulation/sedimentation-BAC process (BAC3). The amounts of organic matter was measured as dissolved organic carbon (DOC), ulta-violet radiation at 254 nm wavelength ($UV_{254}$), total nitrogen (T-N), ammonia nitrogen (NH_3$-N), and total phosphate (T-P). As a results, 30.7% DOC was removed by BAC2 process, which showed higher removal efficiency than BAC1 or BAC3 processes. The removal yield of $UV_{254}$ in BAC1, BAC2, and BAC3 processes were observed as 45.3%, 44.6%, 58.4%, respectively. And the removal yield of ammonia nitrogen were 66%, 81%, 29% in each BAC processes. The optimal empty bed contact time (EBCT) of BAC processes was estimated as 10 minute. This study has shown that BAC process combined with ozone treatment was efficient for removing dissolved organic matter in water.
We isolated Actinomycetes strain GMT 1155 and purified the active compound, MT 1155, on the morphological reversion of ts/NRK cell from the isolate. MT 1155 was identified as toyocamycin having antifungal and antitumor activities from physico-chemical properties and UV, IR, $^1H$-NMR, $^13C$-NMR and mass spectrum. MT 1155 showed the morphologically reversional activity on ts/NRK cell and the cytotoxicity on CTLL cell at the final concentrations of 1.7 JlM and 0.2 11M, respectively and its $IC_{50}$ value on protein kinase A enzyme was 2.3 $\mu$M. Also it had strong antifungal activity against several pathogenic fungi but not antibacterial activity. And it did not inhibit both protein kinase C activity and the bleb-formation of K562 cell induced by phorbol esters.
This study was to investigate the physiological activities of bioconversion soybean embryo extract and the effects of sleeping pack containing extract on skin. The aglycone isoflavones of the soybean embryo extract bioconversed with pectinase were identified by HPLC. in vitro tests showed that they were non-toxic, anti-inflammatory, anti-melanin and anti-aging activity. After 4 weeks of human body application with the sleeping pack using extract, the skin surface roughness and eye wrinkles were so improved by low stimulation that the sleeping pack containing bioconversion soybean embryo extract has a positive effect on the skin. We confirmed that the bioconversion soybean embryo extract was effective in enhancing the physiological activity and was useful as a cosmetic material of various formulations.
The study was aimed to saccharify callulosic waste by pectinase produced from strain KL34 isolated from soil. The enzyme activity in the culture using 1%(w/v) fruit waste as carbon source reached to 3.8 U/ml. In the enzymatic hydrolysis of cellulosic waste, we obtained 9.5g/L reducing sugar in the condition of supernatant containing 5 U/ml enzyme and 10%(w/v) apple rind as substrate. Additionally, in enzymatic hydrolysis of food waste using pectinase from KL34, reducing sugar of 12.7g/L was obtained, indicating enhancement of 1.6 fold compared with that of only cellulase employment.

In vitro 상에서 골조직을 원활하게 재생하기 위해서는 3차원 지지체를 이용한 세포 배양과 세포 배양 시 세포의 형태와 기능을 유지/향상시키기 위한 인체 내 미세 환경 재현은 필수적이다. 따라서 본 연구에서는 뼈 성분과 유사한 생체 활성 물질인 hydroxyapatite (HA)와 생분해성 고분자인 poly $\varepsilon$-caprolactone (PCL)를 복합재료로 이용하여 내부 연결성이 우수한 골조직 재생용 3차원 지지체를 제작하였으며, 골 재생 능력 향상을 위하여 인체내 골조직의 기계적 미세 환경을 체외에서 구현한 새로운 형태의 perfusion bioreactor system을 개발/적용하였다. 또한 본 연구에서 개발된 perfusion bioreactor system의 생물학적 평가를 위해 MG63 (osteoblast like cell, 한국 세포주 은행)과 New Zealand White Rabbit에서 분리한 중간엽 줄기세포를 골조직 재생용 3차원 지지체에 파종하였다. 48시간 동안 안정화 후 perfusion bioreactor system을 이용하여 기계적 자극을 파종된 세포에 인가하였으며, 배양 기간 동안 세포의 증식 확인 및 형태학적 관찰을 실시하였다. 본 연구 결과, perfusion bioreactor system을 이용하여 기계적 자극을 인가한 실험군에서 세포의 증식 및 활성도가 대조군에 비해 우수함을 확인 할 수 있었다. 따라서, perfusion bioreactor를 이용한 세포 배양은 세포의 활성 향상 및 골조직 재생에 도움이 될 것으로 사료된다. 차후 perfusion bioreactor를 이용한 다양한 패턴의 자극이 골재생 능력 및 중간엽 줄기세포의 골 분화능에 미치는 영향에 대한 연구가 필요할 것으로 사료된다.
The bacteria which can biodegrade aromatic compounds were screened from soil and wastewater. The isolated Pseudomonas sp. HC107 had high removal rate of COD and phenol. And also this strain grew on m-cresol, salicylate, toluene, 2, 4-D and benzene. When the strain culture (2 ml/day) was treated on continuous reactor at mixed wastewater from chemical, pharmaceutical and dye industry, the treatment rate of COD, BOD and phenol was to be about 92.5%, 95.3% and 93.5%, respectively.
