Isolation of Siderophore-producing Pseudomonas fluorescens GL7 and Its Biocontrol Activity against Root-rot Disease (Siderophore 생산성 생물방제균 Pseudomonas fluorescens GL7의 선발 및 식물근부병의 방제)

  • For the development of a multifunctional biocontrol agent, the siderophore-producing strain GL7 was isolated from a rhizosphere on chrome azurol S agar. The GL7 was identified as a strain of Pseudomonas fluorescents on the basis of their reactions to standard physicochemcial tests from Bergey's manual, API diagnostic test, and fatty acid analysis. P. fluorescents GL7 considerably inhibited spore germination and hyphal growth of phytopathogenic fungus Funsarium solani in a dual culture. In pot trials of bean with P. fluorescens GL7, the disease incidence was significantly reduced down to 5% from 70% of incidence in the untreated control. P. fluorescens GL7 also enhanced plant growth to nearly 1.5 times than that of the untreated control, promoting elongation and development of the roots. These results suggest that the plant growth-promoting P. fluorescens GL7 can play an important role in the biological control of soil-borne plant disease in a rhizosphere.

Biological Control of Soft Rot on Chinese Cabbage Using Beneficial Bacterial Agents in Greenhouse and Field (유용세균(Beneficial Bacterial Agents)을 이용한 배추 무름병의 생물적 방제)

  • Three beneficial bacterial agents, Lactobacillus strain KLF01, Lactococcus strain KLC02 and Paenibacillus strain KPB3 were showed clear zone against plated Pectobacterium carotovorum subsp. carotovorum (Pcc) soft rot pathogen. In greenhouse test, bio-control efficacy was more significantly effective in the treatments by KLC02 and KPB3 as 64%, 50%, 56% and 66%, 57%, 58% according to date of evaluation, respectively. In case of KLF01 control effect was relatively lower than treatments of KLC02 and KPB3 but its efficacy was still significantly observed when compared to control (only water treatment). Furthermore, KLF01, KLC02 and KPB3 showed 55%, 60% and 62% bio-control efficacy, respectively in field test from early March to late July of 2009. Thus, we suggest that these strains can be useful as bio-control agents against soft rot caused by Pcc.

  • Chemical pesticides have been re used for a long time to protect crops from diseases, insects, and weeds ; ample and secure supply of foods in present days would not be possible without them. However, concerns on the risk of using chemical pesticides rising steadily in recent years forced discard many of them and seek an alternate, environment-friendly control strategies. Microbial pesticide is regarded as one of the most promising methods because if properly developed, it will be effective, cheap, and environment-safe. Currently, about 180 microbial pesticides are under development throughout the world and at least 20 of them including B.t. and antibiotics are registered in Korea. This paper discusses some of the successful examples, intriguing problems, and future prospects of microbial pesticides.

  • 포도 생과의 수입 개방으로 1996년에 2,400톤, 1997년에 3,500톤을 수입한 바 있다. 현재까지는 칠레산 포도가 3월부터 5월 사이에 수입되어 그 동안 안정적으로 발전해 오던 시설포도의 수익성이 크게 떨어진 바 있으나 수입종 과실의 품질, 기호성, 신선도, 잔류 농약의 문제 등으로 소비자들이 국산 포도를 선호하고 있어 크게 문제되지 않고 있다. 그러나 앞으로의 포도의 지속적인 수입 개방으로 인하여 좋은 품질의 포도가 수입될 것으로 예상된다. (중략)

  • 최근 수경재배 면적이 급속히 신장하면서 상추의 수경재배 면적은 15.9㏊(1997)를 나타내고 있다. 이러한 녹황색채소는 건강식품으로서 뿐만 아니라 저공해 청정채소로 수요가 급증하면서 재배면적이 증가하고 있다. 그러나 이들 엽채류의 근권환경제어가 최적수준에서 이루어지지 않고 있어 생산성과 품질이 낮은 실정이다. 또한 식물공장과 같이 무농약으로 청정채소를 계획 생산하는 과정에서 작물의 생장속도를 극대화 할 수 있으면서 고품질을 유지할 수 있는 근권환경제어가 필수적이다. (중략)

Assessment of Pre-Harvest Environmental Factors in Domestic Production of Organic Lettuce (국내 유기상추의 생산환경 조사분석)

  • Among pre-harvest environmental factors, increasing attention has been paid to the effects of chemical and microbiological factors on fresh produce. The occurrence and prevalence of these factors have been usually studied with regard to the final products at the post-harvesting stage and/or when they are sold in the market. However, the origin and routes of transmission of both factors remain to be clarified. In the present study, we examined the contamination levels of food-borne pathogens and chemical factors such as pesticide residues and heavy metals in 83 and 43 samples, respectively, including various soil, water, and fertilizer samples, as well as post-harvested and processed samples. Among the organic farming samples, only one pesticide, dimethomorph, was detected in the soil sample, however no pesticides were observed from any other samples in organic farming system. Thus, it was thought that might be contaminated from conventional farm land in the vicinity. Whereas many pesticide residues were detected in conventional farming systems such as soil, fertilizer, water, and fresh produce as expected. Furthermore, heavy metals detected from all tested samples did not shown contamination levels higher than the standard limit. We comparatively assessed the levels of contamination by food-borne pathogens on the samples from organic and conventional farming systems, and found aerobic bacteria at approximately 7 log CFU/g, with no significant differences observed between the two systems. Coliforms were present at lower levels than aerobic bacteria. No human pathogens were present among the coliforms detected, indicating that these bacteria are saprophytes without the ability to cause food-borne illnesses. In contrast, among the high-risk food-borne pathogens, only sporadic cells of Bacillus cereus were found on samples of organic farming system. These data extend previous findings that the most prevalent food-borne pathogen is B. cereus and demonstrate that it spreads to whole living plants via soil.

Subchronic Toxicity of Herbicide Butachlor in Fish , the Medaka (Oryzias latipes) (제초제 Butachlor의 송사리에 대한 아급성(亞急性) 독성(毒性))

  • To establish an evaluation system of aquatic toxicity of chemicals at no-effect level, flow through and early life stage toxicity test were performed on a freshwater fish, the medaka (Oryzias latipes). The characteristics of medaka as a bioassay organism for the chronic toxicity test were discussed. Maximum acceptable toxicant concentration(MATC) of butachlor for the madaka in soft water was estimated using survival, growth, and reproduction as indicators of toxic effects. During a 3-month period, the fry of medaka were exposed to butachor concentrations ranging from 0.16 to 0.0l mg/liter and the DO concentration, temperature, and pH in the exposure chamber were measured to check the test condition. The highest concentration showed slight decrease of growth rate in medaka and reduced hatchability of spawning egg. Survival, growth, and reproductive success of adults in butachlor concentration of 0.04 and 0.01 mg/liter were not different from those of the control. The MATC was estimated to be between 0.04 and 0.16 mg/liter for medaka.

Effect of Cinnamly Derivatives on Crop Growth Inhibition of Brassica campestris. (신나밀계 화합물이 배추의 종자발아와 유묘생장에 미치는 영향)

  • Cinnamyl derivatives are abundant secondary metabolite in biomass, and they have been studied on their biological activities. However, little information was available for plant growth regulation of the cinnamyl derivatives. In here, the acid, amide, alcohol, aldehyde and ester of cinnamyl derivatives were screened for their root growth inhibition properties including germination. The aldehyde, amide and ester derivatives showed better the root growth inhibition than the carboxylic acid, and the meta-positioned electron withdrawing group on cinnamyl derivatives enhanced the inhibition activity. 3-Chlorocinnamic acid, cinnamaimde and 4-methoxycinnamaldehyde were highlighted with the early stage root development inhibition ($GR_{50}$ < 100 mg/L) on Brassica campestris.
