Antioxidant Activity of Korea Traditional Fermented Sauces Made with Asparagus (아스파라거스를 이용한 전통장류의 항산화 효과)

  • Asparagus have anti-cancer, antioxidant, and anti-inflammatory properties. Since the high functionality of Korean traditional sauces was discovered, various studies have been conducted using asparagus. In this study, the antioxidant activity of traditional sauces containing asparagus was confirmed. The antioxidant capacity of asparagus was independent of the concentration and showed the highest activity at 70 mg/ml. For instance, concerning Meju containing asparagus, high antioxidant activity was observed in water and ethyl acetate extracts; the total polyphenol content was equivalent to 1.4 mg/g and 0.7 mg/g of tannic acid in the water and ethyl acetate fractions respectively. The DPPH and ABTS radical scavenging activities were 27.6% and 9.04%, respectively, in the water layer, and 10.7% and 52.4%, respectively, in the ethyl acetate layer. Additionally, the antioxidant activity of asparagus-containing traditional sauces was measured using 95% ethanol extracts; soy sauce showed the highest activity, retained in a wide range of concentrations.

Antioxidant and Anti-Cholesterol Activities of Standardized Ecklonia Stolonifera Extract (표준화된 곰피추출물의 항산화 활성 및 콜레스테롤 개선 효과)

  • Ecklonia stolonifera, which belongs to the family Laminariaceae, is an edible perennial brown marine alga that is widely distributed, and is rich in polyphenols, including dieckol. Here, we investigated the radical scavenging activities of E. stolonifera extract (ESE) using various in vitro models. We further evaluated the effect of ESE on the cholesterol secretion inhibition activity in HepG2 cells, as well as the hydroxymethylglutaryl-coenzyme A (HMG-CoA) reductase activity. Our results showed that the total phenol, total flavonoid, and dieckol contents of ESE were 9.64±0.04 mg GAE/g, 2.72±0.08 mg RE/g and 27.42±0.66 mg/g, respectively. The antioxidant activity of ESE increased in a dose-dependent manner. In addition, the ESE inhibited cholesterol secretion from HepG2 cells with anti-HMG-CoA reductase activity. These results suggested that ESE possesses antioxidant and anti-cholesterol activities, and can therefore be used as a preclinical bioresource for development of health functional foods.

The Nutritive Value of Grains from Barley Cultivars (Wooho, Youngyang, Yuyeon) (청보리 종실의 품종별(우호, 영양, 유연) 사료가치 평가)

  • This study was conducted to investigate the nutritive value of grains from barley cultivars (wooho, youngyang and yuyeon) for substitution of the feedstuffs as corn and soybean meal. In results of proximately analysis, the crude protein concentration was significantly higher (p<0.05) in barley grains than in corn and the crude fat concentration was significantly lower (p<0.05) in barley grains than in soybean meal. The metabolism energy of barley grains was significantly lower (p<0.05) than corn and it was higher (p<0.05) than soybean meal. The in vitro results of pepsin digestibility was significantly higher (p<0.05) in soybean meal than in barley grains and corn, and it was similar barely grains with corn. Barley grains significantly higher (p<0.05) the composition of saturated fatty acids than corn and soybean meal, while lower (p<0.05) the compositions of unsaturated fatty acids and essential fatty acidsin barley grain. The composition of linolenic acid, concentration of lysine and mineral was significantly higher (p<0.05) in barley grains than in corn, while lower (p<0.05) than soybean meal. Therefore, the barley grains is sufficient for substitution of corn. Moreover, the barley grain of yuyeon has high concentrations of crude protein, metabolism energy, lysine and minerals compared with barley grains of wooho and youngyang.

Characterization of Weissella sp. Strains Isolated from Fermented Squid and the Antibacterial Activities of Fermented Rice against Harmful Bacteria (발효된 오징어젓갈에서 분리된 Weisella sp. 균주의 특성 분석 및 쌀발효물의 유해세균에 대한 항균활성 검증)

  • The purposes of this study were to isolate and characterize lactic acid bacteria with antibacterial activity against Methicillin-resistant Staphylococcus aureus (MRSA) from fermented food and to confirm the antibacterial activities of fermented rice products using the isolated lactic acid bacterium. Three bacteria, namely, Weissella sp. ISF-1, ISF-2, and ISF-3, were selected from fermented squid based on the 16S rRNA gene sequence. All three strains grew well in an MRS medium containing 5% (w/v) NaCl and showed antibacterial activity against Bacillus cereus, Staphylococcus aureus, and MRSA. Their growth was excellent at 0% ~ 5% (w/v) NaCl and relatively good up to 7% (w/v) NaCl. The initial pH of 8 was optimal for their growth, and good growth was also observed at pH 6, 7, and 9. The lyophilisates of the fermented rice using Weissella sp. ISF-1 showed antibacterial activities against B. cereus, S. aureus, and MRSA. We inferred that isolated lactic acid bacteria could be useful in the development of probiotics and biopreservatives for foods and in the treatment of MRSA and may increase the value of rice products.

Induction of Apoptosis in AGS Human Gastric Cancer Cells by Platycarya strobilacea Leaf Extract (굴피나무 잎 추출물의 위암세포에 대한 세포사멸 유도 효과)

  • This study investigated the anticancer activity of methanol extract from Platycarya strobilacea leaf in AGS human gastric cancer cells. We determined the cell viability effect of P. strobilacea using MTS assay. Apoptosis induction and cell cycle arrest were confirmed by fluorescein isothiocyanate and propidium iodide staining using cellometer K2. The mRNA expression levels of the Bcl-2 family were confirmed by reverse transcription-polymerase chain reaction. The cell viability was decreased in a dose-dependent manner treated with different concentrations of P. strobilacea. Total, early, and late apoptotic cells were dramatically increased, and the cell cycle was arrested at the sub-G1 phase. The mRNA expressions of Bcl-2 and Bcl-xL were reduced, whereas pro-apoptotic factors, Bax and Bak, were increased in a dose-dependent manner. These results suggested that P. strobilacea leaf extract induced significant apoptotic activity through an intrinsic mitochondria pathway.

Comparative Profiling of Volatiles in Flower Tea of Dendranthema zawadskii var. latilobum, Chrysanthemum morifolium, Tagetes erecta, and Matricaria chamomilla (구절초, 국화, 마리골드 및 캐모마일 꽃차의 향기 성분 비교)

  • 꽃차(Flower tea)는 최근 다양한 제품이 개발되고 소비가 확대되는 등 그 산업적 가치가 증대하고 있다. 꽃차의 향기 특성은 우렸을 때 나타나는 색깔 및 인체에서의 생리활성과 더불어 주요한 꽃차 품질결정 요소이다. 본 연구는 꽃차로의 이용이 활발한 국화과 식물 중 구절초(Dendranthema zawadskii var. latilobum), 국화(Chrysanthemum morifolium), 노랑색 및 주황색 마리골드(Tagetes erecta 'Yellow' and 'Orange'), 그리고 캐모마일(Matricaria chamomilla)의 향기 성분특성을 구명하기 위하여 제조된 꽃차를 headspace-solidphase microextraction/gas chromatography-mass spectrometry (HS-SPME/GC-MS)를 이용하여 분리, 동정하였다. 국화과 꽃차로부터 총 117종의 휘발성 성분이 확인되었는데, 각 꽃차 종류별 동정된 휘발성 성분의 개수, 주요 3개 휘발성 성분과 전체 향기성분 peak중 이들이 차지하는 구성비율(%)은 다음과 같았다: 구절초 64종, camphor (31%), α-pinene(14%), camphene(14%); 국화 60종, camphor(15%), chrysantheny acetate(13%), eucalyptol (11%); 마리골드 '옐로우' 53종, 2,4-heptadienal(26%), trans-isocarveol(21%), cis-β-Copaene(18%); 마리골드 '오렌지' 61종, β-caryophyllene(16%), β-ocimene epoxide(12%), β-ocimene(12%); 캐모마일 50종, β-farnesene(63%), nonane(9%), spathulenol(5%). 국화과 꽃차 5종 모두에서 공통적으로 검출된 성분은 β-caryophyllene, α-pinene, β-farnesene 등 10종이었으며 마리골리 '옐로'는 '오렌지'와 주요 향기성분의 조성에서 뚜렷한 차이를 나타내었다. 비록 그 함량은 낮았으나 구절초, 국화, 마리골드 '오렌지', 그리고 캐모마일은 각각 10종, 12종, 3종 및 13종이었다. 마리골드 '엘로'의 경우 검출된 모든 향기성분은 마리골드 '오렌지'나 다른 국화과 식물의 꽃차에서도 검출된 바, 향기 성분이 다양성이 다소 낮게 나타났다.

Biological Effects of C. tricuspidata Extract in Normal Rodents (꾸지뽕 추출물의 생리활성 연구)

  • In order to use Cudrania tricuspidata extract for the development of health-functional food, biological changes in normal rodents, such as changes in blood constituent cells, blood lipids, and anti-inflammatory activity, were measured after the rodents had been drinking the substance for 2 weeks. A 30 mg daily dose of the extract taken for 2 weeks produced no change in body weight or renal function, and no toxicity was observed. Increased red blood cell levels and white blood cell levels were confirmed, and platelet counts tended to increase; thus, the extract seems to cause an overall increase in blood cells. In the case of blood lipids, it was confirmed that the extract-treated group exhibited an effect of improved blood-lipid levels, demonstrated through an increase in HDL-C of 21%, with a decrease in triglyceride of 27% compared to the control group. In addition, the decreases in both GOT and GPT led to improved liver function through use of the extract. Therefore, this study suggests that drinking Cudrania tricuspidata extract can have various effects, such as promoting blood cell production, improving blood lipids profile, improving liver function, and improving inflammatory action. The extract would be useful in the future in developing high-value industrial products such as functional foods and pharmaceuticals.

WAP-EPO 유전자 도입 형질전환돼지의 계대번식시 유전자 전이효율에 관한 연구

  • 본 연구는 생명공학 관련 기술개발에 의하여 신소재 물질을 생산하고 사람에게 안전하고 생리활성이 높은 고가의 의료용 단백질 생산기술을 체계화하며 형질전환 가축을 이용한 유용물질 생산에 따른 산업화와 부가가치의 극대화에 그 목적이 있다. 유즙으로 사람의 빈혈치료제인 EPO(Erythropoietin)를 분비할 수 있는 형질전환돼지를 생산하기 위하여 WAP(Whey Acidic Protein) Promoter(2.6kb) 하류에 사람의 조혈촉진유전자(EPO: 2.6kb)를 연결시켜 미세주입용 재조합 벡터(WAP-EPO : 약 7.8kb)를 구축하였다. 구축된 재조합 벡터를 1세포기 수정란에 약 2ng/ul 농도로 미세주입한 다음 외과적 방법으로 이식하였다. 이식 후 분만 모돈으로부터 생산된 자돈의 꼬리조직을 이용 게놈DNA를 추출하고 PCR 검정을 한 결과, 유즙으로 사람의 빈혈치료제를 생산할 수 있는 유전자가 도입된 형질전환돼지 $\boxDr$새롬이$\boxUl$ (♂)를 확인하였다. 또한 이렇게 꼬리조직으로부터 확인된 새롬이의 혈액과 정액을 채취, 게놈 DNA를 추출하여 외래 유전자 삽입여부를 PCR 방법으로 검정한 결과 꼬리조직과 마찬가지로 혈액 및 정액에서도 외래유전자가 삽입되었음을 확인할 수 있었다. 이렇게 생산된 형질전환돼지 $\boxDr$새롬이$\boxUl$를 이용 계대번식을 통한 F$_1$ 산자의 생산과 유전자 전이율을 확인하기 위하여 $\boxDr$새롬이$\boxUl$정액을 이용한 인공수정을 실시하였다. 인공수정은 1999년 7월 1일부터 2000년 9월 8일 까지 총 78두의 모돈을 이용하였으며, 그 중 21두의 모돈이 분만하여 인공수정에 의한 분만율은 26.92%로 나타났다.(Table Omitted) Table 1에서와 같이 새롬이 정액을 이용한 인공수정에 의해 형질전환된 F$_1$ 자돈의 형질전환율은 17.98%로 나타났으며, 32두의 형질전환자돈 중 8두(암:4두, 수:4두)는 분만과 동시에 폐사하였거나 사육중 폐사하여 현재 24두(암:12두, 수:12두)가 생존하여 F$_1$ 간 교배계획에 의해 사육되고 있다. 이 중 암컷 4두는 현재 F$_2$ 자돈 생산과 함께 유즙내로 사람 빈혈치료제의 분비 유무를 검정중에 있으며, FISH 법에 의한 외래 유전자 삽입 검정을 확인 중에 있다.

A Comparison of the Components and Biological Activities of the Ethanol Extracts of Achyranthes japonica Nakai and Achyranthes bidentata Blume (토우슬과 회우슬의 에탄올 추출물의 성분 및 생리활성 비교)

  • In the course of a study for the development of functional foods utilizing Woosul (Achyranthis radix), the components and various biological activities of Korean Woosul (AJN: Achyranthes japonica Nakai) and Chinese Woosul (ABB: Achyranthes bidentata Blume) were compared. Woosul in Korea, including AJN and ABB, are regulated and part of the Korean Pharmacopoeia. From AJN and ABB, ethanol extracts and their subsequent organic solvent fractions were prepared and their in-vitro antimicrobial, antioxidant and anti-diabetes activities were evaluated. Although AJN and ABB have no clear distinction in terms of usage in Korea, our results suggest that AJN has higher quantities of lipid-soluble components and lower amounts of water-soluble sugars than does ABB. ABB also appears to possess greater amounts of flavonoid and polyphenol substances than AJN. Analyses of biological activities showed that the fractions of AJN were more active as antibacterial agents, and possessed more pronounced ${\alpha}$-glucosidase inhibitory activities than those of the ABB fractions. However, the antioxidant activities of the ABB fractions, as determined by DPPH anion-, ABTS cation-, nitrite-scavenging activity and reducing power, were higher than those of the AJN fractions. Our results suggest that the components and bioactivity of the extracts and subsequent fractions of AJN and ABB are different. Therefore, usage of either AJN or ABB should be carefully considered, as regards their individual properties, when the active fractions of Woosul are employed in the development of functional foods or novel plant-derived medicines.

The New Technology Development Strategy of Cosmeceuticals with Use Advanced Materials Resources (신소재 자원을 활용한 기능성화장품의 신기술 개발전략)

  • Cosmeceuticals products we not the products simply for moisturizing and protecting skin but the ones for encouraging to make improvements on wrinkles and helping to whiten, tan, and protect skin against ultraviolet light. In a broader sense, they refer to products with aiming to maintain healthy condition of skin such that skin troubles could be improved or aging of skin could be delayed. However, cosmeceuticals should not have any side effects because they are applied to the whole body for a long period of time differing from medical products which are used on specific areas for a short time. The number of such cosmeceuticals has increased from 500 in 2001 to 2300 in 2003. In order to develop and broaden the scope of cosmeceuticals, the concept of cosmetics needs to be changed from caring skin to delaying aging of the skin and reviving cells. For this purpose, high-valued materials should be developed through basic researches related to the biological function of skin, in vivo experiment and reaction of skin in response to various stimuli using biotechnologies and bioengineering methods. At the same time, it should be proceeded to develop new nano materials for overcoming skin barriers and transfer matericals for helping to absorb effective substances and maintaining stability.
