Lipase를 이용한 곤충 pheromone 합성 응용 - 생물 전환 기술

  • 본고에서는 화학적수법으로 다양한 특이한 기질을 제조하여 여기에 입체특이적 성질(stereospecificity)과 입체선택적 성질(steroselectivity)이 높은 생물학적 방법들 중에서 주로 lipase를 이용한 부제가수분해반응(asymmetric hydrolysis)과 그 역반응인 에스테르화반응(esterification)등을 도입한 chiral building block의 제조와 이를 이용한 곤충 생리활성물질인 pheromone의 합성에 대한 최근의 많은 예들중 상품화를 시도하고 있는 매미나방의 pheromne인 (+)-disparlure에 대하여 간단히 소개하고자 한다.

Biological Activity of Acetoxycycloheximide and Its Producing Microoganism (Acetoxycycloheximide의 생리활성 및 과 생산균주)

  • Strain No.77-AG-567 showed antifungal activity against Pyricularia oryzas and Sphaerotheca fuliginea. In the process of purification of active component this strain was found to produce blasticidin S together with another antibiotic. This compound was identified as being acetoxycyc-heximide, also referred to as E-73, by UV and $^1$H NMR spectral data. Identification of this strain led us to conclude that strain No.77-AG-567 is most probably be Streptomyces atratus. It showed different characteristics from S. atbutus, so far known to produce both blasticidin S and acetoxycycloheximide. Particularly worthy of note was the difference in spore surface. In addition, acetoxycycloheximide has been known to have activity only against yeast and tumor cells but we found that it also has activity against Pyricularia oryzae and Sphasrotheca fuliginea.

Fibrinolytic Activity and Chemical Properties of Cordycepin-Enriched Cordyceps militaris JLM 0636 (Cordycepin 고함유 동충하초 수용성 추출물의 이화학적 특성과 혈전 용해활성)

  • The effect of cordycepin-enriched Cordyceps militaris JLM 0636 ($CM{\alpha}$) and Cordyceps militaris (CM) on fibrinolytic activity was investigated. The bioactive compounds and nutritional materials such as polyphenolic compounds, flavonoids, glutathione, minerals, and fatty acids were also measured. Concentrations of polyphenol compounds, flavonoids, and glutathione were higher in $CM{\alpha}$ than that in CM. The major minerals of both materials were K, Ca, Mg, and Na. The major fatty acids of both materials were linolenic acid, linoleic acid, oleic acid, and palmitic acid. Fibrinolytic activity was higher in $CM{\alpha}$ than that in CM. These results may provide the basic data to understand the fibrinolytic activity and bioactive compounds of $CM{\alpha}$.

Effect of NaCl Treatment on Growth and Physiologycal Activity of Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara (NaCl 처리가 눈개승마[Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara]의 생육과 생리활성에 미치는 영향)

  • This study was conducted to investigate the growth and physiologycal activitis of Aruncus dioicus var. kamtschaticus (Maxim.) H. Hara under the NaCl treatment conditions (0, 100, 200, 300 mM). After 30 days treatment, the growth and physiologycal activitis were investigated. In the growth of plants, the plant height, leaf width, leaf length and ion level were reduced at NaCl treatments of more than 100 mM. The total polyphenol content was decreased by NaCl in a concentration-dependent manner compared to the control group. The contents of total flavonoids did not show any difference at the concentration of 200 mM and 300 mM. However, the content of total flavonoid decreased compared with that of control. In antioxidant activity, the DPPH and ABTS radical scavenging activity and reducing power activity were decreased by NaCl concentration compared to the control. When changes in the content of NO production was monitored by ELISA, production inhibitory effect was 94.5%, 70%, 63%, 56.9% in NaCl concentration of 0, 100, 200, 300 mM, respectively. The growth, ion level, antioxidant and anti-inflammation activity of Aruncus dioicus var. kamtschaticus was reduced at NaCl treatments of more than 100 mM.

  • The possible effects in vivo on the duel usage of sinseng saponin and antibiotics were studied in vitro with microorganisms. Streptomycin.sulfate, kanamycin.sulfate and gentamycin.sulfate as being an aminoglycoside-antibiotic substance showed a general synergism by the interaction of ginseng saponin and these antibiotics. But kanamycin.sulfate and gentamycin.sulfate did not show a synergism in their original antimicrobial activity against Er-winia aroideoe. Chloramphenicol as being a benzene derivative displayed an increased antimicrobial activity by the interactions of ginseng saponin and this antibiotic against Salmonella typhi, Aerobacter aerogenes and the genus Serrotia. This antibiotic also showed the decreased antimicrobial activity against Bacillus subtilis, Bacillus megaterium and Escherichia coli, but did not show an uniform antimicrobial activity against others.

Change in Polyphenol Content, Antioxidant Activity, and Antioxidant Enzyme Status of Cowpea During Germination (동부 발아기간 중 폴리페놀 함량, 항산화성 및 항산화효소 활성 변이)

  • A laboratory experiment was conducted to determine the content of phenolics and flavonoids, antioxidant activity and antioxidant enzyme activity for the extracts from cowpea seed and sprouts. Plant length and weight of cowpea sprouts were significantly increased until 7 days after seeding. Total phenolics level [mg chlorogenic acid equivalents (CAE) $kg^{-1}$ DW] was highest in dry seed (DS) extracts of cowpea ($63.9mg\;kg^{-1}$), followed by imbibed seed (IS) ($56.8mg\;kg^{-1}$) and 1-day-old sprout (1DOS) extracts ($46.4mg\;kg^{-1}$), and significantly reduced with increase of sprout age (p < 0.05). The antioxidant activity of the methanol extracts from all the samples showed same tendency to the results of total phenolics level, and dose-dependently increased. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was higher in DS (87.3%) and IS (41.2%) than in cowpea sprouts from 1DOS to 7DOS, ranging from 17.1 to 30.4%. Antioxidant enzymes, APX, POX, and POX activities were highest in 7DOS and lowest in DS. SOD activity showed much higher activity in sprouts and in seeds. Correlation coefficient between physiological-active substance and the activity was highest between APX and CAT activities ($r^2$= 0.9574). Especially, total phenolics content was more highly correlated with antioxidant or with antioxidant enzyme activities than was total flavonoid level.
