Influence of Dichlorprop and MCPB on the Reduced Effect of Fruit Drop and Fruit Quality before and after Storage in Apples (생장조정제(生長調整劑) Dichlorprop 및 MCPB 처리(處理)가 사과의 후기낙과(後期落果) 경감효과(輕減效果) 및 저장후기(貯藏後期) 과실(果實)의 품질(品質)에 미치는 영향(影響))

  • This experiment was conducted to evaluate the effect of plant growth regulators, dichlorprop and MCPB on the reduced effect of fruit drop and fruit quality before and after storage in apples. Dichlorprop was tested with dilution of 1000 at 30, 40, 50 days before harvesting, and MCPB with dilution of 4000 at 15, 25, 35 days before harvesting. The results are summarized as follows : Percentage of fruit drop was appeared to the notable reduction as compared with the untreated control when regulators was applied with dilution of 1000 at 30 days before harvesting by dichlorprop and with dilution of 4000 at 35 days before harvesting by MCPB. Degree of fruit colour showed to the remarkable promotion at all the treatment of 30, 40, 50 days before harvesting by dichlorprop as compared with the untreated control. Sugar contents in flesh was increased a little at the treatment of 30 days before harvesting by dichlorprop, but acid contents in flesh was reduced at all the treatment of 30, 40, 50 days before harvesting by dichloroprop and at 15, 25, 35 days before harvesting by MCPB. Passed firmness of fruit after storage was maintained at the treatment with dilution of 4000 at 35 days before harvesting. Therefore, it was repressed a softening of fruit, but by dichlorprop treatment at 30, 40, 50 days before harvesting, fruit firmmess was appeared to reduce according to the passage of storage period. Amount of ethylene evolution after storage was showed to reduce at all the treatment by early treated time of dichoroprop and MCPB, but carbon dioxide increased at treatment conditions such as the front. Accordingly, these relationship showed to be contrary each other.

Breaking Physical Dormancy with Sulfuric Acid in Seeds of Lespedeza tomentosa (Thunb.) Siebold ex Maxim (황산처리를 이용한 개싸리 종자의 물리적 휴면 타파)

  • Lespedeza species are mainly used for wildlife food and cover and for erosion control. The germination of these species can be enhanced after a fire occurrence in forest, which is known as fire-activated seeds to germinate. While the heat treatment could break seed dormancy of Lespedeza, its germination rate was quite low. We investigated that chemical scarification could promote germination of L. tomentosa. Seeds were soaked in 100% sulfuric acid (H2SO4) for 0, 1, 3, 6, 12, 24, 48, 96, 192, and 384 min, and then washed in distilled water for 24 h. Very few seeds were germinated in control (H2SO4 for 0 min). More than 90% of seeds were germinated in H2SO4 for 24, 48, and 92 min. However, some damage was observed in roots and cotyledons of seedling dipped in H2SO4 for a long time. To search the optimal soaking time in H2SO4 without defects, seeds scarified in H2SO4 for 30, 60, 90, 120, 150, 180, and 300 min were sown the commercial soil medium. Seeds treated with H2SO4 for 90 min and 150 min emerged by about 92% and 84%, respectively. Therefore, H2SO4 treatment could break the seed dormancy of Lespedeza species, and especially in case of L. tomentosa the optimal treatment time in sulfuric acid was one to two hours. Germination of L. tomentosa began promptly following the scarification and was completed within about one month, indicating that seeds has no physiological dormancy, just has physical dormancy.

Effect of Zizyphus vulgaris Supplementation on Growth Performance, Blood Cortisol and Meat Quality Characteristics in Finishing Pig (비육돈 사료내 산조인(Zizyphus vulgaris)의 급여가 성장 혈액내 Cortisol 및 육질 특성에 미치는 영향)

  • This study was conducted to evaluate the effects of Zizyphus vulgaris supplementation on growth performance, blood cortisol and meat quality characteristics in finishing pigs. The total of thirty-six [Duroc${\times}$Yorkshir${\times}$Landrace] pigs ($91{\pm}2.11$ kg average initial body weight) were used in a 30-days assay. Dietary treatments included 1) CON (basal diet), 2) T1 (basal diet for 15 days and 0.1 % Zizyphus vulgaris for 15 days) and 3) T2 (0.1 % Zizyphus vulgaris for 30 days). The ADG (Average daily gain), ADFI (Average daily feed intake) and ADG/ADFI during the feeding period were not significantly differences among the treatments (p>0.05). Backfat thickness of pigs fed CON was higher than those of T1 and T2 (p<0.05). The appearance rate of A or B carcass grade was in T1 (74%) and T2 (84%) was significantly higher than that in CON (58%) (p<0.05). Pigs fed Zizyphus vulgaris 0.1 % for 30 days tended to decrease on blood cortisol compared with pigs fed CON and T1. But, there was not significantly difference among the treatments (p>0.05). The Hunter's L/sup */ (lightness) value of loin in the pork fed CON was higher than that of loin in the pork fed T1 and T2 (p<0.05). After 7 days, the L/sup */ value of loin in the pigs fed T2 was higher increased than that of pigs fed T1 and CON (p<0.05). However, a/sup */ and b/sup */ values were not affected by dietary Zizyphus vulgaris (p>0.05). There were not found remarkable differences in sensory properties (marbling, firmness and color) among the treatments. The results from the present study suggest that Zizyphus vulgaris could be a effective feed additive to improve meat quality of pigs. However, further research is needed to investigate effects of carcass characteristics.

A Study on Microbial Aspects of Korean Human Milk by Collection Methods (수집방법에 따른 한국인 모유의 미생물 분포에 관한 연구)

  • This study was carried out to evaluate the safety of Korean human milk. The microorganisms were identified from human milk of 149 healthy mothers by two collection methods, hand and pump expression. The means of total bacterial counts were 2.33x10$^4$ cfu/mL on the samples collected by the pump expression and 7.83xl0$^3$ cfu/mL on those collected by the hand expression. Therefore, the total bacterial counts of pump expression samples was 9.80xl0$^2$∼3.06x10$^4$ cfu/mL more than that of hand expression samples. The coliform counts of pump expression was 9.36xl0$^3$∼8.57xl0$^4$ cfu/mL more than that of hand expression. However, there was any significant differences of the lactic acid bacterial counts between the two samples collected by each methods. 100 strains of 5 patterns of total bacterial counts were isolated based on the morphology of colony in the standard plate count agar. 13 species were identified among the isolated strains. The dominant species in Korean human milk were Staphylococcus which 7 subspecies identified(81% in the rate of total bacteria, 1.07x10$^4$ cfu/mL). Other species identified were Micrococcus, Bacillus, Providencia, Pseudomonas, Yersinia and Acinetobacter. 36 strains of 6 patterns of lactic acid bacterial counts were isolated based on morphology of colony in the BCP agar. 7 species were identified among the isolated strains. The dominant species of lactic acid bacteria in Korean human milk were Lactobacillus brevis(50.9% in the rate of lactic acid bacteria, 4.72xl0$^4$ cfu/mL). Others species identified(49.1% lactic acid bacteria) were Lactobacillus curvatus, Lactobacillus delbrueckii subsp. lactis, Lactobacillus acidophilus, Leuconostic lactis and Streptococcus salivarius subsp. thermophilus.

Quality Characteristics of Low-fat Emulsified Sausage Containing Tomatoes during Cold Storage (토마토가 함유된 저지방 유화형 소시지의 냉장 저장 중 품질 특성)

  • This study was conducted to evaluate the quality characteristics of low-fat emulsion type sausages containing 0% tomato powder (C), 5.0% ground raw tomato paste (T1) and 0.5% freeze dried tomato powder (T2) during storage at $5{\pm}1^{\circ}C$ for 30 days. The crude protein content of T2 was significantly lower (p<0.05) than that of the other sausage types. Moisture, crude fat and crude ash contents of the sausages during storage were not affected by the addition of tomato. The pH and shear force ($kg/cm^2$) values of C were significantly higher (p<0.05) than those of T1 and T2. There was no significant difference among the different sausages in cooking loss, ranging from $13.00{\sim}14.98%$. The WHC values of T1 and T2 were significantly higher (p<0.05) than that of C. The values of TBARS were significantly (p<0.05) increased for ail sausages following storage. The TBARS value (mg MA/kg) of C was significantly higher (p<0.05) than those of T1 and T2 at 15 days of storage, however T1 was significantly higher (p<0.05) than the other sausages after 30 days of storage. The meat color values tended to decrease with increased storage time. Microorganism analysis revealed that all sausage types did not reach $4.4log_{10}CFU/g$ until 30 days of storage. The texture, brittleness, Hardness, and springiness of each sausage type were not significantly different after 1 day of storage, while the cohesiveness, gumminess and chewiness of T1 and T2 were significantly higher (p<0.05) than that of C. T1 and T2 sausages had a slightly higher score regarding color, aroma, tenderness and overall acceptability, however the sensory evaluation score among the different sausage types was not significantly different (p>0.05). In conclusion, low-fat sausage with added tomato showed higher lipid oxidative stability during storage than sausage to which no tomato was added.

Enhanced PHB Accumulation in Photosystem- and Respiration-defective Mutants of a Cyanobacterium Synechocystis sp. PCC 6803 (Synechocystis sp. PCC 6803의 에너지 대사 결함 돌연변이 균주에서의 Poly(3-hydroxybutyrate) 축적량 증진)

  • Photoautotrophic bacteria are promising candidates for the production of poly(3-hydroxybutyrate) (PHB) since they can address the critical problem of substrate costs. In this study, we isolated 25 Tn5-inserted mutants of the Synechocystis sp. PCC 6803 which showed enhanced PHB accumulation compared to the wild-type strain. After 5-days cultivation under nitrogen-limited mixotrophic conditions, the intracellular levels of PHB content in these mutants reached up to $10-30\%$ of dry cell weight (DCW) comparable to $4\%$ of DCW in the wild-type strain. Using the method of inverse PCR, the affected genes of the mutants were mapped on the completely known genome sequence of Synechocystis sp. PCC 6803. As a result, the increased PHB accumulation in 5 mutants were found to be resulted from defects of genes coding for NADH-ubiquinone oxidoreductase, O-succinylbenzoic-CoA ligase, photosystem II PsbT protein or histidine kinase, which are involved in photosystem in thylakoid inner membrane of the cell. The values of $NAD(P)H/NAD(P)^+$ ratio in the cells of these mutants were much higher than that of the wild-type strain as measured by using pulse-amplitude modulated fluorometer, suggesting that PHB synthesis could be enhanced by increasing the level of cellular NAD(P)H which is a limiting substrate for NADPH-dependent acetoacetyl-CoA reductase. From these results, it is likely that NAD(P)H would be a limiting factor for PHB synthesis in Synechocystis sp. PCC 6803.

Freeze Risk Assessment for Three Major Peach Growing Areas under the Future Climate Projected by RCP8.5 Emission Scenario (신 기후변화시나리오 RCP 8.5에 근거한 복숭아 주산지 세 곳의 동해위험도 평가)

  • This study was carried out to evaluate a possible change in freeze risk for 'Changhowon Hwangdo' peach buds in three major peach growing areas under the future climate projected by RCP8.5 emission scenario. Mean values of the monthly temperature data for the present decade (2000s) and the future decades (2020s, 2050s, 2080s) were extracted for farm lands in Icheon, Chungju, and Yeongcheon-Gyeongsan region at 1km resolution and 30 sets of daily temperature data were generated randomly by a stochastic process for each decade. The daily data were used to calculate a thermal time-based dormancy depth index which is closely related to the cold tolerance of peach buds. Combined with daily minimum temperature, dormancy depth can be used to estimate the potential risk of freezing damage on peach buds. When the freeze risk was calculated daily for the winter period (from 1 November to 15 March) in the present decade, Icheon and Chungju regions had high values across the whole period, but Yeongcheon-Gyeongsan regions had low values from mid-December to the end of January. In the future decades, the frequency of freezing damage would be reduced in all 3 regions and the reduction rate could be as high as 75 to 90% by 2080's. However, the severe class risk (over 80% damage) will not disappear in the future and most occurrences will be limited to December to early January according to the calculation. This phenomenon might be explained by shortened cold hardiness period caused by winter warming as well as sudden cold waves resulting from the higher inter-annual climate variability projected by the RCP8.5 scenario.

A Study on BMI, Diet Perception and Dietary Behavior of Female University Students (여대생의 체질량지수, 다이어트 인식과 다이어트 행동에 관한 연구)

  • The purpose of this study is to identify the BMI, diet perception and diet behavior of female university students. A descriptive survey design was used 165 students from October 29 to November 9, 2018. Data were analyzed using mean, t-test, ANOVA, cross analysis with SPSS 21.0. Total diet perception was 3.44 points, highest sub category was 'practice' with 4.00 points, followed by 'information and education' with 3.58 points, 'necessity' with 3.32 points and lowest category was 'self-recognition' with 2.84. 65 students(43.0%) were 'less than three times' in 'Number of diets' and diet methods was 65(43.0%) for 'food control' and 62(41.1%) for 'exercise'. During in a week, 98 students(84.9%) exercised within three hours, followed by 33(21.9%) who did not exercised at all. 22 students(14.6%) said they experienced damage from dieting, while the types included gastrointestinal disease, menstrual irregularity and hair loss. The total score of diet perception according to BMI was lower in the underweight group than in the normal and overweight groups(p<.001). In diet 'practice' category, low weight group scored lower than normal weight and overweight groups(p<.05). In 'information and education' category, overweight group scored the highest, followed by normal and underweight groups(p<.001). In diet 'necessity' and 'self-recognition' categories, low weight group scored lower than normal weight and overweight groups(p<.001). Diet behavior by diet perception was different in number and method of diet(p<.001). Based on the results, it is thought that attention and education are needed to form a proper diet perception and maintain healthy living for female university students.

Study on the Antioxidative Activities and Anti-Inflammatory Effect of Kaempferol and Kaempferol Rhamnosides (Kaempferol 및 Kaempferol Rhamnosides의 항산화 활성 및 항염 효과에 관한 연구)

  • In this study, to evaluate the antioxidative activities and anti-inflammatory effects of kaempferol and its rhamnosides, we performed the free radical scavenging assay, ROS inhibition assay and TARC (thymus and activation-regulated chemokine) assay. Also, we studied physiological activity of kaempferol and its rhamnosides (${\alpha}$-rhamnoisorobin, afzelin, kaempferitn) by structure-activity relations. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities were determined with kaempferol (62.5 ${\mu}M$) and ${\alpha}$-rhamnoisorobin (50.0 ${\mu}M$) but afzelin and kaempferitrin did not show free radical scavenging activities. Kaempferol showed a 97.5, 57.8, 47.8 % inhibition of ROS (reactive oxygen species) generated at concentrations of 10, 50 and 100 ${\mu}M$, compared to control (100 %). ${\alpha}$-rhamnoisorobin showed a 93.1, 59.1 and 41.4 % inhibition of ROS at the same concentration. We investigated the inhibitory effects of kaempferol and its rhamnosides on TARC expression. Kaempferol showed a 48.8, 5.5 and 4.4 % inhibition of TARC generated at 10, 50 and 100 ${\mu}M$, compared to control. ${\alpha}$-Rhamnoisorobin showed a 88.1, 19.0 and 1.0 % inhibition of TARC generated at the same concentration. In conclusion, these results indicate that kaempferol and ${\alpha}$-rhamnoisorobin have good antioxidative activities and anti-inflammatory effects that could be applicable to new functional cosmetics for anti-aging and anti-inflammation.

The Inhibitory Effects of Pogostemon cablin Bentham Extract on Melanogenesis (광곽향 추출물의 멜라닌 생성 저해 효과)

  • To develop a new natural whitening agent for cosmetics, we investigated the inhibitory effects of Pogostemon cablin Bentham extracts (PCE) and its active component on melanogenesis. PCE showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $24.2{\pm}2.85{\mu}g/mL$ and $IC_{50}=118{\pm}0.43{\mu}g/mL$, respectively. PCE reduced melanin contents of B16 melanoma cells in a dose-dependant manner and decreased to about 23 % at a concentration of $20{\mu}g/mL$ without cell cytotoxicity (below $100{\mu}g/mL$). And the PCE reduced intracellular tyrosinase activity about 18 % at concentration of $50{\mu}g/mL$. We purified one active compound from PCE and identified its structure. It was identified as patchouli alcohol, sesquiterpene family, by 1H-NMR, $13_C$-NMR, and Mass analysis. Patchouli alcohol also inhibited ROS scavenging activities in DPPH radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.14{\pm}0.12{\mu}g/mL$ and $49{\pm}3.24{\mu}g/mL$, respectively. Patchouli alcohol inhibited melanin synthesis in a dose dependent manner ($IC_{50}=3.9{\mu}g/mL$). And the patchouli alcohol reduced intracellular tyrosinase activity about 40 % at concentration of $10{\mu}g/mL$. Patchouli alcohol inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that PCE and patchouli alcohol reduced melanin formation by the inhibited of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that PCE could be used as a useful whitening agent.
