$N_2O$ Emissions on the Soil of Alpine Wetland by Temperature Change (온도 변화에 따른 산지습지 토양의 $N_2O$ 배출 양상)

  • Global warming due to climate change is a problem facing the entire world. Several factors, such as $CO_2O$ concentration, level of warming, soil temperature, precipitation, water content of soil and denitrification by denitrifying bacteria influence the emission of nitrous oxide ($N_2O$) from soil. In this study, we investigated nitrous oxide emissions from the soil of two wetlands, Jilmoineup in Mt. Odae and Moojechineup in Mt. Jungjok, according to temperature change. Soil collected in Jilmoineup in July showed increasing $N_2O$ emissions as temperature increases, but did not show any significant differences at $10^{\circ}C$ (p<0.05). Soil of $15^{\circ}C$ and $20^{\circ}C$ showed increasing pattern of $N_2O$ emissions until 24 h. After that, however, there was no difference in temperature. Overall, $N_2O$ emissions showed significant differences according to temperature (p<0.05). Soil collected from Moojechineup in July showed increasing $N_2O$ emissions according to temperature increase, but did not show any significant differences at $10^{\circ}C$ (p<0.05) as was the case for Jilmoineup soil. On the other hand, two wetland soils showed a slight increase of $N_2O$ emissions by additional nitrogen supply, but did not show any significant differences in the presence of nitrogen or between nitrogen sources. In conclusion, increasing temperature the wetland soil increased the emission of $N_2O$, which is a known greenhouse gas. In order to more clearly identify $N_2O$ emissions, various subsequent studies such as the influence and correlation of several factors are required.

Fractionation of Angiotensin Converting Enzyme(ACE) Inhibitory Peptides from Soybean Paste (된장으로부터 Angiotensin Converting Enzyme(ACE) 저해 Peptide의 분획)

  • Angiotensin converting enzyme(ACE) inhibitory peptides lowering blood pressure were fractionated from a commercial soybean paste(Doenjang). When the freeze-dried sample of soybean paste was extracted with cold water, the recovery yield of total nitrogen(TN) was shown to be 73.3% in 30 minutes. The cold water extract was filtered through PM-10 membrane(Amicon) for 3 hours in order to remove high molecular weight polypeptides. The TN and salt of ultrafiltrate were recovered to 80.8% and 99.2%, respectively, and its ACE $IC_{50}$ was $41.8{\mu}g/ml$. When the ultrafiltrate was divided into 7 fractions by reverse phase prep-HPLC, F5 fraction showed the highest ACE inhibitory activity ($IC_{50}=6.8{\mu}g/ml$) and salt could be collected into F1 fraction. Subsequently, the F5 fraction was divided into another five fractions by ion exchange prep-HPLC, all of which appeared to be high ACE inhibitory activity($IC_{50}=2.5{\sim}8.3{\mu}g/ml$). Among them, F53 fraction had the highest ACE inhibitory activity, and its main amino acid component was found to be histidine.

Characteristics of Cheese Manufactured by Coprecipitation of Whey and Soy Milk (유청 및 두유의 공동침전에 의해 제조된 치즈의 특성)

  • As a way of improving the texture and flavor of soybean cheese, whey-say cheeses were made by coprecipitation of various mixtures of whey, whey powder, soy milk and soy protein powder, and mixed culture of str. lactis, str. cremoris and rennet were added, then the cheeses were cured at $15^{\circ}C$ for up to 10 weeks. Physicochemical characteristics of the cheese were investigated by analyzing pH, titratable acidity(TA), water soluble nitrogen, 10% TCA soluble nitrogen, amino acid composition, beany flavor, color and hardness. The pH of whey-soy cheeses during ripening changed from 5.3 to 4.2 after 5 or 6 weeks and maintained that value while that of soybean cheese maintained a higher pH value. TA of whey-soy milk cheeses was gradually increased to the value of 0.4-0.45 after 8 weeks, but that of soybean cheese reached only 0.2 after the same period. Water soluble and 10% TCA soluble-nitrogen increased steadily during ripening. Hardness of the whey-soy milk cheeses reached maximum after three weeks of ripening and greatest at those made from 3 : 1 mixture of whey and soy milk and that from soymilk. Color of the whey-soy milk chesses was lighter than that of soybean cheese. The bean flavor of soybean cheese was strong and persistent for the whole ripening period. Acid flavor was dominant in the whey-so milk cheese and masked the beany flavor partially.

Effect of Mulberry Extract on the Growth of Yogurt Starter Cultures (오디 추출물 첨가가 요구르트 스타터 생육에 미치는 영향)

  • This experiment was carried out to investigate the effect of mulberry fruit extinct on the acid production and growth of lactic acid bacteria. The extract of mulberry fruit was showed a high level of yield with 60% ethanol extraction. Lactic acid bacteria was used in a starter culture of Lactobacillus casei YIT9018 and 2782, and Lactobacillus acidophilus NCFM. After 24 hr culture in MRS broth added with 1.0, 5.0 and 10% of the extract 1.0% addition of the extract was showed pH $4.04{\sim}4.19$, titratable acidity $1.25{\sim}1.42%,\;and\;1.2{\sim}7.8{\times}10^9\;cfu/mL$ of viable cell count. The additions above 1.0% extract (5.0% and 10.0% addition) showed slightly lower effect than 1.0% addition. However, the addition of the extract showed a high effect on the growth of lactic acid bacteria comparing with the control. In yoghurt preparation with the extract, 1% addition of the extract showed a high effect on the growth of lactic acid bacteria, Therefore, it was suggested to manufacture of the yoghurt with the addition of 1% mulberry fruit extract and the inoculation of culture of Lactobacillus acidophilus for on the stimulation of growth of the lactic culture.

  • This study was carried out to evaluate the effects of whey ferment containing L. acidophilus KCCM 32820 and P. freudenreichii KCCM 31227 on the quality characteristics of white pan bread. Instrumental analysis such as alveograph, gelatinization temperature, texture analysis, retrogradation rate was determined. In an alveograph test, $P_{max}$ value in the treatment was higher than that in the control, but extensibility of dough in the control showed to be higher than in the treatment, so test dough showed more strength than the control. In terms of DSC analysis for gelatinization, temperature there were no significant differences of $T_p$ and ${\Delta}H$ between the control and the treatment. In hardness analysis by rheometer, dough containing whey ferment revealed lower values than the control. From the analysis of the organic acid contents, propionic acid was not detected in the control, however 1.13 mg/g of propionic acid was detected in the treatment. In the retrogradation analysis by DSC, the test delayed slightly compared to the control.

Cold Storage Characteristics of Early Variety of Citrus unshiu Produced in Cheju with Various Treatments (처리조건에 따른 조생온주밀감의 저온저장 특성)

  • The conditions of cold storage of Citrus unshiu Marc. var. miyagawa produced in Cheju were investigated. Compared to the citrus fruits stored at room temperature, the content of soluble solids, total sugar, vitamin C and specific gravity decreased slightly on the fruits stored at $4^{\circ}C$ and 87% relative humidity. Decay ratio and weight loss were below 10% on keeping freshness relatively till late of March during cold storage. Weight loss, decrease of firmness and soluble solids occured gradually during cold storage. Nevertheless seal-packaging with 0.02 mm LDPE film or wax-coating of citrus fruits were effective on weight loss and appearance, decay ratio increased for long-term storage. It was needed to control humidity in cold chamber for preventing from decay and weight loss. Edible part ratio was decreased gradually by respiration, and peel and tissue of fruits were softened slightly by 60 days of cold storage. Decay ratio and weight loss of full-ripened citrus fruits were little during the storage for 100 days, compared to the fruits harvested early. Appearance and taste of citrus fruits stored for 4 month were good relatively, but decay occured rapidly on seal-packaging fruits putting out of cold room for a few days. Because of the difficulty of long-term storage for early variety of Citrus unshiu, the conditions and periods of cold storage would be determined after considering the physicochemical properties of fruits.

  • As a one of the new methods for soybean protein food processings, an attempt was made to decompose steamed soybean protein to amino acids, peptides, and other water soluble forms by the use of protease produced by Aspergillus sojae, and extract them to use as additives of low protein foods or weaning-foods of children. In this paper, as a one part of this subject, the optimum condition, such as optimum pH, temperature, digestion period, raw material's mixing ratio with wheat meal, amount of water added, and effect of sodium chloride addition, to decompose and extract soybean protein were studied. They were obtained when mixing raw materials with wheat meal in the ratio of 10:4 and adding six folds of water at $40-45^{\circ}C$, pH 5-8 for 18 hrs. digestion. Although sodium chloride addition exerted an unfavorable influence upon the decomposition of the aforementioned materials, it was effective to prolongation of digestion period. Under the abovementioned optimum conditions, the maximum extractability of soybean protein as forms of amino acids, peptides, and other water soluble forms, was nearly 70 percent against protein content of raw materials.

  • This experiment was conducted to determine the effect of the feeding value for the wild legumes silage-making with additives. The chopped Kudzu (Puerarie thunbergii Bentham) of wild legumes was ensiled by the conventional method in the small experimental silo of 2 liters. The additives used in the present experiments were water (8%),water (8%) plus corn starch (3%) and water (8%) plus glucose (3%). These additives as well as that of control without additive were set up and these silage were compared with the conventional corn silage. The fermentative qualities and the characteristic of fiber (neutral detergent fiber, NDF; acid detergent fiber, ADF; acid detergent lignin, ADL) of silage produced was determined by chemical analysis, and dry matter digestibility (DMD) was evaluated by pepsin-cellulase technique. The results obtained are summarized as follows: 1 The weight of Kudzu silage was lost under the 10% for all the treatments. 2 In degree of pH for the Kudzu silages with glucose, starch, control and water treatments were high 3.80,4.04,4.57 and 5.34, respectively.

Petrology of the Volcanic Rocks in the Paekrogdam Crater area, Mt. Halla, Jeju Island (제주도 한라산 백록담 분화구 일대 화산암류의 암석학적 연구)

  • The Paekrogdam summit crater area, Mt. Halla, Jeju Island, Korea, composed of Paekrogdam trachyte, Paekrogdam trachybasalt, and Manseidongsan conglomerate in ascending order. Joint systems show concentric and radial patterns around the summit crate wall. The Paekrogdam crater is a summit crater lake which erupted the tuffs, scorias and lava flows of Paekrogdam trachybasalt after the emplaceent of Paekrogdam trachyte dome. SiO$_2$ contents of mafic and felsic lavas are respectively, 48.0∼53.7 wt.% and 60.7∼67.4 wt.%, reflecting bimodal volcanism. And lavas with SiO$_2$ between 53.7 wt.% and 60.7 wt.% are not found. According to TAS diagram and K$_2$O-Na$_2$O diagram, the volcanic rocks belong to the normal alkaline rock series of alkali basalt-trachybasalt-basaltic trachyandesite and trachyte association. Oxide vs. MgO diagrams represent that the mafic lavas fractionated with crystallization of olivine, clinopyroxene, magnetite and ilmenite and felsic trachyte of plagioclase and apatite. The characteristics of trace elements and REEs shows that primary magma for the trachybasalt magma would have been derived from partial melting of garnet peridotite mantle. In the discrimination diagrams, the volcanic rocks are plotted at the region of within plate basalt (WPB).

Bacterial Leaf Spot of English Ivy Caused by Xanthomonas hortorum pv. hederae (Xanthomonas hortorum pv. hederae에 의한 헤데라 세균성점무의병)

  • English ivy (Hedera helix) in Araliaceae family is an evergreen climbing vine. A severe bacterial disease of English ivy was observed and investigated in January 2005. Initial symptoms on the leaves appeared as small water-soaked lesions on the lower surface. As the spots enlarged, the center became brown to brownish black and greenish-brown water-soaked, irregular margins surrounded the center, The spots developed into large irregular blotches, sometimes 5$\sim$10 mm in diameter, then coalesced. Finally, the water-soaked margins raised, dried out, became corky and broke in the center. A bacterial organism, isolated from the advancing margins of the lesions, was tested for its pathogenicity according to the Koch's postulates and biochemical and physiological tests identified the isolated bacterium as a Xanthomonas. The representative Xanthomonas strains (SL4821 and SL4822) isolated from English ivy were compared with a reference strain X. hortorum pv. hederae for fatty acid profiles, metabolic fingerprints and 16s rDNA sequences, showing that all outcomes were indistinguishable between the representative and reference strains. This is the first report of bacterial leaf spot of English ivy in Korea.
