Characteristics of Culture of Oyster Mushroom (Pleurotus ostreatus) on Addition Rate of Acer tegmentosum (산겨릅나무(Acer tegmentosum) 톱밥의 첨가량에 따른 느타리버섯의 재배적 특성)

  • This study was carried out to investigated optimum mixing ratio of Korean natural Acer tegmentosum for production of functional oyster mushroom. Total nitrogen and carbon sources of Acer tegmentosum were 0.2% and 44.4%, respectively and C/N ratio was 234. Total nitrogen source and pH of substrate mixed with Acer tegmentosum were 2.7~2.9 and 4.8~5.0, respectively. The contents of $P_2O_5$, $K_2O$ and MgO at Acer tegmentosum media were higher than that of the control. Mycerial growth was the fastest at Acer tegmentosum 10%, and slower by increase of Acer tegmentosum substrate. Yields of fruiting body was the highest to 159 g/850 mL at 10% of Acer tegmentosum and dimeter and thick of pileus were the highest, too. The L value of pileus and stipes were decreased by increase of Acer tegmentosum substrate, but there was no significant difference in the a-value and the b-value. The contents of $P_2O_5$ and $K_2O$ of fruiting body were increased at Acer tegmentosum substrate, but there was no significant difference in contents of CaO, MgO and $Na_2O$.

Anti-Diabetic, Alcohol Metabolizing Enzyme, and Hepatoprotective Activity of Acer tegmentosum Maxim. Stem Extracts (산겨릅나무 줄기 추출물의 항당뇨, 알코올 대사 효소 및 간 보호 활성)

  • This study was carried out to investigate the antidiabetic, alcohol metabolism, anti-inflammatory, and hepatoprotective effects of Acer tegmentosum extracts (ATE). A. tegmentosum has been traditionally used as a folk medicine to treat hepatic disorders. The antioxidative activities of ATE were measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide (SOD) assay. DPPH radical scavenging and SOD activities of ATE were about 89% and 82.9% at $0.5{\mu}g/mL$, respectively. Alcohol dehydrogenase and acetaldehyde dehydrogenase activities were 118.0% and 177% at 2 mg/mL, respectively. ${\alpha}-Glucosidase$ inhibitory activity of ATE was 75% higher at $50{\mu}g/mL$ and remarkably increased in a dose-dependent manner. Nitric oxide productions in macrophage RAW 264.7 cells stimulated by lipopolysaccharide was reduced to 16.7% by addition of ATE at 1 mg/mL. ATE showed significant protective effects against tacrine-induced cytotoxicity in Hep G2 cells at $100{\mu}g/mL$. Based on our results, we conclude that ATE may be used as a major pharmacological agent and anti-diabetic, anti-hepatitis, and anti-inflammatory remedy.

Effects of Cultured Acer tegmentosum Cell Extract Against Hepatic Injury Induced by D-galactosamine In SD-Rats (산겨릅나무 세포배양 추출물이 D-galactosamine 유도 급성 간손상에 미치는 보호 효과)

  • Here we report the protective activity of cultured Acer tegmentosum cell extract against liver damage in rat intentionally instigated by D-galactosamine. Local fat degeneration and infiltration of inflammatory cells were significantly decreased in cultured A. tegmentosum cell extract administered rat. In addition, acutely increased AST, ALT, LDH, ALP activities and lipid peroxidation and lipid content by liver damage were recovered in experimental rat administrated with A. tegmentosum extract. These results showed that cultured A. tegmentosum cell extract has a role in blood enzyme activation and lipid content restoration within damaged rat liver tissues. Moreover expression rate of TNF-α which accelerates inflammation and induces tissue damage and necrosis was significantly decreased. Also activities of antioxidant enzymes were more effectively upregulated comparing to those of the control group induced hepatotoxicity. All data that cultured A. tegmentosum cell extract has a preventive role against liver damages such as inflammation, tissue necrosis in rats by improving activities of blood enzymes, antioxidant enzymes and modulating expression of inflammation factor, suggest that cultured Acer tegmentosum cell extract is an effective medicinal resource for restoration of hepatotoxicity.

Growth Environment and Vegetation Structure of Habitats of Acer tegmentosum Maxim. (산겨릅나무(Acer tegmentosum Maxim.) 자생지의 서식환경 특성 및 식생구조)

  • The present study was to survey the site environment, vegetation structure and soil characteristics in the wild habitats of Acer tegmentosum Maxim. and offers basic information for habitats conservation and restoration. Most of the wild habitats were located at altitudes between 605~1,413m with inclinations ranged as 8~30°. The bare rock rate were 8~50%. The vegetation structure by the PC-ORD based on the Two Way Cluster Analysis were divided into three groups Community I(Acer tegmentosum - Quercus mongolica), Community II(Acer tegmentosum - Carpinus cordata), Community III(Quercus mongolica - Tilia amurensis). The species diversity(H') was highest in Community II as 1.474, Community I was 1.471, Community III was 1.219. The soil textures were Clay loam, The average soil pH was 4.8, Soil organic matter was 15.15% and available phosphorus was 2.33ppm. Ordination analysis result by soil characteristics, community, characteristic species showed that the greatest effect factors were slope, altitude, tree and shrub's cover rate, organic matter, total-nitrogen, calcium, magnesium. Correlation analysis between environment factor result showed that O.M. - (T.N., K+, Mg2+, CEC, EC), T.N. - (K+, Mg2+, CEC, EC) were positive correlations.

Removal of toxic compounds from Acer tegmentosum using supercritical fluid extraction (초임계유체 추출을 이용한 산겨릅나무로부터 독성성분들의 제거)

  • Acer tegmentosum is a tree used to treat various liver diseases in Korea. There have been some concern regarding the safety of Acer tegmentosum due to some toxic chemical compounds in its stems. Supercritical fluid extraction (SFE) was employed to develop a removing method of toxic compounds from Acer tegmentosum. The toxic compounds were effectively extracted with ethanol modified supercritical fluid $CO_2$. The optimum condition of SFE was 100 bar of pressure, $40^{\circ}C$ of extraction temperature, 3 mL/min of $CO_2$ flow rate, 0.2 mL/min of modifier (ethanol) flow rate.

In Vitro and In Vivo Anti-Oxidative and Anti-Inflammatory Activities of Acer tegmentosum Maxim Extracts (RAW 264.7 대식세포와 염증유도 동물모델에서 산겨릅나무 추출물의 항산화 및 항염증 효과)

  • Acer tegmentosum Maxim (ATM) is known as traditional medicine for treatment of hepatic disorders such as hepatitis, related-inflammatory disease, and hepatic cancer. In this study, we evaluated the antioxidant and anti-inflammatory effects of ATM extracted with $80^{\circ}C$ water or 95% ethanol. Antioxidant activities of ATM extracts were measured based on DPPH and ABTS radical scavenging activities, total polyphenolic compound contents, and ferric reducing antioxidant power. The anti-inflammatory effects of ATM extract were assayed on release of nitric oxide, tumor necrosis factor $(TNF)-{\alpha}$, and interferon $(IFN)-{\gamma}$ from lipopolysaccharide (LPS)-induced macrophages. In these experiments, 95% ethanol extract of ATM showed stronger antioxidant and anti-inflammatory effects than water extract. Therefore, we determined the effects of ATM ethanol extract on an animal model of sepsis. Seven days oral gavage of ATM ethanol extract followed by LPS stimulation reduced the protein levels of $TNF-{\alpha}$ and $IFN-{\gamma}$ in serum as well as mRNA levels of $TNF-{\alpha}$ and interleukin-6 in intestinal epithelial cells. In addition, ATM ethanol extract reduced DNA damage in mouse lymphocytes. These results indicate that ATM extract has strong antioxidant and anti-inflammatory in vitro and in vivo effects and may be developed as a potential food material for prevention of inflammatory diseases.

DPPH Radical Scavenging Activity of Phenolic Compounds Isolated from the Stem Wood of Acer tegmentosum (산겨릅나무 목질부에서 분리한 페놀성 화합물의 DPPH 라디칼 소거활성)

  • There have been few reports on the constituents and biological activity of stem bark of $Acer$ $tegmentosum$, and no phytochemical and biological studies have been reported for stem wood of $A.$ $tegmentosum$. Two flavan 3-ols (1 and 2), three phenolic acid/alcohols (3~5), and two coumarins (6 and 7) were isolated from the stem wood of $A.$ $tegmentosum$ by repeated column chromatography. The structure of isolated compounds were identified as (+)-catechin (1), (-)-epicatechin (2), $p$-hydroxybenzaldehyde (3), syringic alcohol (4), $p$-tyrosol (5), scopoletin (6), and cleomiscosin A (7) on the basis of spectroscopic evidences such as $^1H$-NMR, $^{13}C$-NMR, 2D-NMR and MS spectrum. $p$-Hydroxybenzaldehyde (3), syringic alcohol (4), scopoletin (6), and cleomiscosin A (7) have not been reported from this plant so far. (+)-Catechin (1) and (-)-epicatechin (2) showed the higher 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity than butylated hydroxyanisole (BHA) used as a positive control.

Metabolizing analysis according to the sawdust media of Pleurotus ostreatus (산겨릅나무와 옻나무 톱밥배지에서 재배한 느타리버섯의 함유성분 분석)

  • The transitivity of Chemical constituents by Pleurotus ostreatus cultivated in different raw sawdusts, which are Acer tegmentosum MAX, Rhus verniciflura, was investigated. The HPLC chromatography patterns on the chemical constituents of P. ostreatus showed the similar chromatography patterns in the different raw sawdusts and control sawdust. The unknown chemical constituents of P. ostreatus cultivated in the mixed medium added 10 %, 20% raw sawdusts, respectively, were increased. But the significance results in the mixed medium added 50% raw sawdusts were not showed. The chromatography patterns of mycelia grown in media added the 80% MeOH extracts of A. tegmentosum and R. verniciflura showed the similar patterns in comparison with control mycelia. In the results, the secondary metabolites of functional media were not degrade and changed to other derivatives compounds by P. ostreatus.
