Effects of Soil Amendments on the Early Growth and Heavy Metal Accumulation of Brassica campestris ssp. Chinensis Jusl. in Heavy Metal-contaminated Soil (중금속 오염 토양에서 안정화제가 청경채의 초기 생육과 중금속 흡수량에 미치는 영향)

  • There have been many studies about efficiency of amendments for heavy metal stabilization through chemical assessment. The objective of this study was to evaluate the efficiency of several soil amendments (lime, agric-lime, dolomite, steel slag, fly ash and acid mine drainage sludge) on heavy metals stabilization through not only chemical but also biological assessments (phytotoxicity test) in abandoned mining area soil. In order to achieve the goal, we conducted preliminary screening experiment targeting 12 types of crop plants such as radish, young radish, chinese cabbage, winter grown cabbage, cabbage, bok choy, chicory, crown daisy, carrot, chives, spinach, and spring onion. The results of inhibition rates of early plant growth in metal-contaminated soil against non-contaminated soil and the correlations between inhibitions items showed that the bok choy was appropriate specie with respect to confirm the effect of several amendments. Several amendment treatments on contaminated soil brought about the changes in the root and shoot elongation of bok choy after 1 week. Agric-lime, dolomite and steel slag treatments showed the great efficiency of reducing on mobility of heavy metals using chemical assessment. But in contrary, these treatments resulted in the reduction of root and shoot elongation and only AMD sludge increased that of elongation, significantly. When considering both chemical and biological assessments, AMD sludge could be recommended the compatible amendment for target contaminated soil. In conclusion, biological assessment was also important aspect of decision of successful soil remediation.

Change in Uptake and Tissue Contents of N, P, and K at Different Growth Stages in Hydroponically-Grown Cut Roses (양액재배 절화장미의 생육단계별 N, P, K 흡수 및 체내성분 함량의 변화)

  • During a 35-day growth cycle, N, P, and K uptake was determined by measuring changes in their contents in culture solutions. At harvest, plants were separated into the roots, base organs and shoot, and dried for tissue analysis for N, P, and K. The uptake rates of N, P, and K followed cyclical patterns that was related to shoot development and harvest, but were independent of the transpiration rate. Uptake of N declined from 5.6 mmol $plant^{-1}$ $day^{-1}$ just prior to the cycle initiation to 4.0 mmol $plant^{-1}$ $day^{-1}$ at day 15. Uptake rate steadily increased as flower stems reached maturity up to 10.3 mmol $plant^{-1}$ $day^{-1}$ day 35. Uptakes rates of P and K followed similar patterns of N uptake. Tissue concentrations of N and P steadily decreased since day 15. Content of K was the lowest at day 20 and steadily increased thereafter. In the root tissue, N and K contents were the lowest at day 15, increased to day 30, and then decreased at day 35. Tissue P content was just a reverse of those of N and K.

Effect on Growth and Quality of Oriental Melon (Cucumis melo L. var. makuwa Makino) by Interspace Mulching between Polyethylene Film House (하우스 동간멀칭이 참외의 생육 및 품질에 미치는 영향)

  • Effects of interspace mulching between polyethylene film house on growth and quality of oriental melon were followed. Soil moisture in control was 26.0%. Those of black nonwoven fabrics sheet and black film were less 1.2% and 2.9% than control, respectively. Soil temperature in control plot was $2.4^{\circ}$ Those of black nonwoven fabrics sheet and black film were higher $0.8^{\circ}$ and $1.8^{\circ}$ than control, respectively. Black film was better than control in stem length, leaf number and area, chlorophyll content, xylem exudate, first flowering and fruit riping periods. Black film was better in fruit weight and soluble solid, and lesser in fermented and malformed fruit ratios, and increased 28% in marketable fruit ratio to control, 1,864kg. This results were mainly from vinyl coverage between polyethylene film house to intercept rainfall and to increase soil temperature, which fasted growth of roots after transplanting in winter season.

Bio-Green' Functional Water Supply Influences Mineral Uptake and Fruit Quality In Tsugaru Apples (‘바이오 그린’ 기능수 처리가 사과 쓰가루 품종의 무기성분 흡수와 과실품질에 미치는 영향)

  • Commercial Bio- Green(B.G.) functional water was manufactured through a series of processes : water - ultra-purification - adding catalysts - energy imprinting fermenting with energized water + zeolite and others + photosynthetic bacteria in fermenter longrightarrow filtering. Control(0), 5 or 10 liters per plant of B.G. functional water were supplied to the orchard soil under canopy of 10 year- old ‘Tsugaru’/M26 apple trees on March 20, May 20 and June 20, 1995, respectively. pH and content of Ca and Mg of orchard soil were increased by supply with B.G. functional water. However, P$_2$ $O_{5}$, K, and B contents were not influenced by the treatment. At harvest time soluble solid content of flesh tissue and anthocyanin of fruit skin were increased by the treatment. B.G functional water treatment showed higher root activities, and photosynthesis of leaves than that of control. Also B.G. functional water treatment enhanced Ca content in fruit skin and flesh tissues, whereas not affected N, K, and Mg contents. During storage at 4$^{\circ}C$ cold room, the more volume of B.G. functional water supply showed lower bitter pit symptom. Respiration and ethylene evolution in fruit decreased, while fruit firmness increased by the treatment during storage.

Effects of treatment of Enterobacter ludwigii SJR3 on growth of tomato plant and its expression of stress-related genes under abiotic stresses (비생물적 스트레스 환경에서 Enterobacter ludwigii SJR3 처리 시 토마토의 생장과 스트레스-관련 유전자의 발현)

  • This study examined effects of Enterobacter ludwigii SJR3 showing a high 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity, on growth of tomato plant and its expression of stress-related genes under drought and salt stress. SJR3 strain was inoculated at $10^6cell/g$ soil to 4-week grown tomato plants, and drought and salt stresses were treated. After additional incubation for 1 week, root length, stem length, fresh weight and dry weight of tomato plants treated with SJR3 increased by 37.8, 37.2, 96.8 and 146.6%, respectively compared to those of uninoculated plants in drought stress environment, and they increased by 19.2, 25.4, 19.5, and 105.8%, respectively in salt stress environment. Proline content in tomato leaves increased significantly under stress conditions as one of a protecting substance against stresses, but proline contents in tomato treated with SJR3 decreased by 62.1 and 54.1%, respectively. Relative expression of genes encoding ACC oxidase, ACO1 and ACO4, ethylene response factor genes ERF1 and ERF4, and some other stress-related genes were examined from tomato leaves. Compared to the non-stressed tomato, expressions of all stress-related genes increased significantly in the stressed tomato, but gene expressions in the inoculated tomato were similar to those of no-stressed control tomato. Therefore, E. ludwigii SJR3 may play an important role in mitigating drought and salt stress in plants, and can increase productivity of crops under various abiotic stresses.

Ecological Niche of Quercus acutissima and Quercus variabilis (상수리나무와 굴참나무의 생태적 지위에 관한 연구)

  • In Korea, Quercus acutissima distributed in good condition with high nutrients and moisture content, but Quercus variabilis in dry soil or rock habitate. In order to understand this ecological distribution of Q. acutissima and Q. variabilis, we cultivated the seedlings of two oak species treated with light, soil moisture and nutrient gradients each four level, from May to October in glass house. Then we measured the ecological niche breadth and niche overlap of the two species, and analyzed the relationship of competition using cluster analysis and PCA ordination. Ecological niche breadths of Q. acutissima under moisture and nutrient treatments were slightly wider than those under light one. Among 14 characters measured, 6 characters related with length items were wider in all the environmental treatments, but 8 characters connected with weight terms narrower in light treatment. Ecological niche breadths of Q. variabilis under moisture and nutrient treatment were wider than those of light one. Ecological niche of Q. acutissima was wider than those of Q. variabilis in all the environmental treatments. Ecological overlap between two species was higher with a range of 0.87$\sim$0.92, especially higher in soil moisture factor. These results means that Q. acutissima is more competitive than Q. variabilis, especially in soil moisture condition. Two species were ordinated with distinct group based on 9 characters. From these results, it can be explained that what Q. variabilis distributed in bad soil condition is due to the escape strategy, because of its low competitive ability to Q. acutissima in natural communities.

Appropriate Drainage Position in Coir Bag Culture Using U-type Bed (U자형 베드에서 코이어 자루재배 시 적정 배액구 위치 구명)

  • This study was carried out to investigate damp injury caused by tomato coir bag culture and prevent that. The tomato variety was used 'Minichal', and tomatoes were grown in greenhouse. The nutrient solution based on Yamazaki Tomato Standard Solution was irrigated from one hour after sunrise to two hour before sunset. The slits for darainage were made in three types; I, L, and bottom slit type. The coir bag of I and L type had six slits of 15cm length, that of bottom slit type had three slits of 15 cm length. The weight of coir bag in 24 hours after saturation was 14.2 kg in I type, 13.8 kg in L type, and 12.8 kg in bottom slit type, but there was not significant difference. The weight of coir bag after one day irrigation was 14.5 kg, 14.2 kg, and 13.3 kg at L, I, and bottm slit type, respectively. This means that the moisture content of coir bag during cultivation was lowest in bottom slit type. The number of adventitious root on stem was 160, 170, and 53 at I, L, and bottom slit type, respectively. The dry weight of root and root length were highest at bottom slit type, compared to other treatment. The marketable yield was highest 26.5 kg/20 plant in bottom slit type. For increasing yield and preventing damp injury, bottom slit type was most effective at U type bed coir bag culture.

The Effect of Nutrient Solution Concentration on Growth of Potato Plantlet in Microponic System (Microponic system에서 배양액의 농도변화가 감자 소식물체 생육에 미치는 영향)

  • It was intended to closely examine an effect that a change in the concentration of culture medium had on the potato(Solanum tuberosum L.) plantlet growth in the microponic system so as to mass-produce the virus-free plant of new variety 'Saebong' for potato processing. The adjusted concentration of potato culture medium was 0.2, 0.6, 1.0, 1.4, 1.8, and $14.0dS{\cdot}m^{-1}$. And potato seedling was cut into pieces of 1.5 cm in length, which included 2 growth points and leaves. And each was explanted in glass vial of 50 mL. And experiments were carried out twice for 18 days or 21days. Culture medium of 2ml was put in the container respectively. And 1 mL was added after 10 days. And in terms of cultivation environment, the experiment was carried out at the day length of 16 hours at the temperature of $23{\pm}1^{\circ}C$ under the white LED light of $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$. The concentration of culture medium in the experiment I was EC 0.2, 1.0, $14dS{\cdot}m^{-1}$ and was adjusted to 0.6, 1.0, 1.4, $1.8dS{\cdot}m^{-1}$ in the experiment II. The results showed that the survival rate of plantlet was 90% at $0.2dS^2m^{-1}$, 100% at $0.6dS^2m^{-1}$, 100% at $1.0dS^2m^{-1}$. 0% at $1.4dS{\cdot}m^{-1}$, 0% at $1.8dS{\cdot}m^{-1}$. and 0% at $14.0dS{\cdot}m^{-1}$ after 7 days. With regard to the explanted potato seedling, in case of the treatment where the electrical conductivity of culture medium was adjusted to $1.0dS{\cdot}m^{-1}$, root developed 2 days after transplantation. And the plantlet vigorously grew into strong plant that had 7 leaves, length of 5cm, and fresh weight of 0.5 g after 18 days. In case of the treatment where the concentration of culture medium was adjusted to $0.6dS{\cdot}m^{-1}$, the root plantlets developed 4 days after transplantation. And those grew into plant that had 7 leaves and fresh weight of 0.2 g after 21 days. Therefore, we found that it is effective to control potato culture medium by adjusting its electrical conductivity to $0.6{\sim}1.0dS{\cdot}m^{-1}$ for the mass production of virus-free potato seedling in the microponic system.

Growth and Runner Production of 'Maehyang' Strawberry as Affected by Application Method and Concentration of Cytokinin (딸기 '매향'의 사이토키닌 처리방법과 농도에 따른 생육과 런너 생산)

  • This study was conducted to examine the effect by application method and concentration of plant growth regulator (PGR) on the growth and runner production of strawberry (Fragaria ${\times}$ ananassa Duch. cv. Maehyang) in a velno-type greenhouse. The seedlings of strawberry were transplanted in pot ($64{\times}27{\times}18cm$) filled with commercial mixed medium (Tosilee) on February 22nd, 2016. The 6-benzylaminopurine (6-BAP) was applied with foliar spray or drench, respectively as 900, 1,200 or $1,500mg{\cdot}L^{-1}$ (50 mL per plant) at 3 weeks after transplanting. Nutrient solution was sufficiently supplied by the drip irrigation as EC $0.65dS{\cdot}m^{-1}$ for rooting during 7 days. After rooting, the 450 mL nutrient solution supplied per pot twice a day (10 min). Plant height and crown diameter of 'Maehyang' mother plant appeared no significantly difference. The other growth characteristics, such as root length, number of primary roots, leaf length, leaf width, leaf area and fresh and dry weights of the shoot or root, were significantly the greatest in the control. And, the SPAD value of strawberry was the highest as 44.2 in the drench with $900mg{\cdot}L^{-1}$. The foiler spray was more effective in runner production than drench, and the number of runners appeared high values at the 900 and $1,500mg{\cdot}L^{-1}$. Whereas, the number of strawberry plantlets was effective in the drench. The results indicate that both growth and the number of runners of strawberry plant were the best achieved by foliar spray application at the $900mg{\cdot}L^{-1}$.

Optimum Strength and NH4+:NO3- Ratio of Nutrient Solution for Romaine Lettuce Cultivated in a Home Hydroponic System (가정용 수경재배기에서 재배한 로메인상추의 생육에 적합한 양액 강도와 NH4+:NO3-의 비율)

  • Concentration of nitrogen, one of the major elements, and ratio of two nitrogen forms (NH4+ and NO3-) in the nutrient solution affect the quality and food safety of fresh vegetable produce. This study was conducted to find an appropriate strength and NH4+:NO3- ratio of a nutrient solution for growth and development of a Romaine lettuce (Lactuca sativa L. var. longiflora) 'Caesar Green', a representative leafy vegetable, grown in a home hydroponic system. In the first experiment, plants were grown using three types of nutrient solution: A commercial nutrient solution (Peters) and two strengths (GNU1 and GNU2) of a multipurpose nutrient solution (GNU solution) developed in a Gyeongsang National University lab. Plants grown with the GNU1 and GNU2 had greater shoot length, leaf length and width, and biomass yield than Peters. On the other hand, the root hairs of plants grown with Peters were short and dark in color. Tissue NH4+ content in the Peters was higher than that of the GNU1 and GNU2. The higher contents of NH4+ in this solution may have caused ammonium toxicity. In the second experiment, eight treatment solutions, combining GNU1 and GNU2 solutions with four ratios of NO3- :NH4+ named as 1, 2, 3 and 4 were used. Both experiments showed more growth in the GNU2 group, which had a relatively low ionic strength of the nutrient solution. The growth of Romaine lettuce showed the greatest fresh weight along with low tissue NO3- content in the GNU2-2. This was more advantageous in terms of food safety in that it suppressed the accumulation of surplus NO3- in tissues due to the low ionic trength of the GNU2 subgroup. In addition, this is preferable in that it can reduce the absolute amount of the input of inorganic nutrients to the nutrient solution.
