Effects of Water-Soluble Extract from Leaves of Morus alba and Cudrania tricuspidata on the Lipid Peroxidation in Tissues of Rats (뽕나무(Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata) 잎의 수용성 추출물이 흰쥐 각 조직중의 지질 과산화물 함량에 미치는 영향)

  • 뽕나무 (Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata) 잎으로부터 추출한 수용성 물질을 0.5% 콜레스테롤 투여 식이에 1%(w/w) 수준으로 첨가하여 각 조직에서의 지질 과산화물 생성정도를 나타내는 TBARS 측정법으로 항산화 활성을 검토하였다. 흰쥐의 체중 및 식이 섭취량은 실험군 간의 유의적인 차이는 없었다. 간장 및 뇌 조직 중량은 뽕잎 추출물군에서 증가하였다. 신장의 지질 과산화물 함량은 대조군에 비해서 뽕잎 추출물군에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화물 함량은 대조군에 비교해서 꾸지뽕잎 추출물군에서 유의적으로 증가하였다. 간장 및 비장의 경우에는 각 실험군 간에 유의적인 차이가 없었다. 각 조직의 microsomes 분획으 지질 과산화물은 심장과 비장에서 뽕잎 및 꾸지뽕잎 추출물군에서 감소하였으나, 다른 조직에서는 현저한 차이가 없었다. 한편, 각 조직의 microsomes 분획을 얻어서 Fe2+/ascorbate로 지질 과산화물을 유도하기 위하여 37$^{\circ}C$에서 한시간 반응을 시킨 결과, 뽕잎 및 꾸지뽕잎 추출물군에서 약 절반 정도 감소하였으며, 상대적 증가량은 뇌조직 microsomes에서 현저하였다. 본 실험의 결과 신장조직 및 간장 microsome막 지질 과산화물 함량은 뽕잎 및 꾸지뽕잎 수용성 추출물에 의해 감소되었으며, 뇌 및 심장조직의 지질 과산화물 함량은 꾸지뽕잎 수용성 추출물에 의해 증가됨으로써 봉잎 추출물간의 차이를 나타내었다.

Effects of Mulberry Leaf Tea Fermented by Monascus pilosus on Body Weight and Hepatic Antioxidant Enzyme Activities in Mouse Fed High-Fat Diet (Monascus pilosus 발효 뽕잎차가 고지방 식이 마우스의 체중과 간 조직 항산화계 효소 활성에 미치는 영향)

  • In this study, we investigated the preventive effects of the mulberry leaf tea fermented by Monascus pilosus on high fat-induced obesity, hyperlipidemia, and fatty liver in mice. Non-fermented mulberry leaf tea powder (UM) and fermented mulberry leaf tea powder (FM) were supplemented with high-fat diet at 2% (wt/wt) dosage for 8 weeks. Both UM and FM lowered body weight gain, feed efficiency ratio, epididymal fat, serum triglyceride, total cholesterol and LDL-cholesterol increased markedly with high fat diet (HC) in mice. FM showed more significant effects when it was compared with UM. In addition, Hepatic lipid peroxides and xanthin oxidase activities of the UM and FM were significantly lower than those of HC, despite the lack of a big difference in the amount of hepatic GSH. Activities of ROS scavenging enzymes and serum alanine aminotransferase activity were also examined as a parameter of hepatic damage. The UM and FM groups showed a recovery to NC group from significant changes induced by HC. Finally, histopathological analyses of liver samples revealed a decrease of lipid accumulation in hepatocytes in the UM and FM groups. These results suggest that UM and especially FM can reduce the development of obesity, hyperlipidemia and fatty liver.

Agronomic Characteristics and Anti-oxidant Capacity of Mulberry Genetic Resources conserved by Jeollabuk-Do (전라북도 뽕나무 유전자원의 재배생태적 특성 및 항산화능 분석)

  • This study was carried out to develop the utilization of mulberry resources conserved by Jeollabuk-Do Agricultural Research & Extension Services. Mulberry accessions were tested for agronomic characteristics and antioxidant capacity according to varieties. From that result, three suitable varieties such as 'Sinilppong', 'Suwonsang 2' and 'Ilbongeum' were selected for the production of mulberry leaves. They have strong agronomic characteristics like size, yield and resistance against damages by blight and harmful insects. Whereas, 'Gumunyoung' showed the lowest freezing resistance. In the antioxidant capacity analysis of mulberry leaves, autumn's mulberry leaves showed higher antioxidant capacity (ascorbic acid equivalent) than that of spring's it. According to collection time, antioxidant capacity were 2,109.8 nmol (August), 2,617.8 nmol (September) and 3,311.5 nmol (October), respectively.

  • In the present study,effect of natural mulbelly leaf tea on DMEM MDCK cell line. Mulbelly leaf teas inhibited the proliferation in primery MDCK cells in a dose dependent manner. These results show that mulbelly teas potent inhibits the reactive oxygen species (ROS) and not destruction a component. Therefore, mulbelly teas might improve overall quality of the color and taste. And might be applied newly to development of componant mulbelly teas quality and biochemical change by ROS in living things.

Effects of Dietary Supplementation of Mulberry Leaves and Dandelion Extracts on Storage of Chicken Meat (뽕잎과 민들레 추출물을 급여한 계육의 저장성에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of mulberry leaves and dandelion extracts on pH, meat color, TBARS (thiobarbituric acid reactive substance), and VBN (volatile basic nitrogen). Two hundred broiler chickens were fed diets for five weeks containing 1% mulberry leaves extracts (T1), 2% mulberry leaves extracts (T2), 1% dandelion extracts (T3), and 2% dandelion extracts (T4). At the end of five weeks feeding experiment, broilers were slaughtered, and stored at $4^{\circ}C$ for five weeks. As storage time increased, the presence of mulberry leaves and dandelion extracts resulted in decreased pH, and $L^*$ and increased TBARS, VBN, and $a^*$ value in all treatment groups (P<0.05). The pH value, TBARS, and VBN were significantly decreased by the supplementation of mulberry leaves and dandelion extracts relative to the control (P<0.05). Therefore, mulberry leaves and dandelion extracts had the possibility to improve shelf life of chicken meat. Especially, T4 was significantly more effective in delay lipid oxidation compared to the control group. However, no significantly difference was found in pH, TBARS, and VBN among mulberry leaves and dandelion extracts treatment groups. In conclusion, this study demonstrates that compared to control group, supplementation of mulberry leaves and dandelion extracts were effective in decreasing pH, TBARS, and VBN and increasing $a^*$ value.

Anti-diabetic effect of the mixture of mulberry leaf and green tea powder in rats with streptozotocin-induced diabetes (뽕잎과 녹차혼합분말의 급여가 streptozotocin 유발 당뇨쥐의 항당뇨효과)

  • This study investigated the hypoglycemic effects of mulberry leaf (M), green tea (G), and a mixture of mulberry leaf and green tea (MG) in rats with streptozotocin-induced diabetes. Male Sprague-Dawley rats were divided into the non-diabetic (N) and diabetic groups. The experimental animals were divided into four groups and fed the following for four weeks: a normal diet (N), the STZ+normal diet (STZ), the STZ+5% mulberry leaf (STZ-M), STZ+5% green tea (STZ-G), and the STZ+5% mixture of mulberry leaf and green tea (STZ-MG). The whole blood glucose level of the STZ-M, STZ-G and STZ-MG groups significantly decreased compared to the STZ group two weeks later. The serum glucose and fructosamine, and urine glucose levels in the STZ-M, STZ-G and STZ-MG groups were lower than in the STZ group. The levels of serum insulin were higher in the STZ-M, STZ-G and STZ-MG groups than in the STZ group. However, serum glucose, fructosamine and insulin levels, and urine glucose levels were not significantly different among the STZ-M, STZ-G and STZ-MG groups. These results indicate that mulberry leaf, green tea, and their mixture help prevent or attenuate the progression of diabetes in rats with STZ-induced diabetes.

Effects of Dietary Supplementation of Powdered Mulberry Leaves on Egg Production, Egg Quality and Blood Characteristics in Laying Hens (산란계 사료에 뽕잎 분말 첨가 급여가 산란율, 계란 품질 및 혈액 성상에 미치는 영향)

  • This study was conducted to investigate the effects of supplementation diets of powdered mulberry leaves on egg production, egg quality and blood characteristics in laying hens. Laying hens were fed diets for 30 days containing 0% powdered mulberry leaves (Control), 1% powdered mulberry leaves (T1), 2% powdered mulberry leaves (T2), and 5% powdered mulberry leaves (T3). The egg production, yolk weight and albumen weight were no significant difference among treatment groups. The albumen weight was significantly decreased by the supplementation of powdered mulberry leaves in all treatment groups (P<0.05). The albumen high, yolk color score, egg sell thickness and egg shell breaking strength were no significant difference. The total cholesterol and triglyceride concentration of control was higher than T3, and the high density lipoprotein (HDL)-cholesterol concentration and glucose of control were lower than treatment groups (P<0.05). The white blood cell (WBC) was significantly increased by the supplementation of powdered mulberry leaves compared to the control (P<0.05). In conclusion, these data indicate that 5% powdered mulberry leaves supplementation was most effective in decreasing total cholesterol and triglyceride and increasing HDL-cholesterol, glucose and WBC.

Antimicrobial Effect of Mulberry Leaves Extracts Against Oral Microorganism (뽕잎 추출물의 구강미생물에 대한 항균효과)

  • In the current research for natural product with antimicrobial effects, various extracts of Mulberry Leaves against microorganisms were evaluated in terms of the minimum inhibitory concentrations(MIC). In general, Candida albicans was more antimicrobial activity than the other microorganisms such as Streptococcus mutans, Staphylococcus epidermis, and Staphylococcus aureus. The maximum activity was exhibited by ethanol extract of the leaves of Mulberry Leaves against Candida albicans (MIC, $1600{\mu}g/ml$). These results suggest that ethanol and water extracts of Mulberry Leaves have a potential antimicrobial activity.

Effects of Number of Days After Urea Foliar Spray on Mulberry Leaf Components and Silkworms (요소 엽면시비후 경과일수가 뽕잎 화학성분과 누에에 미치는 영향)

  • The effect of short term duration(1, 2 and 3 days) after urea foliar spray on leaf quality was investigated by chemical analysis of leaves and silkworm rearing with leaves treated with urea in autumn. Foliar spray increased water content by 0.4~0.7% and total nitrogen by 0.19~0.51% in leaves compared to the conventional treatment. Foliar spray increased NO3-N concentration in leaf 503~1107 ppm more than the conventional treatment. NO3-N concentration in leaf decreased with days after foliar spray. Whole cocoon weight and cocoon shell weight increased by 0.02~0.14g and 0.5~3.1 cg, respectively in foliar spray than in the conventional treatment. Though foliar spray raised NO3-N concentration in leaf, leaves harvested one day after foliar spray were no harmful to silkworm.

Effects of Dietary Mulberry Leaf on Loperamide-Induced Constipation in Rats (식이뽕잎이 흰쥐의 Loperamide로 유도된 변비에 미치는 영향)

  • We investigated the effect of dietary mulberry leaf powder (MP) on loperamide-induced constipation in rats. Male Sprague-Dawley rats were given MP in their diets at a concentration of 0% 5% and 10% for 33 days. Rats were divided into 4 groups: normal diet group (NOR), normal diet and loperamide treated group (MPL0), 5% MP and loperamide treated group (MPL5), and 10% MP and loperamide treated group (MPL10). Constipation was induced by subcutaneous injection of loperamide (1.5 mg/kg body weight/day) for the final 5 days of the experiment Supplemental MP had no effect on the food efficiency ratio, but it reduced body weight gain and food intake in a concentration dependent manner. Administration of loperamide decreased food intake. MP had a concentration-dependent effect on decreasing total cholesterol, LDL-cholesterol, and triglycerides and on increasing HDL-cholesterol. Loperamide had no significant effect on serum lipid profiles. Loperamide decreased the number and wet weight of fecal pellets and fecal water content MP increased the number and wet weight of fecal pellets and fecal water content in a dose-dependent manner. In addition, MP increased gut transit time and transit speed, and the guts of mts treated with MP plus loperamide were longer than those of mts treated with loperamide alone. These results indicate that MP is an effective treatment for constipation.
