Effect of Betaine on Immune Response in Laying Hens (비태인이 산란계의 면역 반응에 미치는 영향)

  • This experiment was conducted to determine the effect of betaine on immune response in laying hens. A total of 72 ISA-brown laying hens were divided into four groups of 18 hens each and fed corn-soybean meal based diets with addition of 0, 300, 600 and 1,200 ppm betaine for four weeks. The effect of betaine on splenocyte proliferations with mitogens, concanavalin A(Con A) and pokeweed mitogen(PWM), were assayed after incubation using [3H] thymidine uptake. Proliferations of splenocyte were significantly increased by activation of mitogen Con A or PWM. Mitogen effects of Con A were increased by Con A plus betaine injection(0.1 mM), whereas PWM effects did not affect in PWM plus betaine injection(0.1 mM) in vitro. Splenocyte of laying hens fed betaine tended to proliferate in the presence of PWM, but appeared to be slightly suppressed in the presence of Con A in vivo. Proliferation of splenocytes which were stimulated by Con A or Con A+betaine injection(0.1 mM) were increased in dietary 600 ppm betaine, but inhibited in dietary 1,200 ppm betaine supplementation. Spleen weights and sheep red blood cell(SRBC) titers of hens fed betaine tended to increase compared to those of control, but were not significantly different. These results suggested that betaine could increase splenocyte proliferation in vitro.

Influence of Dietary Supplemental Betaine on Performance and Egg Quality of Laying Hens during the Heat Stress (고온 스트레스기에 비태인의 사료첨가가 산란계의 생산성 및 계란의 품질에 미치는 영향)

  • Betaine functions as an osmoregulators in the cells and its inclusion in diet can spare the choline and carcass fat reduction in chicken. Thus, two hundred eighty eight laying hens were fed with 0, 500, 1,000, 2,000 ppm of betaine from seventy eight to eighty six weeks of age during the environmentally high temperature stress. Com and soybean basal diets contained 2,800 kcal/kg ME and 16% CP. Egg production, feed intake, and feed conversion were examined for eight weeks. Egg quality characters, serum cholesterol, liver betaine, and lower ileal osmolality were measured at the end of experiment. Egg Production rates of hens fed with 500 or 2,000 ppm of betaine were 75.06 and 75.02%, respectively and tended to increase compared to the control. The feed conversion(FCR) of these treatments was significantly(P<0.05) improved compared to that of control although it did not significantly differ in the e99 Production rates between 500 and 2,000 ppm of betain groups. Eggshell breaking strength of hens fed betaine was significantly(P<0.05) higher than those of control. However, betaine supplements did not influence to improving the albumen height and Haugh unit. Liver betaine in hens fed with betaine was linearly increased unto 2,000 ppm. The birds fed with 2,000 ppm betaine showed significantly(P<0.05) higher in the liver betaine than the control birds. Total cholesterol and triglyceride tended to be increased by dietary betaine supplement. The lower ileal osmolality in betaine supplement group tended to be slightly decreased. As a result, dietary betaine supplement tended to improve the egg Production and eggshell Quality in laying hens during heat stress.

  • Two experiments were conducted to investigate the effect of betaine on egg production, lipid metabolism, and osmoregulation in 18-to 42-week-old ISA Brown laying hens. In experiment 1, three hundred and sixty one hens were fed a com-soy basal diet contailing 16% crude protein (CP), 2800 kcal/kg metabolizable energy (ME), 0.33% methionine, and 0, 300, 600, or 1200 mg betaine per kg diet. Egg production, egg weight, feed consumption, feed conversion, and egg quality were measured every eight weeks. Betaine concentration in live and egg were determined along with serum cholesterol, abdominal fat, total serum protein and albumin levels. In experiment 2, twenty thirty-three-week-old laying hens were fed the same diets as those used in experiment 1 in individual cages and the amount of feed and water consumption were measured for two weeks. At the end of experiment 2, all birds were killed to determine blood plasma and ileal osmopressure, arginine vasotocin (AVT), and liver moisture content. In experiment 1, egg production between the treatments during the first eight weeks were not different, whereas the significant increment of egg production were noticed in the birds fed more than 600 ppm betaine after reaching the peak egg production stage (p<0.05). The egg weight was reduced significantly by the betaine supplementation for the first 8 weeks (p<0.05). Feed conversion tended to improve by betaine supplement. Egg quality was not enhanced by betaine supplementation. Liver betaine level increased with betaine feeding compared to the control but betaine concentration in eggs decreased with betaine supplementation. Betaine supplementation elevated the level of serum total cholesterol and triglyeerides compared to the control. Abdominal fat content was increased by betaine supplementation, whereas liver fat content decreased. In experiment 2, water consumption significantly increased in hens fed diets containing 300 and 600 mg betaine/kg (p<0.05) and osmotic pressure of ileal digesta increased with betaine supplement. Liver moisture content was not affected by betaine, but AVT increased in hens fed betaine. The overal results suggested the possibility of using betaine as a feed additives in the laying hens beacuse of its positive contribution to improving egg production and other metabolic parameters related to lipid metabolism.

단백질 수준에 따른 비태인의 급여가 산란계의 생산능력과 난품질, 혈액 성상에 미치는 영향

  • 비태인의 급여(0, 600 ppm)가 사료의 단백질 수준(14, 16 %)에 따라 산란계의 생산성과 난품질, 혈액성상, 간과 가슴육의 일반성분에 미치는 영향을 구명하기 위하여 83주령 하이라인 산란계 192수를 이용하여 12주간 사양실험을 실시하였다. 기초사료는 에너지 수준이 2,800 kcal/kg, methionine과 lysine, cystine의 수준은 단백질 수준에 비례하도록 하였다. 조사항목으로 산란율과 난중, 사료섭취량, 사료요구율, 계란품질은 4주 간격으로 측정했다. 실험 종료시 복강지방과 혈액중 total protein과 albumin, BUN, triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol 함량, 간의 methionine, choline 함량, 간과 가슴육의 수분, 조단백질, 조지방 함량을 측정하였다. 산란율은 단백질 수준에 따라 증가하였으며(P<0.05) 비태인 급여에 의한 차이는 없었다. 난중은 단백질과 비태인 급여로 증가하였으며(P<0.05) 산란양은 단백질 수준이 증가할수록 감소하였다(P<0.05). 비태인 급여로 사료 요구율은 현저하게 개선되었지만(P<0.05), 계란의 품질은 차이가 없었다. 혈청 total protein은 비태인 급여로 현저히 증가하였으며 특히 단백질 14 % 급여구에서 크게 증가하였다(P<0.05). 복강지방 함량은 단백질 수준과 비태인 급여에 따라 감소하는 경향을 보였다. 가슴육과 간의 조단백질 함량은 사료의 단백질 수준에 따라 증가하였다(P<0.05). 간의 조지방 함량은 비태인의 급여로 감소하는 경향을 보였다. 간의 methionine 함량은 단백질과 비태인 수준에 따라 증가하는 경향을 보였으며 choline 함량은 비태인의 급여에 의해서만 증가하는 경향을 보였다. 따라서 비태인의 급여는 단백질 수준이 높은 조건에서 산란율을 개선시키고 난중을 증가시키며 사료 요구율을 개선한다.록 산가는 증가하는 것을 볼 수 있다. 홍국주의 전자 공여능에 의한 항산화력은 25.6%, 아질산염 소거능은 27.6%, Total phenolic compound 함량은 12.34mg%, ACE저해작용은 38%의 항산화력을 나타냈다.과 $O_2$$CO_2$의 농도에서 평균오차 0.2%로 정밀한 것으로 나타났으며 호흡속도 측정용 챔버의 혼합기체 공급측과 배기측의 가스 농도를 3회 반복 측정한 결과 재현성에서는 0.1%이하의 편차로 나타났다. 개방계 호흡속도 자동 측정 시스템을 이용하여 환경기체조성하에서 토마토의 호흡속도를 측정하는 실측 실험을 수행한 결과 2$0^{\circ}C$에서 12.7~42.1mg$CO_2$/kg.hr였으며 12$^{\circ}C$에서 2.5~8.2mg$CO_2$/kg.hr로 일반적으로 보고되고 있는 토마토 호흡속도와 일치하는 결과를 나타내었다.다.환원당인 sucrose 함량은 계속 증가하였고 fructose, glucose, sorbitol의 함량(추황의 sorbitol을 제외)은 생장이 촉진됨에 따라 증가하다가 다시 점차적으로 감소하였다. 이러한 결과는 총당과 환원당의 측정결과와 일치한 것으로 나타났다. 결론적으로 배의 성장에 따라 산 함량은 감소하였고 당 함량은 증가하였다.luco-pyranoside, quercetin 7-O- -glucopyranoside, acacetin 7-O-$\beta$-D-glucuronide and apigenin-6-C-$\beta$-D-glucopyranosyl-8-C-$\beta$-D-glucopyranoside were first isolated and identified from safflower leaf. Effects of Dietary Betaine and Protein Levels on Performance, Blood Composition, Abdominal Fat and Liver Amino Acid Concentration in Laying Hens (사료내 비태인과 단백질 수준이 산란계의 생산 능력, 혈액의 조성, 복강지방 및 간의 아미노산 함량에 미치는 영향)

  • An experiment was conducted to investigate the effects of dietary betaine and protein levels on performance, internal e99 Qualify, blood composition, abdominal fat and liver amino acid concentration in laying hens. A total of 432 ISA-Brown laying hens were randomly allotted to four treatments with six replications for twenty few weeks. Treatments were factorially designed with two levels of crude protein(14.5 and $16.5\%$) and two levels of betaine(0 and 600ppm). The egg production was increased by feeding $16.5\%$ protein-fed group compared to that of $14.5\%$ protein-fed groups(P<0.05), but was different from betaine supplementation. Increasing protein and betaine supplementation to diets improved egg weight, egg mass and fred conversion(P<0.05). Eggshell breaking strength, thickness and Haugh unit were not influenced by either dietary betaine or protein levels. Dietary betaine did not affect serum total protein, albumin and uric acid concentration, whereas uric acid concentration was increased in $16.5\%$ protein-fed groups(P<0.05). The deposition of abdominal fat was lower in 600ppm betaine groups compared to those fed on 0ppm betaine groups(P<0.05). Concentration of the most hepatic amino acids were affected by dietary protein levels and betaine supplementation. There were synergistic effect between dietary protein and betaine. These results suggested that dietary betaine can improve the egg weight, mass, feed conversion and decrease the abdominal fat, but increase the hepatic amino acid contents.

Effect of Dietary Betaine and Energy Levels on Liver Fats and Cholesterol in Laying Hens (사료내 비태인과 에너지 수준이 산란계의 간지방과 콜레스테롤에 미치는 영향)

  • Two experiments were conducted to examine the effects of betaine intake on blood and yolk cholesterol, abdominal fat, liver fat, tissue triglyceride(TG) and liver HMG-CoA reductase In laying hens. In Expt. 1, a total of 72 ISA-brown laying hens were individually assigned into four treatments from 18 to 21 weeks old. Com-soybean meal based diet were fed with the addition of 0, 300, 600 and 1,200ppm. In Expt. 2, 72 ISA-brown laying hens were housed into individual cage to evaluate the effect of dietary betaine(0, 600ppm) and energy(ME, 2,800, 2,900kca/kg) from 70 to 74 weeks. Serum total cholesterol, LDL and HDL-cholesterol and TG concentration in blood of hens fed betaine tended to increase compared to those of the control, but were not significantly different. However, betaine supplementation showed a statistically significant decrease in yolk cholesterol(P<0.05). There was no significant difference in abdominal fat among the treatments. Liver fats and 7c of birds 130 betaine was decreased compared with control. Serum total cholesterol, LDL-cholesterol and triglyceride concentration were significantly inc.eased by ffeding a diet containing 600ppm betaine in Expt. 2(P<0.05), but were not influenced by the dietary energy levels. Yolk cholesterol, abdominal fat and HMG-CoA reductase activity were affected neither by dietary energy nor betaine level.

Effects of Feeding Betaine on Performance and Blood Hormone in Laying Hens (산란계에 비태인의 급여가 생산 능력과 혈중 호르몬에 미치는 영향)

  • An experiment was conducted to investigate the effects of dietary betaine on egg production, estradiol, progesterone, and melatonin secretion in laying hens. Seventy two ISA-Brown laying hens were placed into individual cage and fed four different levels of betaine (0, 300, 600, 1,200 ppm) in diets for four weeks. Basal diets were mainly mixed with corn-soybean and contained 2,800 kcal/kg ME and 16% CP. Dietary supplementation of betaine showed significantly higher (p<0.05) egg weight and daily eggmass than controls, but did not have consistent influence on the egg production. The serum estradiol was statistically different in betaine treatments compared to that of control (p<0.05). No significant changes in serum progesterone concentrations were observed by the addition of betaine in diets. Serum melatonin concentrations also increased with betaine supplementation in diets (p<0.05). The results of this study indicate that the dietary betaine may contribute to increase daily eggmass, serum estradiol and melatonin concentration in laying hens.

산란계 사료에 비태인의 수준별 급여가 하절기 생산성 및 계란의 품질에 미치는 영향

Basal diets contained 16% CP and 2,800 ME. Egg production, feed intake, feed conversion were examined for eight weeks. Egg qualities, liver betaine, blood osmolarity, AntiDiuretic Hormone(ADH) were measured at the end experiment. Egg production of hens fed 500, 2,000 ppm dietary betaine and showed significance between control and 2,000 ppm betaine treatment(P<0.05). ADH of blood sera tended to increase as dietary supplemental betaine increased. The results of this experiments indicated that dietary supplemental betaine was able to improve the performance, eggshell breaking strength, liver betaine in this experiment.

EFFECT OF 5-FLOROURACIL ON THE POSTANATAL DELEVELOPMENT OF THE CONDYLE IN MICE

  • 항암 화학요법제의 하나인 5-Fluorouracil 이 과상돌기의 성장에 미치는 영향을 연구하기 위하여 20두의 백서를 반씩 나누어 실험군에는 동약물을 체중 kg당 25mg씩 2일간 복강내주사하고 대조군에는 생리식 염수를 주사한 후 각각 제1,3,7,14 및 21일에 두 마리씩 도살하여 과상골디연골부의 조직학적 변화를 관찰하였다. 또한 도살 2시간 전에 5μCi/kg mM 의 ³H-thymidine을 주사하여 표본의 일부는 자기방사법적으로 처리 관찰 하였다. 관찰 결과는 다음과 같다. 1. 과상돌기 연골부의 후경에 상당한 감소가 일어났으며 제7일에서 가장 현저하였다. 2. 이러한 영향은 연골제거대와 세포증식대에서 가장크고 그 다음이 비대층이었다. 3. 제 7일의 변화 증식대엣는 기질 호 염색성의 증가, 핵 형태의 난원형화가 오고 소와내의 세포수는 1~2개였다. 비태층의 소와는 현저하게 작아지고 기질의 호염기성이 뚜렷하며 세포의 배열은 츹어지고 약간의 핵은 그 배 열이 불규칙하며 숫자고 대조군보다 적었다. 4. 위와같은 변화는 제 14일에서 감소하고 점차 회복적인 조직소견을 보여 제21일에는 성장이 거의 대조군에 근접한 조직상을 보였다. 5. 자기방사법적으로 세포증식대 및 연골제거대에서 세포분열에 대한 억제효과를 관찰한 바 제 7일 에서 가장 시하였고 특히 연골제거대에서 심하였다. 대체적으로 자기방사법적 관찰소견은 조직학적 소견과 일치하였다. 결론적으로 하악골 과상돌기의 성장은 5-Fluorouracil의 영향하에서 세포의 증식 및 분화상태로 보아 일정기간동안 저해되며 제 7일의 극기를 지나 점차 회복되어감을 나타내었다.
