Effects of dietary coenzyme Q10 on adriamycin-induced myocardial ultrastructural changes in rats (식이중의 Coenzyme Q10첨가가 Adriamycin을 투여한 흰쥐의 심근 미세구조에 미치는 영향)

  • The present study was designed to evaluate whether supplementation of dietary coenzyme Q10 protects the ADR-induced cardiotoxicity in rats. Experiment was undertaken under the condition of simultaneous adminstration of ADR and coenzyme Q10 for 4 weeks. Adriamycin treatment significantly decreased growth performance of rats. But this decrement was not modified by dietary supplementation of conzyme Q10. In the plasma creatine phosphokinase activity, there was no significant difference among experimental groups. Electron microscopic examination revealed a progression of myocardial lesions were dependent upon the level of ADR injection. The most frequently observed fine structural alterations in rat myocardium were mitochondrial swelling, dilation of the sarcoplasmic reticulum and the appearance of a perinuclear vacuolization. But these structural changes were somewhat lesser in defree by dietary supplementation of coenzyme Q10.

Changes in Serum Lipid Components and Blood Glucose by Krill (Euphausia superba) Meal and NaF in Rats (크릴 분말 및 NaF 첨가 식이가 흰쥐의 혈청 지질성분 및 혈당 변동에 미치는 영향)

  • This study was conducted to examine the changes in serum lipid components and blood glucose by krill (Euphausia superba) meal and sodium fluoride (NaF) in rats fed on experimental diets and administered orally to NaF 10 mg for 5 weeks. Body weight of rats decreased as the amount of krill meal diet increased, it was observed the basal diet plus NaF group (BF group) compared to the lower basal diet group (BD group). The serum concentrations of total cholesterol, LDL (low-density lipoprotein) cholesterol, free cholesterol, triglyceride (TG), phospholipid (PL) and blood glucose in serum were higher in the BF group than the 10% krill meal group plus NaF 10 mg (KF10 group) or BD group, the 20% krill meal plus NaF 10 mg group (KF20 group), the 30% krill meal plus NaF 10 mg group (KF30 group). Conversely depending on the content of krill meal for the HDL (high-density lipoprotein) cholesterol level, it showed higher results. The concentration of total protein was no significant difference among the groups (p<0.05). The results indicate that a krill meal diet effectively inhibited increases in lipid elevation and blood glucose level in the sera of rats.

  • Leptin, the product of obese (ob) gene, is an adipocyte-derived satiety factor that plays a major role in the regulation of food intake, energy homeostasis, body weight, reproductive physiology and neuropeptide secretion. The present study was designed to generate transgenic mice expressing antisense mouse ob (mob) gene. Total RNA was extracted from the adipose tissues of mouse, then reverse transcription was performed. The 303 and 635 bp fragments of anti I and II cDNAs were amplified from mob cDNAs by PCR. The two mob cDNAs were reversely ligated into between adipose tissue specific aP2 promote and SV40 poly(A) site. Transgenic mice carrying two different kinds of antisense mob transgenes were generated by DNA microinjection into pronucleus. Total 14 transgenic mice were born, and the 4 and 5 founder lines of the transgenic mice with anti I and II transgenes were respectively established. Antisense mRNA expression was detected in transgenic F$_1$ mice by RT-PCR analysis. This result suggests that the transgenic mice expressing antisense mob mRNA may be useful as an animal disease model to be obesity caused by decreased amount of leptin secretion.

  • A pot experiment was conducted to find out the effect of application amounts of silicates on the fatty acid and sterol composition of paddy rice at various growing stages. The results obtained were as follows; 1. At maximum tillering stage and heading stage, significant positive correlations were obtained between the ratio of unsaturated fatty acid to saturated fatty acid and silica absorption, dry matter or grain yield, respectively. But significant correlation was not observed between them at harvesting time. 2. At maximum tillering stage and heading stage, the ratio of unsaturated fatty acid to saturated fatty acid in paddy rice was increased with increasing silica application from 80 to 180 ppm of soil, but these ratio was decreased at the plot of 200Kg/10a of silicate. 3. The ratio of sitosterol to stigmasterol in paddy rice at maximum tillering stage was increased with increasing silica application of soil, but these ratio was increased with increasing silica application up to 180 ppm of soil at heading stage, and was decreased at the plot of 200Kg/10a (410 ppm in soil) application of silicates.

  • The effects of ethanol and dietary protein levels on the calcium and phosphorus metabolism were investigated in 15-week-old male rats. Rats were divided into 4 groups ; control group(16% protein, 16 PC) and 8%(8 PE), 16%(16 PE), and 24% protein groups (24 PE) to which was given 5% ethanol mixed with their drinking water. Body weigh gain, organ weight, hemoglobin content, and hematocrit value were not affected by either ethanol or dietary protein levels. Calcium concentrations in spleen were significantly decreased in the ethanol groups than those in control group. Calcium and Phosphorus levels in femur, serum, liver, kidney, and muscle were normal. Among ethanol treated groups, fecal excretions of calcium were a little more decreased, but urinary excretions, balances and apparent absorption rate of calcium were a little more increased in higher percentage of protein group than lower percentage. Urinary phosphorus excretions in the ethanol treated groups were significantly decreased compared with the control group. Among ethanol treated groups, phosphorus balance and apparent phosphorus absorption rate of 24 PE group were significantly higher than those of 8 PE and 16 PE groups.

Effect and Safety of Calorie Restriction and Complex Herbal Medicine Short-term Treatment ('Oil-rescue' Program) for Weight Reduction of Obesity or Overweight Patients: A Practice Based Research (과체중 및 비만 환자의 체중감량을 위한 식사량 조절 및 복합 한약 단기 치료(5일 레스큐 프로그램)의 효과와 안전성: 진료기반 연구)

  • Objectives: This study is a practice based research conducted to investigate the effectiveness and safety of a 5-day short-term diet program ('Oil-rescue' program) designed to reduce the adverse events and initial dropout rate in obesity treatment. Methods: We retrospectively analyzed 4 Korean medicine clinic patient data who completed 'Oil-rescue' program which is consisting of Gambi-hwan, Bium-hwan, Butgiban-hwan and Jayoon Kyungokgo. The weight change before and after participating 'Oil-rescue' program was primary outcome of our study. Changes in body mass index, body fat mass, skeletal muscle mass, body water content, lean body mass, protein, and minerals were also measured. Results: A total of 35 patients who satisfied the eligible criteria were finally included. The body weight decreased from 69.45±11.86 kg to 67.43±11.58 kg, a total of 2.02±1.03 kg (P<0.001). Body fat mass decreased from 25.77±7.45 kg to 24.98±7.26 kg, a total of 0.78±1.21 kg (P<0.001). Body mass index decreased from 26.39±3.64 kg/㎡ to 25.64±3.49 kg/㎡, a total of 0.75±0.41 kg/㎡. (P<0.001). A total of 15 patients had side effects such as diarrhea, abdominal pain, and insomnia, but all were mild. 16 out of 35 people switched to long-term obesity treatment programs. Conclusions: Through this retrospective practice based research, it was found that the 'Oil-rescue' program effectively reduced body weight, body fat, and body mass index, and the other obesity related parameters. It was a relatively safe and effective short-term obesity treatment program.

Leptin, adiponectin, interleukin-6 and tumor necrosis factor-α in obese adolescents (비만아에서의 leptin, adiponectin, interleukin-6, tumor necrosis factor-α에 대한 연구)

  • Purpose : Obesity is associated with insulin resistance. Insulin resistance and the presence of pro-inflammatory mediators are thought to cause a state of vascular endothelial dysfunction, an abnormal lipid profile, hypertension, and vascular inflammation. These chronic inflammatory responses, which are characterized by abnormal cytokine production, lead to activation of a pro-inflammatory signaling pathway. Leptin is an important mediator of inflammatory processes and immune-mediated diseases. The purpose of this study was to investigate the relationship between leptin and various cytokines associated with obesity in adolescents. Methods : Sixty-six obese adolescents (between 16-17 years of age, obesity index >130%) and 26 normal controls were included in this study. Obesity index and body mass index (BMI) were calculated. Serum lipid profile, AST and ALT were tested after 10 hours of fasting. Tumor necrosis factor alpha (TNF-${\alpha}$) and Interleukin-6 (IL-6) levels were measured by ELISA. Insulin, adiponectin, and leptin levels were estimated by radioimmunoassay. Results : Leptin was significantly higher in the obese adolescents compared to the control adolescents ($12.0{\pm}6.8ng/mL$ vs $6.3{\pm}1.0ng/mL$). TNF-${\alpha}$, IL-6, and insulin were significantly higher in the obese adolescents. Adiponectin was significantly lower in the obese group than the control group ($3.3{\pm}1.9{\mu}g/mL$ vs $5.0{\pm}1.4{\mu}g/mL$). Leptin had positive correlations with obesity index, BMI, and IL-6. Conclusion : In obese adolescents, leptin, TNF-${\alpha}$, IL-6, and insulin might be important mediators of obesity. Further clinical research is necessary to ascertain leptin as a predictor of cardiovascular diseases and to develop a guideline for clinical intervention.

A Survey on Dietary Habits in Gyeongnam and the Development of the Nutrition Education Curriculum with Teacher's Guide for Obese Elementary School Children (경남지역 일부 초등학교 비만아동의 식습관 분석 및 영양교육을 위한 교수학습과정안 개발)

  • The purpose of this study was to develop a nutrition education curriculum with teacher's guide which includes discretionary activities for obese children. A survey was carried out to investigate the recognition of body image and food behaviors according to the obesity index (mild, moderate, severe) in school children (4~6th grade, 158 boys and 60 girls) who were selected based on a physical examination in May, 2006 in the Gyeongnam province. Next, a nutrition education curriculum with teacher's guide was developed on the basis of the findings from the survey and from preceding researches. The results are summarized as follow. The results of this study showed the existence of some nutritional problems such as overeating, prejudice, skipping meals, snacking patterns, etc, which indicate the need for nutritional management for obese children. Most overweight children (80.3%) showed the most interest in the nutrition education program, particularly with regards to dieting for weight control (64.7%). The developed nutrition education curriculum consisted of 8 main subjects and 13 subtitles. The curriculum was prepared for 13 lessons and included songs and singing, making-up lyrics, games about nutrition, discussions of the experience of eating (satiety, thirst, hunger), debates on dietary habits, writing and others to promote the interest for learning. We aimed to develop this program in an attempt to improve the dietary habits of obese school children. This is very important because once a dietary habit is formed in adults, it is difficult to change and the best adjustable stage is during childhood. Therefore, early nutrition education during elementary school can change and build-up the awareness of health in young elementary school children.

Effects of Chicory Inulin and Oligosaccharides on Lipid Metabolism in Rats Fed a High-Cholesterol Diet (고콜레스테롤 식이 섭취 흰쥐에서 치커리 이눌린과 올리고당이 지질대사에 미치는 영향)

  • The present study was aimed at investigating effects of chicory inulin and three kinds of oligosaccharides on lipid metabolism in rats fed a high-cholesterol diet. Nine Sprague-Dawley male rats weighing, about 190g were given one of five experimental diets, which were basal cholesterol diet (Control) isomaltooligosaccharide diet (IMO), Iructooligosaccharide diet (FO), chicory inulooligosaccharide diet (CIO) and chicory inulin diet (CI) for 5 weeks. In the oligosaccharide and inulin diets, 6% was added at the expense of sucrose. Rats were pair-fed to the intake of FO group which consumed the least amount, and their feces were collected during the last 4 days. Body weight gain was lower in Fo and CI groups compared with the Control group. Plasma glucose levels of FO and CIO groups were lower and plasma triglyceride concentrations of FO, CIO, and CI groups were lower than those of IMO group. Plasma cholesterol concentration did not differ among groups. Relative liver weight was lower in CIO group. Hepatic triglyceride and cholesterol did not differ among. groups. Fecal excretion of neutral steroid and bile acid were not different among groups, but fecal triglyceride excretion was significantly increased in FO and CI groups compared with the Control group. In conclusion, supplementation of oligosaccharides and chicory inulin at 6% of diets showed no significant hypolipidemic effect in rats fed a high cholesterol diet.

Effect of Supplementary Feeding Levels on Productivity and Grazing Intensity in Grazing Elk stags(Cervus canadensis) (보충사료 급여수준이 엘크 수사슴의 생산성 및 방목강도에 미치는 영향)

  • This study was conducted to investigate the effect of supplementary feeding levels on livestck and forage productivity and grazing intensity in Elk stags (Cervus canadensis). A fifteen 2-year-old Elk stags about 195 kg were randomly assigned to one of three dietary treatments (five animals per treatment). The dietary treatments consisted of a feeding concentrate of 1.0% of body weight (T1), 1.5% of body weight (T2) and 2.0% of body weight. Total dry matter intake (TDMI) was increased with increased with an increasing supplementary feeding levels. Average daily gain (ADG) were significantly increased with an increasing supplementary feeding levels (p<0.05) and reached a maximum on July and was lower in spring than autumn. The velvet antler production was no differences among treatment groups. Forage productivity of pasture and crude protein content were highest on May and decreased thereafter, however, crude fiber content was the reversed. The grazing intensity of Elk stags was increased in spring (38 to 59 head per ha) than summer and autumn (13 to 32 head per ha). The average grazing intensity of Elk stags ranged from 21 to 34 head per ha, which is affected by supplementary feeding levels. This result suggests that feeding supplementary diet at 1.5 % of body weight was needed to maintain the stable wight gain in antler growing periods and control the proper grazing intensity of Elk deer stags.
