Structural Characteristics of Novel Branched Oligosaccharides Syntesized by a Maltose Acceptor Reaction with Dextransucrase from Leuconostoc mesenteroides M-12 (Leuconostoc mesenteroides M-12 덱스트란수크라제의 말토스 억셉터 반응으로 합성된 새로운 분지 올리고당의 구조 특성)

  • The structures of novel branched oligosaccharides synthesized by the acceptor reaction with dextransucrase from Leuconostoc mesenteriodes M-12 were proposed in accordance with the results obtained from enzymatic hydrolyses and a partial acid hydrolysis. The structure of branched oligosaccharide B4 was shown to be 62-O-$\alpha$-D-kojibiosylmaltose. Branched oligosaccharide B5 was shown to be 63-O-$\alpha$-D-kojibiosylpanose. By reacting the acceptor reaction products with endodextranase a novel branched oligosaccharide (D4) could be produced. D4 was derived from the result of endodextranase hydrolysis of oligosaccarides synthesized by the second acceptor reaction with dextransucrase and was resistant to endodextranase and glucoamylase. The proposed structure of D4 was 62-O-$\alpha$-D-kojibiosylisomaltose. Formation pattern of the acceptor reaction products smaller than d.p. 6 with linear or branched linkage was also shown.

Characterization of the high mannose asparagine-linked oligosaccharides synthesized by microfilariae of Dirofilaria immitis (심장사상충 자충이 합성한 high mannose asparagine-linked oligosaccharides의 분자화학적 분석)

  • This report describes the structures of high-mannose-type N-linked oligosaccharides in glycoproteins synthesized by the microfilariae of Diroflcrio immitis. Microfilariae of D. immitis were incubated in vitro in media containing 2-(3H) mannose to allow metabolic radiolabeling of the oligosaccharide moieties of newly synthesized glycoproteins. Glycopeptides were prepared from the radiolabeled glycoproteins by digestion with pronase and fractionation by chromatography on concanavalin A Sepharose. Thirty eight percent of 2- (3H) mannose incorporated into the microalariae of D. immitis glycopeptides was recovered in high mannose-type asparagine-linked oligosaccharides which were bound to the immobilized lectin. Upon treatment of 2-(3H) mannose labeled glycopeptides with endo - β- N- acetylglu co saminidase H , the high mannose type chains were released and their structures were determined by high performance liquid chromatography and exoglycosidase digestion. The major species of high mannose-type chains synthesized by microfilariae of D. immitis have the composition Man5GlcNAc2, Man6ClcNAc2, Man7GlcNAc2, and Man8GlcNAc2. Structural analyses indicate that these oligosaccharides are similar to high mannose-type chains synthesized by vertebrates.

Growth Effect of Branched Oligosaccharides on Principal Intestinal Bacteria (분지 올리고당이 장내 주요 세균의 생육에 미치는 영향)

  • To investigate the growth effect of branched oligosaccharides on the principal intestinal microorganisms, Bifdobacterium, Lactobacillus, Clostridium, Escherichia, Eubactemum, Enterucoccus, Staphylococcus and Bacteroides were cultivated on a medium containing branched oligosaccharides and panose. B. adolescentis, B. logum and L. aciduphilus grew effectively on the medium containing panose, while C. pe@igens, C. paraputrificum, Bac. fragilis and S. aurezds did not. The content of panose decreased greatiy in the culture broth of branched oligosaccharides of B, adolescentis, but it remained in the culture of C. perfringens. The results indicated that panose was consumed effectively by B. adolescentis, but not utilized by C. perfringens. 3. adolescentis still grew on the panose remained in the broth of mixed cultivation of B. adolescentis and C. perfn'ngens. Therefore, panose and branched oligosaccharides seem to promote selectively the growth of B. adolescentis in the human intestine.

Changes of the Textural Properties of the Sweet Potato Starch Gels using Maltogenic Amylase (Maltogenic amylase를 이용한 고구마 전분겔의 텍스쳐 특성 변화)

  • We investigated the texture of the sweet potato starch gels treated with maltogenic amylase. Effects of branched gluco-oligosaccharides and acorn starch on the texture of the sweet potato starch gel were also investigated. Hardness and cohesiveness of gels were measured by using Instron and sensory evaluation on the gel properties was performed. From the results of the instrumental analysis, it was found that the overall textural properties as Mook could be improved by adding branched glucooligosaccharides, maltogenic amylase or acorn starch to the sweet potato starch gel. As a result, there was a decrease in the cohesiveness of gels while the hardness of gels increased. The sensory evaluation study indicated that the sweet potato starch gels treated with 0.02% maltogenic amylase, or added with 12.5% branched gluco-oligosaccharides, or mixed with 50% acorn starch had preferable quality as Mook.

Optimization of Growth Medium Composition for Overproduction of Bacillus licheniformis Amylase in Recombinant Escherichia coli (Bacillus licheniformis amylase(BLMA)의 생산성 향상을 취한 재조합 대장균의 배지 최적화)

  • The research is concerned with optimization of growth medium composition in an attempt to improve the product yield of Bacillus licheniformis amylase (BLMA) in recombinant E. coli containing the BLMA gene. BLMA has the catalytic activity of producing branched oligosaccharides from starch. The medium optimization was performed in flask cultures based on the Box and Wilson method. The optimized medium is composed of tryptone 18.0 g/l, yeast extract 22.4 g/l, NaCl 5.3 g/l and glucose 2.1 g/l. In a jar fermenter culture with the conventional LB medium, the recombinant E. coli yielded 1.39 g/l of final dry cell mass and 5.11 U/ml of enzyme activity. In the optimized medium, however, the final cell mass was increased to 6.01 g/l and the enzyme activity to 23.2 U/ml. Medium optimization improved cell mass by 4.3 times and enzyme activity by 4.5 times. Such an increase in enzyme activity is mainly due to an enhancement of cell mass.

Variations in Growth Characteristics and Seed Qualities of Korean Soybean Landraces (한국 재래종 콩의 수집지대별 생태적 특성 및 품질 차이)

  • This study was carried out to examine agronomic characteristics and seed qualities of 827 Korean soybean landraces according to the collecting areas. Agronomic characteristics and seed qualities was examined according to the localities by weather characteristics. Days to flowering was the longest (mean 70.9 days) in the northwest seashore, and the most short in the south inland (mean 63.8 days). Growth days was the longest in the Gyeongbuk inland (mean 136.0 days), and the most short in the mountain-high (mean 132.8 days). The stem height was the most short in the north-west seashore (mean 81.3 cm), and in the south seashore (mean 58.8 cm). The seed weight of 100 seeds was the lightest in the north-west seashore (mean 17.1 g), and the heaviest in the south-west seashore (mean 21.5 g). The crude protein contents was the lowest in the north-west seashore (mean 40.1%), the highest in the south seashore (mean 42.8%), and vice versa in the crude oil. The contents of sucrose and oligo-saccharide was the highest 5.8% and 8.6%, respectively, in the mountain-high, and the lowest 4.8% and 7.6% in the Gyeongbuk inland and south seashore, respectively. Among the iso-flavone contests, the contents of genistein and total was the highest, $635.6\;{\mu}g/g$ and $1258.1\;{\mu}g/g$, respectively, in the mountain inland, and the lowest $319.2\;{\mu}g/g$ and $835.3\;{\mu}g/g$ in the south-west seashore. The contents of glycitein was the highest $124.3\;{\mu}g/g$ in the south seashore.
