A high molecular weight natural sodium alginate (HMWSAs) was depolymerized by hydrogen peroxide ($H_2O_2$) with ultrasonic irradiation. The effects of the reaction conditions such as reaction temperature, reaction time, hydrogen peroxide concentration and ultrasonic irradiation time on the molecular weights and the end groups of the depolymerized alginates were investigated. It was revealed that depolymerization occurred through the breakage of 1,4-glycosidic bonds of sodium alginate and the formation of formate groups on the main chain under certain conditions. The changes in molecular weight were monitored by GPC-MALS. The molecular weight of 2 wt% alginate solution decreased from 450 to 15.9 kDa for 0.5 hrs at 50 $^{\circ}C$ under an appropriate ultrasonic irradiation. The PDI(polydispersity index)s of the alginate depolymerized in this study were considerably narrow in comparison with those obtained from the other chemical degradation method. The PDIs were in the range of 1.5~2.5 in any reaction conditions employed in this study.
We evaluated the effects of various factors (e.g., serum, inhibitors of protein synthesis, and cytoskeleton and protein kinases) on early PMA-induced THP-1 cell adhesion using an adhesion assay with Sulforhodamine B (SRB) staining, which was used to assess the proliferation of the attached cells. THP-1 cell adhesion to a plastic substrate was detected 1 hr after exposure to Phorbol 12-Myristate 13-Acetate (PMA) and peaked after 18 hr. At concentrations > 25 nM PMA, the level of adhesion did not change. Based on our preliminary results, we used 25 nM PMA and 5 hr of culture as standard assay conditions. Early PMA-induced cell adhesion was not affected by the presence of serum or PD 98059 in the culture medium, but was affected by the addition of PKC inhibitors and cycloheximide. In the presence of actin inhibitor with PMA, the cell adhesion increased when comparing with PMA treatment only. Thus, early PMA-induced adhesion of THP-1 cells does not require serum in the culture medium, MAP-kinase activation, or actin polymerization, but does require de novo protein synthesis and PKC activation. Our SRB-based cell adhesion assay may be used to screen other PKC inhibitors.

Several types of scavenger receptors, including the Collectin-Placenta 1 (CL-P1) receptorthat is present in mammals, are molecules that are expressed on the surfaces of endothelial cells, macrophages and smooth muscle cells. These molecules are cell surface glycoproteins that can be conjugated to oxidized low density lipoprotein (oxLDL). Among these molecules, the effect of quercetin on CL-P1 activation has been confirmed. Quercetin is known as an antioxidant that stops oxidation because it acts to remove free radicals that are responsible for the oxidation reaction. In this study, fragments from the transcription start site of the mouse CL-P1 gene promoter to the -500th base were cloned using DNA polymerase. These fragments were then introduced into macrophage like RAW264.7 cells and fibroblast-like NIH3T3 cells to study the effect of quercetin on the CL-P1 gene expression. As a result, we found that bases ranging from -250 to -350 in the anterior part where gene expression starts are important for producing CL-P1 protein. Among them, the DNA mutation experiments we performed confirmed that the E2F binding sites are critical for producing the CL-P1 protein? In addition, when quercetin was added to the RAW264.7 culture medium, which was a culture of adherent cells, observedthe phenomenon of the cells falling off from the surface of the culture container.
Human cytomegalovirus (HCMV) stimulates the expression of intercellular adhesion molecule (ICAM-l) on the surface of monocytic THP-1 cells. Stimulation of ICAM-l did not require HCMV gene expression since UV-inactivated HCMV (UV-HCMV) was able to induce ICAM-l expression. ICAM-l expression was also stimulated in uninfected THP-l cells which were fed with culture supernatant of HCMV-infected THP-l cells. Co-culture experiment using trans-well insert supported that HCMV-infected THP-l cells secreted some cytokine(s) stimulating ICAM-l expression. The stimulation of ICAM-l by HCMV-infected cell culture supernatant appears to involve $NF-{\kappa}B$ pathway. Culture supernatant from THP-l cells infected with UV-HCMV, whose gene expression was abrogated, failed to stimulate ICAM-l expression on naive THP-l cells. Thus, HCMV gene expression seems to be required in secretion of cytokine(s) stimulating ICAM-l expression.
Plasma treatment and corona treatment have been used for surface modification of polydimethylsiloxane (PDMS) film by activating its surface with the -OH group. Adhesion promoter or coupling agent was also used to improve adhesion of PDMS film with various materials. However, obtained hydrophilicity onto the surface of PDMS films with those processes was transient and vulnerable. In this study, a new alkoxysilane-functionalized acrylic polymer precursor was first synthesized by copolymerization process, and then was reacted with HO-terminated PDMS through condensation reaction to prepare a new surface modifier for PDMS film. The structure and molecular weight of the prepared surface modifier were confirmed by 1H-NMR and GPC measurement. Surface properties of surface modifier-coated PDMS films were also investigated by using XPS, ATR and WCA analysis. The adhesion between the PDMS film and the surface modifier was tested using cross-cut test.

Purpose : Kawasaki disease(KD) is known as an acute multi-systemic vasculitis with various immunologic abnormalities. Adhesion of leukocyte to endothelial cells is a key event in the sequence of inflammatory response. This study was performed to investigate the clinical significance of serum soluble intercellular adhesion molecule-1(sICAM-1) and soluble vascular cell adhesion molecule-1 (sVCAM-1) in acute and subacute stages of typical KD for diagnostic and prognostic value. Methods : A typical KD group was 32 patients who were hospitalized from Jan. 2002 to Jun. 2004 was enrolled. Control was 16 non-KD patients with febrile illness. sICAM-1 and sVCAM-1 were measured and compared by Echocardiographic and clinical findings and cardiac troponin T and I. Results : sICAM-1 and sVCAM-1 levels of acute KD were significantly elevated over control(P=0.019 vs. P=0.049, respectively) and sICAM-1 was significantly decreased in subacute stage(P=0.0015). sICAM-1 and sVCAM-1 had positive correlation with each other in both stages(P=0.0067, P=0.015, retrospectively). Neither sICAM-1 nor sVCAM-1 correctly reflected the coronary abnormalities and responsiveness to intravenous gammaglobulin(IVGG) in both stages. But sVCAM-1 was significantly increased in the carditis group in both stages(P=0.025, P=0.014, retrospectively) and had a positive correlation with troponin T(r=0.63, P=0.00063). Conclusion : The levels of sICAM-1 and sVCAM-1 were not very useful tools for detecting and predicting subsequent coronary abnormalities and responsiveness to IVGG in KD patients. However, sVCAM-1 appears to play a significant role in carditis of KD. Further studies are needed about various adhesion molecules and cytokines in the pathogenesis of KD.
Background: In this study, we investigated the early time course of expression of the major histocompatibility(MHC) antigens, intercellular adhesion molecule-1(ICAM-1), vascular cell adhesion molecule-1(VCAM-1), interleukin-6 and the histopathological changes in the coronary arteries of cardiac allografts exchanged between inbred mice strains that differ in one loci of class I major histocompatibility antigen (B10.BR to B10.A). Material and Method: No immunosuppressive therapy was used. Both allografts and the hearts of the recipients were harvested at 7(group 1, n=6), 15(group 2, n=6), 21(group 3, n=6), and 30(group 4, n=6) days after transplantation. They were examined by immunohistochemistry, microscopy and morphometry. All allografts had contractions at the time of harvest. Result: A strong MHC class I antigen expression was present on the endothelial and medial cells of the coronary arteries in group 1 and remained unchanged in the rest of the groups. However, MHC class II reactivity was none or very little at any time. Mild to moderate ICAM-1 expression was observed on the endothelial cells, but not on the medial cells at any time by 30 days. VCAM-1 expression was strong both on the endothelial and medial cells at any time. Moderate degree expression of interleukin-6 was observed from 7 to 30 day specimens. Histopathologically, percentage of affected vessels(vessels with intimal thickening) was less than 10 % in 7 day group and increased up to 50 % at 30 days. Mean percent narrowing of the lumen of the affected vessels revealed less than 20 % at 7 days and 40 % at 30 days. The area occupied by tropomyosin positive cells in the intimal lesion, graded from 0 to 3, showed gradual increase but remained between grade 0 to 1 by 30 days. Medial integrity was also well preserved at any time. Moderate perivascular mononuclear cell infiltration was observed at 7 days and it was progressively increased upto 30 days. Recipients' heart revealed no positive immunopathologic findings. Conclusion: In this study, the early time course of progression of the transplantation vasculopathy was demonstrated in the murine heterotopic heart transplant model.
Uniform silica nanobeads were synthesized by St$\ddot{o}$ber method and assembled in the form of monolayer on glass substrate using sonication method. Before the assembly of silica nanobeads, glass substrates were treated with molecular linkers, such as CP-TMS and PEI, and nanobeads were dispersed in toluene. In attachment test, SO (sonication without stacking) method and SS (sonnication with stacking) method was used and sonication time was controled. After the experiment, microbalance was use to measure deviation between before and after the attachment experiment then calculate percent of coverage. Minutely observe with SEM (Scanning Electron Microscopy) then select the most close-packed and monolayer assembled cover glass and calculate DOC (Degree of Coverage). In SO method, DOC increased very sharply and reach over 140 percent point, also got lots of multi-layer region. On the other hand, in SS method DOC increased slower than SO method but more close-packed and monolayer assembled.
In this study is performed to investigate the pumping characteristics of three-stage disk-type molecular drag pump (DTDP). The experiments are measured using five vacuum pressure gauges in the positions for rotors of DTDP. The experimented DTDP is consisted of three rotors and four stator. In the DTDP, spiral channels of three rotors are cut on the both upper surface and lower surface of a rotating disk, and corresponding stator is a planar disk. The experiments are performed in the outlet pressure range of $0.2{\sim}533\;Pa$. The pressure of each rotors are measured under the various condition of outlet pressure and throughputs, and nitrogen gas is used for test gas. In the numerical study, the pumping characteristics of each rotor are studied for the variation of throughputs in the all rotating channel. Pressure contour and velocity are obtained by the numerical simulation.

Particulate matters (PM) are small particulate pollution that decrease the function of skin barrier, which causes inflammatory skin diseases and extrinsic aging. In this study, we evaluated the effect of preventing the adherence of PMs from several cosmetic products applied to human skin using iron oxide black. The PM floating chamber consists of skin exposure area, PM inlet, floating power device, and an outlet so that PM can be naturally attached to the skin while floating in the chamber. The change in skin brightness according to the floating concentration of alternative fine dust was checked to confirm the optimal floating concentration conditions. The intensity difference (before-after intensity, Δ) before and after adhesion of iron oxide black was proportional to the amount of PM adhered. The anti-adherence effect of iron oxide black on five cosmetic products were evaluated through 20 each subjects by comparing the amount of iron oxide black adhered on the control and treatment. The difference in brightness before and after the iron oxide black attached to the skin was calculated and compared with the control group(p < 0.05). When over 150 mg of iron oxide black was adhered on the skin, the interference of intensity was low and clearly showed the skin adhered pattern. According to the application of the five cosmetics, the intensity difference was significantly lower than the control group. This means that depending on the product, it prevented the attachment of iron oxide black. This study is a safe and useful method to confirm the prevention of PM skin adherence. In conclusion, cosmetics can prevent the adherence of PM on the skin according to the formulation or ingredients characteristics.
