해조류 추출물로부터 홍합(Mytilus edulis)의 부착 저해물질 정제

  • 연안 해역에서 해조류와 부착패류는 양식장 시설 뿐만아니라 수취구 어선바닥 등에서 그 피해는 심각한 실정이다. 해조류에 의한 부착은 잎파래 가시파래 등이 주가 되고 패류로는 홍합과 따개비가 주를 이룬다 이들의 부착방지를 위해서 염소살균, TBTO (Tributyltin oxide), CuSO$_4$등의 처리를 하고 있지만 이러한 처리에 의해서는 또다른 환경 문제가 야기되고 있다. 이같은 부착방지 물질 탐색의 일환으로 흥합을 탐색 대상종으로 선택하여 각종 해조류 추출물들로부터 부착 방지 효과를 조사하였다. 그 결과 고리매(Sytosimphon lomentaria)로부터 가장 높은 활성을 확인 하였고, TLC, silica-gel chromatography, Ion-exchange, HPLC를 통해 정제 하였다. Effect of Copper toxicant on Suspended and Attached Growth Nitrifying Bacteria (부유 및 부착성장 질화균에 미치는 구리 독성의 영향)

  • The effect of toxicant on the inhibition of nitrification was investigated, using concentrated nitrifying bacteria of both attached and suspended growth. This nitrifying organism was originally obtained from the activated sludge of sewage treatment plant and cultivated for more than three months. The object of this experiment is to determine the effect of the specific surface area and the growth condition of nitrifying bacteria on toxicity of heavy metal. The results of this study were as follows. The specific surface area of both attached and suspended growth of nitrifying organism was proven to be a major factors in determining the inhibition of nitrification of heavy metal such as $Cu^{++}$ion. When the condition of attachment and detachment was compared in an experiment using attached growth nitrifier, the effect on toxicant was 1.12 times less in attached condition than in detached condition for Nitrosomonas, and 1.09 times less for Nitrobacter. In case of suspended growth nitrifier, the effect on toxicant was 1.46 times less in non-ground condition than in ground condition for Nitrosomonas, and 1.35 times less for Nitrobacter. Also, similar results were obtained in a set of experiments, without adding nitrite to the substrate. In an experiment that compared attached condition using attached growth nitrifier with detached condition using attached growth nitrifier, the effect on toxicant was 1.83 times less in attached condition than in detached one for Nitrosomonas, and 1.78 times less for Nitrobacter. In case of suspended growth nitrifier, the effect on toxicant was 1.27 times less in non-ground condition than in ground condition for Nitrosomonas, and 1.32 times less for Nitrobacter.

Alum Floc Attachment in Granular Media Filtration (입상여과에서 액반플럭의 부착)

  • Granular media filtration is used almost universally as the last particle removal process in conventional water treatment plants. Therefore, superb particle removal efficiency is needed during this process to ensure a high quality of drinking water. However, every particle can not be removed during granular media filtration. Besides the pattern of particle attachment is different depending on physicochemical aspects of particles and suspension. Filtration experiments were performed in a laboratory-scale filter using spherical glass beads with a diameter of 0.55 mm as collectors. A single type of particle suspension (Min-U-Sil 5) and alum coagulation was used to destabilize particles. The operating conditions were similar to those of standard media filtration practice: a filtration velocity of 5 m/h. More favorable particles, i.e., particles with smaller surface charge, were well attached to the collectors especially during the early stage of filtration when zeta potential of particles and collectors are both negative. This selective attachment of the lower charged particles caused the zeta potential distribution (ZPD) of the effluent to move to a more negative range. On the other hand, the ZPDs of the effluent moved from more positive to less positive when the surface charge of particles was positive and this result was thought to be caused by ion transfer between particles and collectors.

A Study on Indirect Attachment Method of Compensation Materials to Increase Signal Intensity in Magnetic Resonance Imaging (자기공명영상검사 시 신호강도를 높이기 위한 보상물질의 간접부착 방식에 관한 연구)

  • Previously, studies on compensation material to increase the signal intensity have been conducted which does not affect the reading of images. However, the compensation material has a concern on patient infection as it is attached directly on the skin. Therefore, in this study, we tested an indirect attachment of the compensation material as an alternative method of the direct attachment. The silicon compensation material was fabricated in the form of a cylindrical bar and attached to each element of the 8 channel head coil. Then the signal intensities of the water phantom pre and post application of the silicon were measured. T1 and T2-weighted images were acquired using an 8-channel head coil and a 3.0T superconducting MRI. Signal intensities were measured by using an image measuring program. Paired t-test was used to verify if there were significant differences. The signal intensity before application of the silicon was significantly increased by 3.39% and 2.62% in T1 and T2 weighted images, respectively. Although the indirect attachment method had a limitation to completely replace the existing method, it was considered to be useful in patients with infectious diseases such as diabetic complications since it had a meaningful improvement in signal intensity based on the filling factor increase.

  • Marine algae는 주된 fouling organism으로 전세계에 널리 분포되어 있고, 경제적 손실을 가져온다. 특히나 container ships와 large tanker에서 자라는 algae는 마찰력의 증가로 인한 speed 감소와 연료비의 증가와 같은 문제점을 가진다. 그래서 현재 antifouling paint로 TBT화합물을 사용하고 있지만, 이것은 해양생물에 나쁜 영향을 미치는 것으로 나타나 TBT 화합물 사용에 대해 제한하고 있다. 따라서 본 연구에서는 해조류 추출물을 이용하여 무독성 천연 방오물질의 탐색을 목적으로 우선 선박이나 양식장 그물등에 많이 부착 성장하는 가시파래를 대상으로하여 이의 antifouling 효과를 나타내는 물질을 찾고자 한다. Studies on Adherance Inhibition and Detachment of Helicobacter pylori Using Egg Yolk IgY and Additives (난황항체 및 첨가제를 이용한 헬리코박터 파이로리의 부착 억제)

  • H. pylori is known to be a key pathogen of chronic gastric and duodenal ulcers. Bacterial adhesion to hosts is an essential step for bacterial infection and the inhibition of this adhesion provides a possible method for the treatment of the infection. The inhibitory effect of antibody lgY, produced from immunized hens with H. pylori antigen, was studied in vitro. The inhibition of H. pylori adhesion to AGS was as high as 90% using 0.5mg/ml of lgY, and almost 80% of the detachmentwas also achieved. The inhibitory effect of adhesion-inhibition candidates was investigated. Additives in combination with lgY increased the adhesion-inhibiting effect by about 30-50%. However, the adhesion molecules of H. pylori were varied and complex, therefore the further studies are necessary to develop an adhesion inhibitor and effective enough to be employed for the treatment of H.pylori, in vivo.

Inhibitory Effects of Asadisulphide on the TEA-induced Adherence of HL-60 Cells (TPA로 야기된 HL-60 세포의 기질부착에 대한 Asadisulphide의 억제 효과)

  • Asadisulphide were purified from Ferrula assafoetida by organic solvent extraction and chromatography. Its inhibitory effects on the TPA-induced adherence of HL-60 cells was analyzed. Since ethyl acetate extracts of F. assafoetida has the strongest inhibitory effect on adherence of HL-60 cells, it was re-extracted with ethyl acetate, hexane, and ethyl ether and chromatographed three times to isolate asadisulphide. At the minimum concentration of 2 $\mu\textrm{g}$/ml, asadisulphide inhibited adherence of 98% of HL-60 cells that have been treated with TPA. It also showed anti-cancer effect with no cytotoxity in the ED$_{50}$ value of 20 $\mu\textrm{g}$/ml.

  • A UHF band RFID passive tag antenna, which can be attached on metallic objects, is proposed. The proposed tag antenna is more insensitive to the size of the metallic object than conventional PIFAs or IFAs. It is constructed on a multi-layer and its size is reduced by using a 'L' slit. Performances of the antenna is analyzed by Ansoft's HFSS and measured using a back-scattering method. The simulated and measured data are obtained with varying the size of metallic object.
