Growth inhibitory and antioxidative effects of crude methanolic extract from Euonymus alatus (Thunb.) Sieb on SKBR3 human breast cancer cell line (SKBR3 유방암세포주에 대한 귀전우 메탄올 추출물의 성장억제 및 항산화효과)

  • 이 연구는 SKBR3 인간 유방암세포주에 대한 귀전우 메탄올추출물(CME)의 증식억제 효과, 세포사 유발 효과 및 항산화 활성을 확인하기 위해 이루어졌다. SKBR3 유방암세포주는 48시간동안 다양한 농도($0{\sim}20{\mu}g/m{\ell}$)의 CME가 제공된 곳에서 배양되었고, MMT 측정법을 이용하여 세포생존율을 평가하였다. CME의 50%에서 효과를 나타내게 하는 약물농도인 $ED_{50}$ (effective dose 50%)은 $6.5{\pm}0.3{\mu}g/m{\ell}$) 이며, 투여량이 증가함에 따라 농도에 의존하여 세포증식이 억제되는 것으로 나타났다. 또한 CME의 증식억제 효과는 유방암세포주의 세포사와 관련됨의 세포의 형태학적 변화와 올리고뉴클레오솜 DNA 파편의 확인을 통해 알 수 있었다. 또한 다양한 농도와 배양시간에서 CME가 ROS의 생산을 억제한다는 것을 확인할 수 있었다. 이런 결과들은 귀전우의 메탄올추출물이 SKBR3 인간 유방암세포주에 대해 강력한 증식억제 효과와 강한 항산화 효과를 나타낼 뿐만 아니라 세포사를 유도하는 효능을 가지고 있음을 시사한다. 이러한 효능은 약물에 대한 노출시간과 투여량에 의존하였다. 따라서 귀전우는 다양한 기전에 의해 유방암 세포에 대한 억제효과를 가질 수 있을 것으로 인식할 수 있다.

Effects of Kamikwibitang on Serum Levels of Corticosterone and Histamine and Immune Response after Immobilization Stress in Mice (가미귀비탕(加味歸脾湯)이 생쥐에 Immobilization Stress 부하후(負荷後) 혈중(血中) Corticosterone과 Histamine 함량(含量) 및 면역능(免疫能) 변화(變化)에 미치는 영향(影響))

  • The purpose of this research was to investigate the effects of Kamikwibitang water extract (KKT) on immobilization stress in C57BL/6J mice. KKT decreased the serum level of histamine and corticosterone increased by immobilization stress. In addition, KKT decreased the cell viability of thymocytes and enhanced the cell viability of splenocytes decreased by immobilization stress. Also, KKT decreased the viability of thymocytes and splenocytes in vitro. KKT decreased DNA fragmentation of splenocytes increased by immobilization stress. KKT decreased the population of thymic $CD4^+CD8^-$ cells increased by immobilization stress, and did not affect the population of $B220^+$ cells and the population of $Thy1^+$ cells changed by immobilization stress and enhanced the population of splenic $CD4^-CD8^+$ cells increased by immobilization stress. KKT enhanced the production of ${\gamma}-interferon$ and did not affect the production of interleukin-2 and interleukin-4 decreased by immobilization stress. Also, KKT decreased the phagocytic activity and the level of nitric oxide decreased by immobilization stress. These results indicate that KKT may be useful for the prevention and treatment of stress via suppression of serum histamine and corticosterone level and enhancement of specific-immune response.

Vasodilatory Activities of the Water Extracts of Seventeen Medicinal Remedy (활혈효능(活血效能)을 가진 한약재 17종의 내피세포 의존성 혈관이완작용에 대한 연구)

  • The pharmacological effects of medicinal remedies traditionally used in Asian countries for improving the blood circulation were examined on isolated rat thoracic aorta strips in organ baths. Each experimental medicine was consecutively extracted under reflux with water. Of 17 plants, Curcuma longa (CL) having the strongest acute relaxant activity in endothelium-intact arteries, Mucunae caulis (MC), Cirsium pendulum (CP), Rumex longiflius (RL), Paeonia suffruticosa (PS), Curcuma zedoaria (CZ), Scirpus maritimus (SM), Siphonostegia chinensis (SC), Leonurs sibiricus (LS) and Typha orientalis (TO) were showing dose-dependent relaxant activity. Long-term relaxant effects were showed in Curcuma aromatia (CA), MC, CP, RL, PS, Potulacae grandiflorae (PG), CZ, Panax notoginseng (PN), Achyranthes japonica (AJ), CL, SC, Lycoppus lucidus (LL) and Corydalis turtschaninovii (CT). In endothelium-injury test using carbachol, CL, SC, MC, RL and PS which are having the acute vasorelaxing activity and CA and CT which are not showing vasorelaxaing activity were damaged to endothelium. As a result of this study, the possibility that a part of medicinal remedy may contribute to the beneficial effects in blood circulation was proposed, but inter-individual variation has been observed. Also, further studies on the vasorelaxant effects of these remedies are still required.

  • Purpose : With the development of society, traffic accident(TA) rate is increasing accordingly. And the number of women in pregnancy who are damaged by TA is increasing as more women are involved in economical and social activities. TA victim in pregnancy cannot get proper treatment(Tx) in western medicine. Oriental medicine Tx is noted as alternative in treating TA victims recently. The main purpose of this clinical study is to make a survey of effective way of the oriental gynecological Tx about TA victim in pregnancy. Methods : The patient in this case, 28-year-old, was admitted for 14days(27th/May/2004 - 9th/June/2004) to cure severe neck pain and limitaion of cervical ROM(range of motion) occured from TA on the 4th week of pregnancy. This patient was treated with the acupunture, herbal medicine and physical treatment. Result and Conclusion : The symptoms of patient in this case are remarkable alleviated. And we think that the TA victims in pregnancy can be managed effectively by oriental gynecological Tx due to consider preservation of fetus and pregnant woman. After this, further study, approach and case will be needed about oriental gynecological Tx of TA victims in pregnancy.

Herba Portulacae induced Apoptosis in Human CervicalCarcinoma HeLa Cells (마치현(馬齒莧)이 자궁경부암세포(子宮頸部癌細胞)(HeLa Cell)에 미치는 영향(影響))

  • To address the ability of Herba Portulacae(HP) to induce cell death, we investigated the effect of HP on cell viability. Twenty-four hours later, loss of viability occurred following HP exposure in a dose-dependent manner. The treatment of HP, a commonly used herb formulation in Korea, Japan and China, caused a decrease in cell viability. HP also resulted in apoptotic morphology a brightly blue-fluorescent condensed nuclei by Hoechst 33258-staining, and reduction of cell volume. Our results show that 2mg/ml HP induces mitochondria membrane potential collapse. Immunoblotting data also shows that the expression of Bcl-2, antiaoptotic protein, decrease by the addition of HP. This GFP-Bax overexpression system shows that an important pro-apoptotic Bcl-2-family protein, Bax is translocated to mitochondria by the addition of 2mg/ml HP. Inerestingly, MAPK inhibitor study shows that p38 MAPK inhibitor, SB203580 inhibits HP-induced cell death and caspase-3 activation in HP-treated HeLa cells. Furthermore, HP transiently but significantly induces p38 activation. But P38 MAPK inhibitor does not have any effect on the translocation of Bax. Considering these results, HP induces apoptosis via p38 MAPK activation. But the pathway does not involve the translocation of Bax.

  • To address the ability of Kung-Kyung-Ilho-Jeon(KK) to induce cell death, we investigated the effect of KK on cell viability. Forty-eight hours later, loss of viability occurred following KK exposure in a dose-dependent manner. The treatment of KK, a commonly used herb formulation in Korea and China, caused a decrease in cell viability. KK also resulted in apoptotic morphology a brightly blue-fluorescent condensed nuclei by Hoechst 33258-staining, and reduction of cell volume. Our results show that KK induces caspase-3 and -9 activation in a time-dependent manner. In addtion, the translocation of cytochrome c release into cytoplasm has been observed under the presence of $5mg/m{\ell}$ KK. The subsequent loss of mitochondria membrane potential is collapsed by the addition of KK. Our immunoblotting data show that PARP, a well known caspase-3 and -6 substrate, is cleaved by KK. We show that a pro-apoptotic protein, Bax is increased in the presence of KK but that the amount of Bcl-2 is not changed. We suggest that Bax, a critical protein which can regulate channel of mitochondria to release cytochrome c, is a key protein in KK-induced apoptosis of Hela human cervical carcinoma cells

Dose Dependent Effects of Epimedii Herba on the Reproductive Function in the Mice (농도별(濃度別) 음양곽(淫羊藿) 투약(投藥)이 수컷 생쥐의 생식능력(生殖能力)에 미치는 영향(影響))

  • Objective: This study was undertaken to evaluate the dose dependent effects of Epimedii Herba extract solution on the spermatogenic abilities such as concentration, motility and morphological normality of sperm from the testis, and the activities of spermhyaluronidase and antioxidants. Materials and Method: We choose the 2-month-old mice, and administered the extract powder of Epimedii Herba in the different concentration once in a day for 60 days. The control group was administerde to normal water in the isolated testis tissue. Also we observed changes of isolated testis at the before and after administration of Epimedii Herba extracts in the mice. And we compared to the testicular tissue especially seminiferous tubules between control and treated group by histochemical methods. Results: The significant dose dependent differences were observed in the concentration of total sperm, the motility and normality of spermatozoa of the Epimedii Herba extract administered groups compared than that of control group, respectively. In the histological analysis of the testicular lobes were observed in the Epimedii Herbaextract administered groups than control group, respectively. Also, the activity hyluronidase was significantly increased in the Epimedii Herba extract administered groups than that of the control group. In case of antioxidant activity analysis, the activity of peroxidase and catalase were significantly increased in the Epimedii Herba extract administered groups than that of control group, respectively. Conclusion: This study shows that Epimedii Herba can effect the count and motility of sperm, the important ractor in male fertility and also promote the activity of antioxidants, catalase and peroxidase, which is the important factor in spermatogenesis.

  • Purpose : ${\beta}$-sitosterol is kind of phytosterols or plant which are structurally similar to cholesterol. This study was aimed to investigate the inhibitory effect of the ${\beta}$-sitosterol on the proliferation of human uterine leiomyoma cells and the expression of gene related the mechanism of cell apoptosis. Methods : We counted the number of death cells treated with indicated time of the ${\beta}$-sitosterol and investigated cell death rate by cell count assay. Furthermore, flow cytometry analysis and DNA fragmentation assay were used to dissect between necrosis and apoptosis. and then we observed the differential gene expression by western blot analysis. Results : 1) The inhibitory effect on the growth of uterine leiomyoma cell treated with the ${\beta}$-sitosterol $16{\mu}M$ was increased in a time dependent. 2) The result of flow cytometry analysis, subG1 phase arrest related cell apoptosis was investigated 16.97% in uterine leiomyoma cell treated with the ${\beta}$-sitosterol $16{\mu}M$ and showed the fashion of proportional time dependent. 3) The gene expression of p27, p21 related cell cycle was increased according to increasing time interval but cyclin E-CDK2 complex was decreased expression. 4) The character of apoptosis, DNA fragmentation was significantly observed on the time dependent. 5) The expression of pro-caspase 3 and PARP were decreased dependent on treatment with time dependent. Conclusion : This study showed that the ${\beta}$-sitosterol have the inhibitory effect on the proliferation of human uterine leiomyoma cell and the effect was related with apoptosis.

A Study on the Correlation of Cold Hypersensitivity and Common Health Indexes in Young Women (젊은 여성의 냉증과 건강지표들의 상관성에 관한 연구)

  • Objectives: The purpose of this study is to investigate the relationship between cold hypersensitivity and common health indexes including dyspepsia, headache, defecation, leucorrhea, and menstruation in young women. Methods: 40 outpatients who visited Kangnam Kyung-Hee Oriental Hospital from January, 2010 to November, 2011 were analyzed. The experimental group consisted of 20 patients diagnosed with cold hypersensitivity, while the control group consisted of 20 patients who were not. All patients were subjected to Digital Infrared Thermal Imaging(DITI), and those with thermal difference greater than $2^{\circ}C$ between anterior thigh and top of feet were diagnosed with cold hypersensitivity. The subjective characteristics of dyspepsia, headache, defecation, leucorrhea, and menstruation were obtained through inquiry by residents at the hospital. The relationship between cold hypersensitivity, dyspepsia, headache, defecation, leucorrhea, and menstruation was analyzed with Independent T-test using SPSS for Windows (version 17.0). Results: Means of defecation, leucorrhea, dysmenorrhea in cold hypersensitivity group were higher than control group without significant statistical difference. Dyspepsia appeared statistically higher in experimental group with significance while headache was the same in both groups. Means of menstrual cycle and menstrual coagulation were higher in non-cold hypersensitivity group, but there was no statistical difference. Conclusions: Results suggest that dyspepsia is common in cold hypersensitivity and treatment of cold hypersensitivity may relieve accompanying digestive problems.

A Case Report of Adenomyosis Treated with Gamiseokhong-jeon Gamibang (가미석홍전(加味惜紅煎) 가미방(加味方)으로 호전된 자궁선근증 1례)

  • Objectives: The most common symptom of adenomyosis is dysmenorrhea and menorrhagia. Therefore, final treatment of this disease is hysterectomy. If dysmenorrhea and menorrhagia can be controlled, It doesn't need the surgical treatment. so, this case study was designed to evaluate the effect of Gamiseokhong-jeon on dysmenorrhea, menorrhagia due to adenomyosis. Methods: A 45-year-old female patient who was diagnosed with adenomyosis has been troubled by dysmenorrhea and menorrhagia. She was taken Korean herbal medicine(Gamiseokhong-jeon gamibang) about 3 months. Results: During this treatment period, dysmenorrhea and menorrhagia were relieved. Also after all treatment, the effect has been continuing. Conclusions: Gamiseokhong-jeon gamibang relieve dysmenorrhea and menorrhagia due to adenomyosis.
