Objectives: This study was performed to evaluate effects of Cordyceps militaris(CM) on osteoclast differentiation and its related gene expression. Methods: We used mouse myeloid cells RAW 264.7 stimulated by receptor activator of nuclear factor kappa-B ligand(RANKL) to induce osteoclast differentiation. There are four groups of which RAW 264.7 cells are not stimulated by RANKL (Normal), stimulated by RANKL without CM(Control), stimulated by RANKL with 0.1 ${\mu}g/ml$ of CM(CM 0.1), stimulated by RANKL with 1 ${\mu}g/ml$ of CM(CM 1). Osteoclastogenesis was measured by counting Tartrate-resistant acid phosphatase-positive multinucleated cells [TRAP(+) MNC]. RT-PCR was performed to evaluate the inhibitory effect of CM on gene expression(TRAP, AKT1, JNK1, NFATc1, c-Fos, MITF). Results: 1. CM decreased the number of TRAP(+) osteoclast in RANKL-stimulated RAW 264.7 cell at the concentration of 0.1 ${\mu}g/ml$ and 1 ${\mu}g/ml$. 2. CM decreased the expression of TRAP in osteoclast at the concentration of 1 ${\mu}g/ml$. 3. CM decreased the expression of AKT1, JNK1 in osteoclast at the concentration of 1 ${\mu}g/ml$. 4. CM didn't affect the expression of NFATc1, c-Fos, MITF in osteoclast. Conclusions: Cordyceps militaris has inhibitory effects on osteoclast differentiation and its related gene expression. These results suggest that Cordyceps militaris has a potential as a treatment of osteoporosis.
Objectives: Recently Ciprofloxacin, used in the treatment of mastitis, showed many serious side effects. The object of this study was to recognize whether TNS and KWT can be used in the treatment of mastitis by observing the in vitro antibacterial effects of TNS and KWT aqueous herbal extracts against S. aureus. Methods: Antibacterial activities of TNS and KWT aqueous extracts against S. aureus ATCC 25923 were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were monitored at MIC and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using Raw 264.6 and MCF-7. The results were compared with Ciprofloxacin, a second generation of quinolone antibiotics in the present study. Results: MIC of aqueous extracts of TNS and KWT against S. aureus were detected as ($0.313{\pm}0.107$) and ($0.137{\pm}0.053$) mg/ml, respectively. MIC of Ciprofloxacin was detected as ($0.469{\pm}0.297$) ${\mu}g/ml$ at same conditions. In addition, TNS, KWT aqueous herbal extracts and Ciprofloxacin were also showed marked dosage-dependent inhibition of bacterial growth, and dramatical inhibitions on the both intracellular killing assays and bacterial invasion using Raw 264.6 and MCF-7 cells were detected. Conclusions: The results obtained in this study suggest that TNS and KWT aqueous herbal extracts showed antibacterial effects against S. aureus, and they also showed dosage-dependent inhibitory effects on the bacterial growth. And they showed the significant intracellular killing and bacterial invasion effects. It means, KWT and TNS may show more potent anti-infectious effects against S. aureus in vivo.
Objectives: We have studied the syndrome differentiation of ultrasound of retroverted uterus(RU) as the inspection in traditional Korean medicine. Methods: Eighty-four patients, who visited H clinic for infertility treatment, were recruited and evaluated by questionnaires and ultrasound, from April, 2012 to May, 2012. We used ultrasound(Logiq C3; GE Health care) for examination of RU. For statistics, we used Fisher's extract test to evaluate the RU and symptoms, and Chi-square test to evaluated the RU and syndrome differentiations, and Student T-test for the relationship between RU and each score of syndrome differentiation, using PASW Statistics(version 18.0.0) and GraphPad Prism(version 5.01). Results: 1. RU was correlated with moderate or severe dysmenorrhea and premenstrual lower abdominal pain. 2. In syndrome differentiation, blood stasis questionnaire was correlated with RU. 3. RU was related with dysmenorrhea, mass of low abdomen, pressure pain, clod of menstrual blood, dyspareunia. Conclusions: The ultrasound of RU might be related with the syndrome of blood stasis.
Purpose: The purpose of this study is to define the cut off values of cold hypersensitivity of hands by using digital infrared thermographic imaging(DITI). Methods: Thermographic images of 130 patients with cold hypersensitivity of hands(CHHG, n=65) and non-cold hypersensitivity of hands(NCHHG, n=65) were retrospectively reviewed. We used the temperature difference the palm(PC8) and the upper arm(LU4) for diagnosing cold hypersensitivity of hands. The temperature differences of between two groups were analysed using independent samples t-tests. The cut off values were calculated by ROC curve analysis. Analyses were undertaken using SPSS version 17.0. P value of < 0.05 was considered significant. Results: The temperature difference the palm(PC8) and the upper arm(LU4) were significantly different between groups(p < 0.001). Using receiver operating characteristic curve analysis, the sensitivity, specificity, and area under the curve were 70.8%, 73.8%, respectively both hands. The AUC was 0.822 on right hand and 0.818 on left hand. The optimum cut-off value was defined as $-0.05^{\circ}C$. Conclusions: These results suggest that DITI is a reliable instrument for estimating the cold hypersensitivity of hands.
Objectives: This study was conducted to investigate the acute toxicity of Pyungwi-san(Pingwei-san in Chinese) in ICR mice, according to KFDA and OECD guideline. Methods: In the present study, 15 male and female ICR mice administrated singly by gavage at dose levels of 0 and 2000 mg/kg of Pyungwi-san. During the experimental period, no treatment-related death was observed. There were no adverse effects on clinical signs, body weight, and gross findings at all treatment groups. Results: These results showed that the single oral adminstration of Pyungwi-san (Pingwei-san) did not cause any toxic effect at the dose levels of 2000 mg/kg in rats. Conclusions: Taken together, the median lethal dose($LD_{50}$) of Pyungwi-san (Pingwei-san) was considered to be over 2000 mg/kg body for both sexes.
Purpose: The aim of this study is to determine associated factors affecting the utilization of Medical Care for an overactive bladder. Methods: Total 114 women with overactive bladder participated in this study. Patients were categorized in two groups, the patients with treatment experiences for an overactive bladder(n=40) and without treatment experiences for an overactive bladder(n=74). Patients completed Overactive Bladder Severity Score, Beck Depression Inventory, King's Health Questionnaire, and questionnaires on treatment experience. The results were analysed using SPSS for Windows 12.0k. All statistical tests were two-sided with a p-value < 0.05 being significant. Results: Patients with treatment experience had a greater score on KHQ, OABSS, BDI and had longer symptom duration period than those without treatment experience. And only Impact of Life(P=0.004), Role Limitation(P=0.004), Social Limitation(P=0.007), Emotion(P=0.035), Sleep/Energy(P=0.001), Incontinence Severity Measures(P=0.012) among KHQ domains were statistically significant difference between two groups. Conclusions: The most influencing factor associated with utilization of Medical Service in overactive bladder is the impact of the patient's quality of life.
Objectives: The purpose of this study is to find out the patterns of teenage girls who is easy to suffer from Menstrual Disorders. Methods: The data from the 1681 participants were collected using a structured menstrual history questionnaire. Based on the survey responses, we had 97 adolescents with menstrual disorder as the test group and 97 adolescents without menstrual disorder as the control group. The clinical trials subjects were asked to respond to another questionnaire for identifying Blood Pressure Pulse Analyzer tests. We identified significant variables for dysmenorrhea from comparison between the test and control group; a binary response variable from the test group's responses for ever experiencing dysmenorrhea and explanatory variables from Blood Pressure Pulse Analyzer. Results: The reselts of Blood Pressure Pulse Analyzer test showed that the score of Energ_R, EIX_R were lower in dysmenorrhea group than the control group. The reselts of Blood Pressure Pulse Analyzer test showed that the score of Energ_R, EIX_R were lower, and T4_R were higher in hypomenorrhea group than the control group. The reselts of Blood Pressure Pulse Analyzer test showed that the score of Energ_R, EIX_R were lower in Irregular Menstruation group than the control group. Conclusions: Blood Pressure Pulse Analyzer test showed that the score of Energ_R, EIX_R were lower in the test group than the control group.
Objectives: To observe randomized controlled trials(RCTs) related to investigating the effects of acupoint treatments on nausea and vomiting in pregnancy to establish the basis of korean medical treatments, and suggest the problems of future clinical research. Methods: After searching for national and international papers published after 2000 and restricting the parameters to 'clinical trial', 'controlled clinical trial', 'randomized controlled trial', then comparing the subjects, methods, research tools, and results to understand the effects and re-comparing the conflicting results to assume the cause. Results: Of the 9 research papers that applied acupressure, four of them used control and experimental groups, and three(75%) research papers indicated significant reduction in vomiting and nausea, and of the seven research papers that utilized the experimental and placebo groups, five(71.4%) reported significant reducing effects. Of the four research papers the used acupuncture treatments, four research papers using control and placebo groups, two showed improvements in symptoms but did not have significant results compared to sham acupuncture groups, and two showed significant reduction of symptoms compared to the control groups. The one research paper that used electrical stimulation showed significant results in the reduction of vomiting and nausea compared to the control group. Conclusions: Acupressure and electrical stimulation showed efficacy reducing the symptoms. Acupuncture treatments did not indicate efficacy some experiments. Therefore it is thought that future acupuncture treatments should use methods that resemble actual clinical medicine to establish efficacy to produce significant results.
Objectives: The purpose of this study was to investigate whether Lonicera japonica(LJ) could inhibit LPS-induced type I IFN production. Methods: To evaluate inhibitory effect of LJ on type I IFN, we examined type I IFN, IRF-1, 7 and IL-10 production on LPS-induced macrophages using real time RT-PCR. Next, we observed the interaction of type I IFN, IRF-1, 7 and IL-10 using IL-10 neutralizing antibody. Finally we examined the activation of STAT-1, 3 using western blot. Results: LJ inhibited Type I IFN expression of mRNA and increased IL-10 expression of mRNA. Also LJ inhibited the level of IRF-1, 7 mRNA and the nuclear translocation of IRF-3. Further more, LJ reduced the activation of STAT-1, 3 which are involved in continuous secretion of immune cytokines. Blockade of IL-10 action caused a significant reduction of type I IFN and IRF-1, 7 than LPS-induced LJ pretreatment. Conclusions: LJ inhibits LPS-induced production of type I IFN by IL-10. This study may provide a clinical basis for anti-inflammatory properties of LJ.
Objectives: The purpose of this study was to identify the tendency toward depression and fatigue in early postpartum women and to determine the relationship between the symptoms of these conditions, according to the women's Sasang constitutions. Methods: In this study, 73 healthy postpartum women were analyzed according to Sasang constitution: 33 Taeeumin, 22 Soyangin, and 18 Soeumin. Symptoms of depression and fatigue were assessed using the Edinburgh Postnatal Depression Scale-K(EPDS-K) and the Fatigue Continuum Form(FCF), respectively, on the fourth(time 1), seventh(time 2), and fifteenth(time 3) days postpartum. Sasang constitutional types were determined by a medical specialist, using results from the Two Step Questionnaire for Sasang Constitution Diagnosis(TS-QSCD). Results: 1. The overall depressive symptoms and fatigue scores decreased significantly over time in early postpartum women. 2. The mean scores for the EPDS and FCF were higher in the Soeumin group than in other groups. The scores for EPDS and FCF dropped sharply in comparison to a former level in the Soyangin group. And the mean EPDS score was lower in the Soyangin group than in other groups. The EPDS and FCF scores in the Taeeumin group changed slightly over time. However, these results were not statistically significant according to Sasang constitution. 3. The relationship between postpartum depression and fatigue showed a significant positive correlation. Conclusions: These results suggest that, in early postpartum women, depressive symptoms and fatigue differ according to Sasang constitution.
